An experimental study was carried out to obtain the fundamental data about the effects of radicals induced injection on premixture combustion. A constant volume combustor divided to the sub-chamber and the main chamber was used. The volume of the sub-chamber is set up to occupy less than 1.5% of that of whole combustion chamber. Radial twelve narrow passage holes are arranged between the main chamber and the sub-chamber. The products including radicals generated by spark ignition in the sub-chamber will derive the simultaneous multi-point ignition in the main chamber. While the equivalence ratio of pre-mixture in the main chamber and the sub-chamber is uniform. We have examined the effects of the sub-chamber volume, the diameter of passage hole, and the equivalence ratio on the combustion characteristics by means of burning pressure measurement and flame visualization. In the case of radical ignition method(RI), the overall turning time including the ignition delay became very short and the maximum burning pressure was slightly increased in comparison with those of the conventional spark ignition method(SI), that is, single chamber combustion without the sub-chamber. The combustible lean limit by RI method is extended to more ER=0.25 than that by SI method. Therefore the decrease of every emission including NOx and the improvement of fuel consumption is anticipated due to lean burn.
A prior fundamental study was executed using a constant volume chamber to improve the burning properties of lean pre-mixture by the injection of active radicals generated in the sub-chamber. In consequence, RI method shows remarkable progress in the aspects of burning velocity and combustible lean limit compared with SI method. In this study, the necessary additional works have been performed to be based on the former results. We changed parameters as the initial temperature and the initial pressure of mixture. And the effects of residual gas at issue in a real engine were investigated. As a result, the effects of initial temperature were significant, but on the other hand, those of initial pressure were slight. The correlation of passage hole number between overall passage hole area was grasped. And the more detailed analysis is required on residual gas.
In an attempt to define the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals, we studied malondialdehyde (MDA) level and the activities of catalase and superoxide dismutase in the renal cortex of rats, and the concentrations of blood urea nitrogen(BUH) and serum creatinine of rats at 24hr after the injection of a 50% solution of glycerol. Sprague-Dawley albino rats weighing 240 to 260 mg were injected intramuscularly with a 50% solution of glycerol(2 mι/kg, 4 mι/kg and 8 mι/kg). The group treated with glycerol showed significantlv higher MDA level and catalase activity, lower SOD activity and higher BUN and serum creatinine concentrations at 24 hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity.
Wistar albino rats were injected subcutaneously with mercuric chloride (5 mg/kg) to define the early biochemical determinants that participate in the pathogenesis of mercuric chloride-induced hepatotoxicity, especially focusing on oxygen free radicals, we studied malondialdehyde(MDA) level and the activities of catalase and superoxide dismutase in the liver of rats at 24, 48 and 72 hr after the injection of mercuric chloride. MDA levels at 24, 48 and 72 hr after the injection of mercuric chloride increased as compared with that of control group. The activities of catalase and superoxide dismutase at 24, 48 and 72hr after the injection of mercuric chloride decreased as compared with that of control group. These results suggest that the depression of the activities of catalase and superoxide dismutase resulting from excessive oxygen free radicals is an important determinant in pathogenesis of mercuric chloride-induced hepatotoxicity.
Toluene is mainly metabolized in liver by oxidative pathway. Oxigen free radicals occur through the process of toluene metabolism Therefore it causes tissue and cell min by the oxygen free radicals from the metabolism of toluene. Melatonin acts as a highly efficient free radical scavenger that protects cells from damage by oxygen free radicals. To test this hypothesis, toluene hepatotoxicity was induced by an abdominal injection of toluene. To see if the melatonin protects the rat's liver, melatonin was administrated orally, at the time of each toluene injection. Aspartate aminotransferase(AST), alanin aminotransferase(ALT), latic dehydrogenase(LDH) and alkaline phosphatase(ALP) levels in serum were measured to estimate hepatic function. Malondialdehyde(MDA), which gives an indirect index of oxidative injury was also measured. Hippuric acid is the last metabolic Production of toluene was measured by HPLC. There were significantly higher in AST, ALT, LDH, MDA and hippuric acid in toluene group, but there were no significant difference in melatonin group except ALT and hippuric acid. There was significantly lower in ALP level in toluene group, but there was no significant difference melatonin group, suggesting a significant hepatotoxicity due to oxygen free radicals through the process of toluene metabolism Melatonin treatment significantly protected hepatic function and free radical-mediated injury in the liver against toluene-induced changes. Accordingly, this study shows that melatonin is helpful in protecting liver injury by acute toluene intoxication.
