A study on the safety improvement of above ground membrane LNG storage tank (상지상식 멤브레인 액화천연가스 저장탱크의 안전성 향상 방안)

  • RMembrane LNG storage tanks have been recently investigated to replace full-containment LNG storage tanks because of safety and cost aspects. Quantitative Risk Analysis (QRA) and Finite Element Method (FEM) were used to evaluate safety of membrane LNG storage tanks. In this study, structural safety evaluation results via FEM analysis showed that both membrane type and full-containment type cryogenic LNG storage tanks with 140,000 $m^3$ capacity were equivalently safe in terms of strength safety and leakage safety of a storage tank system. Also, Fault Tree Analysis (FTA) was used to improve the safety of membrane LNG storage tanks and membrane LNG tanks were modified by adding three safety equipments: impact absorber structure for the low part of the membrane, the secondary barrier to diminish the thermal stress of the corner part of the outer tank, and a pump catcher in case of falling of a pump. Consequently, the safety of the modified membrane LNG storage tanks were proved to be equivalent to that of full-containment LNG storage tanks.

Natural hybridization of Iris species in Mt. Palgong-san, Korea (팔공산 금붓꽃 계열의 자연 잡종 현상)

  • Series Chinensis, Genus Iris, endemic to the far regions of East Asia, consists of four species and related varieties. This series is divided into two major groups (I. rossii and I. minutiaurea complex). In this study, the ITS region and matK gene sequences within nuclear ribosomal DNA and plastid DNA were analyzed in order to investigate the phylogenetic relationships among the I. minutiaurea complex (I. minutiaurea, I. odaesanensis, and I. koreana) and the taxonomic identities of a putative hybrid in Mt. Palgong. In the internal transcribed spacer (ITS1, 5.8S, and ITS2) region, a total of 106 cloned genomic sequences from three taxa were obtained to study the intragenomic polymorphisms of the ITS regions. Three taxa revealed high levels of intragenomic polymorphisms, indicative of incomplete nrDNA concerted evolution. This incomplete ITS concerted evolution in the series Chinensis may be linked to the recent species divergence and frequent interspecies hybridization of the series Chinensis. In the matK gene, three taxa were fairly separated by eleven variable sites. In eight individuals collected on Mt. Palgong, putative hybrids between I. odaesanensis and I. minutiaurea were clustered in the I. minutiaurea clade in the NJ (neighbor-joining) tree based on the matK gene. However, in the ITS tree, some of them were clustered in the I. odaesanensis clade and others were clustered in the I. minutiaurea clade. Therefore, the individuals on Mt. Palgong were formed by the hybridization between two taxa (I. odaesanensis and I. minutiaurea) and not through the lineage of I. koreana.

Combined Image Retrieval System using Clustering and Condensation Method (클러스터링과 차원축약 기법을 통합한 영상 검색 시스템)

  • This paper proposes the combined image retrieval system that gives the same relevance as exhaustive search method while its performance can be considerably improved. This system is combined with two different retrieval methods and each gives the same results that full exhaustive search method does. Both of them are two-stage method. One uses condensation of feature vectors, and the other uses binary-tree clustering. These two methods extract the candidate images that always include correct answers at the first stage, and then filter out the incorrect images at the second stage. Inasmuch as these methods use equal algorithm, they can get the same result as full exhaustive search. The first method condenses the dimension of feature vectors, and it uses these condensed feature vectors to compute similarity of query and images in database. It can be found that there is an optimal condensation ratio which minimizes the overall retrieval time. The optimal ratio is applied to first stage of this method. Binary-tree clustering method, searching with recursive 2-means clustering, classifies each cluster dynamically with the same radius. For preserving relevance, its range of query has to be compensated at first stage. After candidate clusters were selected, final results are retrieved by computing similarities again at second stage. The proposed method is combined with above two methods. Because they are not dependent on each other, combined retrieval system can make a remarkable progress in performance.

The Growth and EPA Synthesis of Shewanella oneidensis MR-1 and Expectation of EPA Biosynthetic Pathway

