Modulation of Activator Protein-1 (AP-1) and MAPK Pathway by Flavonoids in Human Prostate Cancer PC3 Cells

  • In last couple of decades the use of natural compounds like flavonoids as chemopreventive agents has gained much attention. Our current study focuses on identifying chemopreventive flavonoids and their mechanism of action on human prostate cancer cells. Human prostate cancer cells (PC3), stably transfected with activator protein 1 (AP-1) luciferase reporter gene were treated with four main classes of flavonoids namely flavonols, flavones, flavonones, and isoflavones. The maximum AP-1 luciferase induction of about 3 fold over control was observed with $20\;{\mu}M$ concentrations of quercetin, chrysin and genistein and $50\;{\mu}M$ concentration of kaempferol. At higher concentrations, most of the flavonoids demonstrated inhibition of AP-1 activity. The MTS assay for cell viability at 24 h showed that even at a very high concentration $(500\;{\mu}M)$, cell death was minimal for most of the flavonoids. To determine the role of MAPK pathway in the induction of AP-1 by flavonoids, Western blot of phospho MAPK proteins was performed. Four out of the eight flavonoids namely kaempferol, apigenin, genistein and naringenin were used for the Western Blot analysis. Induction of phospho-JNK and phospho-ERK activity was observed after two hour incubation of PC3-AP1 cells with flavonoids. However no induction of phospho-p38 activity was observed. Furthermore, pretreating the cells with specific inhibitors of JNK reduced the AP-1 luciferase activity that was induced by genistein while pretreatment with MEK inhibitor reduced the AP-1 luciferase activity induced by kaempferol. The pharmacological inhibitors did not affect the AP-1 luciferase activity induced by apigenin and naringenin. These results suggest the possible involvement of JNK pathway in genistein induced AP-1 activity while the ERK pathway seems to play an important role in kaempferol induced AP-1 activity.

Effect of Defective Onion Extract on the Onion Productivity by Organic Farming (양파 파치 추출물이 유기농 양파성에 미치는 영향)

  • This study evaluated the close crop recycling method by using the extract from defective onions. The mixture of the diluted extract (1/500) and nutrients was applied seven times on the onion leaves without spilling it onto the field. The yield and quality of the onions produced by this method (recycle) were compared to those grown organically (control) and conventionally (normal). The yield from the recycling field was increased significantly by 9% compared to that of the control field, while it was decreased by 11% compared to the normal field (p < 0.05). This lower yield was explained by the differences of the mulching vinyl and the fertilizers between the treatments which effect the onion growth during the winter. The content of quercetin in the onions from the recycling filed was increased significantly by 34% and 44% compared to those of the control and normal field, respectively (p < 0.05). It seemed that minerals and biologically active substances in the defective onion extract were effective in increasing the onion growth. In terms of soil microbial biomass, arbuscular mycorrhizal fungi was increased significantly in the recycle field by 40.1% and 30.6% compared to those of the control and normal fields, respectively (p < 0.05). On the other hand, microbial stress (cy19:0/18:1w7c) was decreased in the recycle field by 21.0% and 14.1% compared to those of the control and normal fields, respectively.

Antioxidant Properties of Water Extracts from Lentinula edodes Cultivars Grown on Oak Log (원목재배 표고버섯 물추출물의 항산화 특성)

  • We investigated the antioxidant activities of nine Lentinula edodes varieties cultivated on oak log. The total polyphenol content (TPC), total flavonoid content (TFC), and antioxidant properties of L. edodes (varieties: Dasanhyang, Chunbaegko, Poongnyunko, Soohyangko, Baekwhahyang, Chunjang 1ho, Chunjang 2ho, Sanlim 5ho, and Sanlim 7ho) were evaluated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay and the ferric reducing antioxidant power (FRAP) assay. The TPC and TFC of mushroom extracts ranged from 5.10 to 10.88 mg gallic acid equivalent (GAE)/g and 0.76 to 5.78 mg quercetin equivalent (QE)/g, respectively. In addition, mushroom extracts exhibited DPPH radical scavenging activity of 24.22~90.41%, 2.0 mg/mL and FRAP of 0.056~0.231, 2.0 mg/mL. The Chunbaegko variety showed the highest TPC (10.88 mg GAE/g), TFC (5.78 mg QE/g), DPPH activity (38.19~90.11%, 0.2~2.0 mg/mL), and FRAP (0.075~0.231, 0.2~2.0 mg/mL).