In an attempt to dofine the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals and $N-acetyl-{\beta}-D-glucosaminidase$ (NAG) activity, we studied 24-hours urine outflow, 24-hours urinary protein excretion and urinary NAG activity after the injection of glycerol and also we studied malondialdehyde(MDA) level and superoxide dismutase(SOD) activity in the kidney of rats at 24hr after the injection of glycerol. Sprague-Dawley albino rats weighing 240 to 260 gm were injected intramuscularly with a 50% solution of glycerol(2ml/kg, 4ml/kg and 8ml/kg). The group treated with glycerol showed significantly lower urine outflow level and urinary protein excretion level and higher urinary NAG activity after the injection as compared to those of control group. Also the group treated with glycerol showed significantly higher MDA level and lower SOD activity at 24hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity and higher urinary NAG activity is an index of renal tubular cell damage in the glycerol-induced nephrotoxicity.
In an attempt to define the effects of Biphenyldimethyl dicarboxylate(DDB) on the lipid peroxidation, oxygen free radical scavenging enzymes activities and hepatic functions in ethanol-induced hepatotoxic rats, we studies malondialdehyde(MDA) level and the activities of catalse, superoxide dismutase(SOD), glutamic-oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) in liver of the rats at 24, 48 and 72 hr after the injection of ethanol and DDB. Sprague-Dalwey albino rats weighing 250 to 280gm were injected intraperitoneally with ethanol(2.5 gm/kg ) only and ethanol plus DDB(300mg/kg ). The result obtained can be summarized as follows : 1) The group treated with ethanol showed significantly higher MDA level and lower catalase and SOD activities at 24, 48 and 72hr after the injection as compared with that of control group. 2) The group treated with ethanol showed significantly higher GOT and GPT activities at 24, 48 and 72hr after the injection as compared with that of control group. 3) The group treated with ethanol plus DDB showed significantly lower MDA level and higher catalase and SOD activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. 4) The group treated with ethanol plus DDB showed significantly lower GOT and GPT activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. These results suggest that the excessive oxygen free radicals resulting from the depression of the activities of catalase and superoxide dismutase is an important determinant in pathogenesis of ethanol-induced hepatotoxicity and DDB has antioxidant effects.

Overstimulation of both kainate (KA) and N-methyl-D-aspartate (NMDA) receptors has been reported to induce excitatoxicity which can be characterized by neuronal damage and formation of reactive oxygen free radicals. Neuroprotective effect of melatonin against KA-induced excitotoxicity have been documented in vitro and in vivo. It is, however, not clear whether melationin is also neuroportective against excitotoxicity mediated by NMDA receptors. In the present work, we tested the in vivo protective effects of striatally infused melatonin against the oxidative stress and neuronal damage induced by the injection of KA and NMDA receptors into the rat striatum. Melatonin implants consisting of 22-gauge stainless-steel cannule with melatonin fused inside the tip were placed bilaterally in the rat brain one week prior to intrastriatal injection of glutamate receptor subtype agonists. Melatonin showed protective effects against the elevation of lipid peroxidation induced by either KA or NMDA and recovered Cu, Zn-superoxide dismutase activities reduced by both KA and NMDA into the control level. Melatonin also clearly blocked both KA- and NMDA-receptor mediated neuronal damage assessed by the determination of choline acetyltransferase activity in striatal monogenages and by microscopic observation of rat brain section stained with cresyl violet. The protective effects of melatonin are comparable to those of DNQX and MK801 which are the KA- and NMDA-receptor antagonist, respectively. It is suggested that melatonin could protect against striatal oxidative damages mediated by glutamate receptors, both non-NMDA and NMDA receptors.
Accumulated evidence indicate that Ginkgo biloba extract (EGb 761) acts as an antioxidant and scavenger of free radicals as well as influencing apoptotis. Earlier studies have employed the inflammatory agent lipopolysaccharide (LPS) to induce severe sepsis. In the present study, we examined whether the intraperitoneal injection of EGb 761 increases the survival rate of mice in the LPS-induced severe sepsis model. The survival rate was significantly increased by 30% in mice administered with 100 mg/kg of EGb 761 but not in mice administered with 50 mg/kg EGb 761. In addition, pre-treatment with EGb 761 increased the survival rate (30%) but post-treatment with EGb 761 did not. These results suggest that EGb 761 may have clinical potential in preventing sepsis induced mortality.
