율무쌀이 쥐의 혈장콜레스테롤 및 지질대사에 미치는 영향 (Effect of Coix on Plasma Cholesterol and Lipid Metabolism in Rats)

  • 율무쌀이 흰쥐의 혈장과 조직의 콜레스테롤, 중성지방, 이지질 및 총담즙산 수준에 미치는 영향을 알아보자 흰쥐에 1%의 콜레스테롤을 첨가시킨 실험식이를 자유급식하여 사양한 결과를 요약하면 다음과 같다. 1) 식이섭취량, 식이효율, 증체량은 각 군간에 유의한 차이가 없었다. 2)형장콜레스테롤 함량은 라이드 급여시 현저히 높았으며 라이드-ㅠ율무쌀을 급여시에는 가장 낮았다. 혈장의 중성지방 역시 라이드 급여시 높았다. 혈장의 인지질 역시 라아드 급여서 현저히 높았고 대두유군이 가장 낮았다. 혈장의 총담즙산 함량은 각군간에 차이가 없었으며 혈장 담즙산은 glycoursodeoxy cholicacid, tauroursodeoxy choilic acid와 cholic acid이었다. 3) 간의 단위무게당 총지질, 콜레스테롤, 중성지방역시 라이드- 율무쌀 급여시 현저히 낮았다. 4) 소장의 단위무게당 총지질, 콜레스테롤, 중성지방 함량도 역시 라이드-율무쌀을 급여시 낮았다. 5)실험식의 소화율을 모든식이 처리간에 차이가 없었으나 콜레스테롤 보유율은 라이드-전분군이 가장 높았고 대두유-율무쌀군이 가장 낮았다.

알긴산과 셀룰로오스 및 펙틴 수준이 흰쥐의 대장기능에 미치는 영향 (Effects of Alginic Acid, Cellulose and Pectin Level on Bowel Function in Rats)

  • The purpose of this article is to know the effects on bowel function of the kind of fiber and the amount of fiber in SD-rats. To do this experiment, we select of $\alpha$-cellulose as n insoluble cellulose source and alginic acid and pectin as soluble cellulose source. The rats diets contained callolose camcentrations of 1.0%, 3.6%, 6.0% and 10.0%. After that, we raised the SD-rats for 4weeks and measured the amount of food intake, body weight, the food effciency ratio, the length of liver and stomach the weight of the intestines, the transit time through the intestines, pH in feces, and the amount of bile acid and Ca, Mg, pp. 1) The amount of food intake was 15.75-31.00g/day. It was highest in the 10.0% cellulose group and the lowest in the 3.6% and 6.0% alginic acid group (p<0.05). The body weights of rats were 277.50-349.809. It was highest in the 1.0% pectin group and lowest in the 3.6% alginic acid group, 6.0% cellulose group, and 10.0% pectin group. It had differences according to the content fiber and the kind of dietary(p<0.01). The food efficiency ratio was (p<0.01). The higher the content of dietary fiber, the lower the calory and the food efficiency ratio. 2) Transit time was 446.0-775.0 minutes and it showed signidicant ifferences according to the content and kind of dietary fiber(p<0.01). It was long in the 1.0% cellulose group and 1.0% pectin group but short in the 10.0% alginic acid group. As the content of dietary fiber increased, the transit time through the intestines was shortened. The length of small intestine was 101.03-120.40cm and there were no difference cegardloss of the content and kind of fiber. The length of the large intestine was 20.92-25.42cm and there were significant differences according to the content and kind of the fiber. High-fiber diets resulted in increases in the length of the large intestine. 3) The weight of the liver was 8.68-10.96g and there were no differences according to the content and kind of fiber. The weight of stomach was 1.28-1.74g and there were no differences resulting from the kind of dietary fiber, but it was highest in the 10.0% alginic acid group. The weight of the small intestine was 5.52-8.04g with no difference resulting from to the kind of fiber. It was highest in the 10.0% the alginic acid group and lowest in the 1.0% alginic acid group(p<0.05). The weight of large intestine was 2.50-3.30g with no differences related to the kind of dietary fiber. It was heaviest in the 6.0% and 10.0% alginic acid groups and in the 10.0% pectin group with differences related to the content of fiber(p<0.05). 4) The pH of the feces was 5.82-6.86 according to the kind of dietary fiber, alginic acid group was high at 6.66, the cellulose group was 6.26. but the pectin group was low at 6.30. There were difference according to the content of fiber, but no consistency. The content of bile acid was 6.25-34.77umol per 1g of dry feces. According to the kind of dietary fiber, the alginic acid group was low at 12.91umol, cellulose group was 18.64umol and, the pectin group was the highest at 27.78umol(p<0.001). Based on the content of dietary fiber, alginic acid group was low at 1.0%, but high at 3.6% pectin group(p<0.001). 5) The amount of feces was 1.00-5.10g/day. The weight of rat feces was 2.23g/day in the alginic acid goup, 2.75g/day in the cellulose group, and 1.82g/day in the pectin group. According to the content of fiber, cellulose group was high at 10.0% but alginic acid group was 1.0%, and there were significant difference according to the dietary fiber. The more the content of fiber, the more increase the content of feces in alginic acid, cellulose and pectin group. The content of Ca in the feces was 80.10-207.82mg/1g of dry feces. In the dietary fiber, alginic acid group was 193.08mg, cellulose group was 87.5mg, pectin group was 138.16mg. In the content of fiber, alginic acid group was high at 1.0% and 3.6% but low at 10.0% of Pectin group. The content of Mg was 19.15-44.72mg/1g of dry feces. According to the kind of dietary fiber, alginic acid group was 35.33mg, cellulose group was 23.60mg, and pectin was 36.93mg. According to the content of fiber, pectin group was high at 1.0% and low at 10.0% of cellulose group. The content of P was 1.65-4.65mg/1g of dry feces. According to the kind of dietary fiber, alginic acid group 2.23mg/g dry feces, cellulose group was 2.29mg/g, pectin group wa 4.08mg/g dry feces. In the content of fiber, pectin group was high at 6.0% and low at 6.0% alginic acid group, but there were significant difference among the analysis value. The conetnt of Ca and MG was higher in soluble alginic acid group and pectin group than in insoluble cellulose group. The high the content of the dietary fiber, the lower the food efficiency ratio and the short the transit time through intestine with the increase of the length of large intestin as well as the higher level of the stomach, the small intestine and the large intestine. According to the content of the dietary fiber, the amount of the feces, Ca, Mg and P was increased but the length the small intestin, the weight of liver, pH of the feces and the amount of bile acid showed no differences and consistency.