Production of High-Level Polyphenol Powders from Young Grape Leaves (어린 포도 잎을 이용한 폴리페놀 고함유 분말 제조)

  • Polyphenols are naturally occurring antioxidants and play many important roles in human health. Grape leaves have not yet been reported to contain the antioxidants. In this study, the physicochemical properties of powders from young grape leaves, obtained after hot-air or vacuum freeze drying, were analyzed. Weight loss rangedfrom 76-80%, but no difference was found between samples prepared by hot-air orvacuum freeze drying. The L-, a-, and b- values of grape powder all fell upon hot-air drying. Quercetin, resveratrol, catechin, and epicatechin were detected in all cultivars. The contents of the four polyphenols were generally higher in powders obtained by vacuum freeze dryingthan in those prepared by hot-air drying, indicating that vacuum freeze drying is better for optimal preservation of polyphenols. None of three polyphenols assayed was found in some cultivars or after certain drying treatments. Based on physiochemical properties, our findings suggest that powders obtained after vacuum freeze drying are optimal for industrial use.

Comparison of the Flavonoid and Urushiol Content in Different Parts of Rhus verniciflua Stokes Grown in Wonju and Okcheon (원주산과 옥천산 참옻나무의 부위별 flavonoid 및 urushiol 함량 비교)

  • Fustin comprised >98 and 73.0-86.7% of the total flavonoid content in the bark, and the stems and lignum of Rhus verniciflua, respectively. The butein, fisetin, and sulfuretin content varied between 0.31-2.17, 0.27-3.32, and 0.15-0.80 mg/g on a dry weight basis, respectively, in different parts of Rhus verniciflua. The urushiol content was 5.09-6.29, 55.05-56.30, and 0.38-0.39 mg/100 g on a dry weight basis in the stems, bark, and lignum, respectively. This showed that the bark of the tree had the highest urushiol content. C15:3 (pentadecatrienyl catechol), C15:1 (pentadecenyl catechol), and C15:2 (pentadecadienyl catechol) comprised 63, 33-35, and 2-3% of urushiol congeners in the tree bark, respectively.

Quality Characteristics of Mackerel Immersed in Sea Wind Mugwort Extract (해풍쑥 추출물에 침지 처리한 고등어의 품질특성)

  • This study examined the influence of sea wind mugwort extract treatment on quality characteristics of mackerel during storage. Mackerel were packaged individually and then immersed in 5% sea wind mugwort extract for 2, 3, or 4 hr and stored at $-20^{\circ}C$. The salinity of a control (no treatment) and that of mackerel immersed in sea wind mugwort for 2 hr was lowest (0.07%). pH of 5.90-6.23, and the change in acidity was in the opposite range. Immersion for 2, 3, and 4 hr led to a decrease in the tensile strength of the samples following storage, whereas the tensile strength of the control increased. The volatile basic nitrogen (VBN) content of the mackerel immersed for 2 hr was significantly lower than that of the control (5.6-15.4 mg% vs. 4.2-50.7 mg%). In the mackerel immersed for 2 hr, the total polyphenol and total flavonoid content was 286.3-497.0 mg gallic acid equivalents (GAE)/100 g and 177.5-385.6 mg quercetin equivalents (QE)/100 g, respectively. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethyl-benzothizoline-6-sulfonic acid) (ABTS) radical scavenging activities of the mackerel immersed in sea wind mugwort for 2 hr were 50.6% and 61.3%, respectively. Overall, the immersion of mackerel in sea wind mugwort for a short time significantly improved quality characteristics, such as salinity, pH, acidity, hardness, antioxidant activity, and perceptible quality, following storage.

