Application of artificial insemination and pregnancy diagnosis kit for Korea native black goats (한국 재래흑염소의 계통별 인공수정과 임신진단 키트의 활용)

  • This study investigated the application of artificial insemination and pregnancy diagnosis kit for Korean native black goats. Semen was collected by electrical ejaculation, followed by semen analysis and artificial insemination in three goat strains (Dangjin, Jangsu and Tongyoung). Pregnancy was confirmed using a cow pregnancy test kit (IDEXX Rapid Visual Pregnancy Test kit) and ultrasound diagnosis. Analysis revealed that semen collected from male Korean native black goats by electrical ejaculation was about 1~1.5 ml in volume, $18{\sim}25{\times}10^8/ml$ concentration, and having > 97% motility. Furthermore, confirmation of pregnancy by pregnancy test kit and ultrasound diagnosis after artificial insemination were similar. In addition, the efficiency of pregnancy was 20~40% for all three strains: Tongyoung was the highest with 44%, followed by Dangjin (%), and Jangsu (20%). This study determines the artificial fertilization efficiency and the feasibility of using a cow pregnancy test kit for early pregnancy diagnosis in Korean native black goats. Although further research is required for validation, the results of the current study contribute to the breeding and improvement of Korean native black goat in research institutions as well as in general farms.

A New Early-Maturing, High Quality Rice Cultivar 'Joami' (조생 고품질 벼 신품종 '조아미')

  • A new rice cultivar, 'Joami', was developed by the rice breeding team of Sangju Substation, National Institute of Crop Science (NICS), Rural Development Administration. It was selected by a bulk and pedigree methods from a cross-combination among 'Sambaegbyeo', 'Yukara', and 'Tonggae112'. A promising line of YR20557-1-1-3-B-3 was designated as 'Sangju 36' in 2006. Local adaptability test of 'Sangju 36' was conducted at ten sites throughout the Korean peninsula during three years from 2006 to 2008. 'Sangju 36', thereafter, was registered as 'Joami' in 2008. The cultivar headed on July 30 in the test of local adaptability. Endosperm of 'Joami' is translucent with clear chalkiness and has 5.4% higher head rice ratio than that of 'Odaebyeo'. The yield potential of 'Joami' in milled rice is about 5.40 MT/ha under ordinary fertilizer level of local adaptability test, which was 6% higher than that of 'Odaebyeo'. In an alpine area of Korea, the rice variety needs a cold tolerance and a resistance to blast disease. 'Joami' showed a tolerance reaction at Chuncheon cold tolerance screening nursery and exhibited resistance reaction to blast disease in nation-wide disease screening nursery. Therefore, 'Joami' would be well adaptable to mid-mountainous area at central and southern part of Korean peninsula.

Impact of environmental factors on milk β-hydroxybutyric acid and acetone levels in Holstein cattle associated with production traits

  • The objective of this study was to estimate the environmental factors affecting milk ${\beta}$-hydroxybutyric acid (BHBA) and acetone (Ac) concentrations in Holstein cattle. A total of 264,221 test-day records collected from the Korea Animal Improvement Association (KAIA) during the period of 2012 to 2014 were used in this study. Analysis of variance (ANOVA) was performed to determine the factors significantly affecting ketone body concentrations. Parameters considered in the model were season of test, season of calving, parity, lactation stage, and milk collecting time (AM and PM). According to the ANOVA, the $R^2$ for milk BHBA and Ac were 0.5226 and 0.4961, respectively. 'Season of test' showed a considerable influence on ketone body concentration. Least square (LS) means for milk BHBA concentrations was the lowest ($39.04{\mu}M$) in winter while it increased up to $62.91{\mu}M$ in summer. But Ac concentration did not significantly change along with 'season of test'. The means of milk BHBA and Ac concentrations were high at first lactation stage, low around second lactation stage, and then gradually increased. Cows milked in the morning had lower mean BHBA and Ac concentrations ($48.49{\mu}M$ and $121.69{\mu}M$, respectively) in comparison to those milked in the evening ($53.46{\mu}M$ and $130.42{\mu}M$, respectively). The LS means of BHBA and Ac slightly increased over parities. These results suggest that proper maintenance of milk collection, herd management programs, and evaluation of ketone body levels in milk should be considered for the efficient management of resistance to ketosis.

