Effect of Feeding Levels and Periods of Dietary Rhus verniciflua Stokes at Finishing Stage on the Meat Quality of Pigs during Refrigerated Storage (옻나무 사료의 급여수준과 기간이 비육돈의 냉장중 육질에 미치는 영향)

  • This study was carried out to investigate the effect of feeding levels and periods of Rhus verniciflua Stokes (RVS) on the quality of M. longissimus from gilts (Landrace×Yorkshire×Duroc) at finishing stage. The gilts were fed diets containing 0 (control), 2 and 4% RVS for 5 weeks (the latter feeding periods) and 8 weeks (the first feeding periods+the latter feeding periods) before slaughtering, respectively. Samples were stored at 3±0.2℃ for 8 days. Crude fat content was decreased by feeding diets of 4% RVS (p<0.05). Water-holding capacity (WHC) was higher in RVS treatments than in control during storage (p<0.05). L*(Lightness) value was higher in 5 weeks-RVS treatment than in control during storage (p<0.05). a*(Redness) and b*(yellowness) were not affected by the levels and periods of RVS supplementation. Thiobarbituric acid reactive substances (TBARS) value was lower in RVS treatment than in control (p<0.05), and the 5 weeks with 4% RVS treatment showed the highest antioxidative activity. Fatty acid compositions of pork were changed by RVS feeding levels and periods. The meat supplemented with RVS showed high levels of unsaturated fatty acid (UFA) composition and monounsaturated fatty acid (MUFA)/saturated fatty acid (SFA) ratio compared with control. The dietary 8 weeks-RVS treatments had lower stearic acid(18:0) and higher polyunsaturated fatty acid (PUFA) contents than in other treatments. Melting point of backfat also decreased by increasing the level and period of RVS supplementation (p<0.05). Therefore, RVS fed pork decreased fat content and melting point, increased WHC and UFA composition, and delayed lipid oxidation during storage.

  • Rhus verniciflua Stokes (RVS) is a widely used herbal plant with various biological properties. Our previous study using in vitro platelet aggregation in whole blood showed that the fractions of RVS had strong anti-aggregatory activity. In this study, using in vitro platelet aggregation in PRP and coagulation parameters, to investigate the anti-platelet activity and anticagulant effects of RVS ethyl acetate layer, the layer was subsequently fractionated by ODS columm chromatograph (50% MeOH).   • This study was carried out to investigate the effect of dietary RVS on the WHC, color stability, and lipid oxidation of Hanwoo (Korean cattle) beef during cold storage after thawing, RVS fed Hanwoo (Korean cattle) beef was more effective in increasing WHC, color stability and delaying lipid oxidation than that without RVS in diet. And 4% RVS fed Hanwoo (Korean cattle) beef showed the greatest effects.

In this study. to investigate the anti-aggregatory activity and antioxidative effects of RVS ethyl acetate layer. the layer was subsequently fractionated by ODS columm chromatograph (50% MeOH). As a result. the fraction 3 was most inhibited the aggregation of platelet in rat whole blood induced by thrombin and all fraction of RVS was detected strong antioxidative effect. (omitted)

  • The Rhus verniciflua Stokes (乾漆-RVS) has been used in traditional East Asia medicine for the therapy of gastritis, stomach cancer, although the mechanism for the biological activity is unclear. In the present study aims to investigate RVS extract contributes to growth inhibitory effect and it's the molecular mechanism on the human gastric cancer cells. AGS (gastric cancer cells) and RIEI (normal cells) were treated to different concentrations and periods of RVS extract $(10{\;}{\sim{{\;}100{\;}ug/mil)$. Growth inhibitory effect was analyzed by measuring FACS study and MTS assay. Cell cycle inhibition was confirmed by measuring CDK2 kinase activity by immunoprecipitation and kinase assay. And apoptosis was confirmed by surveying caspase cascades activation using a pan caspase inhibitor Exposure to RVS extract (50 ug/mll) resulted in a synergistic inhibitory effect on cell growth in AGS cells. Growth inhibition was related with the inhibition of proliferation and induction of apoptosis. The extract induces Gl -cell cycle arrest through the regulation of cyclins, the induction of p27kip1, and the decrease CDK2 kinase activity. And upregulated p27kip1 level is caused by protein stability increment by the reduction of S-phase kinase-associated protein 2 (Skp2), a key molecule related with p27kip1 ubiquitination and degradation, and do novo protein synthesis. Besides, 乾漆 extract induces apoptosis through the expression of Bax, poly(ADP-ribose) polymerase (PARP) and activation of caspase-3. RVS extract induces Gl -cell cycle arrest via accumulation of p27kip1 and apoptosis in human gastric cancer cells but not in normal cells, therefore we suggest that the extract can be used as a novel class of anti-cancer drugs.

