인삼 사포닌에서 Ginsenoside-$Rg_2$와 -$Rg_3$의 이성질체인 20(R&S) Prosapogenin들의 역상 고속 액체 크로마토그래피에 의한 분리 (Separation of 20(R&S) Prosapogenin Isomers of Ginsenoside-$Rg_2$ and -$Rg_3$ from Ginseng Saponins by Reversed-Phase High Performance Liquid Chromatography)

  • Using a reversed-phase high performance liquid chromatography, the separation of 20(S)-, 20(R)-prosapogenin stereo-isomers of ginsenoside-$Rg_2$ and of ginsenoside-$Rg_3$ in ginseng saponins has been carried out with binary solvent system. The optimum conditions for the isomer separation are as following: Nova-$Pak^{(R)}C_{18}$ (Waters, $3.9{\times}150mm$) column, $CH_3CN/CH_3CN$ (100:8, v/v) binary solvent system and the flow rate was 1.7 mL/min. The stereoisomers were separated with change of the mixture ratio of the solvent system, the solvent elution by gradient program, and then detected at 203 nm of UV detector. The simultaneous separation of mixture that were the $Rg_2$, $Rg_3$ isomers was easily performed in nonpolar solvent for $Rg_2$, polar solvent for $Rg_3$ at the same optimum conditions.

Ginsenoside Rg3이 흰쥐 척수압박손상의 초기 염증반응에 미치는 영향 (Effects of Ginsenoside Rg3 on Early-stage Inflammatory Response in Spinal Cord Compression of Rodents)

  • Objectives : In present study, we investigated the effects of ginsenoside Rg3 on early-stage inflammatory response in spinal cord compression of rodents. Methods : Spinal cord injury(SCI) was induced by a vascular clip method(30 g, 5 min) on the spinal cord of mice. Rg3 was treated orally at 1 hour prior to the SCI induction. Messenger ribonucleic acid(mRNA) expression of tumor necrosis factor-${\alpha}$(TNF-${\alpha}$), interleukin-1${\beta}$(IL-1${\beta}$), interleukin-6(IL-6) and cyclooxygenase-2(COX-2) was measured by the real-time polymerase chain reaction(RT-PCR). Microglia in the spinal cord tissue, neurophils and COX-2 in the peri-lesion and inducible nitric oxide synthase(iNOS) expression in the ventral horn of SCI induced rats were measured by immunohistochemical stain. Results : 1. Rg3 significantly reduced the mRNA expression of TNF-${\alpha}$, IL-1${\beta}$, and COX-2 in the spinal cord tissue compared with SCI group(p<0.05, p<0.01). 2. Rg3 significantly reduced the total number of activated microglia and proportion of phagocytic form in the total activated microglia compared with SCI group(p<0.05, p<0.01). 3. Rg3 significantly reduced myeloperoxidase(MPO) positive neurophil in the peri-lesion compared with SCI group(p<0.05). 4. Rg3 reduced the COX-2 expression in the tissue and motor neurons compared with SCI group. 5. Rg3 significantly reduced iNOS positive motor neurons in the ventral horn compared with SCI group(p<0.01). Conclusions : In conclusion, we demonstrated at first that treatment of ginsenoside Rg3 could reduce significantly the levels of inflammatory mediators in a spinal cord compression model of rodents. Therefore, these results suggested that ginsenoside Rg3 may be a useful antimiflamatory therapeutic candidate for SCI.