한국산 녹차, 우롱차 및 홍차 음료의 Cadmium 제거작용에 관한 연구 (Effect of Korean Green Tea, Oolong Tea and Black Tea Beverage on the Removal of Cadmium in Rat)

  • This study was to investigate the cadmium removal effect of Korean green tea, black tea and oolong tea beverage on Cd administered rat, tissues and their excretions. Male Sprague-Dawley rats weighing 143±3.2g were divided into control and experimental groups. The control group were fed standard diet without cadmium . The experimental groups, which were fed standard diet containing 40 ppm Cd, were divided into 4 subgroups again , which were the groups given distilled water (CD group), 5% black tea (BT group), oolong tea (OT group ) and green tea (GT group), respectively. Five days before to sacrifice the rats, all 4 cadmium fed groups were supplied 1 ml of water with 600ppm Cd and control group were fed 1 ml of distilled water without Cd under the same dietary condition. After that, their excretion were collected separately for 3 days. In rat liver and kidney, accmulation of cadmium in 4 Cd administered groups were more than in control group and that of GT group was significantly less than CD group. In bone , also, accumulation of cadmium in 4 Cd administered groups was more than in control group and that of GT, OT,BT groups were much less than that of CD group. GT group was excreted more Cd in urine than Cd group. In feces, 3 tea feeding groups (BT, OT, GT group) were excreted Cd 1.7, 2.1, 2.4 times more than that of the CD group, respectively. We conclude that cadmium accumulations of GT feeding group in rat's liver, kidney and bone were much less than CD group , and the absorption and retention rate of GT group was significantly lower than CD group.

Purple Rice Extract Supplemented Diet Reduces DMH-Induced Aberrant Crypt Foci in the Rat Colon by Inhibition of Bacterial β-Glucuronidase

  • Background: Purple rice has become a natural product of interest which is widely used for health promotion. This study investigated the preventive effect of purple rice extract (PRE) mixed diet on DMH initiation of colon carcinogenesis. Materials and Methods: Rats were fed with PRE mixed diet one week before injection of DMH (40 mg/kg of body weight once a week for 2 weeks). They were killed 12 hrs after a second DMH injection to measure the level of $O^6$-methylguanine and xenobiotic metabolizing enzyme activities. Results: In rats that received PRE, guanine methylation was reduced in the colonic mucosa, but not in the liver, whereas PRE did not affect xenobiotic conjugation, with reference to glutathione-S-transferase or UDP-glucuronyl transferase. After 5 weeks, rats that received PRE with DMH injection had fewer ACF in the colon than those treated with DMH alone. Interestingly, a PRE mixed diet inhibited the activity of bacterial ${\beta}$-glucuronidase in rat feces, a critical enzyme for free methylazoxymethanol (MAM) release in the rat colon. These results indicated that purple rice extract inhibited ${\beta}$-glucuronidase activity in the colonic lumen, causing a reduction of MAM-induced colonic mucosa DNA methylation, leaded to decelerated formation of aberrant crypt foci in the rat colon. Conclusions: The supplemented purple rice extract might thus prevent colon carcinogenesis by the alteration of the colonic environment, and thus could be further developed for neutraceutical products for colon cancer prevention.