  • Skin contact allergy (SCA) is not life-threatening, but a large number of people have been suffered from the reactions caused by various kinds of chemicals and products. Thus, in this study, radiation technology was employed to improve the traditional herb addition method on the SCA reduction of Rhus verniciflua sap (RVS). Rhus verniciflua has traditionally been used as an herbal medicine plant, but its urushiol derivatives are known as a major allergen for the SCA. The present study was commenced to assess the allergenicity of both gamma-irradiated and non-irradiated RVS by using guinea-pig maximization test (GPMT) in order to probe the mechanism of an SCA. In the acute dermal irritation assays, non-irradiated RVS caused erythema, but the irradiated RVS did not provoke any erythema on the abdominal skin of the guinea pigs. From the result of the GPMT, urushiols, the main chemical components of RVS, were identified as an extreme skin sensitizer, and the removal of urushiols by irradiation extremely reduced the erythema. These results suggest that radiation technology is a novel method to reduce SCA through the removal of urushiols of RVS.

Antioxidant activities of Rhus verniciflua seed extract and quality characteristics of fermented milk containing Rhus verniciflua seed extract (옻씨 추출물의 항산화 활성과 옻씨 추출물을 첨가한 발효유의 품질특성)

  • This study was carried out to investigate the quality characteristics of fermented milk containing Rhus verniciflua seed (RVS) extracts (0.0%, 0.1%, 0.3% and 0.5%) and antioxidant activity of RVS extracts. The proximate composition of RVS was moisture (4.76%), crude ash (1.40%), crude fat (5.33%) and crude protein (11.10%). A total polyphenol content of 70% ethanol extract of RVS ($554{\pm}2.64mg/g$) was higher than that of RVS water extract ($145{\pm}3.47mg/g$). 70% ethanol extract ($1103{\pm}6.42mg/g$) of RVS showed higher content in the total flavonoids ($37{\pm}2.30mg/g$) and activities on DPPH free radical scavenging (73.23%) and ABTS free radical scavenging (83.47%) when compared with water extracted of RVS. The quality characteristics, such as pH, titratable acidity, and the number of lactic acid producing bacteria were not remarkably different between the fermented milk samples subjected to treatments with and without the addition of RVS extracts.

  • This report documents a case in which the administration of an herbal product, an extract of the lacquer tree, Rhus verniciflua Stokes, as sequential and concurrent treatment with chemotherapy was associated with a long term survival and good quality of life in a patient with metastatic non-small cell lung cancer(NSCLC). A 51-year-old Korean female was referred to the $M{\cdot}{\mu}$ Integrative Cancer Center, East-West Neo Medical centrer, Kyung Hee University for stage IV, metastatic NSCLC. She was treated with aRVS alone for 19 months and then received 1st line paclitaxel/carboplatin combined with aRVS, 2nd line gefitinib, and 3rd line pemetrexed. The number of cycles of pemetrexed administered was seventeen. aRVS was restarted as the 13th pemetrexed was administered. Pemetrexed with aRVS is currently ongoing. This patient has been alive for 41 months, and has been keeping a good performance status so far. We suggest aRVS as sequential and concurrent treatment with chemotherapy is an effective alternative treatment strategy.
