Marginal Zinc Deficiency Affects Biochemical and Physiological Parameters in Beef Heifer Calves

  • A study detennined whether certain biochemical and physiological variables were altered during marginal Zn deficiency. Ten weaned crossbred Hereford Angus heifer calves, weighing $163{\pm}2kg$, were utilized. Five calves were fed a Zn - deficient (- Zn) brome-alfalfa hay diet containing 17 mg Zn/kg diet DM, and five calves were fed a Zn-adequate (+Zn) diet with 23 mg Zn/kg diet DM from $ZnSO_4$ added to the - Zn diet (total diet, 40 mg Zn/kg diet DM), for 32 d. At 21 d the - Zn calves had a reduction (p < .05) in feed efficiency. By 25 d, plasma Zn and alkaline phosphatase concentrations were reduced (p < .05) in the - Zn calves. Blood urea nitrogen, glucose, insulin, IGF-I, Cu plasma concentration and Zn and Cu concentrations of red blood cell (RBC) and liver were not altered (p > .05) by the - Zn diet through 25 d. In response to a single i. m. injection of dexamethasone (20 mg) on d 25, calves fed the two dietary Zn amounts showed no changes (p > .05) in plasma or RBC Zn and Cu concentrations, serum IGF-I, insulin, and glucose when measured at 6, 12, 24, 48, 72, and 96 h after injection. In response to an intradermal injection of phytohemagglutinin on d 30, cell mediated immune (CMI) response was reduced (p < .05) in the - Zn calves. These observations indicate that during a marginal Zn deficiency in calves, there was a decrease in feed efficiency, plasma Zn, serum alkaline phosphatase, and CMI response.

  • The level and activity of critical regulatory proteins in cells are tightly controlled by several tiers of post-translational modifications. HIF-1α is maintained at low levels under normoxia conditions by the collaboration between PHD proteins and the VHL-containing E3 ubiquitin ligase complex. We recently identified a new physiologically relevant mechanism that regulates HIF-1α stability in the nucleus in response to cellular oxygen levels. This mechanism is based on the collaboration between the SET7/9 methyltransferase and the LSD1 demethylase. SET7/9 adds a methyl group to HIF-1α, which triggers degradation of the protein by the ubiquitin-proteasome system, whereas LSD1 removes the methyl group, leading to stabilization of HIF-1α under hypoxia conditions. In cells from knock-in mice with a mutation preventing HIF-1α methylation (Hif1αKA/KA), HIF-1α levels were increased in both normoxic and hypoxic conditions. Hif1αKA/KA knock-in mice displayed increased hematological parameters, such as red blood cell count and hemoglobin concentration. They also displayed pathological phenotypes; retinal and tumor-associated angiogenesis as well as tumor growth were increased in Hif1αKA/KA knock-in mice. Certain human cancer cells exhibit mutations that cause defects in HIF-1α methylation. In summary, this newly identified methylation-based regulation of HIF-1α stability constitutes another layer of regulation that is independent of previously identified mechanisms.

Is It Necessary to Use Dextran in Free Flap Surgery? (유리피판술에서 덱스트란의 사용이 필요한가?)

  • Purpose: Low - molecular - weight dextran is one of the most frequently used antithrombotic agents in microvascular surgery, but there is controversy if it has the real benefit in the clinical aspects. The purpose of this study was to evaluate the effect associated with postoperative use of low - molecular - weight dextran in breast reconstruction by free TRAM flap patients. Methods: From January 2002 to October 2008, we reconstructed 88 cases of postmastectomy deformity using the free TRAM flap. The 88 cases were divided into two groups : a group with no use of dextran(66 patients, control group, Group A) and a postoperative low - molecular weight dextran loaded group(22 patients, Group B). We assessed number of flap survival, rate of complication like hematoma or seroma, total amount of drainage from operative wound, duration of drainage, and amount of transfusion in each group. Results: There was no total flap loss and every flap was survived. Total amount of drainage for post - operative 5 days were 857 ml in group A and 1101 ml in group B. Drain was kept for average of 7.3 days in group A and 8.7 days in group B. Packed red cell transfusions were made in average of 3.3 units for group A and 3.0 units for group B. Group B showed significantly higher values in former 2 comparative parameters than group A. Conclusion: There was no definitive advantage of anticoagulants in elective free - flap surgery in terms of success rate. However, groups with using anticoagulants had the increased bleeding tendency in immediate postoperative period. The routine use of anticoagulants in elective free - flap surgery should be reconsidered with postoperatively less bleeding and early recovery.

