Pleiotropic regulator 1 (PLRG1), a highly conserved element in the spliceosome, can form a NineTeen Complex (NTC) with Prp19, SPF27, and CDC5L. This complex plays crucial roles in both pre-mRNA splicing and DNA repair processes. Here, we provide evidence that PLRG1 has a multifaceted impact on cancer cell proliferation. Comparing its expression levels in cancer and normal cells, we observed that PLRG1 was upregulated in various tumor tissues and cell lines. Knockdown of PLRG1 resulted in tumor-specific cell death. Depletion of PLRG1 had notable effects, including mitotic arrest, microtubule instability, endoplasmic reticulum (ER) stress, and accumulation of autophagy, ultimately culminating in apoptosis. Our results also demonstrated that PLRG1 downregulation contributed to DNA damage in cancer cells, which we confirmed through experimental validation as DNA repair impairment. Interestingly, when PLRG1 was decreased in normal cells, it induced G1 arrest as a self-protective mechanism, distinguishing it from effects observed in cancer cells. These results highlight multifaceted impacts of PLRG1 in cancer and underscore its potential as a novel anti-cancer strategy by selectively targeting cancer cells.
Nebulin is a giant ($600{\sim}900$ kDa), modular sarcomeric protein proposed to regulate the assembly, and to specify the precise lengths of actin filamints in vertebrate skeletal muscles. Recently, There is an evidence that the nebulin also expressed in non muscle tissue, brain and liver. We identified a new isoform of nebulin from adult brain library by PCR screening. It contains two simple-repeats exon 165, 166 and linker-repeats exon $154{\sim}161$ except exon 159. The nebulin modules M160 to M170 (exon 150 to exon 161) has been shown to bind desmin. In mature striated muscle, desmin intermediate filaments surround Z-discs and link individual myofibrils laterally at their Z-discs and to other intracellular structures, including the costameres and the intercalated discs of the sarcolemma, sarcoplasmic reticulum, mitochondria, T-tubules, and nuclei. Therefore, it is an interesting possibility that the differential splice pathways within the linker region of nebulin modify the affinity of nebulin's interaction with desmin. The specific interactions of nebulin and desmin were confirmed in vivo by yeast two hybrid experiments. To verify in the cellular level the interaction between nebulin isoform and desmin, we transfected COS-7 cell with EGFP-tagged nebulin and DsRed-tagged desmin. Based on evidence showing that despite exon 159 was deleted, the new isoform of nebulin was interact with desmin. This suggest that nebulin in brain may interact with another intermediate filament. The conservation of these ligand-binding capacity in brain and skeletal nebulins suggest that nebulins may have conserved roles in brain and skeletal muscle.

Purpose : To detect differentially expressed genes in the patients with uterine cervical cancer during the radiation therapy. Materials and Methods : In patients with biopsy proven uterine cervical cancer, we took tumor tissue just before radiation therapy and at 40 minutes after external irradiation of 1.8 Gy. Total RNAs isolated from non-irradiated and irradiated tumor tissue samples were analyzed using the differential-display reverse transcription-polymerase chain reaction (DDRT-PCR). Complementary DNA (cDNA) fragments corresponding to differentially expressed messenger RNAs(mRNAs) were eluted, and cloned. The differential expression of the corresponding mRNAs was confirmed by reverse northern blot. Differentially expressed cDNA bands were sequenced. Nucleotide sequence data were analyzed in the Gene Bank and EMBL databases via the BLAST network sewer to identify homologies to known genes or cDNA fragments. Expression pattern of down-regulated clone was examined using RT-PCR in S patients undergoing radiotherapy. Results : We identified 18 differentially expressed bands by DDRT-PCR, which were eluted and cloned. There were 10 up-regulated clones and 1 down-regulated clone in reverse northern blot. One cDNA fragment had homology to chemokine receptor CXCR4, four were identified as Human ESTs in the EMBL database in EST clones. Down-regulated CxCa-11 was also down regulated in all patients. Conclusion : Using the DDRT-PCR, we have identified 10 up-regulated and 1 down-regulated clone(s) in the patients with uterine cervical cancer during the radiation therapy. The clinical relevance and the functions of these genes will be further investigated.

