Effect of UV-C Irradiation on the Quality of Imported Dried Fish during Storage (UV-C 조사가 수입 건어포류의 저장 중 품질에 미치는 영향)

  • The effect of UV-C irradiation on microbial growth and the quality of imported dried filefish and octopus during storage was examined. Samples of imported dried filefish and octopus were irradiated at 0, 5, 10 and $20\;kJ/m^2$ and stored for 3 months at $20^{\circ}C$ or for 6 days at $4^{\circ}C$. Exposure times of 5 min 33 sec, 11 min 6 sec, and 22 min 12 sec were used. UV-C treatment of the imported dried filefish and octopus decreased the populations of aerobic bacteria, yeasts and molds in proportion to radiation dose. Compared to the control, total aerobic bacteria, and yeast and mold populations were significantly lower (1-2 log CFU/g) with UV-C treatment of $20\;kJ/m^2$. UV-C irradiation caused negligible changes in the Hunter color L, a and b values. These results indicate that UV-C irradiation could be useful in inhibiting microbial growth on imported dried fish without impairing quality during storage.

A Study on Improvement of Image Quality Decrease due to Tooth Restoration in Facial CT (안면부 CT 검사 시 치아 충전물에 의한 화질 저하 개선 방안에 관한 연구)

  • The purpose of this study was to investigate the degree of image degradation and the improvement of image quality caused by the density difference between the orthodontic filling material and the surrounding anatomical structure during the examination of the facial CT by quantitative and qualitative analysis. The teeth were scanned using 64-MDCT (Discovery 750 HD, GE HEALTH CARE, Milwaukee, USA). The teeth were scanned and compared according to tube voltage, silicone application, and MAR application. As a result, 10.36% CT value decreased at 140 kVp and 5.81% decrease at the application of silicon material. As a result of the qualitative evaluation, it was evaluated that 7 of the 10 observers and 3 of the acceptors were applied to the MAR algorithm. Therefore, it is possible to reduce the unnecessary burden on the radiation exposure dose as well as to reduce the loss of image data by reducing the high density artifacts, as well as the inspection parameters used in the current clinical application and various algorithms that can reduce the high density artifacts. It can be expected to provide a lot of image information.

Performance Assessment of Low- and Intermediate-Level Radioactive Waste Disposal Facility in Korea by Using Complementary Indicator: Case Study with Radionuclide Flux (보조지표를 활용한 중·저준위 처분시설 성능평가: 방사성 핵종 플럭스 사례연구)

  • The use of complimentary indicators, other than radiation dose and risk, to assess the safety of radioactive waste disposal has been discussed in a number of publications for providing the reasonable assurance of disposal safety and convincing the public audience. In this study, the radionuclide flux was selected as performance indicator to appraise the performance of engineered barriers and natural barrier in the Wolsong low- and intermediate-level waste disposal facility. Radionuclide flux showing the retention capability by each compartment of the disposal system is independent of assumptions in biosphere model and exposure pathways. The scenario considered as the normal scenario of disposal facility has been divided into intact or degraded silo concrete conditions. In the intact silo concrete, the radionuclide flux has been assessed with respect to the radionuclide retardation performance of each engineered barrier. In the degraded silo concrete, the radionuclide flux has been explored based on the performance degradation of engineered barriers and the relative significance of natural barrier quantitatively. The results can be used to optimally design the near-surface disposal facility being planned as the second project phase. In the future, additional complimentary indicators will be employed for strengthening the safety case for improving the public acceptance of low- and intermediate-level waste disposal facility.

Flow Characteristics Evaluation in Reactor Coolant System for Full System Decontamination of Kori-1 Nuclear Power Plant (고리1호기 계통제염을 위한 원자로냉각재내 유동 특성 평가)

  • The Kori-1 Nuclear Power Plant (NPP), WH 2-Loop Pressurized Water Reactor (PWR) operated for approximately 40 years in Korea, was permanently ceased on June 18, 2017. To reduce worker exposure to radiation by reducing the dose rate in the system before starting main decommissioning activities, the permanently ceased Kori-1 NPP will be subjected to full system decontamination. Generally, the range of system decontamination includes Reactor Pressure Vessels (RPV), Pressurizer (PZR), Steam Generators (SG), Chemical & Volume Control System (CVCS), Residual Heat Removal System (RHRS), and Reactor Coolant System (RCS) piping. In order to decontaminate these systems and equipment in an effective manner, it is necessary to evaluate the influence of the flow characteristics in the RCS during the decontamination period. There are various methods of providing circulating flow rate to the system decontamination. In this paper, the flow characteristics in Kori-1 NPP reactor coolant according to RHR pump operation were evaluated. The evaluation results showed that system decontamination using an RHR pump was not effective at decontamination due first to impurities deposited in piping and equipment, and second to the extreme flow unbalance in the RCS caused deposition of impurities.