Preparation and evaluation of proliposomes formulation for enhancing the oral bioavailability of ginsenosides

  • Background: This research main objective was to evaluate a proliposomes (PLs) formulation for the enhancement of oral bioavailability of ginsenosides, using ginsenoside Rg3 (Rg3) as a marker. Methods: A novel PLs formulation was prepared using a modified evaporation-on-matrix method. Soy phosphatidylcholine, Rg3-enriched extract, poloxamer 188 (Lutrol® F 68) and sorbitol were mixed and dissolved using a aqueous ethanolic solution, followed by the removal of ethanol and lyophilization. The characterization of Rg3-PLs formulations was performed by powder X-ray diffractometry (PXRD), transmission electron microscopy (TEM) and in vitro release. The enhancement of oral bioavailability was investigated and analyzed by noncompartmental parameters after oral administration of the formulations. Results: PXRD of Rg3-PLs indicated that Rg3 was transformed from crystalline into its amorphous form during the preparation process. The Rg3-encapsulated liposomes with vesicular-shaped morphology were generated after the reconstitution by gentle hand-shaking in water; they had a mean diameter of approximately 350 nm, a negative zeta potential (- 28.6 mV) and a high entrapment efficiency (97.3%). The results of the in vitro release study exhibited that significantly more amount of Rg3 was released from the PLs formulation in comparison with that from the suspension of Rg3-enriched extract (control group). The pharmacokinetic parameters after oral administration of PLs formulation in rats showed an approximately 11.8-fold increase in the bioavailability of Rg3, compared to that of the control group. Conclusion: The developed PLs formulation could be a favorable delivery system to improve the oral bioavailability of ginsenosides, including Rg3.

Immunogenic Cell Death Induced by Ginsenoside Rg3: Significance in Dendritic Cell-based Anti-tumor Immunotherapy

  • Cancer is one of the leading causes of morbidity and mortality worldwide; therefore there is a need to discover new therapeutic modules with improved efficacy and safety. Immune-(cell) therapy is a promising therapeutic strategy for the treatment of intractable cancers. The effectiveness of certain chemotherapeutics in inducing immunogenic tumor cell death thus promoting cancer eradication has been reported. Ginsenoside Rg3 is a ginseng saponin that has antitumor and immunomodulatory activity. In this study, we treated tumor cells with Rg3 to verify the significance of inducing immunogenic tumor cell death in antitumor therapy, especially in DC-based immunotherapy. Rg3 killed the both immunogenic (B16F10 melanoma cells) and non-immunogenic (LLC: Lewis Lung Carcinoma cells) tumor cells by inducing apoptosis. Surface expression of immunogenic death markers including calreticulin and heat shock proteins and the transcription of relevant genes were increased in the Rg3-dying tumor. Increased calreticulin expression was directly related to the uptake of dying tumor cells by dendritic cells (DCs): the proportion of CRT+CD11c+cells was increased in the Rg3-treated group. Interestingly, tumor cells dying by immunogenic cell death secreted IFN-γ, an effector molecule for antitumor activity in T cells. Along with the Rg3-induced suppression of pro-angiogenic (TNF-α) and immunosuppressive cytokine (TGF-β) secretion, IFN-γ production from the Rg3-treated tumor cells may also indicate Rg3 as an effective anticancer immunotherapeutic strategy. The data clearly suggests that Rg3-induced immunogenic tumor cell death due its cytotoxic effect and its ability to induce DC function. This indicates that Rg3 may be an effective immunotherapeutic strategy.

Biosurfactant Production by Marine Actinomycetes Isolates Streptomyces althioticus RG3 and Streptomyces californicus RG8 as Promising Sources of Antimicrobial and Antifouling Effects

  • Two marine actinobacterial isolates, RG3 and RG8, were identified using 16Sr DNA as Streptomyces althioticus RG3 and Streptomyces californicus RG8 and submitted to the database of genetic information with accession numbers MW661230 and MW661234, respectively; they were found to have emulsification indexes of 60 ± 2.5% and 53 ± 2.2%, respectively. The biosurfactants obtained were stable at a temperature of 35℃ for both strains; they were stable at 10% NaCl, in the case of S. althioticus RG3 and at 10-15% NaCl in the case of Str.californicus RG8; both strains produced the most biosurfactant when exposed to alkaline conditions. We characterized the biosurfactants, including features such as their chemical composition, using Fourier transform infrared spectroscopy analysis. The antimicrobial activity of the biosurfactant extracts was evaluated using the well diffusion method against Vibrio alginolyticus MK170250, Escherichia coli ATCC 8739, Pseudomonas aeruginosa ATCC 4027, and Staphylococcus aureus ATCC 25923. S. althioticus RG3 biosurfactants were found to have better antimicrobial activity than those of Str. californicus RG8, indicating that they may be used in pharmaceutical industries and in the manufacture of antifouling products.

