Active Immunization Study of Colon Cancer Derived 1-8D Peptide in HHD Mice

  • Background: 1-8D gene is a member of human 1-8 interferon inducible gene family and was shown to be overexpressed in fresh colon cancer tissues. Three peptides 1-6, 3-5 and 3-7 derived from human 1-8D gene were shown to have immunogenicity against colon cancer. Methods: To study tumor immunotherapy, of three peptides we established an active immunization model using HHD mice. $D^{b-/-}{\times}{\beta}2$ microglobulin $({\beta}2m)$ null mice transgenic for a chimeric HLA-$A2.1/D^{b-}\;{\beta}2m$ single chain (HHD mice) were challenged with B16/HHD/1-8D tumor cells and were immunized with irradiated peptide-loaded RMA- S/HHD/B7.1 transfectants. In therapy model tumor growth was retarded in HHD mice that were injected with 3-5 peptide-loaded RMA-S/HHD/B7.1. In survival test vaccination with 1-8D-derived peptide protects HHD mice from tumor progression after tumor challenge. Results: These studies show that peptide 3-5 derived from 1-8D gene can be the most effective candidate for the vaccine of immunotherapy against colon cancer and highlight 1-8D gene as putative colon carcinoma associated antigens. Conclusion: We demonstrated that RMA-S/HHD/ B7.1 loaded with 1-8D peptides, especially 3-5, immunization generates potent antitumor immunity against tumor cells in HHD mice and designed active immunization as proper immunotherapeutic protocols.

Stream Flow Analysis of Dry Stream on Flood Runoff in Islands (도서지역 건천의 홍수유출 시 흐름 해석)

  • In this study, compared with the result of water surface elevation and water velocity on the establishment of river maintenance basic plan and result of HEC-GeoRAS based GIS, and after use the result of water surface elevation and velocity were observed in the Han stream on Jeju island, analysis 2 dimensional stream flow. the lateral hydraulic characteristics and curved channel of the stream were analyzed by applying SMS-RMA2 a 2 dimensional model. The results of the analysis using HEC-RAS model and HEC-GeoRAS model indicated that the distribution ranges of water surface elevation and water velocity were similar, but the water surface elevation by section showed a difference of 0.7~2.18 EL.m and 0.63~1.16 EL.m respectively, and water velocity also showed differences of maximum 1.58m/sec and 2.67m/sec. SMS-RMA2 analysis was done with the sphere of Muifa the typhoon as a boundary condition, and as a result, water velocity distribution was found to be 1.19 through 3.91 m/sec, and the difference of lateral water velocity in No. 97 through 99 the curved channel of the stream was analyzed to be 1.59 through 2.36 m/sec. In conclusion it is anticipated that the flow analysis of 2 dimension model of stream can reflect the hydraulic characteristics of the stream curved channel or width and shape, and can be applied effectively in the establishment of river maintenance basic plan or management and designing of stream.

  • 홍수시 저수지 상류로부터 유입되는 다량의 오염물, 협잡물은 저수지의 경관 및 수질에 악영향을 미친다. 이러한 오염물질은 홍수시 일시에 유입되어 표층에 부유하므로 수거를 위한 시간 및 비용이 비교적 크게 발생된다. 따라서 이러한 부유쓰레기를 효과적으로 차단할 수 있는 시설물 설치는 필연적이다. 이렇게 부유쓰레기 수거를 위한 방법 중, 저수지 내부에 선 차단 시설을 설치하여 부유쓰레기를 홍수터로 유도하여 수거하는 방법은 일시에 많은 양의 부유쓰레기를 차단하게 되는데 유지관리, 경제적 측면에서 유리하다. 본 연구에서는 부유쓰레기 수거를 위한 선 차단시설 운영 방안을 검토하기 위하여 홍수시 저수지 내로 유입되는 유속흐름을 2차원 수치해석 모형인 RMA-2 모형으로 해석하고, 저수지의 내 외부 지형분석을 통하여 시범 지구의 선 차단시설 설치 위치를 선정하였고, 3차원 수치해석 모형인 FLOW-3D 모형으로 검증하였다. 선 차단시설 설치 시범지역 위치 선정을 위하여 저수지의 규모, 접근성, 시공 장비의 대여 편이성 등을 종합적으로 고려하여 기흥저수지를 시범예정 구역으로 선정하였다. 저수지 내로 유입되는 유속특성을 2차원 수치해석 모형인 RMA-2 모형으로 해석하여 시범 지구의 선 차단시설 설치 위치를 제안하고 홍수터 부지를 결정하였다. 부유쓰레기 유도를 위한 선차단시설은 저수지 급확대로 인한 유속 감소구간과 와류발생 구간이 적절한 것으로 분석되었으며 시범설치 지역인 기흥저수지 상류 급확대부에 선 차단시설을 제안하였다. 홍수터의 규모는 Armitage와 Rooseboom(2000)이 제안한 경험식을 바탕으로 $5,300m^2$을 제안하였으며, 기흥저수지 상류부 우안에 항아리 모양으로 설치하도록 계획하였다. 또한, 3차원 수치모형실험을 통하여 검토한 결과, 차단막 설치로 부유 쓰레기의 하류 이송은 차단되고, 원활하게 홍수터로 유도됨을 확인하였다.

