Effect of Prechilling, Light Quality and Daily Irradiation Hours on Seed Germination in Three Campanulan Plants (저온처리(低溫處理), 파종후(播種後) 광질(光質) 및 일중조명시간(日中照明時間)에 따른 도라지, 더덕, 만삼의 발아율(發芽率))

  • Campanulaceae having the most growing areas among medicinal crops cultivated in Korea occasionally failed to establish a reasonable standing in practice. The experiment was carried out to examine the effect of prechilling (0 : 4 : 8 days), light quality (red : white : dark) and daily irradiation hours (8 : 12 : 16) after sowing on seed germination and radicle elongation of Campanulaceae (Platycodon grandiflorum : Codonopsis lanceolata : C. pjlosula) to give an information on their earlier standing establishment. Mean germination rate of P. grandiflorum was the highest but that of C. pilosula was the lowest regardless of all the treatments. 12 hours irradiation or prechilling increased to 8 days enhanced their earlier or later germination, respectively. White light increased the rate of P. grandiflorum but alleviated that of C. lanceolata regardless of the daily irradiation hours. Although prechilling eliminated such effect of white light, light quality treatment effect on their mean germination rates was influenced by period after sowing, daily irradiation hours or prechilling. On the 9th day after sowing, C. lanceolata showed the greatest radicle length, and both daily 8 hours irradiation and 8 days prechilling enforced to elongate their radicles, while P. grandjflorum and C. lanceolata more lengthened their radicles in all prechilling treatments than in no chilling but C. pilosula showed the similar result only in the 8 days prechilling.

Priming Effect of Rice Seeds on Seedling Establishment under Adverse Soil Conditions

  • An experiment was carried out to find out the priming effects of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) on. the seedling establishment and early emergence under excess soil moisture conditions. Seeds were primed by soaking in -0.6 MPa polyethylene glycol (PEG) solution at $25^{\circ}C$ for 4 days. The primed seeds were sown in soils with various soil moistures (60, 80, 100, 120, and 140% field capacity) at 17 and $25^{\circ}C$, respectively. Germination and emergence rates, plumule height, and radicle length of primed seeds were higher than those of untreated seeds at any soil moisture and temperature examined. The time from planting to 50% germination ($T_{50}$) of primed seeds was less than that of untreated seeds by 0.9~3.7 days. Germination rate, emergence rate, plumule height, and radicle length were highest at the soil moisture of 80% field capacity among the soil moistures. Priming effects of rice seeds on germination and emergence rates were more prominent under the unfavorable soil moistures (60, 100, 120, and 140% field capacity) than those under the optimum soil moisture condition (80% field capacity). However, priming effects on seedling growth were greater at near optimum soil moisture compared with too lower or higher soil moistures. Therefore, these findings suggest that priming of rice seeds may be a useful way for better seedling establishment under the adverse soil conditions.

  • Anatomical changes in the forming and germinating processes of tobacco seeds were investigated to obtain basic information on the ecological characteristics of tobacco seeds. Seed development studied through the longitudinal section of the fertilized ovule clarified that the cell division of the zygote was initiated after 7 days of flowering. After 12 days of flowering, perfect seed constituents such as cotyledon, epicotyle and radicle were formed and those were expressed to recognizable level of germinability. After 15 days of flowering germination rate reached higher than 30% and 17 and 21 days after flowering a perfect seed which have 70% or higher germinability were produced. Seed size was ranged between 0.3-0.6 mm and varietal differences were noted in the given seed size range. Under the light treatment, the morphological changes were observed by elongation of radicle after 2 days of imbibition and apparent germination after 3 days of imbibition. But no responses of the seeds imbibed 6 days under the dark condition were observed.

Effect of $GA_3$ and Light Quality on Seed Germination in Three Campanulan Plants ($GA_3$ 및 파종(播種) 후(後) 광질(光質) 처리(處理)에 따른 도라지, 더덕, 만삼의 발아율(發芽率))

  • Gibberellin (GA) has been widely used to break seed dormancy for better stand establishment. The experiment was done to clarify the effect of $GA_3$ (concentration; period) and light quality (red; white; dark) after sowing on seed germination and radicle elongation of Campanulaceae (Platycodon grandiflorum; Codonopsis lanceolata; C. pilosula) to get an information on their field emergence. The germination test was carried out with 12 hours irradiation for 9 days after priming treatment. In the darkness, the mean germination rate of all the species was decreased in the order to P. grandiflorum, C. lanceolata, C. pilosula. Their mean germination rates and radicle lengths were increased with increased concentration to 0.1mM of $GA_3$. Earlier germination rate was higher but later one was less in 4-day $GA_3$ treatment than in 1- or 2-day $GA_3$ treatment. Light treatment. especially red light given after $GA_3$ treatment. eliminated the $GA_3$ treatment effect. Red light done after $GA_3$ treatment nearly blocked the germination of P. grandiflorum and C. pilosula but delayed that of C. pilosula compared to the other light quality treatments having the similar rate. In addition. the radicle elongation of all three species affected by light quality treatment showed the same result as the germination rate.