Evaluation and Experimental Production of Single-Phase Full-wave Rectification Type for X-ray Equipment of High Precision (고정밀도의 단상전파정류형 X선 장치의 제작 및 평가)

  • Diagnosis X-ray equipment localized at 1950's but it is developed suddenly at 1960's with demand together. Manufacture of Diagnostic X-ray equipment is controled by the KS regulation and the Ministry of Health and Welfare because of hazardous element etc. exposure by radiation. Most of diagnostic X-ray equipment ware single phase and three phase full-wave rectification but from 1980's it transforms it was exchanged in inverter type X-ray equipment. Inverter type X-ray equipment produces approximately 50~80% more average photon intensity then single phase full-wave rectification and the accuracy is high. But from a clinic it dose not use because expensive therefor the efficiency improvement of single phase full-wave rectification is necessary. We produced single phase full-wave rectification X-ray equipment control unit, high tension transformer, filament heating transformer, rectification circuit, high tension cable and others and evaluated efficiency, in result which is excellent compare with Rule of Safety Management and KS regulation.

Effects of 2-deoxy-D-glucose and quercetin on osteoblastic differentiation and mineralization in irradiated MC3T3-E1 cells (2-deoxy-D-glucose와 quercetin이 방사선조사 MC3T3-E1 골모세포주의 분화와 석회화에 미치는 영향)

  • Purpose: To investigate the in vitro response of MC3T3-E1 osteoblastic cells to X-ray in the presence and absence of 2 deoxy-D-glucose (2-DG) and quercetin (QCT). Materials and Methods: The MC3T3-E1 cells were cultured in an ${\alpha}-MEM$ supplemented with 5 mM 2-DG or $10{\mu}M$ QCT and then the cells were incubated for 12 h prior to irradiation with 2, 4, 6, and 8Gy using a linear accelerator (Mevaprimus, Germany) delivered at a rate of 1.5 Gy/min. At various times after the irradiation, the cells were processed for the analyses of proliferation, viability, cytotoxicity, and mineralization. Results: Exposure of the cells to X-ray inhibited the tritium incorporation, 3-(4, 5-dimethylthiazol-2yl-)-2, 5-diphenyl tetrazolium bromide (MTT)-reducing activity, and alkaline phosphatase (ALP) activity, and caused cytotoxicity and apoptosis in a dose-dependent manner of the X-ray. This effect was further apparent on day 3 and 7 after the irradiation. RA+2-DG showed the decrease of DNA content, cell viability, and increase of cytotoxicity rather than RA. ALP activity increased on day 7 and subsequently its activity dropped to a lower level. 2-DG suppressed the calcium concentration, but visual difference of number of calcified nodules between RA and RA+2-DG was not noticed. RA+QCT showed the increase of DNA content, cell viability, but decrease of cytotoxicity and subG1 stage cells in the cell cycle, and increased calcified nodules in von Kossa staining rather than the RA. ALP activity showed significant increases on day 7 and subsequently its activity dropped to a lower level. Conclusion: The results showed that the 2-DG acted as a radiosensitizing agent and QCT acted as a radiosensitizing agent respectively in the irradiated MC3T3-E1 osteoblast-like cells.

Comparative evaluation of computed tomography for dental implants on the mandibular edentulous area (하악 무치악 부위의 임플란트 이식을 위한 전산화단층촬영 영상의 비교 평가)

  • Purpose: The purpose of this study was to evaluate the clinical usefulness of the recently developed multi-detector computed tomography and cone beam computed tomography in pre-operative implant evaluation, by comparing them with the single detector computed tomography, already confirmed for accuracy in this area. Materials and Methods: Five partially edentulous dry human mandibles, with $1{\times}1mm$ gutta percha cones, placed in 5mm intervals posterior to the mental foramen on each side of the buccal part of the mandible, were used in this study. They were scanned as follows: 1) Single detector computed tomography: slice thickness 1mm, 200mA, 120kV 2) Multi-detector computed tomography: slice thickness 0.75mm, 250mA, 120kV 3) Cone beam computed tomography: 15mAs, 120kV Axial images acquired from three computed tomographies were transferred to personal computer, and then reformatted cross-sectional images were generated using V-Implant $2.0^{(R)}$ (CyberMed Inc., Seoul, Korea) software. Among the cross-sectional images of the gutta perch a cone, placed in the buccal body of the mandible, the most precise cross section was selected as the measuring point and the distance from the most superior border of the mandibular canal to the alveolar crest was measured and analyzed 10 times by a dentist. Results: There were no significant intraobserver differences in the distance from the most superior border of the mandibular canal to the alveolar crest (p>0.05). There were no significant differences among single detector computed tomography, multi-detector computed tomography and cone beam computed tomography in the distance from the most superior border of the mandibular canal to the alveolar crest (p>0.05). Conclusion: Multi-detector computed tomography and cone beam computed tomography are clinically useful in the evaluation of pre-operative site for mandibular dental implants, with consideration for radiation exposure dose and scanning time.

