This paper describes a design process of end-milling cutters: solid model of the designed cutter is constructed along with computation of cutter geometry, and the wheel geometry as well as wheel positioning data f3r fabricating end-mills with required cutter geometry is calculated. In the process, the main idea is to use the cutting simulation method by which the machined shape of an end-milling cutter is obtained via Boolean operation between a given grinding wheel and a cylindrical workpiece (raw stock). Major design parameters of a cutter such as rake angle, inner radius can be verified by interrogating the section profile of its solid model. We studied relations between various dimensional parameters and proposed an iterative approach to obtain the required geometry of a grinding wheel and the CL data for machining an end-milling cutter satisfying the design parameters. This research has been implemented on a commercial CAD system by use of the API function programming, and is currently used by a tool maker in Korea. It can eliminate producing a physical prototype during the design stage, and it can be used for virtual cutting test and analysis as well.

Functional gene studies in the cultivated white button mushroom Agaricus bisporus have been constrained by the absence of effective gene-silencing tools. Using two endogenous genes from A. bisporus, we have tested the utility of dsRNA hairpin constructs to mediate downregulation of specific genes. Hairpin constructs for genes encoding orotidine 5'-monophosphate decarboxylase (URA3) and carboxin resistance (CBX) were introduced into A. bisporus using Agrobacteriummediated transfection. Although predicted changes in phenotype were not observed in vitro, quantitative-PCR analyses indicated unambiguously that transcripts in several transformants were substantially reduced compared with the non-transformed controls. Interestingly, some hairpin transformants exhibited increased transcription of target genes. Our observations show that hairpin transgenic sequences can mediate downregulation of A. bisporus endogenous genes and that the technology has the potential to expedite functional genomics of the mushroom.

Oriented Strand Board (OSB) is an important engineered wood product used in housing construction which has a lower environmental impact or "carbon footprint." In this paper, reliability and statistical tools are applied to gain insights on the strand thickness of OSB panels. An OSB panel consists of several hundred wood strands that are resinated and pressed. The variability of OSB strand thickness for six manufacturers in the Eastern United States is examined as a whole, as well as individually. Little research exists on OSB strand thickness across mills even though strand thickness variability has been documented in laboratory experiments to greatly influence the dimensional stability of OSB panels. Our aims are to quantify and characterize strand thickness, plus apply reliability techniques, such as Kaplan-Meier curves, to characterize the probability of strand thickness. We further explore graphically and statistically the thickness of the strands.
This research was performed to develop the simple techniques to predict the copper concentration in alkaline copper quat (ACQ), copper azole (CUAZ), and bis-(N-cyclohexyl-diazeniumdioxy)-copper (CB-HDO) solutions. Two simple methods measuring the color due to copper compounds were evaluated by using a spectrophotometer. One is to directly measure the color of the preservative solutions. The other is to measure the color developed on the surface of a treated sample with the preservatives. The $L^*$ of the measured color values appeared to be the most sensitive to the change of copper concentration. The $a^*$ values of the preservative solutions tended to be decreased at above a certain concentration condition, and the $b^*$ values showed no trend with the concentration of copper compounds in preservative solutions. The surface color of the treated samples were changed from bluish to greenish as time passed. Both methods showed the high $R^2$ values of the regression models determined by using the lightness, which suggested that the methods might be applicable in preservative-treatment mills for the easy and fast prediction of the copper concentration.
Effects of acid and ${\alpha}$-amylase on resistant starches including retrograded RS3 and cross-linked RS4 prepared from normal maize starch were investigated. Acid and ${\alpha}$-amylase hydrolytic patterns of RS3 were similar, while those of native starch and RS4 differed. Acid hydrolysis rate of RS3 was markedly higher at initial stage, then slowly decreased up to 20 days, whereas that of RS4 increased continuously. The sizes of acid- and ${\alpha}$-amylase-treated RS3 residues decreased, but those of RS4 remained unchanged. X-ray patterns of all treated residues did not change; however, the peak intensities increased. Swelling power of RS3 increased to 150% at $95^{\circ}C$, whereas that of RS4 differed depending on the treatment condition. Swelling power of acid-treated RS4 residue increased markedly, but that of ${\alpha}$-amylase-treated one remained constant. Gel filtration chromatography profiles of untreated RS3 and RS4 residues were similar, whereas that of acid-treated RS4 residue was different from them. RS showed different hydrolytic behavior by acid and ${\alpha}$-amylase depending on the type, and susceptibility of RS3 was higher than that of RS4.
