The Comparisons between RIA and CIA in CA19-9 Monitoring for Diagnosis of Pancreaticobiliary Tumors

  • The CA(carbohydrate antigen)19-9 is complex protein that can be used as an important marker which aids the clinical diagnosis and prognosis of various pancreaticobiliary tumors. However, it was also reported that there were some CA19-9 positive patients with benign disease as using RIA method. The purpose of this study is to evaluate the clinical usefulness of serum level of CA19-9 with RIA(radioimmuno assay), CIA(chemiluminescence immuno assay), and conventional liver function tests. The correlation between CIA and RIA in CA19-9 of pancreatobiliary disease was 0.9833(P<0.01). Also, the correlations between CIA and RIA in CA19-9 of benign and malignant pancreaticobiliary tumor patients was 0.8714(P<0.01) and 0.9727(P<0.01) respectively. The correlation between CA19-9 and ALP was 0.5140(P<0.01) and CEA was 0.3385(P<0.05) as using CIA. The measurement of serum CA19-9 levels by CIA method may be useful in differentiating patients with malignant disease from those with benign disease in pancreaticobiliary tumors.

잉어중 Endosulfan의 잔류분석(殘留分析)을 위한 Radioimmunoassay(RIA)의 개발(開發) (Development of Radioimmunoassay(RIA) for Residue Analysis with Endosulfan in Water and Carp(Cyprinus carpio L.))

  • Endosulfan의 잔류분석(殘留分析)을 위한 RIA(radioimmuno assay)는 endosulfan alcohol(EA)-BSA conjugate를 항원(抗原)으로 하여 집토끼에서 면역항체(免疫抗體)를 생산(生産)하였다. 생산(生産)된 면역항체(免疫抗體)의 역가(力價)는 1 : 32,000 으로 endosulfan과 EA 이외(以外)의 유사화합물(類似化合物)에서는 거의 반응성(反應性)을 나타내지 않았으며 RIA를 위한 최적(最適) 희석배수(稀釋倍數)는 1 : 2,000 으로 나타났다. RIA의 최적(最適) 배양온도(培養溫度)는 $4\;^{\circ}C$ 이었으며 endosulfan의 검출범위(檢出範圍)는 $1\;ng-20\;{\mu}g$ 이었고 최소검출량(最小檢出量)은 1 ng 이었다. RIA 기법(技法)을 이용(利用)하여 잉어의 각(各) 조직(組織)과 공시수(供試水)에서 실시한 회수율(回收率) 시험(試驗) 결과(結果)는 GLC/ECD나 ELISA를 이용(利用)한 회수율(回收率)보다 우수하게 나타났다. 시료(試料)에서 RIA에 의한 endosulfan의 검출한계(檢出限界)는 0.1 ppb 였다.

  • Fluoride is one of the most potent stimulators of bone formation in vivo. But its direct effects on osteoblast is not yet clear This study was to investigate the effects of Sodium fluoride on alkaline phosphatase(ALP) activity, cAMP formation responsive to parathormone(PTH) and type I $\alpha$ 2 collagen ribonucleic acid (mRNA) level in Murin osteoblast-like (MC3T3-E1) cells. The cells were cultured in $\alpha-Minimal$ essential medium $(\alpha-MEM)$ supplemente with $10\%$ fetal bovine serum (FBS) and then changed to $0.1\%$ FBS with various concentration of Sodium fluoride. The ALP activity was assayed by the method of Lowry with disodium phenyl phosphated as substrate. cAMP formation was measured by Radioimmuno Assay(RIA). Type I $\alpha$ 2 collagen ribonucleic acid(mRNA) expression was studied by Nothern blot analysis. The results were as follows: 1. cAMP level was increased by PTH in MC3T3-E1 cells. 2. Sodium fluoride showed the tendency of inhibitory effects on cAMP responsiveness to PTH in MC3T3-E1 cells. 3. Sodium fluoride increased ALP activity at cocentration of $2{\mu}M,\;4{\mu}M,\;and\;10{\mu}M$ significantly different from control at the 0.001 level. ALP activity revealed maximum value at $10{\mu}M$ in this study. 4. Nothern blot analysis of Sodium fluoride treated cells, using Type I $\alpha$ 2 collagen prove, revealed significant increase at $10{\mu}M$ in MC3T3-E1 cells.

