The Effect of Yongyukjowitang Distillate on the Immune Activity of Spleen Cells of Aged Rats (용육조위탕(龍肉調胃湯) 증류액이 노화 흰쥐 비장 세포의 면역활성에 미치는 영향)

  • Objectives The purpose of this study was to investigate the effect of Yongyukjowitang(YJT) distillate on the immune activity of spleen cells of aged SD rats. Methods Spleen cells from 10w, 52w, 72w old rats were stimulated with Concanavalin A(Con A) and treated with Vitamin C(Vit.C) or Yongyukjowitang distillate(YJT). After 24 hours, levels of IL-2, IL-4, IL-10, IFN-${\gamma}$ were measured using ELISA in spleen cells. Results and Conclusions 1. The concentration of IL-2, IL-4 in spleen cells of 52 weeks old SD rats(YJT group) significantly decreased in comparison with ConA group. 2. The concentration of IFN-${\gamma}$ in spleen cells of 72 weeks old SD rats significantly increased in comparison with 10 weeks old SD rats(NT group), but decreased in comparison with 10 weeks old SD rats(YJT group). These results suggest that Yongyukjowitang distillate has an effect on the immune activity of spleen cells of aged SD rats.

The Effect of Angiotensin II on the Hypertension Immune Mechanism in Salt-Sensitive Rats (염 민감성 쥐에서 안지오텐신 II가 고혈압 면역 기전에 미치는 영향)

  • Hypertension caused by high-fat and high-salt diets is is a well-known significant risk factor for cardiovascular and cerebrovascular diseases. In this study, to confirm the relationship between hypertension and immune cells, angiotensin (Ang) II was administered to Dahl salt-sensitive (SS) rats and Dahl salt-resistant (SR) rats. Then the expression of immune cells and the proinflammatory cytokines were compared between the SS and SR rats. It was observed that after administration of Ang II (50ng/kg/min) for three weeks, blood pressure was increased in the SS rats, but there was no significant change in the SR rats. In addition, the expression of T helper (Th) cells and Th 17 cells in the spleen and the expression of Th cell Rorγt and regulatory T regulatory (Treg) cells in the peripheral blood mononuclear cells did not show a significant difference between the two experimental groups even after the administration of Ang II.IL-1β expression was significantly increased in the kidney tissue of the SS rats, while there was no significant difference in the IL-6 expression in all the experimental groups. The results of this study suggest that Ang II induces hypertension by stimulating IL-1β secretion from renal macrophage in SS rats.

Differential Expression of Metabolism-related Genes in Liver of Diabetic Obese Rats

  • The Otsuka Long-Evans Tokushima Fatty (OLETF) rat, a model of spontaneous type 2 diabetes (T2D), develops hyperglycemic obesity with hyperinsulinemia and insulin resistance after the age of 25 weeks, similar to patients with noninsulin-dependent diabetes mellitus (DM). In the present study, we determined whether there are differences in the pattern of gene expression related to glucose and lipid metabolism between OLETF rats and their control counterparts, Long-Evans Tokushima (LETO) rats. The experiment was done using 35-week-old OLETF and LETO rats. At week 35 male OLETF rats showed overt T2D and increases in blood glucose, plasma insulin, plasma triglycerides (TG) and plasma total cholesterol (TC). Livers of diabetic OLETF and LETO rats also showed differences in expression of mRNA for glucose and lipid metabolism related genes. Among glucose metabolism related genes, GAPDH mRNA was significantly higher and FBPase and G6Pase mRNA were significantly lower in OLETF rats. For lipid metabolism related genes, HMGCR, SCD1 and HL mRNA were substantially higher in OLETF rats. These results indicate that gluconeogenesis in OLETF rats is lower and glycolysis is higher, which means that glucose metabolism might be compensated for by a lowering of the blood glucose level. However, lipid synthesis is increased in OLETF rats so diabetes may be aggravated. These differences between OLETF and LETO rats suggest mechanisms that could be targeted during the development of therapeutic agents for diabetes.

