In this study, we evaluated the anti-melanogenesis effects of Caffeoylserotonin (CaS) in B16 melanoma cells. Treatment with CaS reduced the melanin content and tyrosinase (TYR) activity in B16 melanoma cells in a dose-dependent manner. CaS inhibited the expression of melanogenesis-related proteins, including microphthalmia-associated transcription factor (MITF), TYR, and tyrosinase-related protein-1 (TRP-1), but not TRP-2. ${\alpha}$-MSH is known to interact with melanocortin 1 receptor (MC1R) thus activating adenylyl cyclase and increasing intracellular cyclic AMP (cAMP) levels. Furthermore, cAMP activates extracellular signal-regulated kinase 2 (ERK2) via phosphorylation, which phosphorylates MITF, thereby targeting the transcription factor to proteasomes for degradation. The CaS reduced intracellular cAMP levels to unstimulated levels and activated ERK phosphorylation within 30 min. The ERK inhibitor PD98059 abrogated the suppressive effect of CaS on ${\alpha}$-MSH-induced melanogenesis. Based on this study, the inhibitory effects of CaS on melanogenesis are derived from the downregulation of MITF signaling via the inhibition of intracellular cAMP levels, as well as acceleration of ERK activation.
Parathyroid hormone (PTH) is known to exert its effects on bone cells through the mediation of adenosine 3', 5'-monophosphate (cAMP). Orthodontic forces have also been shown to alter the cAMP content of paradental cells, particularly the alveolar bone osteoblasts. The objective of this experiment was to determine whether a combined orthodontic treatment-PTH administration regimen would have an additive effect on cAMP content in paradental cells in sites of periodontal ligament (PDL) tension. Seven groups of 4 one year old female cats each were treated for 1,3,6,12,24 h, 7 and 14 d by tipping one maxillary canine. PTH was administered twice daily, 30u/kg. Maxillary horizontal sections were stained immunohistochemically for cAMP and the degree of cellular staining intensity was determined microphotometrically as per cent light transmittance at 600nm. Alveolar bone osteoblasts, progenitor cells, PDL fibroblasts and cementoblasts in tenion sites were measured and the data were analyzed statistically by a mixed model analysis of variance. PTH administration increased the cAMP staining of nonorthodontically treated paradental cells in comparison to cells untreated by force or hormone. Cells in PDL tension sites of PTH-treated cats demonstrated significantly darker cAMP staining than cells in non-orthodontically-treated sites. Osteoblasts demonstrated the greatest response in terms of cAMP elevation, while in PDL fibroblasts orthodontic force did not increase cAMP levels above those measured in non-stretched hormonally-treated cells. These results demonstrate that PTH increases cAMP levels in paradental cells, particullarly in osteoblasts, and that the effects of PTH and orthodontic forces on paradental target cells may approach additivity.
혈소판 응집의 조절은 정상적인 지혈을 유지하는 데 중요하지만 비정상적이거나 과도한 혈소판 응집은 뇌졸중, 죽상동맥 경화증 및 혈전증과 같은 심혈관 질환에 기여할 수 있다. 따라서 혈소판 응집을 제어하거나 억제할 수 있는 물질을 식별하는 것은 이러한 상태의 예방 및 치료를 위한 유망한 접근 방식이다. Artemisia 또는 Scopolia 속 식물에서 추출한 artemisinin은 항암 및 알츠하이머병 연구와 같은 다양한 분야에서 가능성을 보여주었다. 그러나 artemisinin이 혈소판 활성화 및 혈전 형성에 영향을 미치는 구체적인 역할과 메커니즘은 아직 완전히 밝혀지지 않았다. 이 연구는 혈소판 활성화 및 혈전 형성에 대한 artemisinin의 효과를 조사하였다. 그 결과, cAMP 생성과 cAMP 의존성 kinase에 대한 기질인 VASP 및 IP3R의 인산화가 artemisinin에 의해 유의미하게 증가되었다. IP3R의 인산화는 조밀한 관형 시스템에서 정상적으로 동원되는 Ca2+를 억제하였고, VASP의 인산화는 αIIb/β3 혈소판 막 불활성화를 통한 fibrinogen 결합을 억제하였다. 마지막으로, artemisinin은 thrombin이 유발하는 혈전 형성을 농도의존적으로 억제하였다. 따라서 우리는 artemisinin이 혈소판 활성화의 효과적인 예방 및 치료제로 작용하여 비정상적인 혈소판 응집 및 혈전 형성으로 인해 유발되는 심혈관 질환의 개선에 기여할 수 있음을 제안한다.
