지형요소를 이용한 홍수범람해석 모형의 비교 (Comparison of Flood Inundation Models using Topographic Feature)

  • 본 연구의 목적은 소하천 유역에 대한 홍수범람해석 모형을 비교하는 것이다. 검청천을 대상으로 1차원 수리모형인 HEC-RAS와 범람해석을 위한 HEC-GeoRAS, RAS Mapper, RiverCAD 모형을 적용하여 홍수범람해석을 수행하였다. 실측 강우량과 빈도별 홍수량을 적용하여 3가지 모형에 따른 홍수범람해석을 모의하였으며, 해석결과와 당시 조사된 침수흔적도를 비교하고 각 모형들의 빈도별, 침수심별에 따라 홍수범람면적을 비교하였다. 실측 강우량을 적용하여 해석된 홍수범람면적은 HEC-GeoRAS 모형의 결과가 침수흔적도와 비교적 유사하게 나타났다. 또한, 빈도별 설계홍수량을 적용하여 해석된 범람면적은 RAS Mapper 모형이 가장 크게 나타났으며, 3가지 모형의 전체 범람면적 차이가 10% 이내로 비교적 유사한 결과를 나타내었다. 침수심별 범람면적을 비교한 결과 침수심 2.0 m 이하 구간에서는 3가지 모형의 결과가 비교적 유사하게 나타났으며, 침수심 2.0 m 이상의 구간에서 RiverCAD 모형의 범람면적이 비교적 큰 차이를 나타내었다.

FPGA를 이용한 single rate Rate Adaptive Shaper 설계 (Design of single rate Rate Adaptive Shaper Using FPGA)

  • 본 논문은 RFC2963에서 제안된 single rate Rate adaptive Shaper (srRAS)를 설계한 것이다. srRAS는 RFC2693에서 제안된 하향의 single rate Three Color Marker (srTCM)과 함께 사용된 쉐이퍼이다. 그것은 가변 속도로 출력되는 tail-drop FIFO (First Input First Out) 큐이다. srTCM은 srRAS로부터의 IP 패킷 스트림을 측정한 후 그 패킷을 green, yellow, 또는 red로 마킹해 준다. 이 쉐이퍼는 AF PHB (Per Hop Behavior)를 제공하는 DS (Differentiated Service) 네트워크의 입력에서 사용되도록 제안되었다. 그리고 srRAS는 srTCM의 상향 트래픽의 버스트성을 줄여줄 수 있다. 본 논문은 srRAS의 알고리즘, 구조, 그리고 FPGA 및 관련 기술을 통하여 구현할 수 있는 방안을 언급하였다.

Induction of MAP kinase phosphatase 3 through Erk/MAP kinase activation in three oncogenic Ras (H-, K- and N-Ras)-expressing NIH/3T3 mouse embryonic fibroblast cell lines

  • Ras oncoproteins are small molecular weight GTPases known for their involvement in oncogenesis, which operate in a complex signaling network with multiple effectors. Approximately 25% of human tumors possess mutations in a member of this family. The Raf1/MEK/Erk1/2 pathway is one of the most intensively studied signaling mechanisms. Different levels of regulation account for the inactivation of MAP kinases by MAPK phosphatases in a cell type- and stimuli-dependent manner. In the present study, using three inducible Ras-expressing NIH/3T3 cell lines, we demonstrated that MKP3 upregulation requires the activation of the Erk1/2 pathway, which correlates with the shutdown of this pathway. We also demonstrated, by applying pharmacological inhibitors and effector mutants of Ras, that induction of MKP3 at the protein level is positively regulated by the oncogenic Ras/Raf/MEK/Erk1/2 signaling pathway.