One of the issues currently facing nuclear power plants is how to store spent nuclear waste materials which are contaminated with radionuclides such as $^{134}Cs$, $^{135}Cs$, and $^{137}Cs$. Bioremediation processes may offer a potent method of cleaning up radioactive cesium. However, there have only been limited reports on $Cs^+$ tolerant bacteria. In this study, we report the isolation and identification of $Cs^+$ tolerant bacteria in environmental soil and sediment. The resistant $Cs^+$ isolates were screened from enrichment cultures in R2A medium supplemented with 100 mM CsCl for 72 h, followed by microbial community analysis based on sequencing analysis from 16S rRNA gene clone libraries(NCBI's BlastN). The dominant Bacillus anthracis Roh-1 and B. cereus Roh-2 were successfully isolated from the cesium enrichment culture. Importantly, B. cereus Roh-2 is resistant to 30% more $Cs^+$ than is B. anthracis Roh-1 when treated with 50 mM CsCl. Growth experiments clearly demonstrated that the isolate had a higher tolerance to $Cs^+$. In addition, we investigated the adsorption of $0.2mg\;L^{-1}$$Cs^+$ using B. anthracis Roh-1. The maximum $Cs^+$ biosorption capacity of B. anthracis Roh-1 was $2.01mg\;g^{-1}$ at pH 10. Thus, we show that $Cs^+$ tolerant bacterial isolates could be used for bioremediation of contaminated environments.
The chlorophyll fluorescence and photosynthetic $CO_2$ fixation capacity of leaves from three major crop trees found on Jeju Island, Camellia sinensis L., Camellia japonica L., and Citrus unshiu M., were analyzed. The photosynthetic $CO_2$ fixation rate of C. sinensis was similar to that of C. unshiu, and much higher than that of C. japonica which belongs to the same genus. Stomatal conductance in the three species was high at dawn and low during daytime. The intercellular $CO_2$ concentration of the three species was also high at dawn and decreased at midday. The transpiration rate showed an opposite trend from the intercellular $CO_2$ concentration. The photochemical efficiencies of PSII (Fv/Fm) in C. sinensis were slightly lower at midday compared to the level at dawn and/or dusk. The decline in Fv/Fm of C. sinensis at midday was much smaller than that of C. japonica. These results indicate that C. sinensis is better acclimated to high levels of radiation under natural conditions in late summer, although its PSII reaction center was inhibited by strong radiation. Of the chlorophyll fluorescence parameters in the species, the RC/CS decreased significantly while the ABS/RC, TRo/RC, ETo/RC, and DIo/RC increased significantly at midday in late summer. However, C. unshiu did not show significant changes in these values depending on the time of day. Among the three species, the daily $CO_2$ fixation rate in C. sinensis ($320.1mmol\;m^{-2}d^{-1}$) was the highest, followed by that of C. unshiu ($292.5mmol\;m^{-2}d^{-1}$) and C. japonica ($244.8mmol\;m^{-2}d^{-1}$). Thus, C. sinensis may be a valuable crop tree in terms of the uptake of $CO_2$ under natural field conditions.

3D histology is a imaging system for the 3D structural information of cells or tissues. The synchrotron radiation propagation phase contrast micro-CT has been used in 3D imaging methods. However, the simple phase contrast micro-CT did not give sufficient micro-structural information when the specimen contains soft elements, as is the case with many biomedical tissue samples. The purpose of this study is to develop a new technique to enhance the phase contrast effect for soft tissue imaging. Experiments were performed at the imaging beam lines of Pohang Accelerator Laboratory (PAL). The biomedical tissue samples under frozen state was mounted on a computer-controlled precision stage and rotated in $0.18^{\circ}$ increments through $180^{\circ}$. An X-ray shadow of a specimen was converted into a visual image on the surface of a CdWO4 scintillator that was magnified using a microscopic objective lens(X5 or X20) before being captured with a digital CCD camera. 3-dimensional volume images of the specimen were obtained by applying a filtered back-projection algorithm to the projection images using a software package OCTOPUS. Surface reconstruction and volume segmentation and rendering were performed were performed using Amira software. In this study, We found that synchrotron phase contrast imaging of frozen tissue samples has higher contrast power for soft tissue than that of non-frozen samples. In conclusion, synchrotron radiation propagation phase contrast cryo-microCT imaging offers a promising tool for non-destructive high resolution 3D histology.