Anti-Metastasis Effects of Ginsenoside Rg3 in B16F10 Cells

  • Ginsenoside Rg3 is a bioactive ginseng constituent that has been reported to have diverse pathological and physiological effects, including anti-inflammatory and anti-metastatic activities. Metastasis is one of the most important factors involved in patients with melanoma. However, the molecular mechanism underlying the anti-metastatic activities of Rg3 in malignant melanoma cancer has not been fully elucidated. In this study, we have evaluated that Rg3 effectively inhibits metastasis of B16F10 melanoma cancer cells. We found that Rg3 significantly suppresses the migration, invasion, wound healing, and colony-forming abilities of B16F10 cells in a dose-dependent manner. Mechanistically, we demonstrate that Rg3 suppresses B16F10 cell metastasis by inhibiting MMP-13 expression. These results indicate that Rg3 suppresses the metastasis of B16F10 mouse melanoma cancer cells via MMP-13 regulation. Importantly, MMP-13 downregulation may influence the migration and invasion capabilities of melanoma cells and has been correlated with melanoma progression. Therefore, Rg3 is a potential therapeutic candidate that could be used to treat patients with metastatic melanoma.

고려인삼으로 제조된 홍삼중의 화학적 암 예방성분 (Cancer Chemopreventive Compounds of Red Ginseng Produced from panax ginseng C.A. Meyer)

  • 발암물질을 투여하여 발생하는 마우스 폐선종은 홍삼추출믈의 투여에 의하여 그 발생율이 억제되나 수삼을 투여하면 발생율이 억제되지 않는다. 또한 암환자-대조군연구 결과에 있어서도 수삼즙 또는 수삼절편을 복용한 사람에서는 암의 위험비가 감소되지 않으나 수삼열탕 또는 홍삼을 복용하면 현저한 위험비의 감소를 볼 수 있었다. 이와 같은 결과는 열로 처리된 홍삼중에 암예방 유효성분이 있을 것이라고 추정되어 왔다. 저자들은 4종의 홍삼중의 진세노사이드 즉 Rh$_1$, Rh$_2$, Rg$_3$ 및 Rg$_{5}$ 를 고려홍삼으로부터 분리합성하여 윤의 9주 중기 항발암실험법에 의하여 항발암성을 관찰한바 진세노사이드 Rg$_3$와 Rg$_{5}$의 투여시에는 통계학적으루 유의한 폐선종 발생율이 감소되었으나 Rh$_2$에서는 폐선종발생율이 약간 감소되는 경향을 보였고 Rh$_1$에서는 전혀 영향을 주지 않았다. 이와 같은 소견으로 홍삼에 의한 항발암작용 또는 암예방작용은 홍삼중의 진세노사이드 Rg$_3$및 Rg$_{5}$가 유효성분임을 파악하였으며 이들 진세노사이드 Rg$_3$ Rg$_{5}$ 및 Rh$_2$가 단독 또는 복합적으로 작용할 것으로 추정된다.