Spillway Design by Using Hydraulic and Numerical Model Experiment - Case Study of HwaBuk Multipurpose Dam (수리 및 수치모형실험을 이용한 여수로 설계 - 화북다목적댐)

  • This study on the HwaBuk Multipurpose Dam showed that two- and three- dimensional numerical model experiments, as well as hydraulic model experiments, can be useful analysis tools for engineers. A commercially available RMA2, which solves the shallow water equations, and FLOW-3D, which solves the Reynolds averaged Navier-Stokes equations, were used to simulate the hydraulic model setup. Numerical simulation results on the following were compared with the hydraulic model results: the flow in the reservoir basin and the approaching channel; the discharge in the overflow weir; the water surface profiles in the rollway, chute, and stilling basin; and the pressure distributions in the rollway. It was shown that there is a reasonably good agreement between the numerical model and the hydraulic model for the most of computations. There were, however, some differences between the numerical simulation results and hydraulic model results for the hydraulic jump in the stilling basin because of air entrainment effect.

Two-Dimensional Flow Analysis of Approach Channel for the Design of Spillway Guidewall (여수로 유도벽 설계를 위한 접근수로의 2차원 흐름해석)

  • Numerical simulations were performed to analyse the flow pattern of the approach channel and to design the spillway guidewall for stable flow conditions. RMA-2, two dimensional finite element model which can easily represent complicated geometry was used, and model parameters were estimated from the observation data of hydraulic model test. Numerical experiments were made separately for the approach region and for the upstream region, and upstream boundary position of the hydraulic model beyond which the boundary effects are negligible was determined from the numerical results. For the stable flow condition in approach channel, alternative designs for guidewall were developed, and flow analysis for alternative designs was done through the numerical simulation. From the analysis of alternative design, we can see that the flow pattern in the approach channel is stable and the lateral stage difference disappears mostly before the spillway crest.

  • In this study, virtual confluence of $30^{\circ}$, $45^{\circ}$, $60^{\circ}$, $75^{\circ}$ type flowing into main channel was designed to comprehend occlusion phenomenon of the mouth of river, then performed numerical analysis using the RMA-2 and SED2D model of SMS according to application of 40m, 60m, 80m, 100m Training levee corresponding to main channel width ratio. Results of simulations analysis are summarized as follows. Applying appropriate Training levee length determined by numerical analysis results, Training levee dimension for occlusion reduction was studied through the analysis of velocity, water level, bed variation at the Gamchen confluence and Wichen confluence.

  • The objective of this study is to examine validation of Grid Reconstruction Method, which is developed to simulate drying/wetting in complex natural rivers with wetting and drying domain areas. To verify application of the developed model, the model was applied to natural rivers with wetting and drying domain areas such as Han river and Nakdong river. The simulation results have shown good agreements with observed data and the results for the developed model were more accurate and improved stability of numerical computation than those of RMA-2 model. If the analysis of contaminant advection-diffusion and sediment transport are performed with the study results, the results can be effectively applied to river flow analysis and ecological hydraulics.

Prediction of River Bed Change due to Yongdam Dam Discharge (용담댐 방류에 따른 하상변동 예측)

  • The purpose of this study is to identify the downstream influences due to the dam discharge by using 2-dimensional model, SMS(Surface water Modeling System). RMA-2 and SED-2D in SMS were applied to Yongdam multipurpose dam watershed located in Gum river basin. Through the simulation, erosion and deposit quantitative analysis of sinuous channels and scour pattern analysis of bridges have been done. A differences erosion depths between deposit are simulated as $-102.4 mm{\sim}54.2 mm$ at No.176(1.4 km) and $-104.1 mm{\sim}28.9 mm$ at No.146(7.4 km), sinuous channel. The river bed at Kamdong bridge in straight channal is simulated as uniform erosion. However, the river bed at Dumdul bridge in sinuous channal has been shown as different erosion depths at each sides. Consequently, the parts that could not be simulated on the existing 1-dimensional model, can be improved results by using a 2-dimensional model, about weakness points for hydraulic modeling such as extreme bend, tributary confluence.

Analysis of Behavior Characteristics of Instantaneous Input of Pollutant in River (하천에 순간 유입된 오염물질의 거동 특성 분석)

  • In case of continuous input of a pollutant, dispersion characteristics do not change much with changing dispersion coefficient, but that of an instantaneous input is very sensitive to the changes of dispersion coefficient. The characteristics of behavior of instantaneous input of a pollutant at the downstream of Han river were analyzed in this paper Field measurement of hydraulic and water quality factors at the downstream of Han river were conducted at low flow condition. The hydraulic factors were used to estimate the longitudinal dispersion coefficient, and the reasonable empirical equations for longitudinal dispersion coefficient at the downstream of Han river were suggested. The measured concentrations of BOD were closely matched with the calculated ones from RMA-4 model. In case of instantaneous input, range of dispersion, transport pathway and the traveltimes of the first and maximum concentration with variation of the longitudinal dispersion coefficients and water levels of downstream boundary were evaluated in this paper.

Flow Characteristics and Riverbed Changes Simulation for the Upstream and Downstream Sections of Gongju Bridge (공주대교 상.하류구간에 대한 흐름특성과 하상변동 모의)

  • The flow characteristics and the aspects of riverbed changes were analyzed for the upstream and downstream sections of Gongju Bridge. The upstream and downstream had complex topography, and the sections had the confluence of tributaries and several structures. In order to simulate the flow characteristics of the target sections, 1D HEC-RAS and 2D RMA2 were applied. As a result, the longitudinal water level of the target sections matched the results of simulated 1D and 2D samples. Also, 2D SED2D were applied to predict riverbed changes. As a result of the simulation, quantitative analysis was able to be performed for longitudinal riverbed changes from the sections of sudden change, bridges, the confluence of tributaries, and bends. Also, the distribution of riverbed changes on the main sections was in close relation to flow velocity. As a result of evaluating the sensitivity of SED2D, the concentration of suspended sediment, the thickness of sand beds, and the size of sand grains affected riverbed changes sensitively. These results will be used to apply the models of riverbed changes in the future.
