Background: The purpose of this study is to evaluate the immunosuppressive and antioxidant effects of a novel radioprotective agent using the vitamin E derivative 2-(alpha-D-glucopyranosyl)methyl-2,5,7,8-tetramethylchroman-6-ol (TMG) and its effect on tumors, and to study its usefulness. Materials and Methods: In this study, C57BL/6NCrSlc mice were divided into four groups (control, TMG, radiation therapy [RT], and RT+TMG), using 10 mice in each group. In the TMG and 2 Gy+TMG groups, 500 mg/kg TMG was administered. Two groups (2 Gy and 2 Gy+TMG) among RT and RT+TMG groups were irradiated with 2 Gy in a single fraction, while the other two groups (6 Gy and 6 Gy+TMG) were irradiated locally with 6 Gy in three fractions. Results and Discussion: TMG positively affected CD4+ and CD8+ T lymphocytes. Tumor volumes and growth inhibition rates were compared. In order to evaluate how TMG administration affected tumor growth, Ehrlich cancer cells were injected into the thigh of mice, and the tumor volume and growth suppression rate were compared. Not only RT but also TMG alone inhibited tumor growth. If RT conducted to the mice with TMG, TMG could increase the number of leukocytes, primarily that of lymphocytes. TMG also inhibited tumor growth in addition to RT. Tumor growth was significantly inhibited in the 6 Gy+TMG group. Conclusion: In conclusion, TMG exerted an immunopotentiating effect mainly by increasing the white blood cell numbers including that of lymphocytes. In addition to RT, TMG also inhibited tumor growth. Therefore, TMG is considered to be a useful radioprotective agent in radiotherapy without tumor growth induction.
The effects of red ginseng extracts (5.5 mg/mouse: i.p.) on the activities of antioxidant enzymes (superoxide dismutase, catalase and peroxidase) and lipid peroxidation were studied in the cytosol fraction of kidney. The experiments were carried out with whole-body irradiated (6.0 Gy, $^{60}Co$) and non-irradiated ICR mice. In the red ginseng extract-treated and irradiated mice, the activities of Cu, Zn- SOD, Mn-SOD, catalase and peroxidase were significantly enhanced by 27.8, 31.9, 17.9 and 15.0%, respectively, but the contents of malondialdehyde were considerably decreased (81.OfS) after 21 days, compared with those of non-treated mice. The enhanced activities of antioxidant enzymes inhibited the increase of malondialdehyde product resulted from the ionizing radiation. These results suggest that red ginseng extracts probably play an important role in radioprotective effect. Key words Red ginseng, SOD, catalase, peroxidase, lipid peroxidation.
본 연구는 in vitro에서 나린진과 나린제닌의 항산화 활성을 평가하고, in vivo에서 BALB/c 마우스에 나린진과 나린제닌을 7일 동안 경구투여 한 후 6Gy의 감마선을 조사시켜 24시간 뒤 감마선 조사로 인한 세포 손상 및 산화적 스트레스에 대한 방사선 예방효과를 검토하였다. 나린진과 나린제닌의 항산화 활성평가에서 나린제닌이 나린진보다 ABTS 제거 활성 및 FRAP을 유의적으로 증가시켜 아글리콘 형태인 나린제닌이 글리코사이드 형태의 나린진보다 항산화 능력이 뛰어난 것을 확인하였다. 감마선 조사 전 나린제닌을 투여 한 군이 감마선 조사군에 비해 비장 지수, 흉선 지수 및 백혈구 수치가 증가하여 조혈 면역계 손상에 대해 보호효과가 있음을 확인하였다. 나린제닌을 투여 후 감마선을 조사한 군의 ALT와 AST가 감마선 조사군에 비해 유의적으로 감소하여 간세포 손상에 대한 예방효과를 확인하였다. 감마선 조사 전에 나린제닌을 투여한 군의 지방질과산화는 감마선 조사군에 비해 유의적으로 낮게 나타남을 보였으며, XO는 감마선 조사군에 비해 유의적으로 감소하여 방사선에 의한 장해를 감소시킨 것으로 판단할 수 있다. 감마선 조사 전 나린제닌을 투여한 군의 GSH와 항산화 효소의 활성은 감마선 조사군에 비해 유의적으로 증가하여 생체 내에 항산화 활성을 회복시켰다. 이 상의 결과를 통해 방사선 조사 전에 나린제닌의 투여는 방사선에 의한 세포 손상과 산화적 스트레스에 대해 보호 효과가 있어 방사선보호제로 유용하게 사용될 수 있다.