Effect of Lactobacillus casei fermented Senna tora L. seeds and its active compound via muscarinic M3 signaling on the improvement of intestinal function in rats

  • We previously reported the potential of Senna tora L. seeds fermented by Lactobacillus casei (FSL) as a laxative agent in a loperamide-induced constipation rat model. Here, we examine the mechanism of action of FSL and its bioactive compound, revealed herein, on loperamide-induced constipation Sprague Dawley rat model. We identified the compound aurantio-obtusin (AO) using HPLC quantitative analysis. Rats were randomly assigned to six experimental groups (eight rats each)-normal and constipated groups (loperamide, FSL [100, 300, 500 mg/kg], and AO [1 mg/kg]). The FSL and AO-treated group showed an increase in the frequency, amount, and water content of feces in the constipated rat. Moreover, FSL and AO increased the intestinal transit speed in the constipated rat. Histological analysis revealed that FSL and AO recovered the intestinal mucus, the number of goblet cells, as well as thickness of the mucosa layer and muscle. Furthermore, the protein levels of the muscarinic acetylcholine receptor M3, which is involved in intestine contraction, were recovered in the FSL and AO-treated group. Its downstream signaling pathway (p-protein kinase C) was recovered by FSL and AO treatment. In conclusion, fermentation of S. tora L. seeds increases AO, which improves intestinal function, indicating that FSL is effective for treating constipation.

  • The absorption, distribution and excretion of $^{14}C$ labeled YH1885 {5,6-Dimethyl-2(4-fluorophenylamino)-4-(1-methyl-1,2,3,4-tetrahydroisoquinolin-2-yl)pyrimidine hydroc hloride), a new proton pumpinhibitor, were investigated in rats after a single administration of $^{14}C$-YH1885. 1. After intravenous administration of 5mg/kg, the blood level of radioactivity declined in a biphasic fashion with the mean terminal elimination half-life of 12.4hr. 2. After oral administration of 20mg/kg, the maximum blood level of radioactirity was reached at 4.0hr in female rats. The blood level of radioactivity-time profiles in male and female rats were similar, and the absorptionof $^{14}C$-YH1885 was not affected by food. 3. Appproximately 89% and 1% of radioactivity of the total dose were excreted in feces and urine, respectively. 4. Biliary excretion of radioactivity was 47.9% of the dose. Enterohepatic circulation of radioactivity was 49.6%. 5. Radioactivity was excreted maily into feces via bile. 6. The concentration of radioactivity in most tissues reached the peak level at 4.0hr after dosing, and then declined. Autoradiograms of male rats showed that the radioactivity levlels in the fat, harder's gland, liver and G-Itract were higher than those in the other tissues and the elimination of radioactivity from fat and liver was slow. 7. Autoradiograms of a pregnant rat showed that radioactivity was transferred to mammary gland, placenta and fetus. The radioactivity level in the mammary gland was higher than that in the blood.

미세결정화키틴과 팜유의 수준을 달리한 식이가 흰쥐의 지방대상에 미치는 영향 (The Effects of Micro Crystalline Chitin and Palm Oil on the Lipid Metabolism in the Rat)

  • The effects of MCC and palm oil at different levels on lipid metabolism were assessed in adolescent rat. Dietary fat levels were 20% and 40% (kcal/kcal) and MCC level were 0%, 2%, and 4% (wt/wt). The experimental period took 8 weeks. During the experiment, weight gain and food efficiency ration were not affected by dietary factors. The weight of thymus however, was lower in high fat groups than in middle fat groups. The contents of total lipid and total cholesterol in plasma and HDL-cholesterol in plasma and HDL-cholesterol were significantly affected by dietary factors. The concentrations of cholesterol in LDL and VLDL , and the triglyceride content of VLDL was higher in high fat groups than in middle fat groups. Adding MCC lowered the LDL triglyceride in the high fat groups. The analysis of plasma fatty acids generally reflected the composition of the dietary palm oil. MCC had significant effects on total lipid and triglyceride of feces, but not on total cholesterol . In conclusion, the level of MCC had little hypolipodemic effects on lipid metabolism in rats fed a high fat diet. the middle fat group that contained 4% MCC showed increased contents of fecal triglyceride than the others, indicating that MCC had an effect on lipid absorption. Therefore, the other physiological functions of MCC need to be tested for their useful applications.