Effect of Onion Consumption on Cardiovascular Disease in Human Intervention Studies: A Literature Review (국내.외 인체중재연구를 통해 살펴 본 양파의 심혈관계 질환 개선효과에 관한 고찰)

  • Onion (Allium cepa L.) production in Korea has increased gradually over the past 15 years, placing second in food consumption survey with 20.6 g daily intake in 2006. Onions, used as an ingredient in many dishes and accepted by almost all traditions and cultures, have been reported to have a range of health benefits which include anticarcinogenic, antiasthmatic, antibiotic, and antioxidative effects. These effects may be attributable to a powerful flavonoid pigment-containing compounds, such as quercetin and alk(en)yl cysteine sulphoxides (ACSOs). Although antiplatelet and antithrombotic activities of onion have been confirmed by many of in vitro or animal studies, only a few human intervention studies have been examined. The majority of human studies identified that onion improves some cardiovascular markers such as lipid profile and platelet coagulant. With regard to antioxidative effects, somewhat positive effects are confirmed through strengthening the resistance of oxidative DNA damage in lymphocyte and urine, while most studies failed to find inhibitory effects on LDL oxidation. The discrepancies among studies might be ascribed to producing area, processing methods of onion, dosage, subject characteristics, study duration, and measurement methods. In this review, we focused on the preventive effect of cardiovascular disease through onion consumption in human intervention studies.

Composition of Buckwheat (Fagopyrum esculentum Moench) Cultivars from Korea (한국산 메밀의 성분)

  • To clarify the values and varieties of the buckwheats as a dietary source of nutritional and functional components, thirteen different samples of buckwheat were analyzed for this investigation. Six developed seeds were given by RDA, Korea or RDA branch of Kangwondo, and seven land race seeds were collected from a farmhouse. Amino acid analysis showed that glutamate, arginine and asparagine were major amino acids, whereas tryptophan, methionine and cysteine were minor ones of buckwheat. In addition, tryptophan content of buckwheat cultivars from Korea was 195 mg% on average. The content of rutin tended to be higher in developed cultivars than land races. On the other hand, the contents of phytic acid in buckwheats were in the range of 7.0 to 13.6 mg/g. In the tocopherol homologues of the buckwheats analyzed by HPLC, mean ${\gamma}-tocopherol$ contents were 6.16 mg/100 g with the actual range of $4.67{\sim}8.58\;mg/100g$, whereas ${\beta}-form$ was very low or zero. There were a big variations in the iron content of the buckwheats of the minerals. SDS-PAGE showed that total proteins from buckwheats exhibited a relatively similar electrophoretic patterns on the whole. The results show that CV Suwon 1 has good quality, judged from the distribution of the components of buckwheats analyzed.