Activation of Phosphatidylinositol 3-kinase(PI3K) is Required for Invasiveness and Motility in H-ras MCE10A Cells

  • 인간유방상피세포에서 H-ras가 침윤성과 세포 이동성을 유도한다는 것을 이 전연구에서 밝혔다. Phosphatidylinositol 3-kinase(PI3K)는 세포 이동성에서 중요한 역할을 하는 것으로 보고되고 있다. 본 연구에서 인간유방상피세포인 MCF10A에서 H-ras에 의해 유도된 침윤성에 PI3K가 어떠한 영향을 미치는지 살펴보고자 하였다. PI3K의 활성은 PI3K의 downstream molecule인 Akt의 인산화를 Western blot으로 확인하였다. Akt는 MCF10A, H-ras, N-ras MCF10A 세포에서 같은 정도로 발현되는 반면, 인산화된 Akt는 MCF10A 세포에 비해 H-ras MCF10A 세포와 N-ras MCF10A 세포에서 현저히 높게 나타났다. 이상의 결과로서 H-ras, N-ras 둘 다 PI3K를 활성화시키며, 침윤성과 세포이동성이 없는 N-ras MCF10A 세포에서도 PI3K가 활성화되었으므로, PI3K의 활성은 세포침윤성과 이동성을 유도하는데에 있어서 충분하지는 않음을 말해준다. PI3K의 저해제인 LY294002와 wortmannin을 세포에 처리하였을 때 세포침윤성과 이동성이 유의성 있게 감소되었다. 이상의 결과는 MCF10A 세포의 침윤성과 이동설에 있어서 PI3K의 활성이 충분하지는 않지만 반드시 필요하다는 것을 알 수 있었다.

Effect of preparation based on Paenibacillus ehimensis on the content of alkaloid lappakonitine in harvested rhizomes of Aconitum septentrionale

  • We performed an analysis of the effect of postharvest treatment of rhizomes of Aconitum septentrionale Koelle with the biological preparation of Bacispecin based on a cytokinin producing strain of Paenibacillus ehimensis IB-739 on the content of alkaloid lappaconitine in rhizomes. The total alkaloid content was assayed based on exhaustive extraction method. The lappaconitine content was measured by HPLC. The efficiency of the preparation was dependant on either the concentration or intensity of the growth processes in rhizomes. Both parameters varied depending on the stage of development of plants and the amount of precipitation. In the years with normal precipitation, concentrations of Bacispecin ranging from 5 to 10 g/l were found to be most effective for increasing lappaconitine content in rhizomes of A. septentrionale. Whereas, under local drought conditions, the concentration of preparation should be less than 2 g/l.

Chronic Treatment of Ethanol Inhibits Proliferation of Normal Fibroblasts, but Not Oncogenic ras-Transformed Cells

  • The adverse effects of ethanol on cell proliferation have been described for a variety of tissues and cells. In the present study, we investigated whether chronic ethanol intoxication impairs the cell proliferation and DNA synthesis induced by oncogenic $H-ras^{V12}$ - and $v-K-ras^{V12}$-transformed cells. Ethanol treatment inhibited the cell proliferation and the DNA synthesis of control parental fibroblasts in a time- and dose-dependent manner. In contrast, ethanol did not suppress the proliferation of either oncogenic $H-ras^{V12}$ - or $v-K-ras^{V12}$ -transformed fibroblasts. Microinjection of oncogenic $H-Ras^{V12}$ protein induces DNA synthesis and ethanol treatment did not interfere with the DNA synthesis. The antiproliferative toxicity of ethanol was rescued by antioxidants, such as N-acetylcysteine and 4-methlpyrazole. These results indicate that the antiproliferative action site of ethanol toxicity lies upstream or is independent of Ras and ethanol exerts its toxicity through a free radical formation.

  • The signal transduction pathways through the RAS gene product and adenyl cyclease play a critical role in regulation of the cell cycle in yeast, Saccharomyces cerevisiae. We examined the genetic relationship between the spt3 gene and ras/cAMP pathway. A mutation in the SPT3 gene suppressed cell cycle arrest at the G1 phase caused by either an inactivation of the RAS or CYR1 gene which encodes a yeast homologue of human ras proto-oncogene or adenyl cyclase, respectively. The phenotypes such as sporulation and heat shock resistancy, that resulted from a partial inactivation of the RAS or CYR1 genes, were also suppressed by the spt3 mutation. Expression of the SSA1 gene encoding one of th heat shock proteins (Hsp70) can be induced by heat shock or nitrogen starvation. Expression of this gene is derepressed in cry1-2 and spt3 mutants. The bcy 1 mutation repressed by the bcy1 mutation, but not in spt3 mutants. These results suggest that the SPT gene is involved in expression of genes that are affected by the RAS/cAMP pathway.