Here we report essential features of the draft genome of an AmpC-β-lactamase-producing bacterial isolate obtained from farm tomatoes in South Africa. The isolate designated strain Tom1 featured a genome of 4950426 bp with a G+C% of 59.83. It was identified as Serratia marcescens by ribosomal multilocus sequence typing (rMLST), digital DNA-DNA hybridization (dDDH), average nucleotide identity (ANI), and phylogenetic analysis using reference genomes. Its genome encoded an AmpC-β-lactamase (blaSST-1), an efflux pump providing tetracycline resistance (tet(41)), and an aminoglycoside acetyltransferase (aac(6')-Ic). Additionally, genes encoding proteins involved in prodigiosin biosynthesis and associated with adherence, biofilm formation, virulence, and pathogenicity were detected.

The lutropin/choriogonadotropin receptor (LHR) is a member of the rhodopsin-like subfamily of G protein coupled receptor (GPCRs), that has been shown to mediate the internalization of its two naturally occurring agonist, lutropin and choriogonadotropin (CG). The clustered agonist-receptor complex is internalized by a dynamin-dependent pathway and traverses the endosomal compartment without agonist dissociation Dissociation of the agonist-receptor complex occurs in the lysosomes, where both the agonist and receptor are degrade. Recently, constitutively activating mutations of the receptor have been identified that are associated with familial male-precocious puberty (FMPP). A FMPP is a form of sexual precocious puberty in boys in which testosterone levels are elevated independent of changes in luteinizing hormone-releasing hormone and serum luteinizing hormone levels, We have now analyzed two naturally occurring, constitutively active mutants of the human LHR. These mutations were introduced into the rat LHR (rLHR) and are designated L435R and D556Y. Cells expressing rLHR-D556Y bind human choriogonadotropin (hCG) with normal affinity, exhibit a 25-fold increase in basal cAMP and respond to hCG with a normal increase in cAMP accumulation. Cells expressing rLHR-L435R also bind hCG with normal affinity, exhibit a 47-fold increase in basal cAMP, and do not respond to hCG with a further increase in cAMP accumulation. This mutation enhances the internalization of the free and agonist-occupied receptors ~2- and ~17- fold, respectively We conclude that the state of activation of the rLHR can modulate its basal and/or agonist-stimulated internalization. Since the internalization of hCG is involved in the termination of hCG actions, we suggest that the lack of responsiveness detected in cells expressing rLHR-L435R is due to the fast rate of internalization of the bound hCG. The finding that membranes expressing rLHR-L435R respond to hCG with an increase in adenylyl cyclase activity supports this suggestion. Autonomous Leydig cell activity in FMPP is caused by a constitutively activating LH/CGR.
The cyclic AMP receptor protein (CRP) homolog, GlxR, controls the expression of several genes involved in the regulation of diverse physiological processes in Corynebacterium glutamicum. In silico analysis has revealed the presence of glxR binding sites upstream of genes ptsG, adhA, and ald, encoding glucose-specific phosphotransferase system protein, alcohol dehydrogenase (ADH), and acetaldehyde dehydrogenase (ALDH), respectively. However, the involvement of the GlxR-cAMP complex on the expression of these genes has been explored only in vitro. In this study, the expressions of ptsG, adhA, and ald were analyzed in detail using an adenylate cyclase gene (cyaB) deletion mutant and glxR deletion mutant. The specific activities of ADH and ALDH were increased in both the mutants in glucose and glucose plus ethanol media, in contrast to the wild type. In accordance, the promoter activities of adhA and ald were derepressed in the cyaB mutant, indicating that glxR acts as a repressor of adhA. Similarly, both the mutants exhibited derepression of ptsG regardless of the carbon source. These results confirm the involvement of GlxR on the expression of important carbon metabolic genes; adhA, ald, and ptsG.
CRP (cAMP receptor protein)은 cAMP와 결합하여 cAMP-CRP 복합체를 형성하여 전사조절의 조절인자로서 작용한다. crp 유전자에 변이를 도입하여 cAMP의 비존재 상태에서 cAMP-CRP와 비슷한 기능을 가진 crp 유전자가 도입된 대장균 MK2001 (crp, cya::km)을 숙주로 사용하여 cAMP 혹은 cGMP의 비존재하에서도 mal 유전자의 발현을 촉진시키는 유전자 sfs (sugar fermentation stimulation) 수 종을 클로닝 하였다. 본 실험에서는 이미 밝혀진 nlp (Ner like protein) 유전자와 같이, sfs의 새로운 유전자를 탐색하여, 그 중 sfs4의 2126 bp 전 염기배열을 결정하고, 잠정적인 sfs4의 promoter 영역에는 CRP 단백질과의 결합 DNA 공통 염기배열(5' AAT TGTGA ACACCA TCACC CGT 3')이 존재함을 확인했다. lacZ 융합 유전자를 작성하여 TP2010R1 MK2001의 균주에서 cAMP를 첨가할 경우 각각 2.3배, 1.8배의 ${\beta}-galactosidase$ 활성이 증가하는 것으로 보아 sfs4는 cAMP-CRP에 의해 발현 조절을 받는 것으로 나타났다.
