The Unique Mechanism of SNX9 BAR Domain for Inducing Membrane Tubulation

  • Sorting nexin 9 (SNX9) is a member of the sorting nexin family of proteins and plays a critical role in clathrinmediated endocytosis. It has a Bin-Amphiphysin-Rvs (BAR) domain which can form a crescent-shaped homodimer structure that induces deformation of the plasma membrane. While other BAR-domain containing proteins such as amphiphysin and endophilin have an amphiphatic helix in front of the BAR domain which plays a critical role in membrane penetration, SNX9 does not. Thus, whether and how SNX9 BAR domain could induce the deformation of the plasma membrane is not clear. The present study identified the internal putative amphiphatic stretch in the $1^{st}$ ${\alpha}$-helix of the SNX9 BAR domain and proved that together with the N-terminal helix ($H_0$) region, this internal putative amphiphatic stretch is critical for inducing membrane tubulation. Therefore, our study shows that SNX9 uses a unique mechanism to induce the tubulation of the plasma membrane which mediates proper membrane deformation during clathrinmediated endocytosis.

Selective Stimulating Effect of Flavonoids on the Antioxidant Defense System in Normal and Transformed Hepatic Cell Lines

  • Previously, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein, here named RCMF $({\underline{R}}VS\;{\underline{c}}hloroform-{\underline{m}}ethanol\;{\underline{f}}raction)$, was prepared from a crude acetone extract of Rhus verniciflua Stokes (RVS) which is traditionally used as a food additive and as an herbal medicine. In this study, we evaluated the effects of RCMF on the antioxidant defense system using embryonic normal hepatic cell line (BNL CL.2) and its SV40-mediated transformed cell line (BNL SV A.8). This study demonstrates that RCMF selectively stimulated the antioxidant defense system of normal cells, as BNL CL.2 cells proved to be more sensitive to RCMF-mediated increases of superoxide dismutase, catalase, glutathione, and glutathione reductase than BNL SV A.8 cells. In particular, RCMF caused a significant increase in the malonaldehyde content of BNL SV A.8 cells, which is believed to be closely associated with cytotoxicity of RCMF and RCMF-mediated growth inhibition. Collectively, our findings suggest that the flavonoid fraction, RCMF, selectively stimulates the antioxidant defense system in normal rather than hepatic tumor cells.

Cumulus Oocyte Complex Expansion Inhibiting Ingredient in Porcine Follicular Fluid (돼지 난포액내 난구세포 난자복합체 팽창 억제 성분)

  • The objective of this study was to identify a follicular fluid ingredient inhibiting the cumulus oocyte complex (COC) expansion. Thus, follicular fluid or liquid chromatographic fractions of follicular fluid was supplemented in COC culture medium. And COCs were incubated for 48 hours to investigate about cumulus expansion and also the first polar body extrusion. The results obtained were as follows; 1. The fluid of medium follicle significantly inhibited the COC expansion. 2. The fluid of large follicle inhibited the COC expansion. 3. Follicular fluid showed six major fractions at retention volumes (RVs) 1.83, 1.91, 2.15, 2.34, 2.53 and 2.74 ml after separation with Superose 12 column. Of the major fractions, fractions RV2.15, RV2.34, RV2.53 and RV2.74 inhibited both COC expansion and polar body extrusion. Especially, fractions of RV2.15 and RV2.53 significantly inhibited COC expansion, oocyte denudation and polar body extrusion. In conclusion, porcine follicular fluid contained a COC expansion inhibiting ingredient (CEI) that may be contained largely in fractions RV2.15 and RV2.53. And CEI may inhibit oocyte maturation by inhibition of oocyte denudation and extrusion of the first polar body.

Effect of Rhus verniciflua Stokes Extract and Fisetin on UVB-induced Apoptosis in NIH3T3 Cells (NIH3T3 세포에서 UVB에 의한 세포고사에 미치는 옻 추출물과 fisetin의 효과)

  • For a long time Rhus vemiciflua Stokes (RVS) has been traditionally used as a herbal plant in Asia. In this study, we have investigated the effect of acetone extract of Rhus verniciflua Stokes (RVSE) and fisetin, a component of RVSE, on DNA damage response in NIH3T3 cells. Exposure of cells to DVB light $(200 J/m^2)$ and postincubation in growth medium for 48 hr resulted in a decrease of cell viability to about $10-20\%$ of nontreated control. Addition of various concentrations of RVSE in the postincubation medium, however, significantly increased the cell viability as compared with the values expected. The genotoxicity-decreasing effect was also demonstrated in cells exposed to UVB light and incubated in medium containing fisetin. The genotoxicity-decreasing effect of RVSE and fisetin was further demonstrated by various analyses including cell morphology studies, trypan blue exclusion assay and DAPI staining. By Annexin V binding analysis, RVSE and fisetin were shown to decrease the early apoptosis induced by UVB exposure. These results suggest the RVSE contain components that either increase the DNA repair or decrease the apoptosis in UVB-exposed cells.

  • We investigate multivariate volatilities based on high frequency time series. The PCA (principal component analysis) method is employed to achieve a dimension reduction in multivariate volatility. Multivariate realized volatilities (RV) with various frequencies are calculated from high frequency data and "optimum" frequency is suggested using PCA. Specifically, RVs with various frequencies are compared with existing daily volatilities such as Cholesky, EWMA and BEKK after dimension reduction via PCA. An analysis of high frequency stock prices of KOSPI, Samsung Electronics and Hyundai motor company is illustrated.