생쥐에 5.5mg의 홍삼추출물을 투여한 후 6.5Gy의 감마선을 전신 조사하여 생쥐 간에서의 항산화 효소들(Superoxide dismutase, Catalase, Peroxidase)의 활성도와 지질 과산화 수준의 변화를 측정 비교하여 이들의 상호 관련성과 인삼이 방사선 장해에 미치는 영향을 검토하였다. 방사선 조사로 인하여 SOD, Catalase 그리고 Peroxidase의 활성은 계속적으로 감소하였으나 홍삼추출물 투여군은 방사선 조사 후 21일에 생리적 식염수 투여군보다 Cu, Zn-SOD($40.7\%$), Mn SOD($26.9\%$), Catalase($20.0\%$) 그리고 Peroxidase($20.1\%$)의 활성이 증가되었으며, 또한 방사선 조사로 인하여 상승되는 지질 과산화의 함량이 억제되었다($70.5\%$). 이러한 결과는 인삼 성분이 방사선 조사로 인하여 생성이 촉진된 free radicals를 제거시킴으로서 지질 과산화물의 생성을 억제시켰거나 방사선에 의하여 손상된 기관들의 회복능력을 강화시킨 결과로 보이며 이는 인삼이 방사선 장해에 대하여 회복 또는 방호효과도 있음을 보여 주는 것이라 생각된다.
Patt등이 1949년 처음으로 cysteamine과 히utathione등이 정상조직의 방사선 내성을 증가시킨다는 사실을 규명한 이후로 많은 방사선 방어제가 연구되어 왔으나, 그들의 독성때문에 임상적적용이 매우 제한되었다. 본 실험에서는 3개의 sulfur기를 지닌 환상구조의 Trithioformaldehyde(TTFA)를 합성하여 방사선 조사 후 생존율에 대한 영향과 마우스 공장 소낭선 세포에 대한 방호 효과를 관찰하였다. TTFA 2.0 g/kg을 방사선 조사 15분 전에 복강내로 투여하고 800 rad를 조사한 후 30일간 관찰했을 때 78%의 생존율을 보였다. 또, 공장 소낭선에 대한 실험에서 TTFA는 1.0g/kg과 2.0g/kg에서 공장 소낭선에 대한 protection factor가 각각 1.17와 1.23 임을 나타냈다.
We performed this study to determine the effect of Bu-Zhong-Yi-Qi-Tang, as a prescription of traditional Oriental medicine, and its major ingredients on jejunal crypt survival, endogenous spleen colony formation, apopotosis in jejunal crypt cells, lethality and hematological change of mice irradiated with high and low dose of Y-radiation. Bu-Zhong-Yi-Qi-Tang administration before irradiation protected the jejunal crypts (p<0.0001), increased the formation of endogenous spleen colony (p<0.05) and reduced the frequency of radiation-induced apoptosis (p<0.05). The survival rate and mean survival time of the groups treated with Bu-Zhong-Yi-Qi-Tang within 30 days after the treatment were far better than the irradiation control group. In the experiment on the effect of ingredients of Bu-Zhong-Yi-Qi-Tang, the result indicated that the extract of Rensan (Panax ginseng), Danggui (Angelica sinensis), Shengma (Cimicifuga heracleifolia) and Chaihu (Bupleurum falcatnosa) might have a major radioprotective effect. Although the mechanisms of this inhibitory effect remain to be elucidated, these results indicated that BU-Zhong-Yi-Qi-Tang might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the protective nature of Bu-Zhong-Yi-Qi-Tang extract and its ingredients.