폐결핵의 진행정도 및 치료에 따른 혈청내 Tumor Necrosis Factor-Alpha, Gamma-Interferon 및 Soluble-Intercellular Adhesion Molecule-1의 변화 (The Changes of Serum Level of Tumor Necrosis Factor-Alpha, Gamma-Interferon and Soluble-Intercellular Adhesion Molecule-1 Relating to the Progression and Treatment of Patients with Pulmonary Tuberculosis)

  • 연구배경 : 결핵은 우리나라에서 아직도 흔하게 발생하는 질환으로 세포면역반응에 의해 육아종이 형성되며, 이 과정에서 대식세포가 분비하는 tumor necrosis factor-alpha (TNF-$\alpha$), Th1 세포가 생성하는 gamma-Interferon (INF-$\gamma$), 내피 세포가 표현하는 intercelluar adhesion molecule-1 (ICAM-1)이 중요한 역할을 할 것이라고 생각되었다. 방 법 : 저자들은 결핵의 경중(輕重)과 이러한 물질들이 관련이 있는가를 알아보기 위하여, 환자의 혈액을 채취하여 TNF-$\alpha$, INF-$\gamma$를 radioimmuno assay(RIA)로, ICAM-1이 혈액으로 유리된 형태인 sICAM-1을 enzyme linked immunosolvent assay(ELISA)로 각각 측정하였다. 또한 화학 요법에 따른 변화를 알기 위해 치료 시작후 6개월 시점에서 다시 추적검사를 실시하였다. 결 과 : TNF-$\alpha$, INF-$\gamma$, sICAM-1은 중등증과 중증의 결핵에서는 의미 있게 증가하였고, 경증에서는 의의가 없었다. 6개월간의 항결핵 치료후, sICAM-1은 임상경과에 동반하여 의미있는 감소를 보였지만. TNF-$\alpha$, INF-$\gamma$에서는 감소는 있었지만 의의는 없었다. 결 론 : 본 실험 결과 결핵의 세포 면역 매개과정에는 TNF-$\alpha$와 INF-$\gamma$가 매우 중요한 역할을 하며, 그 반응 정도가 질병의 병기에 따라 심할수록 많이 증가함을 관찰하였다. ICAM-1은 TNF-$\alpha$와 INF-$\gamma$ 농도와 비례하여 sICAM-1이 증가하였고, 질병의 병기에 따라 농도의 차이가 있을뿐 아니라, 치료경과에 비례하여 농도변화를 보여, 질병의 할동성을 나타내는 것 외에 치료 경과를 나타내는 지시자로서의 가능이 있을 것으로 생각된다.

임신 중 모체와 신생아 제대 혈청의 엽산과 비타민 $B_{12}$ 농도 변화 (A Change of Serum Folate and Vitamin $B_{12}$ Concentrations of Maternal and Umbilical Cord Blood during Pregnancy)

  • Folate and Vitamin $B_{12}$ are essential nutrients important during pregnancy. This study was conducted to evaluate the folate and vitamin $B_{12}$ nutritional status of Korean pregnant women and to investigate the relationship between maternal­umbilical cord serum folate and vitamin B12 levels and pregnancy outcomes. Dietary intakes of the pregnant women were estimated by 24 hour-recall (3 times). Serum folate and vitamin B12 levels in maternal blood and umbilical cord of 27 pregnant women at 1'st-, 2'nd-, 3'rd-trimester and delivery were measured by RIA (radioimmuno assay), respectively. Means of folate and vitamin $B_{12}$ intake were $283.53\pm58.01{\mu}g/day\;and\; 2.99\pm1.32mg/day$, respectively. Maternal mean serum folate levels of the trimester and delivery were $9.75\pm3.60ng/ml,\;10.46\pm4.63ng/ml,\;10.71\pm4.14ng/ml\;and\;15.05\pm7.04ng/ml$. Those maternal levels were significantly lower than that of umbilical cord blood $(23.99\pm9.42ng/ml)$. Serum vitamin $B_{12}$ levels of maternal trimester and delivery were $479.07\pm137.56 pg/ml,\;310.96\pm137.56pg/ml,\;308.22\pm74.65pg/ml,\;and\;295.67\pm93.36pg/ml$, which were significantly lower than those of umbilical cord blood $(500.13\pm185.60ng/ml)$. This finding indicates that the uptake of folate and vitamin $B_{12}$ in the fetus may be due to an active placental transport mechanism. Maternal serum level correlated positively with those of umbilical cord blood, showing that folate and vitamin $B_{12}$ concentration of umbilical cord blood might be affected by maternal status. There was no significant correlation between the serum folate levels in maternal-umbilical cord blood and the pregnancy outcomes. However, maternal vitamin $B_{12}$ level at l'st trimester was significant positive correlation between the gestational age except for birth weight and weight gain.