  • Reactive dyes waste water, a toxic and refractory pollutant, was treated by an electrochemical method using $Ti/IrO_2$ as anode and Stainless Steel 316 as cathode. In this technique, sodium chloride as an electrolyte was added. A number of experiments were run in a batch system. Artificial samples (reactive blue 19, red 195, yellow 145) were used. Operation parameters, such as supporting electrolyte concentration, current density, pH and sample concentration have been investigated for their influences on COD and color removal efficiencies during electrolysis. After 5 and 90 minites of eletrolysis, color was reduced by 51.5% and 98.9% respectively. Under the condition of current density $10A/dm^2$, NaCl concentration 12mg/l and pH 3, 62.9% of $COD_{Cr}$ was removed after electrolysis for 90 minites. The optimum condition of color removal and COD reduction in this work was found to be the following : pH 3, sodium chloride concentration 20g/l, current density $10A/dm^2$. As a result, we confirmed to be effective to color removal and reduction of refractory organic material.

Effects of Continuous Application of CO2 on Fruit Quality Attributes and Shelf Life during Cold Storage in Cherry Tomato

  • 'Unicon' cherry tomato (Solanum lycopersicum) is one of the most highly perishable horticultural crops due to its high water content and respiration rate. This study was carried out to assess the effect of continuous application of $CO_2$ (control [air], 3%, and 5%) on the quality and shelf life of cherry tomato fruits stored at $10^{\circ}C$ and $85{\pm}5%$ relative humidity (RH) at two maturity stages (pink and red). Continuous application of $CO_2$ did not affect the soluble solids content (SSC) or titratable acidity (TA) of the fruit at either maturity stage during storage. However, there was a significant difference among treatments in terms of flesh firmness, cell wall thickness, pectin content, vitamin C content, skin color, lycopene content, weight loss, ethylene production rate, respiration rate, and acetaldehyde and ethanol production. Fruits treated with 5% $CO_2$ maintained their high quality with regards to vitamin C, skin color ($a^*$), lycopene content, weight loss, physiological parameters (ethylene production rate, respiration rate, and volatile compounds), flesh firmness, cell wall thickness, and pectin content at both maturity stages compared with 3% $CO_2$ treatment and the control. Continuous application of $CO_2$ (5%) reduced the ethylene production rate and the production of volatile compounds during storage. Therefore, cherry tomato 'Unicon' fruit can be stored for two weeks without losing fruit quality at both maturity stages under continuous application of 5% $CO_2$ as a postharvest treatment.

Effects of vitamin E and selenium (Selevit) on the orchidectomized rats

  • The present study was devised to determine the effects of body weight, organ weight, hematological values and biochemical parameters by vitamin E and selenium (Selevit) on the Orch rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. Orch group received operation and no treatment. Orch+Selevit received operation and Selevit. The body weights of each group increased, but that of the Orch+Selevit group were significantly lower than those of all the other groups. There were significant differences (P<0.001) of body weights between Orch+Selevit group and all the other groups. Also, organ weights such as heart, liver, spleen and kidney were measured. The heart weights were significantly lower (P<0.001) in the Orch+Selevit group than in Intact and Sham group. The liver weights in the Orch+Selevit group were significantly differences (P<0.001) in comparison with those in the Intact and Sham groups. The kidney weights in the Orch+Selevit group were significantly differences (P<0.01, P<0.001) in comparison with those in all the other groups. On the other hand, there were no significantly differences in the organ weights of spleen between the Orch+Selevit groups and the any other groups. The number of white blood cell (WBC) was significantly higher (P<0.05) in the Orch+Selevit group than in all the other groups. The hematological values of red blood cell (RBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) were not significantly different in any of the groups. The concentrations of serum total protein, albumin and alkaline phosphatase increased significantly (P<0.05, P<0.01) in the Orch+Selevit group as compared to that in the Orch group. However, there were no significant differences in AST and ALT in any other groups. We conclude that Selevit was significantly decreased the body weight in the orchidectomized rats. Our findings suggest that Selevit may influence the process of lipid packaging and absorption in the orchidectomized rats.