Effects of Electrical Stimulation on Wound Healing and Skin Mast Cells in Streptozotocin-Induced Diabetic Rats (스트렙토조토신 유도 당뇨 흰쥐에서 전기자극이 상처치유와 피부 비만세포에 미치는 영향)

  • The aim of this study was to investigate the effect of electrical stimulation on healing of impaired wound and alteration of mast cells in experimental diabetic rats. Thirty male Sprague-Dawley rats were divided into three groups : incision (control), diabetes+incision (diabetes) and diabetes + incision + electrical stimulation (D/ES). Diabetes was induced in rats by streptozotocin (STZ) injection (60 mg/kg, one time) and 20 mm length incision wounds were created on the back after shaving hair. The electrical stimulation rats were treated with a current intensity of 30~50 V at 120 pps and $140{\mu}s$ for 10 days from 3 days after STZ injection. The lesion and adjacent skin tissues were fixed with 10% buffered formalin, embedded with paraffin. For wound healing analysis, hematoxylin-eosin (HE) and picrosirius red staining were performed. Mast cells (MC) were stained with toluidine blue (pH 0.5) and quantified at ${\times}200$ using a light microscope. The density of keratinocyte proliferation and microvessels in skin tissues were analyzed using a computerized image analysis system on sections immunostained with proliferative cell nuclear antigen (PCNA) and ${\alpha}$-smooth muscle actin (${\alpha}$-SMA), respectively. The results showed that the wound healing rate, collagen density and neoepidermis thickness, density of PCNA-positive cells and density of ${\alpha}$-SMA-positive vessels were significantly higher in D/ES rats than in diabetic rats. The density of MCs and degranulated MCs in D/ES rats were also significantly higher than those in diabetic rats. These findings suggest that the electrical stimulation may promote the tissue repair process by accelerating collagen production, keratinocyte proliferation and angiogenesis in the diabetic rats, and MCs are required for wound healing of skin in rats.

Effects Protein Quality, Energy Restriction and Subsequent Rehabilitation on Growth in Young Rats (단백질 급원의 변화와 열량제한 및 식이회복이 흰쥐의 성장발달에 미치는 영향)

  • Effects of dietary protein qualify, energy restriction, and subsequent rehabilitation were studied in the weanling rats. Rats were devided into the six experimental diet groups. Rats in AC (casein- containing diet ), AS (soyprotein-containing diet), and AG(gluten-containing diet) group consumed their diets ad libitum, and rats in PC, PS, PG groups were pair-fed in 60% of the of amounts eaten by corresponding rats in the AC, AS, AG groups, respectively. After 3 weeks of nutritional restriction, all rats were switched to the AC diet for 14 days. At the end of restriction period, body weight, weight gain, food intake, and FER were higher in casein- fed rats than other groups of rats. The differences were, however, disappeared with rehabilitation diet, except body weights which were not fully recovered until the end of experiment. The weights of liver, gastrocnemius muscle, kidney, small intestine, spleen, and lung were the highest in the casein group, and the lowest in gluten group at the end of restriction period. The recovery with rehabilitation diet were differ in different organs. The muscle and liver seemed to be the most affected organs by dietary protein quality and energy restriction. There were no differences in brain weights among the experimental groups during the restriction period, however, after 2 weeks of rehabilitation, rats in AC group showed lower brain weight compared to AC and AS groups, and the brain weight of PC group was lower than AC group. Protein contents in liver and muscle showed the same trends as their weights. Gluten-fed rats showed low serum protein concentration, but recovered fully with rehabilitation diet for 3 days.

Effect of Postnatal Angelicae Gigantis Radix Herb-acupuncture on Cell Proliferation in Offspring Rats with Prenatal Noise Stress during Pregnancy (당귀약침(當歸藥鍼)이 소음 Stress를 받은 새끼 쥐의 신경세포 생성에 미치는 영향)