Virgin bleached kraft pulp, CPO (computer printout) and white ledgers are main raw materials used in tissue mills. The utilization rate of recycled fibers and virgin pulp in South Korea tissue industry are 90% and 10%, respectively. To improve brightness of printing grades the use of 'fluorescent whitening agents (FWAs)' or 'optical brightening agents (OBAs)' has been increased. When recycling these papers for tissue production, it is unavoidable that FWAs contained in recycled papers flow into tissue production lines and remain in the products. And this draws great attention from the public. This study was carried out to develop a technology for the removal of fluorescent whitening agents from recycled fibers. Enzymatic removal of FWAs was evaluated as a method to remove FWAs from the recycled fiber. The ${\alpha}-amylase$ that degrades starched used for surface sizing of fine papers and contained substantial condition is needed to prevent the re-adsorption of FWAs on fibers. The temperature of pulp suspension was another important factor affecting on FWA removal. The higher the temperature, the greater the efficiency of removing FWAs was obtained. Optimum pH and temperature for the effective removal of FWAs were suggested to be pH 8.5 and $65^{\circ}C$, respectively. The enzymatic removal of FWAs showed a great synergistic effect when proper control in pH and temperature was made.
Hydrolysis of biomass for the production of fermentable sugar can be improved by the addition of surfactants. In pulp and paper mills, lignin, which is a by-product of the pulping process, can be utilized as a fine chemical. In the hydrolysis process, lignin is one of the major inhibitors of the enzymatic breakdown cellulose into sugar monomer. Therefore, the conversion of lignin into a biosurfactant offers the opportunity to solve the waste problem and improve hydrolysis efficiency. In this study, lignin derivatives, a biosurfactant, was applied to enzymatic hydrolysis of various lignocellulosic biomass. This Biosurfactant can be prepared by reacting lignin with a hydrophilic polymer such as polyethylene glycol diglycidylethers (PEDGE). In this study, the effect of biosurfactants on the enzymatic hydrolysis of pretreated sweet sorghum bagasse (SSB), oil palm empty fruit bunch, and sugarcane trash with different lignin contents was investigated. The results show that lignin derivatives improve the enzymatic hydrolysis of the pretreated biomass with low lignin content, however, it has less influence on the enzymatic hydrolysis of other pretreated biomass with lignin content higher than 10% (w/w). The use of biosurfactant on SSB kraft pulp can increase the sugar yield from 45.57% to 81.49%.
