파프리카 추출물이 quinone reductase 유도활성에 미치는 영향 (Induction of Quinone Reductase Activity in Hepatoma Cells by Paprika (Capsicum annuum L.))

  • Phase II 효소계는 자연계에 존재하는 다양한 화학물질과 천연 소재들에 의해 유도되며, 이들의 유도는 화학적 발암물질과 그 밖의 여러 가지 독성물질들로부터 생체를 보호하는데 중요한 역할을 한다. 본 연구에서는 파프리카의 메탄올 추출물을 제조하고, 이를 각 용매별로 분획하여 Hepa1c1c7 세포주를 이용하여 대표적인 암예방 효소인 quinone reductase의 유도 활성을 조사하였다. 그 중 QR 유도활성이 가장 높았던 EtOAc 분획물은 $10-500\;{\mu}g/mL$ 농도로 처리했을 때, $200\;{\mu}g/mL$의 농도에서 QR 활성이 약 3.3배로 가장 높게 나타났으며, 파프리카의 추출물의 총 폴리페놀 및 플라보노이드 함량 역시 EtOAc 분획물에서 가장 높게 나타났다. 또한 $BP^rcl$ 세포주에서도 농도 의존적으로 높은 QR 유도활성을 보여 파프리카의 EtOAc 분획물의 항암성분은 monofunctional inducer인 것으로 생각된다.

Computed Tomography Manifestations of Histologic Subtypes of Retroperitoneal Liposarcoma

  • Objective: Liposarcoma (LPS) is the most common soft tissue sarcoma and accounts for approximately 20% of all mesenchymal malignancies, often occurring in deep soft tissue of retroperitoneal space. Accurate preoperative diagnosis is therefore necessary. We explored whether computed tomography (CT) could be used to differentiate between the various types of retroperitoneal liposarcoma (RPLS). Method: Forty-seven cases of RPLS, diagnosed surgically and histologically, were analyzed retrospectively. CT features were correlated with postoperative pathological appearance. Results: The study radiologist identified 29, 11, 2, 2 and 3 RPLS as atypical lipomatous tumor/well-differentiated liposarcoma (ALT/WDL), dedifferentiated liposarcoma (DDL), myxoid/round cell liposarcoma (ML/RCL), pleomorphic liposarcoma (PL) and mixed-type liposarcoma. Analysis of CT scans revealed the following typical findings of the different subtypes of RPLS: ALT/WDL was mainly visible as a well-delineated fatty hypodense tumor with uniform density and integrity margin; DDL was marked by the combination of focal nodular density and hypervascularity. ML/RCL, PL and mixed liposarcoma showed malignant biological behaviour and CT findings need further studies. Conclusions: CT scanning can reveal important details including internal components, margins and surrounding tissues. Based on CT findings, tumor type can be roughly evaluated and biopsy location and therapeutic scheme guided.

Lactobacillus acidophilus가 생산한 항균물질(抗菌物質)에 관한 연구(硏究) (Studies on antimicrobial agent produced by lactobacillus acidophilus)