Exponential rise in the use of mobile communication devices has generated health concerns due to radiofrequency (RF) exposure due to its close proximity to the head. Calcium binding proteins like calretinin regulate the levels of calcium ($Ca^{2+}$) which plays an important role in biological systems. Ginseng is known for maintaining equilibrium in the human body and may play a beneficial radioprotectant role against electromagnetic field (EMF) exposure. In the present study, we evaluated the radioprotective effects of red ginseng (RG) extract in a mouse model. Calretinin (CR) expression was measured using a free-floating immunohistochemical method in the hippocampus of mice after 835 MHz EMF exposure for 5 h/d for 5 d at specific absorption rate=1.6 W/kg for the different experimental groups. The control animals were treated with NaCl while the experimental animals received 10 mg/kg ginseng, or 30 mg/kg; EMF exposed mice were also treated with NaCl, 10 mg/kg ginseng (E10), or 30 mg/kg (E30). Decreases in CR immunoreactivity (IR) along with loss of CA1 and CA3 interneurons and infragranular cells were observed in the ENaCl group while such losses were not observed in the E10 and E30 groups. CR IR significantly increased in the RG-treated group compared to control and EMF-exposed groups treated with NaCl. The study demonstrates that RG extract can serve as a radioprotective agent that maintains $Ca^{2+}$ homeostasis and prevents neuronal loss in the brain hippocampal region caused by RF exposure.
방사선피폭 후 급성조혈계 손상에 대한 Melatonin과 Ge-132의 방어효과를 8 Gy의 선량으로 급성, 전신피폭 된 마우스에서 연구하였다. Melatonin은 피폭 1시간 전에 200 mg/kg의 용량으로 복강 내 주사하였고, Ge-132는 130 - 150 mg/kg/d의 용량으로 피폭 후 5일째부터 20일째까지 경구복용 시켰다. 방사선방어 효과를 평가하기 위해 비장에서 TUNEL assay를 실시하였고 혈액 내 림프구와 총 백혈구 수치를 측정하였다. 피폭 후 30일째까지의 생존분석을 위해 4 실험군들 (피폭단독군, melatonin 전처치군, Ge-132 후처치군, melatonin 전처치 및 Ge-132 후처치군)에서 날짜경과에 따른 사망을 측정하였다. 피폭단독군과 melatonin 전처치군 간에 세포고사지수 (47.8% vs 45.9%, p=0.385)와 림프구 수치 ($97/{\mu}{\ell}\;vs\;101/{\mu}{\ell}$, p=0.898)에서 차이가 없었으나, 총 백혈구 수치는 melatonin 전처치군에서 유의하게 높은 수치 ($147/{\mu}{\ell}\;vs\;306/{\mu}{\ell}$, p=0.010)를 보였다. 피폭 후 30일째의 생존율은 피폭단독군, melatonin 전처치군, Ge-132 후처치군, 그리고 melatonin 전처치 및 Ge-132 후처치군에서 각각 21.4%, 100%, 35.7%, 그리고 85.7%였다. 실험군들 간의 생존분석에서 melatonin 전처치 군은 피폭단독군(p=0.000)이나 Ge-132 후처치군(p=0.0003)에 유의하게 높은 생존을 보였다. 본 연구결과는 방사선피폭 후 급성조혈계증후군에 대하여 melatonin이 방사선방어제로서 효능을 지님을 보여주고 있다.
Radiation protective fraction was Isolated and partially purified from Korean white ginseng. The effect of the fraction was studied on the cell survival of W-damaged CHO-Kl cells. As a result, it was found that the fraction increased the survival rate of damaged cells significantly within the dose range of which cytotoxicity did not appear This fraction was separated into two parts by adding butanol, namely the precipitated protein component and the butanol extract. Damaged cells were treated with each of these components and their survival rates were measured. The protein component demonstrated significant increase in the survival rates, while the butanol extract showed no such increment. These results suggest that the radiation protective effect of the ginseng fraction is originated from the butanol-precipitated protein component, not from the butanol-soluble compounds.
This study aims to summarize and make a reference of Sagunjatang and Samultang by investigating and comparing the effects of prescriptions. In this process, this review surveyed all papers used Sagunjatang and Samultang in PubMed database and finally 36 papers were included. The publication year, the nations of authors, the type of materials, the formation of experiments, the type of herbal medicine, and their action and mechanisms were analyzed. The number of studies on Sagjunjatang and Samultang has increased since 1990's. The main formation of experiments was in vivo study and the portion was 55%. The proportion of the research using prescriptions was 71%. Sagunjatang was investigated in the field of GI track function, anticancer effect, radioprotective effect and etc. Samultang was studied in the field of anticancer effect, brain cognitive function, anti-inflammatory effect, cell protective effect, and etc. Based on these results, Sagunjatang and Samultang showed the different bioactivities. This study will provide the useful information on differentiation and analysis of the effect of herbal prescriptions by the theory of Korean traditional medicine.
