Ginsenoside-Rp1 inhibits radiation-induced effects in lipopolysaccharide-stimulated J774A.1 macrophages and suppresses phenotypic variation in CT26 colon cancer cells

  • This study investigated the inhibitory effect of ginsenoside-Rp1 (G-Rp1) on the ionizing radiation (IR)-induced response in lipopolysaccharide (LPS)-stimulated macrophages and its effects on the malignancy of tumor cells. G-Rp1 inhibited the activation of IR-induced DNA damage-related signaling molecules and thereby interfered with the IR-increased production of nitric oxide (NO) and interleukin (IL)-1β. The inhibitory effect of G-Rp1 increased the survival rate of mice inoculated with CT26 colon cancer cells by suppressing the phenotypic variation of tumor cells induced by conditioned medium obtained from IR- and LPS-treated J774A.1 macrophages.

γ-Irradiation Induced Adhesion Molecules are Reduced by Vitamin C in Human Endothelial Cells

  • Inflammation is a frequent radiation-induced reaction following therapeutic irradiation. Treatment of human umbilical endothelial cells (HUVEC) with ${\gamma}$-irradiation (${\gamma}$IR) induces the expression of adhesion proteins such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. Since the upregulation of these proteins on endothelial cell Surface has been known to be associated with inflammation, interfering with the expression of adhesion molecules is an important therapeutic target. In the present study, we demonstrate that vitamin C inhibits ${\gamma}$IR induced expression of ICAM-1, VCAM-1, and E-selectin on HUVEC in a dose- and time dependent manner. Vitamin C a1so inhibited the production of Nitric oxide (NO) induced by ${\gamma}$IR. These data suggest that vitamin C has therapeutic potential for the treatment of various inflammatory disorder associated with an increase of endothelial leukocyte adhesion molecules.

HMA also inhibited $\gamma$IR induced production of Nitric oxide (NO). These data suggest that HMA has therapeutic potential for the treatment of various inflammatory disorder associated with an increase of endothelial leukocyte adhesion molecules.

사간 물 추출물의 항염증 효과 (Anti-inflammatory Effects of Belamcanda Chinensis Water Extract)

  • The purpose of this study was to investigate the anti-inflammatory effects of aqueous extract from Belamcanda chinensis (BC) on the RAW 264.7 cells. To evaluate the anti-inflammatory effects of BC, we examined the cytokine productions including nitric oxide (NO), interleukin (IL)-1b, IL-6 and tumor necrosis factor-a (TNF-a) in lipopolysaccharide (LPS)-induced RAW 264.7 cells and also inhibitory mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor kappa B (NF-kB) using Western blot. BC inhibited LPS-induced production of NO, IL-6 and TNF-a but not of IL-1b in RAW 264.7 cells. BC respectively inhibited the activation of MAPKs such as c-Jun NH2-terminal kinase (JNK) and p38 but not of extracelluar signal-regulated kinase (ERK 1/2) and NF-kB in the LPS-stimulated RAW 264.7 cells. Taken together, Our results showed that BC down-regulated LPS-induced NO, IL-6 and TNF-a productions mainly through JNK and p38 MAPK pathway.

Allicin Reduces Adhesion Molecules and NO Production Induced by γ-irradiation in Human Endothelial Cells

  • Background: Inflammation is a frequent reaction following therapeutic irradiation. Since the upregulation of adhesion molecules on endothelial cell surface is known to be associated with inflammation, the expression of adhesion molecules is an important therapeutic target. Methods: Treatment of human umbilical endothelial cells (HUVECs) with ${\gamma}$-irradiation (${\gamma}IR$) induces the expression of adhesion proteins such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. Changes in the expression of these proteins on ${\gamma}$-irradiated HUVECs which had been treated previously with allicin were measured by ELISA. Results: In the present study, we demonstrate that allicin inhibits the ${\gamma}IR$ induced expression of ICAM-1, VCAM-1, and E-selectin on HUVEC in a dose-dependent manner. Allicin was also found to inhibit the ${\gamma}IR$ induced production of nitric oxide (NO). Conclusion: These data suggest that allicin has a therapeutic potential for the treatment of various inflammatory disorders associated with increase numbers of endothelial leukocyte adhesion molecules.

Anti-Inflammatory Effects of Streamed Platycodon grandiflorum against UVB Radiation-Induced Oxidative Stress in Human Primary Dermal Fibroblast

  • Ultraviolet B (UVB) exposure is a risk factor for skin damage resulting in oxidative stress, inflammation, and cell death. The purpose of this study was to investigate the physicochemical properties of Platycodon grandiflorum (PG) to improve its biological activities using a three-step steaming process. We investigated the protective effects of PG and steamed PG extracts on human dermal fibroblasts (HDFs) against UVB radiation-induced oxidative stress and inflammation as well as the underlying mechanisms. The antioxidant potential of the PG extracts was evaluated by measuring the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) scavenging activity. ABTS and DPPH were shown by the 0, 30, and 70% ethanol extracts of 2S-PG and 3S-PG ($IC_{50}$, 28~45 and $27{\sim}30{\mu}g/mL$, respectively). Treatment of UVB-irradiated cells with steamed PG ($25{\sim}400{\mu}g/mL$) did not affect their viability. The streamed PG extract suppressed UVB-induced generation of reactive oxygen species (ROS). In addition, streamed PG extract reduced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) protein expression in UVB-irradiated HDF, regulating nuclear factor $(NF)-{\kappa}B$ expression. These findings suggest that steamed PG extract may be potentially effective against inflammation associated with UVB-induced oxidation stress.