  • The research was conducted(1) to confirm the agent(s) responsible for the antimicrobial activity contained in the fermented tomato juice with L. acidophilus(2) to extract and purify the antimicrobial agent(s)(3) to find the biological, physical and chemical properties of the agent(s). The following results were obtained and summarized as followings; 1. The agent responsible for the inhibitory activity was confirmed by both well assay method using fermented tomato juice with L. acidophilus and turbidimetric technique using the cell-free filtrate or neutralized filtrate of tomato acidohilus culture and found exerted antimicrobial agent other than lactic acid. 2. The procedures of purification : The isolation and purification of antimicrobial agent from the lyophilized acidophilus tomato culture were carried out by (1) methanol extraction (2) acetone extraction, (3) Sephadex G-50 gel filtration (4) paper chromatography and (5) thin layer chromatography. 3. The biological, physical and chemical properties of antimicrobial agent: The biological, physical, chemical properties of the purified antimicrobial agent were: (1) The antimicrobial activity was strong against test organisms; Bacillus subtilis(ATCC 6633), Escheichia coli(ATCC 25922), Staphylococcus aureus(ATCC 167), Pseudomonas fluorescens(KFCC 32394), Proteus vulgaris and Shigella dysenteriae. (2) The pH value of the agent was 2.0 and the inhibitory activity was lost when it was neutralized at 7.0 of pH and the agent was heat stable at $121^{\circ}C$ for 60 minutes. (3) The ultraviolet light absorption spectra of methanol-acetone extract and TLC fraction exhibited a maximum absorption at 260nm and 224nm respectively. (4) The most purified agent from TLC plate increased about 130-fold in activity. (5) The agent isolated from TLC plate was free from $H_2O_2$ or lactic acid. 4. Bioautographic assy: By means of bioautography of the agent on silica gel of TLC plate a strong inhibitory activity against B. subtilis was demonstrated. 5. Mass spectrometry: The agent obtained from TLC plate was analyzed by mass spectrometry which show the parent peak at m/e 264 suggesting the molecular weight of the compound and molecular group such as [$C_2H{^+}_4$], [CO], [CH=NH], [$C_3{H^}4_7$], [$\begin{array}{rcl}O\\{\parallel}\\CH_3-C\\\end{array}$], [$C_6-H{^+}_{11}$], [$C_5H{^+}_{11}$], [$\begin{array}{rcl}O\\{\parallel}\\C_5H_7-C^+\\\end{array}$] were suggested.

무지 중수지 관절의 만성 파열된 척측, 요측 측부 인대 봉합술 후 결과 비교 (Comparison between Chronic Ulnar and Radial Collateral Ligament Repairs in the Metacarpophalangeal Joint of the Thumb)

  • 목적: 무지 중수 수지 관절의 척측 측부 인대의 만성 파열에서의 직접 봉합술은 양호한 결과가 보고되고 있으나 요측 측부 인대에서의 결과는 논란이 있다. 본 연구의 목적은 무지 중수 관절의 척측과 요측 측부 인대의 만성 파열에서 인대 재부착술의 결과를 비교하는 것이다. 방법: 무지 중수 수지 관절의 6주 이상된 측부 인대 파열에서 봉합 앵커를 이용해서 재부착술을 시행한 요측 6예 및 척측 8예의 평균 22개월 추적한 방사선적 및 임상 결과를 후향적으로 검토하고, 통계적으로 비교하였다. 결과: 수술 후 평균 척측 변위 각은 요측 측부 인대에서 $13.3^{\circ}$, 척측 측부 인대에서 $2.0^{\circ}$였다(p=0.020). 수술 후 인대 불안정성은 요측 인대 6예 중 4예에서 관찰되었으나, 척측 인대에서는 관찰되지 않았다. 수술 후 경과 관찰에서 관절의 아탈구는 요측 인대 2예에서만 관찰되었다. 결론: 무지 중수 수지 관절의 요측 측부 인대의 지연 봉합은 덜 양호한 결과를 나타내며, 반 이상에서는 수술 후 인대의 불안정성이 관찰된다.

Selective Gene Express Profiles in Rat Uterus during Estrus Cycle

  • The uterus undergoes dynamic changes during the cycle and displays many features typical of developmental process. In order to be prepared for implantation, endometrium undergoes predictable, sequential phases of proliferation and secretory changes. The uterus during estrus cycle synthesize a complex of signaling molecules with specific spatial and temporal modes of expression and which are critical for cell proliferation and differentiation. The purpose of this investigation was to use cDNA microarrays to evaluate the expression of genes of rat uterus in estrus cycle. Animals were sacrificed on proestrus, estrus, metestrus, diestrus. Differential gene expression profiles were revealed(growth-related c-myc reponsive protein RCL, heat shock 47-kDa protein (HSP47), cytochrome c oxidase polypeptide Vlc2 (COX6C2), calreticulin (CALR)). Reverse transcription polymerase chain reaction (RT-PCR) was used to validate the relative expression pattern. Using this approach, we found several genes whose expression in rat uterus was altered with estrus cycle. Our long-term goal is to determine the role of these differentially expressed genes during estrus cycle. 원전 배관용 SA 106 Gr.C의 협개자동용접 적용에 관한 연구 (The Application of Narrow-Gap Welding Process for SA 106 Gr.C in Nuclear Power Plant)