  • Methods : 40 of Female rats were allowed to mate with 40 of male rats. Then, female rats were delivered of offspring rats, After birth 28 days, offspring rats were divided 8 groups, The normal group(Group A), the 10 mg/kg Angelicae gig antis radix~treated group(Group B), the 50 mg/kg Angelicae gigantis radix-treated group(Group C), the 100 mg/kg Angelicae gig antis radix-treated group(Group D), The control (noise-treated) group(Group E). the noise -10 mg/kg Angelicae gigantis radix-treated group(Group F), the noise-50 mg/kg Angelicae gigantis radix~treated group(Group G), and the noise-100 mg/kg Angelicae gigantis radix-treated group(Group H)(n = 5 in each group), From the 15th day of pregnancy, all rats were subcutaneously injected with 50 mg/kg BrdU once a day 30 min before the starting of experimental treatment. Rats of the prenatal noise-treated group were applied with 95 decibel supersonic machine sound for 1h once a day until delivery, After birth 28 days, offspring rats intraperitoneally injected with 50 mg/kg of BrdU and offspring rats were treated Angelicae gigantis radix Herb-acupunture on chungwan(CV12) for 7 consecutive days. For the detection of BrdU-positive cells and Ki-67 positive cells in hippocampus, immunohistochemistry was performed. Results : 1. The number of BrdU-positive cells in the dentate gyrus of noise-treated group was significantly decreased to normal group, and the Group F, G, H were significantly increased to control group. 2. The number of Ki-67 positive cells in the dentate gyrus of noise-treated group was significantly decreased to control group, and the Group G, H were significantly increased to control group. Conclusion : We concluded that postnatal Angelicae gigantis radix administration has effect on cell proliferation in offspring rats with prenatal noise stress during pregnancy.

Effects of Isoflavones on Bone Mineral Density and Bone Mineral Content in Ovariectomized Rats (이소플라본이 난소절제 쥐에서 골밀도와 골무기질 함량에 미치는 영향)

  • A recent study reported that a diet rich in isoflavones is beneficial for bone formation in growing rats. It therefore seemed desirable to find out whether the beneficial effect of isoflavones in ovariectomized rats could also be reproduced with same amount of isoflavones which used for growing rats. To study the effect of isoflavones, an equal amount of isoflavones which used for growing rats, on bone mineral density and bone mineral content in ovariectmized rats were performed. Forty female Sprague-Dawley rats (body weight $210{\pm}5g$) were divided into two groups, ovariectomy and sham groups, which were each randomly divided into two subgroups that were fed casein and casein supplemented with isoflavones diets for 9 weeks after operation. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin) in spine and femur. Serum alkaline phosphatase activity (ALP) and osteocalcin and urinary DPD crosslinks value were measured as markers of bone formation and resorption. The results of this study indicate that body weight gain and food intake were higher in ovariectomy groups than in sham groups regardless of diets. Serum Ca concentration was lower in ovariectomy groups than in SHAM groups. Serum ALP, osteocalcin, and crosslink value were increased in ovariectomy groups. Spine BMD/weight, femur BMD/weight, and femur BMC/weight of ovariectomy groups were significantly lower than SHAM groups after 9 weeks. However, isoflavones supplemented group in ovariectomy groups, serum ALP and osteocalcin concentrations, spine BMD/weight and spine BMC, femur BMD/weight and femur BMC/weight were significantly increased after 9 weeks. In conclusion, the beneficial effect of isoflavones on bone in ovareiectomized rats was shown on 9 weeks after feeding with an equal amount of isoflavones supplementation which used for growing rats.

Gene Expression of Intrarenal Renin-angiotensin System in Streptozotocin-induced Diabetic Rats

  • In humans and many animal models with chronic progressive renal diseases, angiotensin-converting enzyme (ACE) inhibitor markedly attenuates the progression of nephropathy. Several studies have reported augmented gene expression and redistribution of renal renin in partial nephrectomized rats. Although precise mechanism(s) is not known, the renin-angiotensin system (RAS) may play an important role in the progression of renal diseases. Thus, this study was undertaken to examine the gene expression of renal renin, angiotensinogen, and $AT_1$ subtypes ($AT_{1A}$ and $AT_{1B}$) in rats with diabetic nephropathy, and the influences of lipopolysaccharide (LPS)-induced septicemia on the gene expression. Four weeks after streptozotocin (STZ) treatment (55 mg/kg, i.p.), rats were randomly divided into LPS-treated (1.6 mg/kg, i.p.) and control rats. At 6 hours after LPS treatment, the rats were killed and the kidney was removed from each rat. Northern blot and reverse transcription-polymerase chain reaction (RT-PCR)techniques were used to detect mRNA expression. STZ treatment markedly attenuated body weight gain and significantly increased blood glucose level. Renal renin content (RRC) was significantly decreased in the STZ-treated rats compared to that in control rats. The renal ACE activity between STZ-treated and control rats was not significantly different. Renal renin mRNA level was prominently increased, while angiotensinogen and $AT_{1A}$ mRNA levels were slightly decreased in STZ-treated rats compared to those in controls. $AT_1$B mRNA level did not differ in both groups. Acute LPS treatment did not show any significant changes of mRNA levels of intrarenal RAS components in both groups. These results suggest that intrarenal RAS components were differentially regulated in STZ-treated diabetic rats. Further studies are required to evaluate the relationship between intrarenal RAS and other vasomodulatory systems.