To investigate the radiation effect of P-32 on the soybean plants, the seeds treated with various levels of P-32 solution were sown and cultured in the pots. The growth of the plants and the contents of the macroelements were observed and the following results were obtained. 1) The linear growth of the plants at the early stage seemed to have been promoted by the low-level P-32 treatemnt. At the later growing stage, however, this difference among treatments were less conspicuous. 2) The plants of high level P-32 application showed some growth damaging symptom at the early growing stage. Later this damage was recorded as the time went on and these plants showed even better growth than the control. As a result at the late growing stage, they ensued highest growth. 3) The plants showed in general more growth at the low activity level than at the high-level at the early growing stage. At the late stage, however, the high-level activity promoted more growth than the low-level. 4) At the early growing stage P-32 treatment produced in general significantly more lower than control. At the later stage, however, this difference was not clearly seen. 5) The P-32 treatment seemed to have stimulated earlier florescence and this tendency was more clearly observable eapecially at the high activity level. 6) The weight of the air-dried seeds tended to be increased through P-32 treatment by 10-45%. This tendency was clearly observed especially at the low-level activity. 7) As for the contents of the various macroelements in the leaves, the nitrogen showed significantly larger contents at the middle level(S1) P-32 treatments. The phosphorous contents showed also highest at the middle levels activity and lower both at the high and low-activity levels. The potassium contents was proved, on the contrary, higher at the low-level activity and lower at the high-level. 8) The nitrogen contents in the stems was found significantly higher than control, except at the low-activity level. The phosphorous showed higher contents at the low-activity level and no significant difference at the high-activity level. As for the contents of potassium, calcium and magnesium, three seemed no significant difference among treatments. However, the magnesium showed somewhat higher content at the low-activity level, whereas the calcium was proved high than control. 9) The inorganic contents in the root showed that N and P in the P-32 treated plant were significantly higher than the control and the K-contents was, on the contrary, significantly higher at the control than the rest of the treatments. As for the calcium and magnesium there showed no difference among all treatments.

Background: Early onset breast cancer is associated with poor outcomes but variable results have been reported. It is a significant problem in Pakistani women but remains under reported. Breast conservation plays an important role in surgical management of this younger patient group. The objective of this study was to determine the outcome of breast conservative therapy in patients with early onset breast cancer in our population and compare it with their older counterparts. Materials and Methods: A review of patients with invasive breast cancer who underwent breast conservation surgery at Shaukat Khanum Cancer Hospital from 1997 to 2009 was performed. Patients were divided into two groups i.e. Group I age ${\leq}40$ and Group II >40 years. A total of 401 patients with breast cancer were identified in Group I and 405 patients in Group II. Demographics, histopathological findings and receptor status of the two groups were compared. The Chi square test was used for categorical variables. Outcome was assessed on basis of 10 year locoregional recurrence free survival (LRRFS), disease free survival (DFS) and overall survival (OS). For survival analysis Kaplan Meier curves were used and significance was determined using the Log rank test. Cox regression was applied for multivariate analysis. Results: Median follow up was 4.31 (0.1-15.5) years. Median age at presentation was 34.6 years (17-40) and 51.9 years (41-82) for the two groups. Groups were significantly different from each other with respect to grade, receptor status, tumor stage and use of neoadjuvant therapy. No significant difference was present between the two groups for estimated 10 year LRRFS (86% vs 95%) (p=0.1), DFS (70% vs 70%) (p=0.5) and OS (75% vs 63%) (p=0.1). On multivariate analysis, tumor stage was an independent predictor of LRRFS, DFS and OS. Conclusions: Early onset breast cancer is associated with a distinct biology but does not lead to poorer outcomes in our population.