In this study, we prepared cordycepin-enriched (CE)-WIB801C, a n-butanol extract of Cordyceps militaris-hypha, and investigated the effect of CE-WIB801C on collagen-induced human platelet aggregation. CE-WIB801C dose-dependently inhibited collagen-induced platelet aggregation, and its $IC_{50}$ value was $175{\mu}g/ml$. CE-WIB801C increased cAMP level more than cGMP level, but inhibited collagen-elevated $[CA^{2+}]_i$ mobilization and thromboxane $A_2$ ($TXA_2$) production. cAMP-dependent protein kinase (A-kinase) inhibitor Rp-8-Br-cAMPS increased the CE-WIB801C-downregulated $[CA^{2+}]_i$ level in a dose dependent manner, and strongly inhibited CE-WIB801C-induced inositol 1, 4, 5-trisphosphate receptor ($IP_3R$) phosphorylation. These results suggest that the inhibition of $[CA^{2+}]_i$ mobilization by CE-WIB801C is resulted from the cAMP/A-kinase-dependent phosphorylation of $IP_3R$. CE-WIB801C suppressed $TXA_2$ production, but did not inhibit the activities of cyclooxygenase-1 (COX-1) and $TXA_2$ synthase (TXAS). These results suggest that the inhibition of $TXA_2$ production by WIB801C is not resulted from the direct inhibition of COX-1 and TXAS. In this study, we demonstrate that CE-WIB801C with cAMP-dependent $CA^{2+}$-antagonistic antiplatelet effects may have preventive or therapeutic potential for platelet aggregation-mediated diseases, such as thrombosis, myocardial infarction, atherosclerosis, and ischemic cerebrovascular disease.
혈소판의 적절한 활성화와 응집이 필요하지만 과도하거나 비정상적인 응집은 뇌졸중, 혈전증, 동맥경화증과 같은 심혈관 질환을 유발할 수 있다. 따라서 이러한 질병을 예방하고 치료하기 위해서는 혈소판 응집을 조절하거나 억제할 수 있는 물질을 찾는 것이 중요하다. 여러 연구에서 Panax 인삼의 특정 ginsenoside 화합물이 혈소판 응집을 억제할 수 있음이 알려져 있다. 이들 화합물 중 Panax ginseng의 Rk3 (G-Rk3)는 혈소판 응집 억제의 기전이 불확실 하기에 이를 밝히기 위한 연구가 필요하다. G-Rk3는 cAMP의 양을 강하게 증가시켰고 cAMP 의존성 kinase의 기질인 VASP 및 IP3R의 인산화를 유도했다. 또한, G-Rk3의 효과는 PI3K/Akt 인산화의 억제를 일으켜 세포 내 과립의 분비를 감소시켰다. 궁극적으로 G-Rk3는 혈소판 응집을 효과적으로 억제하였다. 따라서 우리는 과도한 혈소판 응집으로 인한 심혈관 질환의 예방 또는 치료제로서의 G-Rk3의 가능성을 제안한다.
파충류 골격근의 근소포체에서 칼슘유리 채널의 존재를 밝히고저 뱀 골격근에서 근소포체를 분리하여 SDS-PAGE 전기영동, RyR의 정제, $[^3H]ryanodine$ 결합실험 및 $^{45}Ca$ 유리 실험으로 아래와 같은 결과를 얻었다. 1) 뱀골격근 소포체도 단일 band의 high molecular weight 단백을 가지고 있고, 그 mobility는 포유류 골격근의 것과 유사했다. 2) RyR의 정제과정에서 얻어진 $[^3H]ryanodine$의 peak 결합 분획에서 high molecular weight의 단백분획이 발견되었다. 3) 뱀 골격근 SR vesicles에 대한 $[^3H]ryanodine$의 maximum binding site와 Kd값은 각각 6.36 pmole/mg protein과 17.62nM이었으며, $[^3H]ryanodine$의 특이성 결합은 칼슘과 AMP에 의해 유의성있게 증가되었고 (P<0.005), tetracaine에 의해 억제되지 않았으나 ruthenium red와 $MgCl_2$에 의해 일부만 억제되었다. 4) 근 소포체로부터 $^{45}Ca$ 유리는 낮은 농도의 칼슘 $(1{\sim}10{\mu}M)$과 AMP에 의해 증가되었고 (P<0.05), 고농도의 칼슘 $(300{\mu}M)$, tetracaine, ruthenium red 또는 $MgCl_2$에 의해 억제되었다 (P<0.05). 이상의 실험성적으로 파충류 (뱀)의 골격근에도 칼슘유리 채별이 있어 근 수축시 세포내 칼슘 농도 조절에 관여할 수 있을 것으로 여겨지며, 채널의 기능적 특징 일부가 포유류의 것과 유사한 것으로 사료된다.