  • Shewanella oneidensis MR-1 has the ability to inhale certain metals and chemical compounds and exhale these materials in an altered state; as a result, this microorganism has been widely applied in bioremediation protocols. However, the relevant characteristics of cell growth and biosynthesis of PuFAs have yet to be thoroughly investigated. Therefore, in this study, we have attempted to characterize the growth and fatty acid profiles of S. oneidensis MR-1 under a variety of temperature conditions. The fastest growth of S. oneidensis MR-1 was observed at $30^{\circ}C$, with a specific growth rate and doubling time of $0.6885h^{-1}\;and\;1.007 h$. The maximum cell mass of this microorganism was elicited at a temperature of $4^{\circ}C$. The eicosapentaenoic acid (EPA) synthesis of S. oneidensis MR-1 was evaluated under these different culture temperatures. S. oneidensis MR-1 was found not to synthesize EPA at temperatures in excess of $30^{\circ}C$, but was shown to synthesize EPA at temperatures below $30^{\circ}C$. The EPA content was found to increase with decreases in temperature. We then evaluated the EPA biosynthetic pathway, using a phylogenetic tree predicted on 16s rRNA sequences, and the homology of ORFs between S. oneidensis MR-1 and Shewanella putrefaciens SCRC-2738, which is known to harbor a polyketide synthase (PKS)-like module. The phylogenetic tree revealed that MR-1 was very closely related to both Moritella sp., which is known to synthesize DHA via a PKS-like pathway, and S. putrefaciens, which has been reported to synthesize EPA via an identical pathway. The homology between the PKS-like module of S. putrefaciens SCRC-2738 and the entire genome of S. oneidensis MR-1 was also analyzed, in order to mine the genes associated with the PKS-like pathway in S. oneidensis MR-1. A putative PKS-like module for EPA biosynthesis was verified by this analysis, and was also corroborated by the experimental finding that S. oneidensis MR-1 was able to synthesize EPA without the expression of $dihomo-{\gamma}-linoleic$ acid (DGLA) and arachidonic acid (AA) formed during EPA synthesis via the FAS pathway.

Stand Structure and Biomass in Wild Populations of Camellia sinensis (야생(野生) 차(茶)나무 집단(集團)의 임분구조(林分構造)와 물질현존량(物質現存量))

  • Twelve Camellia sinensis populations were studied to investigate the biomass distribution related to stand structure. With increasing mean age of the population, mean root collar diameter increased, while mean height, density and root collar area had no correlation with the age. The result of dimension analysis after cutting the sample trees showed that dry weights of stems +branches and root were exponentially increased and dry weights of current leaves and twigs were linearly increased with increasing root collar diameter. However, the dry weight of older leaves was not related to the root collar diameter. The range of total biomass was 1,162~11,474kg/ha and the range of current leaf biomass was 165~1,341kg/ha. The range of T/R ratio was 1.5~2.8. The biomass of stems+branches and root were significantly correlated with the root collar area of the population and were not significantly correlated with the mean age, mean root collar diameter and density of the population. The biomass of current leaves and twigs were significantly correlated with the root collar area and density of the population.

Breeding of a New Japanese Apricot (Prunus mume Siebold et Zucc.) Cultivar 'Okjoo' with High Yields (다수성 매실 품종 '옥주')

  • Japanese apricot (Prunus mume Siebold and Zucc.) is a deciduous tree of the family Rosaceae, and it has long been used as a folk remedy for cough and dyspepsia. A new cultivar 'Okjoo' was developed from a cross between 'Gyokuei' and 'Rinsyu' carried out at the National Institute of Horticultural & Herbal Science in 1993. It w as s elected for good shape, large size and high yield capacity in 2006, and then it was granted official patent No. 4556 in 2013. It blooms 4 days and 2 days earlier than 'Gyokuei' and 'Rinsyu', respectively. Its flower petal color is pink, and the pollen amount is negligible. Its S-genotype, determined using Polymerase Chain Reaction with a S-RNase gene-specific primer pair, is $S_3S_6$. The average optimum harvest time of 'Okjoo' is late June. The fruit is round in shape and its suture is shallow. Average fruit weight is 18.5 g, and it contains total soluble solids $7.66^{\circ}Brix$ and titratable acidity at 4.81%. Fruit skin color is green. Sometimes only the light side of the fruits seems to develop blush. The incidence levels of scab (Cladosporium carpophilum Thumen) and bacterial shot hole (Xanthomonas arboricola pv. Pruni) are quite low. Consequently, 'Okjoo' seems to be a promising new cultivar for Japanese apricot growers.

Ecological Studios on the Bark Beetles on Plum and Apricot (자두와 살구나무에 기생하는 나무좀의 생태에 관한 연구)

  • The ecological studies were conducted to identify some bark-beetles on apricot and plum trees in Jeonnam province. Harmful bark-beetles caught from apricot and plum trees were identified as Scolytus seulensis. Xyleborus atratus., X. germanus, X. rubricollis and X. saxeseni. The seoul barkbeetle emerged during the period from early May to late October $1980\~1981$, it happens twice in a year, with the peak July 10 and August 25 in 1950, July 15 and August 20 in 1981. A daily peak emerged for adults was at 11 AM to 1 PM. Distribution of holes on the apricot stems varied; more than 100 holes per meter in Henderson (Apricot) and Y-49057 (Apricot), 12 holes per meter in Derbyroyal (Apricot). Comparatively more holes were found in the European variety, and the cardinal distribution was in the order of east, south, west and north. The longer the longth of the mother-gallery, the greater number of egg-gallery found. Optimum temperature for mating was $27^{\circ}C$. The time for mating was longer during May-June than July-August. The fruit weight of damaged plum tree reached 12.25g around four weeks after flowering; where as that of normal tree increased up to 63.5g at harvest time. The natural enemy of the seoul bark beetle was Eurytoma sp. They were found as mature larva or pupa and they emerged from early May to the middle of June. Adult longevity was about a week.