An Architecture for Home-Oriented IPTV Service Platform on Residential Gateway

  • In order for end-users in home networks to receive opportunities for useful services that go beyond legacy Internet Protocol TV (IPTV) services, this paper proposes a service platform that resides on the residential gateway (RG) for interworking between the home network and IPTV. This proposed service platform is called the home-oriented IPTV service platform (HISP) on the RG (HISP-RG). The proposed HISP-RG provides open architecture and functionalities to enable 3rd party IPTV service providers to locally and directly deliver home-oriented IPTV services to end-users in home networks. The HISP-RG can be an "add-on" and not a "built-in" solution for the existing standard RG. This paper introduces several home-oriented IPTV services that can be executed and delivered locally through the HISP-RG. Then, the open architecture and functionalities of the HISP-RG are defined and their requirements are specified. Finally, use cases of the HISP-RG for home-oriented IPTV services are presented.

흑삼의 신생혈관 억제활성에 대한 연구 (Study on Antiangiogenic Effect of Black Ginseng Radix)

  • Objectives : This study was performed to investigate the influence of black ginseng radix extracts (BG) and ginsenoside Rg3, Rg5 on basic fibroblast growth factor (bFGF) induced proliferation, migration and capillary tubule-like formation of human umbilical vein endothelial cells (HUVECs). Methods : HUVECs were cultured with BG and ginsenoside Rg3, Rg5 at different concentrations (60, 125, 250, 500, $1,000{\mu}g/m\ell$) for 2 day In the presence of bFGF, respectively. XTT was used to detect the proliferation. Migration and tube formations were examined to detect the antiangiogenesis. Also, the chick embryo chorioallantoic membrane (CAM) assay was performed to detect the antiangiogenesis. Results : BG and ginsenoside Rg3, Rg5 significantly inhibited bFGF-induced endothelial cell proliferation and migration in a dose-dependent manner. Tube formation in bFGF-induced HUVECs were suppressed by BG and ginsenoside Rg3, Rg5. Moreover, BG and ginsenoside Rg3, Rg5 (30-$50{\mu}g$/egg) inhibited new blood vessel formation on the growing CAM. Conclusions:Based on the present results, it can be suggested that BG has a potential chemopreventive agent via antiangiogenesis.

Memory Enhancing and Neuroprotective Effects of Selected Ginsenosides

  • The effects of ginsenosides Rg$_3$(R) , Rg$_3$(S) and Rg$_5$/Rk$_1$ (a mixture of Rg$_5$ and Rk$_1$ 1:1, w/w), which are components isolated from processed Panax ginseng C.A. Meyer (Araliaceae), on memory dysfunction were examined in mice using a passive avoidance test. The ginsenosides Rg3(R), Rg3(S) or Rg$_5$/Rk$_1$, when orally administered for 4 days, significantly ameliorated the memory impairment induced by the single oral administration of ethanol. The memory impairment induced by the intraperitoneal injection of scopolamine was also significantly recovered by ginsenosides Rg3(S) and Rg$_5$/Rk$_1$. Among the three ginsenosides tested in this study, Rg$_5$/Rk$_1$ enhanced the memory function of mice most effectively in both the ethanol­and scopolamine-induced amnesia models. Moreover, the latency period of the Rg$_5$/Rk$_1$­treated mice was 1.2 times longer than that of the control (no amnesia) group in both models, implying that Rg$_5$/Rk$_1$ may also exert beneficial effects in the normal brain. We also evaluated the effects of these ginsenosides on the excitotoxic and oxidative stress-induced neuronal cell damage in primary cultured rat cortical cells. The excitotoxicity induced by glutamate or N­methyl-D-aspartate (NMDA) was dramatically inhibited by the three ginsenosides. Rg$_3$(S) and Rg$_5$/Rk$_1$ exhibited a more potent inhibition of excitotoxicity than did Rg$_3$(R). In contrast, these ginsenosides were all ineffective against the H$_2$O$_2$- or xanthine/xanthine oxidase-induced oxidative neuronal damage. Taken together, these results indicate that ginsenosides Rg$_3$(S) and Rg$_5$/Rk$_1$ significantly reversed the memory dysfunction induced by ethanol or scopolamine, and their neuroprotective actions against excitotoxicity may be attributed to their memory enhancing effects.