Identification and Effects of Phenolic Compounds from Some Plants (수종 식물의 페놀화합물 분석과 효과)

  • The extracts of selected plants and analyzed phenolic compounds were used to study the effects of alleloKDICicals on seed germination and seedling growth. HPLC analysis of the aqueous extracts of seven species identified 15 phenolic compounds including caffeic acid. Among them, protocatechuic acid was detected at 65.87ppm and 6.84ppm, in Erigeron canadensis and Pinus rigida, respectively. And the extract of P. rigida showed the strongest inhibitory effect on seed germination. The extract of P. rigida leaves significantly inhibited germination and radicle growth of Raphanus sativus var. hortensis for. acanthiformis in direct proportion to concentration. However, germination of Cassia mimosoides var. nomame was stimulated by the treated extracts at the same concentrations, but root growth was inhibited at high concentrations. Except chlorogenic acid, eleven of the twelve phenolic compounds inhibited the germination of R. sativus var. hortensis for. acanthiformis. In the case of C. mimosoides var. nomame, some phenolic compounds such as chlorogenic acid, vanillic acid, protocatechuic acid, ferulic acid, gallic acid and ${\rho}-coumaric$ acid stimulated germination, while the others reduced it.

  • Effects of temperature, pH, light-and-dark and degree of oxygen supply on germination of Papaver somniferum L. seeds were investigated. The optimum temperature of the germination was 2$0^{\circ}C$, but the germination was decreased as temperatures changed to the upper and lower. The largest elongation of both radicle and hypocotyl also occurred at 2$0^{\circ}C$. The best germination occurred at pH 7 but this decreased suddenly the above and below of that pH value. Although no effect of light-and-dark on the germination was, this was restricted under limited. oxyten supply in terms of underwater germination as compared with abovewater one.

Esterase Isozyme Banding Pattern in Wild Legume Plants (야생 콩과식물의 Esterase Isozyme Banding Pattern에 관한 연구)

  • Starch gel electrophoresis was used to examine the banding pattern of Esterase isozyme in the leaf, root-nodule and seedling of four wild legume species, Trifolim repense, Glycine soja, Phaseolus nipponensis and Vigna uexillata. The number of band, enzyme activity and migrating rate of esterase isozyme varies depending on the species and tissues of legume plants. The isozyme banding pattern in the cotyledon and radicle of T. repense showed same pattern, however, the number of band were varible among the cotyledon of G. soja, P. nipponensis and V. vexzllata, respectively. Est-1 in the leaf of G. soja, V, vexillata. root-nodule of G. soja and seedling of V. vexillata expreesed the highest enzyme activity. The Est-1 showed the rapidest migrating rate among the isozymes.

  • To verify allelopathic effects of Artemisia princeps var. orientalis, a number of laboratory experiments have been performed. Aqueous extracts of leaves, stems and roots of the above species inhibited the seed germination and seedling growth of experimental species, receptor plants. In general, the higher was the concentration of the extracts, the lower was the germination and the growth ratio. Volatile substances released from leaves of the donor plants also inhibited the seed germination and the radicle elongation of receptor plants. Therefore, to find out the inhibitory substances emitted from the wormwood, gas chromatography was employed. As a result of the analysis, 17 KDICical substances were isolated from the leaves and most of them were identified as phenolic compounds.

  • The esterase isozyme in tissue of wild legume plants were separated by horizontal starch gel electrophoresis. Extracts used in this study were prepared from fully expanded young leaf, cotyledon and radicle of seedling and root-nodule of Glycine sola, Vigna vexillata var. tsuscmensis and Trifoliwn repens. The results are as follows; 1. Each tissue examined had a characteristic banding pattcrn. Number of bands in each species, G. soja, V. vexillata, and T . repens, were 14, 8 and 1 1 bands, respectively. And difference in esterase isozyme bands were greater from tissue to tissue than difference between habitat. 2. Est-I, Est-2. Est-3 and Est-4 in G. soja, Est-I in V. vexillata and Est-l and Est-2 in T. repens showed strong cnzyme activity than other enzyme. 3. Esterase isozyme variation in G. soja and T . repens showed more variety than V. vexillata. This is resulted from many genotypic differences within species. 4. The main enLyme among thc esterase isozyme were Est-I. Est-2, Est-3 and Est-4.