To investigate the effects of internal radiation of various intensityon the test crop(flax), seeds of four levels of activity($15.5{\times}10-4 $\mu$c/grain, 9.0{\times}10-2$\mu$c/grain, 2.2{\times}100$\mu$c/grain and 3.8{\times}10-1$\mu$c/grain)$ which had been obtained by immersing them into various concentrations of P-32 original solution (total activity: 90 mc, To: 3/21) for 24 hours at room temperature, were germinated, transplanted later into pots, and the rate of germination and the successive growth were observed, and the inorganic conents of the plant top were analyzed. The results are as follows: 1. At the early stage of germination, the plants manifested themselves both inhibitory and promotive effects at higher and lower activity levels respectively, compared those of the control. These difference of growth on account of different levels of activity appeared, however, to be gradually narrowed in the course of time after germination, except at the highest activity. 2. Two weeks after transplanting, the plants of the lowest activity showed more vigorous growth than those of control. The plants belonging to the other activity levels on the other hand, tended to be less growing, the higher the activity. However, this growth gap between treatments seemed to be progressively closed one month after transplanting. 3. Most of the leaves and stems of the plants belonging to the highest activity level ($3.8{\times}101$\mu$c/grain$) were withered during the early stage of growth, and this damage did not recover. 4. Practically no difference of growth was observed among treatments(excluding that of highest activity) one and half months after transplanting. 5. The fluorescence tended to be mroe delayed than the control, as the activity decreased. 6. There was a tendency that the number of pods harvested were larger in the plants treated with P-32 than that of the control. 7. The proportion of fiber in the plants at harvest appeared to be larger at lower activity and smaller at higher P-32 concentration than that of control. 8. As for the inorganic contents of the plant top harvested the floowing tendency was observed: (1) Nitrogen content was highest at the highest activity level at which the poorest growth ensued. (2) There was no clear difference of phosphorus content among treatments. (3) The contents of potassium and magnesium were higher than control at the medium levels of activity. (4) Calcium content of all treated blocks was found to be more than that of control.
The hepatoprotective activity of flavonol glycosides rich fraction (F-2), prepared from 70% alcohol extract of the aerial parts of V calcarata Desf., was evaluated in a rat model with a liver injury induced by daily oral administration of $CCl_4$ (100 mg/kg, b.w) for four weeks. Treatment of the animals with F-2 using a dose of (25 mg/kg, b.w) during the induction of hepatic damage by $CCl_4$ significantly reduced the indices of liver injuries. The hepatoprotective effects of F-2 significantly reduced the elevated levels of the following serum enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH). The antioxidant activity of F-2 markedly ameliorated the antioxidant parameters including glutathione (GSH) content, glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), plasma catalase (CAT) and packed erythrocytes glucose-6-phosphate dehydrogenase (G6PDH) to be comparable with normal control levels. In addition, it normalized liver malondialdehyde (MDA) levels and creatinine concentration. Chromatographic purification of F-2 resulted in the isolation of two flavonol glycosides that rarely occur in the plant kingdom, identified as quercetin-3,5-di-O-$\beta$-D-diglucoside (5) and kaempferol-3,5-di-O-$\beta$-D-diglucoside (4) in addition to the three known compounds identified as quercetin-3-O-$\alpha$-L-rhamnosyl- (${\rightarrow}6$)-$\beta$-D-glucoside [rutin, 3], quercetin-3-O-$\beta$-D-glucoside [isoquercitrin, 2] and kaempferol-3-O-$\beta$-D-glucoside [astragalin, 1]. These compounds were identified based on interpretation of their physical, chemical, and spectral data. Moreover, the spectrophotometric estimation of the flavonoids content revealed that the aerial parts of the plant contain an appreciable amount of flavonoids (0.89%) calculated as rutin. The data obtained from this study revealed that the flavonol glycosides of F-2 protect the rat liver from hepatic damage induced by $CCl_4$ through inhibition of lipid peroxidation caused by $CCl_4$ reactive free radicals.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