The Phosphorylation Status of Merlin Is Important for Regulating the Ras-ERK Pathway

  • The neurofibromatosis type2 (NF2) tumor suppressor gene product, merlin, is structurally related to the ezrin-radixin-moesin (ERM) family of proteins that anchor the actin cytoskeleton to specific membrane proteins and participate in cell signaling. However, the basis of the tumor suppressing activity of merlin is not well understood. Previously, we identified a role of merlin as an inhibitor of the Ras-ERK signaling pathway. Recent studies have suggested that phosphorylation of merlin, as of other ERM proteins, may regulate its function. To determine whether phosphorylation of merlin affects its suppression of Ras-ERK signaling, we generated plasmids expressing full-length merlin with substitutions of serine 518, a potential phosphorylation site. A substitution that mimics constitutive phosphorylation (S518D) abrogated the ability of merlin to suppress effects of the Ras-ERK signaling pathway such as Ras-induced SRE transactivation, Elk-mediated SRE transactivation, Ras-induced ERK phosphorylation and Ras-induced focus formation. On the other hand, an S518A mutant, which mimics nonphosphorylated merlin, acted like wild type merlin. These observations show that mimicking merlin phosphorylation impairs not only growth suppression by merlin but also its inhibitory action on the Ras-ERK signaling pathway.

Oncogenic Ras downregulates mdr1b expression through generation of reactive oxygen species

  • T In the present study, we investigated the effect of oncogenic H-Ras on rat mdr1b expression in NIH3T3 cells. The constitutive expression of H-RasV12 was found to downregulate the mdr1b promoter activity and mdr1b mRNA expression. The doxorubicin-induced mdr1b promoter activity of the H-RasV12 expressing NIH3T3 cells was markedly lower than that of control NIH3T3 cells. Additionally, there is a positive correlation between the level of H-RasV12 expression and a sensitivity to doxorubicin toxicity. To examine the detailed mechanism of H-RasV12-mediated down-regulation of mdr1b expression, antioxidant N-acetylcysteine (NAC) and NADPH oxidase inhibitor diphenylene iodonium (DPI) were used. Pretreating cells with either NAC or DPI significantly enhanced the oncogenic H-Ras-mediated down-regulation of mdr1b expression and markedly prevented doxorubicin-induced cell death. Moreover, NAC and DPI treatment led to a decrease in ERK activity, and the ERK inhibitors PD98059 or U0126 enhanced the mdr1b-Luc activity of H-RasV12-NIH3T3 and reduced doxorubicin-induced apoptosis. These data suggest that RasV12 expression could downregulate mdr1b expression through intracellular reactive oxygen species (ROS) production, and ERK activation induced by ROS, is at least in part, contributed to the downregulation of mdr1b expression.

리듬청각자극(RAS)을 사용한 소뇌 별아교세포종(CA) 환아의 보행훈련 사례 연구 (Case Study of Gait Training Using Rhythmic Auditory Stimulation(RAS) for a Pediatric Patient with Cerebellar Astrocytomas)

  • 본 연구는 소뇌 별아교세포종 발병으로 인해 소뇌 수술을 받은 12세 환아의 보행 기능 개선을 위해 RAS 보행훈련을 실시하여 보행인자의 변화를 관찰한 사례연구이다. 객관적 분석을 위해 VICON 370 Motion Analysis System을 이용한 동작분석을 RAS 보행 훈련 사전 사후에 실시하였다. 매 회기 30분씩 총 9회의 훈련을 실시한 결과 시공간적 지표 중 분속수, 보행속도, 보행시간, 보장, 활보장의 증가와 운동형상학적 지표의 움직임이 정상범주의 형상과 근접해가는 경향성을 나타내었다. 이는 RAS 보행훈련 후 보행의 효율성이 증진되는 것으로 해석될 수 있다. 이는 신체적 재활이 요구되는 여러 대상군에게 적용 가능성을 보여주었으며 효율성 검증을 위한 후속 연구가 요구된다.