Effects of Pretreatment of Serotonin Synthesis Inhibitor p-chlorophenylalanine on Lipopolysaccharide-induced Anorexia in Rats

  • In the present study, we investigated the effect of pretreatment of p-chlorophenylalanine (PCPA), inhibitor of serotonin synthesis, on lipopolysaccharide (LPS)-induced anorexia in rats. First of all, effects of PCPA injection on food intake and body weight in rats were investigated. During 4 days of PCPA injection (300 mg/kg BW once a day), food intake was decreased by 33% and daily gain in body weight was inhibited compared with controls. Twenty-four hours after last PCPA injection, food intake and gain in body weight returned toward almost normal. Pair-feeding to PCPA (PCPAP) injection revealed that body weight of rats in PCPA group was not different from rats in PCPAP groups. To quantify the effects of LPS on food intake and body weight, we administered varying doses $(10,\;100,\;500\;{\mu}g/kg\;BW)$ of LPS to rats. LPS induced a reduction of food intake and weight loss in a dose dependent manner compared with controls. To evaluate the effects of PCPA pretreatment on LPS injection, rats were treated with PCPA for 4 days (300 mg/kg BW once a day), which was followed by LPS injection for 2 days $(500\;{\mu}g/kg\;BW\;once\;a\;day)$ (PCPA+LPS group), while rats in LPS group had injections with normal saline instead of PCPA for 4 days, which was followed by LPS administration. Rats in control group received 0.9% NaCl for 6 days. LPS decreased food intake by 80% and inhibited gain in body weight, while PCPA pretreated rats showed normalized food intake and gain in weight during the days of LPS injections compared with controls. In conclusion, pretreatment of PCPA prevented LPS-induced anorexia.

Effect of Dietary Supplementation of Sypjeondaebotang or Jahyulyanggeuntang on Iron Bioavailability in Rats (십전대보탕 및 자혈양근탕 첨가 식이가 흰쥐의 철분 생체이용률에 미치는 영향)

  • The effect of dietary supplementation of the two oriental medicinal prescriptions, Sypjeondaebotang or Jahyulyanggeuntang, on iron bioavailability was evaluated in rats which were depleted of iron by being fed an irondeficient diet for 4 weeks. Seventy two iron-depleted rats were randomly divided into 3 groups (n = 24) , and fld one of the following experimental diets for 4 (n = 8), 12 (n = 8), and 25 days (n = 8) : Control diet (CD), Sypjeondaebotang-supplemented diet (SD), Jahyulyanggeuntang -supplemented diet (JD). The CD contained 20 mg Fe/kg diet as FeSO4, and the SD or JD was identical except that the respective oriental medicinal prescription extract was included at the level of 4 g/kg diet. Animals fed the SD and JD for 25 days consumed significantly less food (p<0.001), but showed no change in food efficiency ratio compared to those that were fed the CD. Serum iron concentration and transferrin saturation values were significantly higher in rats fed the SD for 25 days compared to those that were fed the CD for the same period (p < 0.05). The values of serum UIBC (p < 0.05) and TIBC (p > 0.05) were lower in rats fed the SD compared to those fed the CD. Dietary supplementation of Sypjeondaebotang during the period of iron repletion significantly increased blood levels of hemoglobin (p < 0.05) and hematocrit (p < 0.01) measured at day 12, and increased mean corpuscular volume (p < 0.05) measured at day 25, compared to the values for the CD rats. Regression analyses of hemoglobin-repletion bioassay data from rats fed the SD and JD showed the relative biological values of 123% and 99%, respectively, calculated against the slope for the CD rats. Apparent iron absorption and retention values were also significantly lower (p < 0.05) in rats fed the SD for 25 days than those for the CD rats. Based on the results from diverse biochemical indices of iron status and the chemical balance study, the effect of Jahyulyanggeuntang on iron bioavailability appears to be less prominent than that for Sypjeondaebotang. Taken together, these results indicate that Sypjeondaebotang has a positive effect in restoring iron depletion by increasing the iron bioavailability in rats.