The Effects of Lacca Sinica Exsiccata (LSE) on the Mast Cell-mediated Inflammatory Responses (건칠이 비만세포 매개 염증반응에 미치는 영향)

  • Objective : Lacca Sinic: Exsiccate (LSE) extracted from Rhus vemicitlus Stokes (RVS) has been used traditionally as a remedy for inflammation in Korea, China, and Japan. However, as yet there is no clear explanation of how LSE affects the production of inflammatory cytokines. This study was to determine the effects of LSE on the mast cell-mediated inflammatory responses. Method : We measured the amount of inflammatory cytokine production induced by the phorbol myristate acetate (PMA) plus calcium ionophore(A231S7) in the human mast cell line (HMC-l) incubated with various concentrations of Laces Sinica Exsiccate (LSE). The $TNF-\alpha$, IL-6 and IL-8 secreted protein levels were measured by the ELISA assay. The $TNF-\alpha$, IL-6 and IL-8 mRNA levels were measured by the RT-PCR analysis. Nuclear and cytoplasmic proteins were examined by Western blot analysis. The NF-${\kappa}B$ promoter activity was examined by a luciferase assay. Result : LSE inhibited the PMA + A231S7-induced $TNF-\alpha$, IL-6, and IL-8 expression and suppressed NF-${\kappa}B$ activation in the stimulated-HMC-1. In addition, LSE inhibited induction of NF-${\kappa}B$ promoter-mediated luciferase activity. Conclusion : In this study, we have found that LSE is an inhibitor of NF-${\kappa}B$ and cytokines on the mast cell-mediated inflammatory responses.

  • Epidural anesthesia has significantly advanced in neuraxial anesthesia and analgesia. It is used for surgical anaesthesia and treatment of chronic pain. Hearing loss during or after epidural anesthesia is rare, and it is known to occur by the change of the intracranial pressure. Cerebrospinal fluid is connected with perilymph in the cochlear and vestibule that is important to hearing and balance. If the intracranial pressure is abruptly transferred to the inner ear, perilymph can be leak, that called perilymphatic fistula, dizziness, and hearing loss can occur suddenly. We report a 65-year-old woman who presented with acute onset dizziness and hearing loss during the epidural nerve block for back pain, wherein we speculated a possibility of perilymphatic fistula as the mechanism of hearing loss and dizziness. The mechanism of dizziness and hearing loss was suspected with perilymphatic fistula.

Spectroscopic and Photometric Investigation of BS Cassiopeiae

  • New high-resolution spectra and multi-band photometric data of BS Cas were obtained at the Bohyunsan Optical Astronomy Observatory in 2018 and at the Jincheon Station of the Chungbuk National University Observatory in 2011, respectively. We measured the radial velocities (RVs) for both components, and the effective temperature of the more massive star was determined to be $6262{\pm}56K$. In addition, historical light curves showed strong time-dependant light variations at the total eclipse. These variations were modeled by a cool spot on the more massive component. Finally, the physical parameters of BS Cas by a simultaneous analysis of our RV curves with the photometric light curves were presented. Individual masses and radii of both components were deduced as $M_1= 0.59{\pm}0.07M_{\odot}$, $M_2=1.47{\pm}0.15M_{\odot}$, $R_1=0.94{\pm}0.03R_{\odot}$ and $R_2=1.47{\pm}0.05R_{\odot}$.

Pseudo Complex Correlation Coefficient: with Application to Correlated Information Sources for NOMA in 5G systems

  • In this paper, the authors propose the pseudo complex correlation coefficient (PCCC) of the two complex random variables (RV), because the four real correlation coefficients (RCC) of the corresponding four real RVs cannot be obtained only from the complex correlation coefficient (CCC) of given two complex RV. Such observation is motivated by the general statement; "The complex jointly-Gaussian random M-vector cannot be completely described by the complex covariance matrix, even though the real Gaussian random 2M-vector can be completely descried by the real covariance matrix. Therefore, in order to describe completely the complex jointly-Gaussian random M-vector, we need an additional matrix, namely the complex pseudo-covariance matrix, along with the complex covariance matrix." Then, we apply PCCC to correlated information sources (CIS) for non-orthogonal multiple access (NOMA) in 5G system, and investigate impact of the proposed PCCC on the achievable data rate of the stronger channel user in the conventional successive interference cancellation (SIC) NOMA with CIS. It is shown that for the given same CCC, the achievable data rates with the different PCCC are different, because the corresponding RCC are different. We also show that as the absolute value of the same CCC increases, the impact of the different PCCC becomes more significant.

A Case Report of Non-small Cell Lung Cancer with Brain Metastasis Patient Treated with Banhabaekchulchoenma-tang Gagambang (반하백출천마탕 가감방을 투여하여 비소세포성 폐암의 뇌전이에 의한 두통이 호전된 1례)

  • Lung cancer is the most common primary tumor of brain metastasis. Metastasis to the brain is a frequent complication of non-small cell lung cancer (NSCLC) occuring in 12 ~ 18% of patient. The incidence of brain metastasis is rising because of longer survival of cancer patients as a result of the increase in early diagnosis of primary tumors and aggressive management, and improvements in imaging quality and accessibility such as widespread use of MRI. The most common symptoms are gradual onset of headache, focal weakness, and mental changes. Surgery, chemotherapy, radiotherapy and steroid therapy are treatment of NSCLC with brain metastasis. We report a patient with NSCLC metastasis to the brain. This patient underwent chemotherapy, radiotherapy and steroidtherapy. However, the patient requested oriental medical treatment for the patient's continuous headache and disease. We administered Banhabaekchulchoenma-tang gagambang with Allergen-removed Rhus Verniciflua Stokes(aRVS). The patient showed remarkable improvement in terms of frequency and severity of headache. Further study will be needed in order to determine the long-term effectiveness of oriental medical treatment on cancer patient with headache.