Effects of L-carnitine on the orchidectomized rats

The hematological values of mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) were significantly differences (P<0.05, P<0.01, P<0.001) in comparison with those in all the other groups. On the other hand, the hematological values of white blood cell (WBC), red blood cell (RBC) and mean corpuscular hemoglobin concentration (MCHC) were not significantly different in any other groups. The concentrations of total cholesterol (T-chol), triglyceride (TG) and high density lipoprotein (HDL) decreased significantly (P<0.05) in the Orch+Carn group as compared to those in the Orch group. We conclude that L-carnitine was significantly decreased the body weight in the orchidectomized rats. Our findings suggest that L-carnitine may influence the process of lipid packaging and absorption in the orchidectomized rats.

Evaluation through Photochemical Response Analysis on Growth Enhancing Effect of Decomposed Hatchery Waste Egg for Red Pepper (광화학적 반응 분석을 통한 부화장 폐달걀 분해 액비의 고추 생장촉진효과 평가)

  • In this study, growth enhancing effect of hatchery waste egg decomposed liquid fertilizer in pepper plant cultivation through chlorophyll fluorescence (O-J-I-P) analysis. In a whole growth period, egg decomposed fertilizer treated pepper grew well than non treated plant, though it was not statistically significantly different. Amount of chlorophyll fluorescence of non treated plant was higher thant that of fertilizer treated plant. It is determined that eventually lead to increased photosynthesis. In this study, six parameters, Fo, ABS/RC, RC/ABS, TRo/RC, DI0/RC, and DF Total ABS were the important factors represent efficiency of photochemical responses of pepper plant treated with hatchery waste egg decomposed fertilizer.

Studies on the antimetastasis & antiangiogenesis effects of Gamisoamsan (가미소암산(加味消癌散)의 혈관신생(血管新生) 억제에 관한 항암효과 연구)

  • Soamsan is known as an anti-cancer remedy in the traditional Korean Medicine. To enhance the synergic effects of anti-cancer activity of Soamsan, this study reconstituted the original components of Soamsan with a slight modification and produced a novel herbal remedy, namely Gamisoamsan. Extracts of Gamisoamsan inhibited the growth of cultured CT-26 cells, mouse colon adenocarcinoma, in a dose-dependent manner $(1\;to\;50{\mu}g/ml)$, and $ID_{50}$ was estimated approximately $16.7{\mu}g/ml$. Using tumor-bearing mouse model, in which was produced by subcutaneous injection of CT-26 cells ($1{\times}10^5$cells). the effects of Gamisoamsan on tumor growth and host survival were examined by evaluating tumor volume and increase in life span. When Gamisoamsan extracts in variable doses of 100, 200 and 500mg/kg body weight per day were orally administered to tumor-bearing mice, following results were obtained: Improvement in the hematological parameters following Gamisoamsan treatment such as hemoglobin contents, red blood cells and white blood cells of the tumor-bearing mice have been observed. Gamisoamsan treatment also showed a prolongation of life span and a reduction of tumor volume in the CT-26 tumor hosts. The results of the present study suggest that Gamisoamsan extracts has a potential anti-tumor activity and may be an useful remedy to prevent and/or treat cancer.

The effect of repeated firings on the color change of dental ceramics using different glazing methods

  • PURPOSE. Surface color is one of the main criteria to obtain an ideal esthetic. Many factors such as the type of the material, surface specifications, number of firings, firing temperature and thickness of the porcelain are all important to provide an unchanged surface color in dental ceramics. The aim of this study was to evaluate the color changes in dental ceramics according to the material type and glazing methods, during the multiple firings. MATERIALS AND METHODS. Three different types of dental ceramics (IPS Classical metal ceramic, Empress Esthetic and Empress 2 ceramics) were used in the study. Porcelains were evaluated under five main groups according to glaze and natural glaze methods. Color changes (${\Delta}E$) and changes in color parameters (${\Delta}L$, ${\Delta}a$, ${\Delta}b$) were determined using colorimeter during the control, the first, third, fifth, and seventh firings. The statistical analysis of the results was performed using ANOVA and Tukey test. RESULTS. The color changes which occurred upon material-method-firing interaction were statistically significant (P<.05). ${\Delta}E$, ${\Delta}L$, ${\Delta}a$ and ${\Delta}b$ values also demonstrated a negative trend. The MC-G group was less affected in terms of color changes compared to other groups. In all-ceramic specimens, the surface color was significantly affected by multiple firings. CONCLUSION. Firing detrimentally affected the structure of the porcelain surface and hence caused fading of the color and prominence of yellow and red characters. Compressible all-ceramics were remarkably affected by repeated firings due to their crystalline structure.
