삼묘환(三妙丸)의 LPS에 의해 활성화된 RAW 264.7 cells과 BV-2 Microglia cells로부터 생성되는 nitric oxide 및 $TNF-{\alpha}$의 생성억제효과 (Inhibitory effects of Sam-Myo-San on the LPS-induced production of nitric oxide and $TNF-{\alpha}$ in RAW 264.7 cells and BV-2 Microglia cells)

  • Objectives : Sam-Myo-Whan(SMW) has been known traditional prescription with anti- anthritis activities. We investigated inhibitory effects of SMW on lipopolysaccharide (LPS)-induced nitric oxide(NO), $TNF-{\alpha}$ and inducible nitric oxide synthase(iNOS) production from RAW264.7 cells and BV-2 Microglia cells. Methods : SMW, which had been extracted with 70% MeOH, concentrated and freeze-dried was used for this experiment. After BV2 mouse brain macrophages and RAW264.7 mouse peritoneal macrophages were pretreated with increasing concentrations of SMW extract for 30min, and then activated with LPS. To investigate cytotoxicity of SMW extract, cell viability was measured by MTT assay. NO production was measured in each culture supernatant by Griess reaction. mRNA expression of iNOS in two type cells was investigated by RT-PCR. $TNF-{\alpha}$ production was measured in each culture supernatant by ELISA. Results : SMW extract significantly inhibited LPS-induced NO and $TNF-{\alpha}$ production in BV2 cells and RAW264.7 cells dose-dependently. SMW extract also greatly suppressed mRNA expression of iNOS in both type cells activated with LPS. Conclusion : These data suggests that SMW extract may have an anti-inflammatory effect through the inhibition of iNOS expression.

LPS로 유도한 RAW 264.7 세포의 염증반응에서 마치현(馬齒莧) 70% 에탄올 추출물의 항염증 효과 (The anti-inflammatory effect of Portulaca oleracea 70% EtOH Extracts on lipopolysaccharide-induced inflammatory response in RAW 264.7 cells)

  • Objectives : Portulaca oleracea (PO) have been used as a traditional medicine to treat inflammatory diseases in Korea. However, the anti-inflammatory effect of PO ethanol extract on lipopolysaccharide (LPS)-induced inflammation is not well-known. Therefore, this study was performed to identify the anti-inflammatory effect of PO on LPS induced inflammatory.Methods : Identification of PO was conducted by comparison with purified standards by HPLC. To measure out the cytotoxicity of PO, author performed the MTT assay. To evaluate the anti-inflammatory effects of PO, author examined the inflammatory mediators such as nitric oxide (NO) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-α, interleukin, (IL)-1β and IL-6) on RAW 264.7 cells. Author also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-κB) activation by western blot.Results : Three major components (peaks 1, 2, 3) were detected in both varieties and peak 1 was characterized as caffeic acid, peak 2 as p-coumaric acid, and peak 3 as ferulic acid by comparison of chromatographic properties with authentic standards. Extract from PO itself did not have any cytotoxic effect in RAW 264.7 cells. PO inhibited LPS-induced productions of inflammatory mediators such as NO and pro-inflammatory cytokines in RAW 264.7cells. In addition, PO inhibited the phosphorylation of extracellular signal-regulated kinase1/2 (ERK1/2), c-Jun NH2-terminal kinase (JNK) and NF-κB activation in RAW 264.7 cells.Conclusions : Above experiment data can be an important indicator for the identification of PO and this study suggest that treatment of PO could reduce the LPS-induced inflammation. Thereby, PO could be used as a protective agent against inflammation.

Inhibitory effects of honokiol on LPS and PMA-induced cellular responses of macrophages and monocytes

  • The regulatory effects of honokiol on the cellular responses of macrophages and monocytes were evaluated. Specifically, we investigated the effects of honokiol with respect to lipopolysaccharide (LPS)-induced cytotoxicity, LPS- or phorbol-12-myristate-13-acetate (PMA)-mediated morphological changes, and relevant events (FITC-dextran-induced phagocytic uptake). Honokiol blocked the LPS-induced cytotoxicity of RAW264.7 cells in a dose-dependent manner. In addition, honokiol appeared to block the production of cytotoxic cytokines such as interleukin (IL)-$1{\beta}$ and tumor necrosis factor (TNF)-$\alpha$, nitric oxide (NO), and reactive oxygen species (ROS). Moreover, honokiol strongly prevented the morphological changes in RAW 264.7 and U937 cells that were induced by LPS and PMA. The surface levels of marker proteins, which are up-regulated under the morphological changes of RAW264.7 and U937 cells, were also diminished. The data presented here strongly suggest that the honokiol modulates various cellular responses managed by macrophages and monocytes.