  • Conventionally, SMAW process was applied to join pipes of RCL, which caused lot of loss in time and cost due to excessive heat input and defects in joining section. Recently, narrow-gap welding(NGW) process was introduced to overcome the disadvantages of SMAW. However, the application of NGW to nuclear power plant is not yet common because safety of NGW process is not proven. In present paper, the welded coupons are made of carbon steel. They are manufactured under different processes; general welding(GW), post-weld heat treatment(PWHT) after GW, repair welding after GW and PWHT with repair welding after GW in carbon steel. Performed are various mechanical tests investigation of microstructure, hardness test, tensile test at room and high temperature, bending test, impact test and J-R test. It is verified that the mechanical properties of carbon steel are greatly changed after repair welding process due to applied heat flux, and that the effect of PWHT is beneficial.

석영기판에 증착된 질화탄소막의 유전특성 (Dielectric Characteristics of Carbon Nitride Films on Quartz Substrate)

  • Carbon nitride($CN_x$) thin films were deposited on quartz substrates using reactive RF magnetron sputtering system at uarious deposition conditions and investigated dielectric characteristics. Samples for capacitance measurements were of the MIM(Metal-Insulator-Metal) type devices. Aluminum film electrodes were prepared by a vacuum thermal evaporation method before and after the deposition of carbon nitride films. Capacitances were measured by a FLUKE PM6306 RCL Meter at room temperature. Current-voltage(I-V) characteristics and resistivity were measured by a CATS CA-EDA semiconductor test and analyzer. The carbon nitride films showed ${\alpha}-C_3N_4$ and ${\beta}-C_3N_4$ etc. peaks through Raman and FTIR. Observed surface of film and side structure using SEM(Scanning Electron Microscope), and measured thickness of film by ${\alpha}-step$. We can find that the dielectric constant was the lowest value in 50% nitrogen ratio and the resistivity was the highest value in 70% nitrogen ratio.

$RuO_2$계 후막저항체의 교류 임피던스 특성 (A.C.impedance properties on $RuO_2$-based thick film resistors)

  • 저저항(DuPont 1721, 100.OMEGA./sq.)과 고저항(1741, 10K.OMEGA./sq.)의 두 Ru계 후막저항체를 여러 조건에서 소결하여 소결막의 복소임피던스 특성과 임피던스의 주파수의존성을 1KHz-13MHz의 주파수 범위에서 조사하였다. 저저항 1721계의 경우 600.deg.C이상에서 소결한 모든 시편이 거의 저항성분(R)만으로 구성된 등가회로에 해당되는 복소임피던스 거동을 보였으며 임피던스에 미치는 주파수 의존성은 크게 나타나지 않았는데 5KHz까지는 주파수에 따라 변화가 없다가 그 이상의 주파수에서 주파수 증가에 따라 약간씩 증가하였다. 고저항 1741 후막저항체의 경우는 소결조건에 따라 복소임피던스 거동과 임피던스에 미치는 주파수 의존성이 달리 나타났다. 600.deg.C에서는 용량(C) 성분만으로 구성된 등가회로에 해당하는 복소임피던스 거동을 얻었고 주파수 증가에 따라 임피던스가 직선적으로 감소하였으며 700.deg.C이상 900.deg.C까지는 저항(R)과 용량(C)이 병렬로 연결되는 형태의 등가회로에 해당하는 복소임피던스 거동을 얻었고 이때의 임피던스의 주파수 의존성은 저주파수 영역에서는 임피던스가 주파수에 변함없이 일정하다가 5KHz이상의 주파수에서는 주파수 증가에 따라 임피던스가 직선적으로 감소하였다. 1000.deg.C반응에서의 복소임피던스 거동은 RCL성분이 병렬로 연결된 형태의 등가회로에 해당되는 결과를 얻었으며 임피던스도 작아지고 주차수 의존성도 현저하지 않았다.