Subjective Health Assessment of Elderly Agricultural Worker

  • Objective: The purpose of this study was to survey of the subjective health status of the elderly farmer. Background: With increase of aged population, the health of the elderly farmer has become main concerns of the elderly in the rural community. For the effects of the factors related to quality of life, subjective health condition showed the largest influence. Method: The survey was conducted by 326 elderly farmers aged 50 and older in rural areas through structured questionnaires. Survey questionnaire consisted items of general characteristics, health-related life habits, levels of stress(SF-PWI), symptom of musculoskeletal diseases and general level of health(SF-36). For the data analysis, SPSS 19.0K was used, and analysis was conducted by using frequency, percentage, mean, and standard deviation. For the symptom prevalence of musculoskeletal disorders and comparison of average level of health, chi-square test and t-test were used. Results: The results of the study are as follows. It was shown that the symptom prevalence of musculoskeletal disorders had a significant difference in gender (p<0.01). Frequency analysis on presence of musculoskeletal symptoms in each part of body showed that leg/knee region was 28.1%, back region was 26.6%, shoulder region was 18.0%, arm/elbow region was 11.9%, hand/wrist region was 8.2% and neck region was 7.2%. According to comparison of average level of health, a social functioning were the highest as score 87.3, followed by mental health(65.6) and role limitation-emotional(65.5). Conclusion: Health promotion program for the elderly and female in the rural are needed. Application: The survey of the subjective health status of the elderly should be used for planning factors to make a health improvement plan in elderly farmer.

Breeding of biparental sex-limited larval marking yellow cocoon variety "Hanbyeolnue"

  • All of silkworm varieties reared in farmhouses are first generation hybrids, for the production of these hybrid silkworms precise and labor-saving sex discrimination is necessary. The new variety "Hanbyeolnue" is biparental sex-limited larval marking yellow cocoon variety which was bred from biparental sex-limited strain of Japanese originated Jam 319 and Chinese originated Jam 320. Productivity test of Hanbyeolnue in 2015 showed high healthiness and short larval period. The Hanbyeolnue was evaluated as an excellent variety which can be utilized for special purpose silkworm such as male pupae or Cordyceps production.

The Study on Evaluation Method of Pest Control Robot Requirements for Smart Greenhouse (스마트 온실 방제 로봇의 요구조건을 고려한 평가 방법 연구)

  • Recently, research and development on agricultural robots have been on the rise as the interest in smart farming has increased. Robots used in smart greenhouses should be taken into account with the working characteristics and growing environment. This study examined cleaning robots developed through the environmental analysis of smart greenhouses. This study assessed the evaluation method considering the requirements of the pest control robot applicable to the smart greenhouse. The performance and quality assessment criteria were established to conduct tests through the requirements of robots. The required functions and goals of the pest control robot were derived by referring to the robot-related standards. A driving and working ability test was conducted to assess the performance of the robot. The driving test was conducted on the driving performance of the robot and the work capability was tested on the pest control performance. In addition, a durability test was conducted to assess the quality of the robot. The required factors for smart greenhouse robots were derived from the test results. The study results are expected be a standard for an evaluation of a variety of robots for applications to smart greenhouses.

Cellulose Digestibility Increased with CelD Transgenic Pigs

  • This study was performed to test the cellulose digestibility using the transgenic pigs harboring cellulose degradation gene D (CelD). After delivered offsprings between normal pig and transgenic swine, DNA was isolated from piglets tail for PCR analysis. In first generation, five out of 65 piglets showed CelD positive. Unfortunately, four CelD-positive pigs were died during growing, but one survived pig was used as a transgenic founder to produce F$_1$ descendents. Among 3 F$_1$ transgenic pigs produced, one died and the remaining two pigs were used to test the fiber digest efficiency. An assorted feed was composite of 5% fiber with other ingredients. The feed of 3 kg per day was provided to the pigs including transgenic founders and littermate controls. The manure quantity was measured daily for a month, and all manures were dried for three days to analysis nitrogen, phosphate and fiber concentrations. The fiber digestion efficiencies of the transgenic F$_1$ pigs showed approximately 10% higher than those of control pigs. Fiber digestion was not greatly improved in transgenic pigs as it had been expected approximately 30%. Nitrogen concentration of transgenic pig's manure was slowly decreased compare to the control pigs. Because there were only two transgenic pigs tested, a large number of transgenic pigs may be necessary to obtain more reliable data. Breeding of animals to obtain sufficient transgenic pigs subjected for a further study is on progress. Taken together, this study demonstrated successful production of transgenic pigs with increase of cellulose digestibility in the porcine feed.