  • The purpose of this research is to present necessity of manament system establishment and policy direction for fundmental data, understanding management reality and problem, comparing resident's management attitude to related variables in apartment and raw houses multi-family houses. The major finding were as follows : 1) The management attitude of Apartment inhabitant is very highm especially the attitude of maintenance management is the higest. But they disagree with the raise of management cost necessary to maintenance management. 2) The management attitude of Raw multi-family houses inhbitant is especially high, the maintenance management demand for safety and community-life contol attitude, resident's mutual cooperation is especially high, they think much of autonomy management, desire to achieve under more sysemnatic plan.

Broiler저장의 온도 및 경시에 따른 Methionine 함량 변화에 관한 연구 (A Study on the Methionine Content of Cold Storage Broiler in the Cooking)

  • In the view of the fact that data are scare concerning the change of Methionine composition of poultry meat in function of storage temperature and cooking method. The present study was carried out to change in these compounds and the results were summarized as follows. 1. The result of measuring methionine compound of raw material stored under the condition of 5$^{\circ}C$, 1$0^{\circ}C$ for 6 weeks were increased as temperature increases. 2. The result of change in methionine compound of roasting were decreased than raw material It was greatly decreased in the storage for 42 days at 1$0^{\circ}C$ and slightly decreased in the storage for 14 days at 5$^{\circ}C$ 3. When the boiling in the water, methionine compound was decreased than roasting to compare with raw material 4. By the results of this study, one of the best cooking method is roasting, because of the change in methionine compound of roasting is slightly decrease.

원료의약품 분진의 폭발 위험성 평가 (Hazard Assesment of Dust Explosion Pharmaceutical Raw Material Powders)

  • Dust explosions are occurring in a variety of industries. A dust explosion caused by a specific energy generates huge amount of energy in the ignition and releases decomposition gas. Damages can be increased since this released decomposition gas can cause second and subsequent explosions. In this study, the goal was to obtain practical information on what could affect the explosion by comparing the characteristics of two kinds of dusts with completely different chemical properties. Three kinds of dusts were measured and evaluated for explosion pressure, dust explosion index, explosion limit and minimum ignition energy. It is possible to grasp the characteristics of each dust and use it as useful accident prevention data in the production of raw material powder.

  • RDF(Refuse-Derived Fuel) is a fuel of pelletized form made of combustible solid wastes and can not only be used as alternative energy to fossil fuel but also solve troubles in thermal uses of incinerator. As the first stage for obtaining elementary data required to develop criteria of raw materials appropriate to RDF combustion facilities actively spread recently in Korea, preliminary experiments were conducted on CO, SOx, NOx and HCl production and reduction characteristics in combustion of RDF. RDF samples weighing 2~3 g per a sample were manufactured in a lab-scale way and combustion tests of RDF were carried out in electric furnace with quartz tube of 50 mm inside diameter.

Nutraceutical Properties of Dioscorea opposita Thunb. (Yam) Fermented by Lactobacillus bulgaricus and Streptococcus thermophilus

  • This study was performed to determine by the ability of the mixed culture of Lactobacillus bulgaricus and Streptococcus thermophilus to ferment Dioscorea opposita Thunb. (yam) and to evaluate the nutraceutical value of fermented yam. The titratable acidity (TA) value increased from 2 to 6% with increased concentrations in both raw yam and extracted lactic acid bacteria (LAB) fermented yam (LFY). The viable cell counts and the allantoin and diosgenin contents were higher in raw LFY at large concentrations (6%) than in extracted LFY samples at all fermentation periods up to 32 h. Based on these data, it confirmed that raw yam fermented by the combination culture of L. bulgaricus and S. thermophilus for various fermentation periods favors the symbiotic growth of LAB and results in higher nutraceutical content.

생전분을 당공여체로 한 Stevioside의 당전이 반응의 동력학적 해석 (Kinetic Analysis of Transglycosylation Reaction of Stevioside Using Raw Starch as a Glycosyl Donor)

  • 분쇄마찰매치 함유 불균일상 효소반응계에서 생전분을 당공여체로 한 stevioside의 당전이 반응의 kinetics에 관한 연구를 수행하였다. 생전분으로부터 CD를 생서하는 과정과 생성된 CD와 stevioside가 random sequenrial bireactant ,echanism으로 반응하여 당전이 equation을 유도하였다. 또한 유도된 반응식의 각종 kinetic constants을 평가하였다. 유도된 반응식을 Runge-Kutta integration법으로 계산하였으며, 계산 결과를 실험치와 비교하여 유도식의 효용성을 검토하였다. 유도된 kinetic equations는 당공여체인 생전분의 농도, stevioside의 농도, 그리고 중간산물인 CD의 농도 변화를 비교적 정획히 표시할 수 있었으며, 고효율 당전이 효소반응기 개발에 활용될 것이다.