Phylogeny of Korean Viola based on ITS sequences (ITS 염기서열에 의한 한국산 제비꽃속(Viola)의 계통 유연관계)

  • Molecular phylogenetic studies were conducted to evaluate interspecific relationships in 40 populations of Viola including 35 Korean taxa, four Japanese populations and one outgroup using nuclear ribosomal ITS sequences. The phylogenetic analyses were conducted using parsimony and neighbor-joining methods. Subsection Trigonocarpae of section Nomimium appeared as the most basal clade within the Korean Viola. Section Dischidium and Chamaemelanium was monophyletidbootstrap 100%) and placed between subsect. Trigonocarpae and three other subsections of sect. Nomimium. Sect. Nomimium was paraphyletic. Although each subsectional grouping was in accordance with previous infrageneric classification based on morphological characters, yet discordance remained at the series level. Two evolutionary trends observed in the ITS tree were as follows. First, subsect. Trigonocarpae(x=10) was derived from the outgroup(x=6); Second, subsects. Bilobatae and Vaginatae(x=10 or 12), and subsect. Patellares(x=12) of sect. Nomimium were originated from sects. Dischidium and Chamaemelanium(x=6). Viola albida complex including three very closely related taxa was recognized as independent group within subsect. Patellares in parsimony tree. This result suggested that they should be treated as a taxa in series Pinnatae. Phylogenetic position of a putative hybrid species, Viola woosanensis was not supported with previous morphological hypothesis.

Variation of Seasonal Groundwater Recharge Analyzed Using Landsat-8 OLI Data and a CART Algorithm (CART알고리즘과 Landsat-8 위성영상 분석을 통한 계절별 지하수함양량 변화)

  • Groundwater recharge rates vary widely by location and with time. They are difficult to measure directly and are thus often estimated using simulations. This study employed frequency and regression analysis and a classification and regression tree (CART) algorithm in a machine learning method to estimate groundwater recharge. CART algorithms are considered for the distribution of precipitation by subbasin (PCP), geomorphological data, indices of the relationship between vegetation and landuse, and soil type. The considered geomorphological data were digital elevaion model (DEM), surface slope (SLOP), surface aspect (ASPT), and indices were the perpendicular vegetation index (PVI), normalized difference vegetation index (NDVI), normalized difference tillage index (NDTI), normalized difference residue index (NDRI). The spatio-temperal distribution of groundwater recharge in the SWAT-MOD-FLOW program, was classified as group 4, run in R, sampled for random and a model trained its groundwater recharge was predicted by CART condidering modified PVI, NDVI, NDTI, NDRI, PCP, and geomorphological data. To assess inter-rater reliability for group 4 groundwater recharge, the Kappa coefficient and overall accuracy and confusion matrix using K-fold cross-validation were calculated. The model obtained a Kappa coefficient of 0.3-0.6 and an overall accuracy of 0.5-0.7, indicating that the proposed model for estimating groundwater recharge with respect to soil type and vegetation cover is quite reliable.

Antioxidant Activities of Processed Deoduck (Codonopsis lanceolata) Extracts (가공공정에 따른 더덕 추출물의 항산화 활성)

  • This study investigated the antioxidant activities of processed Deoduck (Codonopsis lanceolata) extracts treated through high-pressure extraction and steaming with fermentation. The antioxidant activities were determined for DPPH and ABTS radical-scavenging activity, SOD-like activity, ferric reducing antioxidant power (FRAP), and $Fe^{2+}$ chelating. Total phenolic and flavonoid contents were also measured. Among eight Deoduck extracts, the S5FDW extract had the highest total phenolic and flavonoid content, 73.9 mg GAE/g and 50.9 mg QUE/g, respectively. The S5FDW extract had the highest DPPH radical-scavenging activity (27%) at a 1.0 mg/mL concentration. The ABTS radical-scavenging activity was highest for S5FDW extract (82.1%) at a 10 mg/mL concentration. The HFDE extract showed the highest SOD-like activity (29.7%) at a 1.0 mg/mL concentration. FRAP was highest in S5FDW extract (140.8 ${\mu}M$) at a 1.0 mg/mL concentration. The DE extract showed the highest $Fe^{2+}$ chelating (46%) at a 1.0 mg/mL concentration. The phenolic and flavonoid contents significantly correlated with the antioxidant activity of several processed Deoduck extracts and was higher in the processed Deoduck extracts compared to the raw Deoduck extracts. Therefore, processing techniques can be useful methods for making Deoduck a more potent and natural antioxidant.