Genome wide association test to identity QTL for dressing percentage in Hanwoo (전장 유전체 관련성 분석을 통한 한우 도체수율 관련 양적형질좌위 탐색)

  • Genome-wide association study was performed on data from 266 Hanwoo steers derived from 66 sire using bovine 10K mapping chip in Hanwoo (Korean Cattle). SNPs were excluded from the analysis if they failed in over 5% of the genotypes, had median GC scores below 0.6, had GC scores under 0.6 in less than 90% of the samples, deviated in heterozygosity more than 3 standard deviations from the other SNPs and were out of Hardy-Weinberg equilibrium for a cutoff p-value of $1^{-15}$. Unmapped and SNPs on sex chromosomes were also excluded. A total of 4,522 SNPs were included in the analysis. To test an association between SNP and QTL, GWAS for five genetic mode (additive, dominant, overdominant, recessive and codominant) was implemented in this study. Three SNPs (rs29018694, ss46526851 and rs29018222) at a threshold p< $1.11{\times}10^{-5}$ were detected on BTA12 and BTA21 for dressing percentages in codominant and recessive genetic mode. The G allele for rs29018694 has 4.9% higher dressing percentage than A allele, while the T allele for ss46526851 has 2.57 % higher dressing percentage than C allele. Therefore, rs29018694 SNP showed a bigger effect than the other two SNPs (ss46526851 and rs29018222) in this study. In conclusion, this study identifies three loci with moderate effects and many loci with infinitesimally small effect across genome in Hanwoo.

Evaluating germination of lettuce and soluble organic carbon leachability in upland sandy loam soil applied with rice husk and food waste biochar (왕겨 바이오차 및 음식물쓰레기 바이오차가 밭 사양토에서 상추발아 및 수용성 유기탄소 용출에 미치는 영향 평가)

  • This study was carried out to evaluate the effect of rice husk (RHB) and food waste biochar (FWB) on upland soil with sandy loam texture, in terms of physico-chemical analysis, lettuce seed germination test, and orgainc carbon leaching experiment. RHB and FWB had different physico-chemical properties each other. Carbon to nitrogen ratio (C/N ratio) of RHB was 32, showing two times higher than that of FWB. FWB had high salt and heavy metal content, compared to RHB. This is probably due to different ingredients and production processing between two biochars each other. Results of germination test with Lettuce showed lower germination rate when FWB was applied because of higher salt concentration compared to control and RHB. Organic carbon leaching test using saturated soil column (${\Phi}75{\times}h75mm$) with $10MT\;ha^{-1}$ biochar application rate, showed higher saturated hydraulic conductivity in rice husk biochar treatment column, compared to control and food waste biochar treatment. The highest total organic carbon concentration in column effluent was lower than those in both of rice husk biochar and food waste biochar, whereas the differences was negligible after 9 pore volumes of effluent. Consequently, biochars from byproducts such as rice husk and food waste in sandy loam textured upland soil could enhance a buffer function such as reduction of leaching from soil, but the harmful ingredient to crops such as high salt and heavy metals could limit the agricultural use of biochars.

Identification and Antibiotic Susceptibility Test of Bacteria from In Vitro Cultures of Pinellia ternata and Tea Plant (반하 및 차나무의 기내배양시 발생하는 세균의 동정 및 항생제 감수성 검정)

  • Contamination of bacterial infection is one of serious problems in in vitro culture system of root crops. From the contaminated tubes over 140 of petiole cultures of Pinellia ternata, a medicinal plant, 4 genera 8species 48 strains of bacteria, including Aeromonas and Pseudomonas, were isolated and identified and another 8 strains were not fully identified. Most of them were motile Gram positive bacteria as in common in early stage of in vitro cultures. Six strains of bacteria, 5 of Gram negative, including Enterobacter, and 1 of Gram positive, were identified from the embryonic axes cultures of tea plant. From the susceptibility test to pre-screened 5 antibiotics, all of the bacteria except for 2 species of Pseudomonas were susceptible to cefotaxime 60∼100mg/L. While 60mg/L erythromycin only was effective to Pseudomonas. Combination of erythromycin 20mg/L and cefotaxime 60mg/L totally suppressed the growth of all bacterial strains tested. Susceptibility test of bacteria from tea embryonic axes cultures showed similar results. Combination of erythromycin 35mg/L and cefotaxime 60mg/L was effective to 15 bacterial strains and partially effective to 1 unidentified.