A study was undertaken to examine the effect of the addition of job's tears(Coixlachryma-jobi L.) chungkukjang(soybeanfermented food) and wheat bran powder on quality attributes of pound cakes. Job's tears chungkukjang and wheat bran powder was added to the batter at a ratio of 10, 12.5 and 15% respectively. The antioxidant activity was estimated by DPPH free radical scavening activity, the total phenolic compounds content and flavonoid content in job's tears chungkukjang & wheat bran powder and pounds cakes. Antioxidative activity was highly correlated with the total phenolic and total flavonoids contents of job's tears chungkukjang and wheat bran pound cakes respectively(r=0.9671, r=0.8335). The quality characteristics of job's tears chungkukjang powder and wheat bran powder pound cakes were estimated in terms of the specific load volume, hardness, hunter value of crumb and crust and sensory quality of cakes. The effects of monensin, Tween 80 and exogenous fibrolytic enzymes on ruminal fermentation and animal performance were studied in vitro and in vivo. In Expt 1, the effects of the surfactant Tween 80 (0.2% wt/wt, DM basis) on ruminal fermentation of alfalfa, corn and orchardgrass silages were investigated using in vitro gas production techniques. Tween 80 did not affect (p>0.05) cumulative gas production at 24 h, but it reduced (p<0.05) the lag in fermentation of all three silages. With corn silage and orchardgrass silage, gas production rates and concentrations of total volatile fatty acids (VFA) were increased (p<0.05) by Tween 80; with alfalfa silage, they were reduced (p<0.05). Tween 80 increased (p<0.05) the proportion of propionate in total VFA, and reduced (p<0.05) acetate to propionate ratios (A:P) with all three silages. In Expt 2, exogenous fibrolytic enzymes (E; at 0, 37.5 or 75 g/tonne DM), monensin (M; at 0 or 25 ppm and Tween 80 (T; at 0 or 2 L/tonne DM) were added alone or in combination to backgrounding and finishing diets fed to 320 crossbred steers in a feeding trial with a $3{\times}2{\times}$2 factorial arrangement of treatments. The backgrounding and finishing diets contained barley grain and barley silage in ratios of 57.8:42.2 and 93.5:6.5 (DM basis), respectively. Added alone, none of the additives affected DM intake (p>0.1) in the backgrounding or in the finishing period, but interactive $M{\times}T$ effects were observed in the finishing period (p=0.02) and overall (p=0.04). In the finishing period, T without M tended to reduce DM intake (p=0.11), but T with M increased (p=0.05) DM intake. Monensin increased average daily gain (ADG) during backgrounding (p=0.07) and finishing (p=0.01), and this ionophore also improved overall feed efficiency (p=0.02). Warm carcass weight was increased (p<0.001) by M, but dressing percentage was reduced (p=0.07). In the backgrounding period, T increased ADG by 7% (p=0.06). Enzymes increased (p=0.07) ADG by 5 and 6% (low and high application rates, respectively) during backgrounding, but did not affect (p>0.10) ADG during finishing, or overall feed efficiency. Whereas T enhanced the positive effects of M on ADG during backgrounding (p=0.04) and overall (p=0.05), it had no impact (p>0.1) on the effects of E. Interactions between M and T suggest that the surfactant may have potential for enhancing the positive effects of monensin on beef production, but this requires further research.
Three hundred and seventy-three steers (approximately 7 mo of age and $247{\pm}19.4\;kg$) were utilized to determine the effects of trace mineral (TM) source and growth implants on trace mineral status. Steers were blocked by ranch, post-weaning treatment within ranch, stratified by initial body weight, and randomly assigned to one of 36 pens (9-12 head/pen). Treatment consisted of: I) control (no supplemental Cu, Zn, Mn, and Co), ii) inorganic trace minerals, and iii) organic trace minerals. Six pens of steers per treatment received a growth implant at the beginning of the experiment and were re-implanted during the finishing phase. The remaining steers received no growth implants. Steers were fed a corn silage-based growing diet for 56 d then were gradually switched to a high concentrate finishing diet. Treatments during the finishing phase consisted of: i) control (no supplemental Zn); ii) inorganic Zn (30 mg of Zn/kg DM from $ZnSO_4$); and iii) organic Zn (iso-amounts of organic Zn). By the end of the growing and finishing phases, implanted steers had greater (p<0.01) plasma Cu concentrations than non-implanted steers. During the growing phase, liver Cu concentrations (p<0.01) and plasma Zn concentrations (p<0.02) were greater in steers supplemented with TM compared to control steers. Steers supplemented with inorganic minerals had greater liver Cu concentrations than steers supplemented with organic minerals at the beginning (p<0.01) and end (p = 0.02) of the growing phase. During both the growing (p = 0.02) and finishing phases (p = 0.05), nonimplanted control steers had greater plasma Cu concentrations than non-implanted steers supplemented with TM, whereas, implanted control steers had similar plasma Cu concentrations than implanted steers supplemented with TM. Non-implanted steers that received inorganic TM had lower plasma Cu concentrations (p = 0.03) during the growing phase and ceruloplasmin activity (p<0.04) during the finishing phase than non-implanted steers that received organic TM, whereas, implanted steers supplemented with either organic or inorganic TM had similar plasma Cu concentrations.