"이전고환극구흡충" 패류중간숙주로서의 "애기물달팽이" (Austropeplea ollular (Pulmonata: Lymnaeidae): a new first intermediate host of Neodiplostomum seoulense ( Trematoda : Diplostomatidae ) in Korea)

  • Some planorbid snails such as Hippeutis cantori and Segmentina hemisphaerula have reported as the molluscan intermediate hosts of Neodiplostomum seoulense, one of important snail-borne human intestinal trematodes in Korea. However, one of the Korean lymnaeid snail species, Austropeplea ollular was also found to be the first intermediate ho of N. seoulense. In field-collected Austropeplea snails from Sorae and Kimpo out of se collected localities, the bifurcated cercariae of N. seoulense were shed (infection rat 0.3%), whereas no Radix auricularia and Fossaria truncatula were found shedding cercariae. Each of 12 tadpoles of Rana nigromaculata, known as the second intermedia host of N. seoulense, were exposed to 200 cercariae shed from field-collected A ollula. F tadpoles of R. nigromaculata were found to be massively infected with metacercariae o N. seoulense (recovery rate: 62.1%). Each of five rats (Sprague-Dowley strain) was or fed with 200 metacercariae, and eggs of N. seoulense were detected in the rat feces on week later. These rats were killed 4 weeks after postinfection and adult worms of N seoulense were recovered from the small intestines (recovery rate: 9%). This is the f report of A. ollula as the first molluscan intermediate host for N. seoulense in Korea.

  • 변비는 우리나라나 서구국가에서 가장 흔한 질환중의 하나로서 생활 습관, 식이, 육체적 활동, 스트레스등과 밀접한 관련이 있다고 한다. 그리고 가임기의 여성에서 남성보다 빈도가 높으며 그 이유는 여성호르몬에 의한 대장운동성 저하때문이라고 한다. 본 연구는 로페라미드 투여에 의한 변비 쥐에서 프로바이오틱스가 장운동과 변비에 미치는 영향을 알아보기 위하여 시행되었다. 로페라미드 투여는 쥐에서 변량은 감소시킨 대신에 변 pellet수는 증가시켰다. 변의 수분함량은 로페라미드 투여에 의하여 감소하였으나 건조 변량은 실험군간의 유의한 차이는 없었다. 따라서 로페라미드는 장 운동 감소에 의하여 변비를 유발하는 것으로 생각되었다. 프로바이오틱스의 투여는 로페라미드에 의하여 발생한 장 운동 저하와 변비를 완화시켜 주는 것으로 나타났다. 이상의 결과로 미루어 보아 프로바이오틱은 로페라미드에 의한 장 운동 감소 및 변비발생을 예방 또는 치료해 줄수 있는 것으로 생각되었다.

다시마 (Laminaria japonicus) Alginate의 가열가수분해에 따른 물리$\cdot$화학적 및 생물학적 특성에 관한 연구 7. 저분자 Alginate에 의한 랫드 분변의 성분 변화 (Studies on Physicochemical and Biological Properties of Depolymerized Alginate from Sea tangle, Laminalia japonicus by Thermal Decomposition 7. Effects of Depolymerized Alginate on Fecal Composition in Rats)

  • 저분자 alginate인 HAG-10, HAG-50, HAG-100 및 alginate를 랫드에 장기간 섭취시켰을 때, 분변의 무게와 수분함량, 단백질과 지방함량, pH, VBN 및 단쇄지방산을 측정하여 소화생리특성에 미치는 영향에 대하여 검토하였으며, 그 결과를 요약하면 다음과 같다. 랫드분변의 무게와 수분함량의 변화는 alginate에서 가장 높은 증가를 보였고, 다음으로 HAG-100, HAG-50이었으며 HAG-10은 투여전과 거의 변화가 없었다. 분변의 단백질과 지방함량은 $5\%$ HAG-50에서 가장 높았고, 그 소화율은 $5\%$$10\%$ HAG-50에서 유의적으로 현저히 저하하였다. 분변의 pH는 $5\%$$10\%$ HAG-50에서 급격하게 저하하였으나, $10\%$ HAG-100과 alginate에서는 오히려 급격히 증가하는 경향을 나타내었다. VBN은 $10\%$ HAG-10, HAG-50 모두 그리고 $1\%$$5\%$ HAG-100에서 뚜렷한 저하를 보였으나, $10\%$ HAG-100 및 $5\%$$10\%$ alginate는 반대로 급격히 증가하였다. 단쇄지방산의 변화는 $5\%$$10\%$ HAG-50에서 n-butyric acid가 증가하였고, propionic acid와 acetic acid는 반대로 현저히 감소하였다. 이상의 결과로부터, HAG-50은 랫드 분변의 pH와 VBN을 현저히 저하시켰으며, 단쇄지방산중 n-butyric acid를 증가시키고 propionic acid와 acetic acid를 유의적으로 감소시켰으므로, 랫드의 소화생리특성을 개선시키는 데 있어서 효과적인 저분자 alginate라고 사료된다.