  • The Recently, large area matrix-addressed image detectors are investigated for X-ray imaging with medical diagnostic and other applications. In this paper, a new flat panel gas detector for diagnostic X-ray imaging is proposed, and its characteristics are investigated. The research of flat panel gas detector is not exist at all. Because of difficulty to inject gas against to atmospheric pressure. So almost gas detector made by chamber shape. We made flat panel sample by display technique. (ex: PDP, Fed, etc.) The experimental measurements, the transparent electrodes, dielectric layer, and the MgO protection layer were formed in front glass. And, the X-ray phosphor layer and address electrodes are formed in the rare glass. The dark current, the x-ray sensitivity and linearity as a function of electric field were measured to investigate the electrical properties. From the results, the stabilized dark current density and the significant x-ray sensitivity were obtained. And the good linearity as a function of exposure dose was showed in wide diagnostic energy range. These results means that the passive matrix-addressed flat panel gas detector can be used for digital x-ray imaging.

Purification and Characterization of Anticarcinogenic Compound from Corni fructus (산수유에 함유된 항암물질의 정제 및 특성)

  • Chloroform layer from methanol extract of Corni fructus (Cornaceae) showed strong antiproliferation effect on human cancer cell lines by SRB assay. Anticarcinogenic-active compound was isolated and purified by silica gel column and thin layer chromatograpies, and identified as ursolic acid ($3{\beta}$-hydroxyrus-12-ene-28-oic acid, MW:456) by mass and IR spectrophotometries, and $^1H-and\;^{13}C-NMRs$. The compound inhibited proliferation of A549 (human lung cancer cell line) and MCF-7 (human breast cancer cell line) cells in dose-dependant manner when treated for 48 hr. Inhibition rates of both cells were over 40% and 90% compared with control cells at the $30\;{\mu}g/mL\;and\;100\;{\mu}g/mL$, respectively. Morphology of cells treated with the compound for 15 hr at $10\;{\mu}g/mL$ was distorted with shrinked cell mass, and cell number was lower than that of control cells. Cell cycle analysis showed sub-G1 phase arrest in both cell lines following 15 hr exposure to the compound; % of cell phase increased to 11.7 and 11.2% compared to the control of 4.0% and 2.1% in A549 and MCP-7 cells, respectively.

Effect of retinoic acid on the radiosensitivity of normal human oral keratinocyte (Retinoic acid가 사람 정상 구강각화세포의 방사선감수성에 미치는 영향에 관한 연구)

  • Purpose : To evaluate the effect of all-trans-retinoic acid (ATRA) on the radiosensitivity of normal human oral keratinocyte (NHOK). Materials and methods: Relative cell survival fraction including SF2 (survival fraction at 2 Gy) was calculated on the basis of colony formation assay. Data were fitted to the linear-quadratic model to establish the survival curve and calculate α and β values. Using flow cytometry at 1, 2, 3, 4, and 5 days after exposure to 2 and 10 Gy irradiation, cell cycle arrest and apoptosis were analysed. To understand the molecular mechanism of the radiosensitization of ATRA on NHOK, proteins related with apoptosis and cell cycle arrest were investigated by Western blot analysis. Results: Treatment with ATRA resulted in a significant decrease of SF2 value for NHOK from 0.63 to 0.27, and increased α and β value, indicating that ATRA increased radiosensitivity of NHOK. ATRA increased LDH significantly, but increasing irradiation dose decreased LDH, suggesting that the radiosensitizing effect of ATRA is not directly related with increasing cell necrosis by ATRA. ATRA did not induce appotosis but increased G2 arrest after 10 Gy irradiation, implying that the increased radiosensitivity of NHOK may be due to a decrease in mitosis casued by increasing G2 arrest. ATRA inhibited the reduction of p53 at 3 days after l0Gy irradiation and increased p21 at 1 day after 10 Gy irradiation. Further study is required to determine the precise relationship between this effect and the radiosensitizing effect of A TRA. Conclusion: These results suggested that ATRA increase radiosensitivity by inhibiting mitosis caused by increasing G2 arrest.
