Expression of Human Thrombopoietin in Insect Cells by Polyhedrin-gp64 Dual Promoter-Based Baculovirus Vector System

  • A new baculovirus transfer vector (pPGP404) was constructed to increase the expression level of human thrombopoietin (hTPO) in insect cells. In pPGP404, hTPO was cloned next to the AcNPV polyhedrin-gp64 dual promoter and the leader sequence of hTPO was substituted with that of gp64. A recombinant baculovirus, AcPGP404, was constructed by using pPGP404 as a transfer vector. hTPO was expressed in AcPGP404-infected TN5 cells and it was observed that the expression levels of hTPO in TN5 cells increased three-fold ($6.0 {\mu}g/ml^{-1}$) compared to the level expressed under the control of the polyhedrin single promoter. These results indicate that the polyhedrin-gp64 dual promoter system would be useful for expression in large quantities of recombinant proteins in insect cells.

대체 프레온 및 자연 냉매를 이용하는 이원 냉동시스템의 성능 예측 (Prediction on Performance of Cascade Refrigeration System using Alternative Freon and Natural Refrigerants)

  • 본 연구에서는 이원냉동시스템에서 사용되던 저온용 규제프레온냉매 R13 및 고온용 규제프레온냉매 R503의 대체냉매로서 프레온혼합냉매 및 친환경적 자연냉매를 사용하는 이원냉동시스템의 성능특성을 규명하고자 하였다. 이를 위해 일반적인 이원냉동시스템의 운전범위에서 과냉각도, 과열도, 응축온도, 증발온도, 캐스케이드 열교환기에서의 온도변화를 고려하였다. 고온시스템 및 저온시스템의 작동유체의 종류에 상관없이 이원냉동시스템의 COP는 캐스케이드의 온도영향을 받고 있으며, 또한 과냉각도가 증가할수록 COP는 높아지지만, 과열도의 영향은 크게 받지 않았다. 따라서 본 연구에서 고온용 시스템 및 저온용 시스템의 작동유체로 대체 프레온혼합냉매 및 자연냉매의 냉매조합 중에서 (R23/R290), (R23/R600), (R23/R600a), (R23/R717), (R744/R404A) 냉매를 사용하는 이원냉동시스템의 COP는 저온측에 R23, 고온측에 R22를 사용하는 시스템에 비해 20 ~ 36% 높게 나타났다.

Bracket bonding to polymethylmethacrylate-based materials for computer-aided design/manufacture of temporary restorations: Influence of mechanical treatment and chemical treatment with universal adhesives

  • Objective: To assess shear bond strength and failure mode (Adhesive Remnant Index, ARI) of orthodontic brackets bonded to polymethylmethacrylate (PMMA) blocks for computer-aided design/manufacture (CAD/CAM) fabrication of temporary restorations, following substrate chemical or mechanical treatment. Methods: Two types of PMMA blocks were tested: $CAD-Temp^{(R)}$ (VITA) and $Telio^{(R)}$ CAD (Ivoclar-Vivadent). The substrate was roughened with 320-grit sandpaper, simulating a fine-grit diamond bur. Two universal adhesives, Scotchbond Universal Adhesive (SU) and Assure Plus (AP), and a conventional adhesive, Transbond XT Primer (XTP; control), were used in combination with Transbond XT Paste to bond the brackets. Six experimental groups were formed: (1) $CAD-Temp^{(R)}/SU$; (2) $CAD-Temp^{(R)}/AP$; (3) $CAD-Temp^{(R)}/XTP$; (4) $Telio^{(R)}$ CAD/SU; (5) $Telio^{(R)}$ CAD/AP; (6) $Telio^{(R)}$ CAD/XTP. Shear bond strength and ARI were assessed. On 1 extra block for each PMMA-based material surfaces were roughened with 180-grit sandpaper, simulating a normal/medium-grit ($100{\mu}m$) diamond bur, and brackets were bonded. Shear bond strengths and ARI scores were compared with those of groups 3, 6. Results: On $CAD-Temp^{(R)}$ significantly higher bracket bond strengths than on $Telio^{(R)}$ CAD were recorded. With XTP significantly lower levels of adhesion were reached than using SU or AP. Roughening with a coarser bur resulted in a significant increase in adhesion. Conclusions: Bracket bonding to CAD/CAM PMMA can be promoted by grinding the substrate with a normal/medium-grit bur or by coating the intact surface with universal adhesives. With appropriate pretreatments, bracket adhesion to CAD/CAM PMMA temporary restorations can be enhanced to clinically satisfactory levels.

