Adipose Tissue-Derived Mesenchymal Stromal Cells from Ex-Morbidly Obese Individuals Instruct Macrophages towards a M2-Like Profile In Vitro

  • Obesity, which continues to increase worldwide, was shown to irreversibly impair the differentiation potential and angiogenic properties of adipose tissue mesenchymal stromal cells (ADSCs). Because these cells are intended for regenerative medicine, especially for the treatment of inflammatory conditions, and the effects of obesity on the immunomodulatory properties of ADSCs are not yet clear, here we investigated how ADSCs isolated from former obese subjects (Ex-Ob) would influence macrophage differentiation and polarization, since these cells are the main instructors of inflammatory responses. Analysis of the subcutaneous adipose tissue (SAT) of overweight (OW) and Ex-Ob subjects showed the maintenance of approximately twice as many macrophages in Ex-Ob SAT, contained within the CD68+/FXIII-A- inflammatory pool. Despite it, in vitro, coculture experiments revealed that Ex-Ob ADSCs instructed monocyte differentiation into a M2-like profile, and under inflammatory conditions induced by LPS treatment, inhibited HLA-DR upregulation by resting M0 macrophages, originated a similar percentage of TNF-α+ cells, and inhibited IL-10 secretion, similar to OW-ADSCs and BMSCs, which were used for comparison, as these are the main alternative cell types available for therapeutic purposes. Our results showed that Ex-Ob ADSCs mirrored OW-ADSCs in macrophage education, favoring the M2 immunophenotype and a mixed (M1/M2) secretory response. These results have translational potential, since they provide evidence that ADSCs from both Ex-Ob and OW subjects can be used in regenerative medicine in eligible therapies. Further in vivo studies will be fundamental to validate these observations.

  • In this work the Multi-Domain model of an electric vehicle is developed. The electric domain model consists on the traction drive and allows including faults associated with stator winding. The thermal model is based on a spatial discretization. It receives the power dissipated in the electric domain, it interacts with the environment and provides the temperature distribution in the induction motor. The mechanical model is a half vehicle model. Given that all models are obtained using the same approach (Bond Graph) their integration becomes straightforward. This complete model allows simulating the whole system dynamics and the analysis of electrical/mechanical/thermal interaction. First, experimental results are aimed to validate the proposed model. Then, simulation results illustrate the interaction between the different domains and highlight the capability of including faults.

  • 본 논문에서는 선박 내 유선 네트워크에서 차등화 서비스를 지원하기 위해, 기존 RIO (RED In and Out) 방식보다 우수한 패킷 폐기 기술을 제안한다. 제안한 패킷 폐기기술은 개별 플로우별로 관리하지 않고 특정한 기준에 따라 몇 개의 플로우 그룹으로 나누어, 그룹별 관리를 통해서 동일 클래스 내 플로우 간 공평성을 제공한다. 시뮬레이션 결과에서는 RIO 방식보다 링크 이용률은 조금 감소하나, 다중 병목 구간을 경유하는 플로우들이 RIO 방식보다 더 많은 전송률을 보장 받음을 알 수 있다.

Numerical simulation by the finite element method of the constructive steps of a precast prestressed segmental bridge

  • The design of segmental bridges, a structure that typically employs precast prestressed concrete elements and the balanced cantilever construction method for the deck, may demand a highly complex structural analysis for increased precision of the results. This work presents a comprehensive numerical analysis of a 3D finite element model using the software ANSYS, version 21.2, to simulate the constructive deck stages of the New Guaiba Bridge, a structure located in Porto Alegre city, southern Brazil. The materials concrete and steel were considered viscoelastic. The concrete used a Generalized Kelvin model, with subroutines written in FORTRAN and added to the main model through the customization tool UPF (User Programmable Features). The steel prestressing tendons used a Generalized Maxwell model available in ANSYS. The balanced cantilever constructive steps of a span of the New Guaiba Bridge were then numerically simulated to follow the actual constructive sequence of the bridge. A comparison between the results obtained with the numerical model and the actual vertical displacement data monitored during the bridge's construction was carried out, showing a good correlation.

  • In this paper, we explore two ways to improve the bandwidth assurance performance of Assured Service(AS). It is well known that AS fails to meet the bandwidth assurance target for large-profile TCP flows competing with many small-profile flows. This flows after the back-offs induced by packet drops. Thus currently proposed solution to this problem naturally focus on modifying TCP's behavior to counter the unfairness in the TCP dynamics. Unfortunately, these proposals lack practicability in terms of the required changes in the incumbent Internet infrastructure. Admitting this difficulty, we instead look to not yet deployed Diff-Serv mechanisms for practical solutions. In particular, we investigate the role of RIO, RED with IN(in-profile) and OUT(out-profile), queue management scheme in the assurance failure for As. Specifically, we identify the inadequacy of RIO that aggravates the bandwidth assurance failure. Then we alleviates the bandwidth assurance failure problem by separately controlling the out-of-profile packet queue length. Through extensive simulations we demonstrate that RI+O extends the regime where AS consistently provides the bandwidth assurance.

  • Gaming is an old humans' habit. Games help in logical development and encourage learning of theoretical and practical concepts. Besides they offer entertainment and challenge. The advent of the personal computer changed this tradition. Every year new challenges arise in a digital format, which lead the young and adults to spend hours in front of a computer or TV screen in an attempt to overcome hurdles and reach an objective. Quality, sophistication, and constant innovation are attained through complex computer software that almost has an obligation to improve as each new title is released, due to this game development becomes a challenge. Considering that a game title is software and thus faces the same restrictions of business applications, this article intends to analyze, under the optics of reuse, if game development resorts to reuse, and where and how this happens.

  • This paper presents a IEEE1588 based clock synchronization technique for a sRIO (Serial RapidIO) network which is applied to a submarine system. Clock synchronization plays a key role in the success of a networked embedded system. Recently, the IEEE1588 algorithm making use of dedicated chipset has been widely used for the synchronization of various industrial applications. However, there is no chipset available for the sRIO network that can offer many advantages, such as low latency and jitter. In this paper, the IEEE1588 algorithm for a sRIO network is implemented using only software without any dedicated chipset. The proposed approach is verified with experimental setup.

  • Steel beam-to-column joints are often subjected to a combination of bending and axial forces. The level of axial forces in the joint may be significant, typical of pitched-roof portal frames, sway frames or frames with incomplete floors. Current specifications for steel joints do not take into account the presence of axial forces (tension and/or compression) in the joints. A single empirical limitation of 10% of the beam's plastic axial capacity is the only enforced provision in Annex J of Eurocode 3. The objective of the present paper is to describe some experimental and numerical work carried out at the University of Coimbra to try to extend the philosophy of the component method to deal with the combined action bending moment and axial force.

Effect of the Size and Carbides Dispersion in the Sintering and Hardness of Samples of Stainless Steel Reinforced with NbC And TaC

  • The present study investigates the behavior of the sintering and hardness of stainless steel samples reinforced with NbC and TaC. Matrixes of pure stainless steel were compacted with addition of up to 3% wt NbC or TaC in a cylindrical die of steel $(\phi\;=\;5,0\;mm)$ at 700 MPa and sintered in an electrical resistance furnace under argon atmosphere. The sintered samples were characterized by density and hardness measurement, optical microscopy and scanning electron microscopy (SEM). The preliminary results show that the size and distribution of carbides influence in the sintering and hardness of the sintered samples.