Berberine에 의한 HepG2 세포의 사멸과정에서 활성기산소와 p38 MAP kinase의 역할에 관한 연구 (The Role of ROS and p38 MAP kinase in Berberine-Induced Apoptosis on Human Hepatoma HepG2 Cells)

  • Berberine은 전통적인 중의약재로 이용되어지는 isoquinoline alkaloid로 황련, 황백과 같은 식물에서 주로 추출되며, 약리효과로는 항암, 항염, 항균과 같은 다양한 효과를 나타내는 것으로 알려져 있다. 그러나 간암세포에서 berberine의 산화적 스트레스에 의한 세포사멸기전에 대해서는 아직 밝혀진 바 없다. 따라서 본 연구는 사람의 간암세포인 HepG2 세포에서 berberien의 세포사멸기전에 reactive oxygen species(ROS)와 MAP kinase의 연관성을 조사하였다. Berberine은 HepG2 세포에서 처리 시간과 농도에 의존적으로 세포독성효과를 보였으며, $LD_{50}$은 berberine(50 ${\mu}M$) 처리 후 48시간에서 관찰 되었고, 세포고사의 특징인 핵의 응축 및 분절, DNA의 분절이 확인되었다. 또한 berberine에 의해 caspase-3, p53, p38 그리고 Bax의 발현이 현저하게 증가된 반면, anti-apoptotic 신호기전인 Bc1-2의 발현은 감소되었다. 이와 더불어 세포 내 nitric oxide(NO)와 ROS의 생성도 증가되었다. 본 연구 결과 HepG2 세포에서 berberine은 산화적 스트레스인 ROS와 NO의 생성을 유발하고 p38 MAP kinase와 p53의 인산화를 유도하였으며 미토콘드리아에서 Bcl-2의 감소와 bax의 증가, caspase-3의 활성을 경유하여 DNA의 손상을 통한 세포고사가 이루어지는 것을 확인 하였다.

방사선 육종 차조기와 백출 복합추출물의 항관절염 효과 (Anti-Arthritic Effect of Radiation Mutant Perilla frutescens var. crispa and Atractylodes macrophala koidz.)

  • 본 연구에서는 방사선 육종 차조기와 백출의 복합추출물의 항관절염 효능을 평가하였다. 방사선 육종 차조기와 백출 복합추출물이 세포에 미치는 독성을 확인하기 위해 RAW 264.7 세포에서 MTT 기법으로 세포 생존율을 평가하였다. 방사선 육종 차조기와 백출 복합추출물의 항염증 효능을 확인하기 위해 LPS로 염증을 유도한 RAW 264.7 세포에 방사선 육종 차조기와 백출 복합추출물을 5, 10, 25 ㎍/㎖ 농도로 처리한 후 ROS와 NO level, 염증성 사이토카인의 분비, 염증성 인자인 NF-κB, COX-2, iNOS 등의 발현을 측정하였다. 또한 type II collagen으로 유도한 관절염 모델 동물실험에서 방사선 육종 차조기와 백출 복합추출물을 33.5, 66, 133 mg/kg/day로 처리한 후 항관절염 효능을 확인하였다. 그 결과, 방사선 육종 차조기와 백출 복합추출물은 25 ㎍/㎖ 농도까지 세포독성이 없었으며, LPS로 유도된 RAW 264.7 세포에서 ROS 생성 및 NO의 생성을 5, 10, 25 ㎍/㎖ 농도에서 대조군 대비 유의성 있게 감소시켰으며, 사이토카인(IL-1β, IL-6, TNF-α)의 생성을 유의성 있게 억제시키고, NF-κB, COX-2, iNOS의 발현을 유의성 있게 감소시켜 세포 내에서 뛰어난 항염증 효과를 보였다. 관절염 모델 동물실험에서는 방사선 육종 차조기와 백출 복합추출물이 66.5, 133mg/kg 농도에서 관절염을 유의성 있게 억제시키는 효능을 나타내었다. 본 연구는 방사선 육종 차조기와 백출 복합추출물이 뛰어난 항염증 효능을 나타내는 것을 제시하며, 관절염 질환을 개선하기 위한 건강기능식품 및 치료제의 원료로 개발될 수 있다는 것을 제시한다.

Improvement of Skin Photoaging by Polysaccharide Extract Derived from Tremella fuciformis (White Jelly Mushroom)

  • Chronic ultraviolet (UV) radiation causes photoaging, which represents skin damage, disrupts skin barrier function, and promotes wrinkle formation. We investigated that the polysaccharide extract of an edible basidiomycetous white jelly mushroom, Tremella fuciformis, (TF-Glucan®) exhibited statistically photoprotective activity by inhibiting matrix metalloproteases (MMPs) and increasing collagen synthesis, and an anti-inflammatory activity by inhibiting nitric oxide and pro-inflammatory cytokines at the concentrations of less than 1000 ㎍/ml, which is not cytotoxic (p < 0.05). Additionally, TF-Glucan® increased the expression of involucrin and filaggrin to prevent the disruption of UVB-induced barrier function (p < 0.05). TF-Glucan® was assessed as a safe material by the human primary skin irritation (1, 3, 5%), human repeated insult patch test (no sensitization at 5%), 3T3 NRU phototoxicity assay (no phototoxicity, PIF < 2, MPE < 0.1), eye irritation test test by BCOP (no category, IVIS ≤ 3) and local lymph node assay (negative at 10, 25, 50%) for identifying potential skin sensitizing. These results suggest that TF-Glucan® may be useful as an anti-photoaging ingredient for developing cosmeceuticals.