Flavone 유도체들의 Tyrosinase 저해활성화 반응에서 Hydroxyl 치환기들의 역할 (The Roles of Hydroxyl Substituents in Tyrosinase Inhibitory Activation of Flavone Analogues)

  • Tyrosinase 저해활성화 반응에 대한 polyhydroxy 치환된 flavone 유도체(1-25) 중, hydroxyl-치환기($R_1-R_9$)들의 역할을 이해하기 위하여 Free-Wilson 분석과 tyrosinase (PDB ID: Deoxyform (2ZMX) 및 Oxy-form; 1WX2)의 활성화 지점에 대한 분자도킹이 연구되었다. Free-Wilson 분석으로부터 $R_1-R_9$ 치환기중에서 $R_1$=hydroxyl 치환기가 tyrosinase 저해활성에 가장 큰 영향을 미치고 있음을 알았다. 기질분자의 hydroxyl 치환기들과 tyrosinase의 반응점 내 아미노산 잔기들 사이의 수소결합들은 안정한 기질-수용체 착 화합물을 형성하는데 기여하였다. 특히, 수소결합성에 기초한 비경쟁적 저해활성화 반응은 기질분자의 hydroxyl 치환기들과 tyrosinase의 반응점 내 peroxide 산소원자(Per404) 사이의 수소결합을 경유하여 일어날 것임을 제안하였다.

Performance of Alternative Refrigerants in Low Temperature Chillers

  • This paper is about the performance of alternative refrigerants for low temperature and transport refrigeration chillers. A breadboard refrigeration chiller was constructed with counterflow heat exchangers. R502, its transitional alternatives of R402A and R402B, and long-term alternatives of R404A and R507 were tested in an attempt to compare the performance of each refrigerant against R502. Measurements were conducted at two condensing temperatures of 43.3$^{\circ}C$ and 52.$0^{\circ}C$ and the evaporating temperature was varied over a range from -$25^{\circ}C$ to -5$^{\circ}C$. The evaporator superheat and condenser subcooling were maintained constant at about 5$^{\circ}C$ for all tests. Test results showed that all alternative fluids tested in this work can be used as 'drop-in fluids' to replace R502 without any major problem. It is also found that in the long run HFC alternatives are to be used due to their favorable environmental characteristics and better performance.

간호대학생의 자아탄력성, 간호전문직 자아개념, 전공만족도가 대학생활적응에 미치는 영향 (Effect of Self-resilience and Professional Self-concept, Major satisfaction on Nursing Student's Adjustment to college life)

  • 본 연구는 간호대학에 재학하는 학생을 대상으로 자아탄력성, 간호전문직 자아개념, 전공만족도가 대학생활적응에 미치는 영향을 파악하여 대학생활적응을 돕는 기초자료를 제공하고자 시행하였다. 연구대상자는 광주, 전남에 소재하는 4년제 간호대학 2개교에서 간호대학생 310명을 대상으로 자아탄력성, 간호전문직 자아개념, 전공만족도, 대학생활적응 정도를 설문지로 측정하였다. 자료수집기간은 2016년 4월 1일부터 4월 15일까지이었다. 설문에 응답한 310명의 자료를 SPSS WIN 20.0을 이용하여 평균, 표준편차, t-test, ANOVA, Scheffe's test, Pearson' correlation coefficients, multiple regression으로 분석하였다. 연구 결과, 간호대학생의 자아탄력성은 4점 만점에 3.80점, 간호전문직 자아개념은 4점 만점에 2.81점, 전공만족도는 5점 만점에 3.86점, 대학생활적응은 5점 만점에 2.74점이었으며, 대학생활적응과 자아탄력성(r=.418, p<.01), 간호전문직 자아개념(r=.404, p<.001), 전공만족도(r=.455, p<.001)는 각각 유의한 양의 상관관계가 있었다. 또한 대상자의 대학생활적응에 영향을 미치는 요인은 자아탄력성(${\beta}=.252$, p<.001), 전공만족도(${\beta}=.309$, p<.001)로 나타났으며, 모델의 설명력은 27.2%였다. 본 연구를 통해 대학생활적응에 영향을 미치는 다양한 요인들을 파악하고 대학생활적응을 높일 수 있는 프로그램의 개발이 필요하다.

대체냉매의 2중관 응축기 열 및 물질전달과 성능평가 (Heat and Mass Transfer Characteristics and Performance Evaluation of a Double-Tube Condenser for an Alternative Refrigerant)

  • This paper deals with heat and mass transfer characteristics and performance evaluation of a counter flow double-tube condenser for a multi-component refrigerant mixture. The local heat and mass transfer characteristics of ternary zeotropic refrigerant mixtures composed of HFC32/HFC125/HFC134a are evaluated for a counter flow double-tube condenser cooled by water. Then, the local values of vapor quality, thermodynamic states at bulk vapor, vapor-liquid interface and bulk liquid, heat flux and condensation mass flux are obtained. The heat exchange performance for ternary zeotropic refrigerant mixtures composed of HFC32/HFC125/HFC134a on the total pressure drop and the heat transfer characteristics are also compared with those for R404A, R410A, R502, R22, R32, Rl23 and R134a.