단백질 수준에 따른 비태인의 급여가 산란계의 생산능력과 난품질, 혈액 성상에 미치는 영향

  • 비태인의 급여(0, 600 ppm)가 사료의 단백질 수준(14, 16 %)에 따라 산란계의 생산성과 난품질, 혈액성상, 간과 가슴육의 일반성분에 미치는 영향을 구명하기 위하여 83주령 하이라인 산란계 192수를 이용하여 12주간 사양실험을 실시하였다. 기초사료는 에너지 수준이 2,800 kcal/kg, methionine과 lysine, cystine의 수준은 단백질 수준에 비례하도록 하였다. 조사항목으로 산란율과 난중, 사료섭취량, 사료요구율, 계란품질은 4주 간격으로 측정했다. 실험 종료시 복강지방과 혈액중 total protein과 albumin, BUN, triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol 함량, 간의 methionine, choline 함량, 간과 가슴육의 수분, 조단백질, 조지방 함량을 측정하였다. 산란율은 단백질 수준에 따라 증가하였으며(P<0.05) 비태인 급여에 의한 차이는 없었다. 난중은 단백질과 비태인 급여로 증가하였으며(P<0.05) 산란양은 단백질 수준이 증가할수록 감소하였다(P<0.05). 비태인 급여로 사료 요구율은 현저하게 개선되었지만(P<0.05), 계란의 품질은 차이가 없었다. 혈청 total protein은 비태인 급여로 현저히 증가하였으며 특히 단백질 14 % 급여구에서 크게 증가하였다(P<0.05). 복강지방 함량은 단백질 수준과 비태인 급여에 따라 감소하는 경향을 보였다. 가슴육과 간의 조단백질 함량은 사료의 단백질 수준에 따라 증가하였다(P<0.05). 간의 조지방 함량은 비태인의 급여로 감소하는 경향을 보였다. 간의 methionine 함량은 단백질과 비태인 수준에 따라 증가하는 경향을 보였으며 choline 함량은 비태인의 급여에 의해서만 증가하는 경향을 보였다. 따라서 비태인의 급여는 단백질 수준이 높은 조건에서 산란율을 개선시키고 난중을 증가시키며 사료 요구율을 개선한다.록 산가는 증가하는 것을 볼 수 있다. 홍국주의 전자 공여능에 의한 항산화력은 25.6%, 아질산염 소거능은 27.6%, Total phenolic compound 함량은 12.34mg%, ACE저해작용은 38%의 항산화력을 나타냈다.과 $O_2$$CO_2$의 농도에서 평균오차 0.2%로 정밀한 것으로 나타났으며 호흡속도 측정용 챔버의 혼합기체 공급측과 배기측의 가스 농도를 3회 반복 측정한 결과 재현성에서는 0.1%이하의 편차로 나타났다. 개방계 호흡속도 자동 측정 시스템을 이용하여 환경기체조성하에서 토마토의 호흡속도를 측정하는 실측 실험을 수행한 결과 2$0^{\circ}C$에서 12.7~42.1mg$CO_2$/kg.hr였으며 12$^{\circ}C$에서 2.5~8.2mg$CO_2$/kg.hr로 일반적으로 보고되고 있는 토마토 호흡속도와 일치하는 결과를 나타내었다.다.환원당인 sucrose 함량은 계속 증가하였고 fructose, glucose, sorbitol의 함량(추황의 sorbitol을 제외)은 생장이 촉진됨에 따라 증가하다가 다시 점차적으로 감소하였다. 이러한 결과는 총당과 환원당의 측정결과와 일치한 것으로 나타났다. 결론적으로 배의 성장에 따라 산 함량은 감소하였고 당 함량은 증가하였다.luco-pyranoside, quercetin 7-O- -glucopyranoside, acacetin 7-O-$\beta$-D-glucuronide and apigenin-6-C-$\beta$-D-glucopyranosyl-8-C-$\beta$-D-glucopyranoside were first isolated and identified from safflower leaf. Growth and Medical Constituents of Saururus chinensis Baill as Affected by Different Amounts of Nitrogen Fertilizer Application (질소 시비량이 삼백초 생육 및 성분함량에 미치는 영향)

  • This study was conducted to investigate the selected chemical properties of soils in Saururus chinensis Baill (Chinese lizard's tail) cultivation fields to provide optimal fertilizer application rates and to examine the growth and pharmaco-consitituents of Saururus chinensis Baill as influenced by different amounts of nitrogen (N) fertilizer applications. Based on the results of selected soil chemical properties in 37 cultivation sites of the plant, soil pH, organic matter content, and exchangeable $K^{+}$ concentration were lower than optimal values for cultivating general medicinal crops even though relatively high standard deviations were found in some of the values. At the harvesting stage of the plant aerial parts, soil pH, electrical conductivity (EC), available $P_2O_5$, and exchangeable $Ca^{2+}$, $Mg^{2+}$, $Na^{+}$ decreased as comparing with those before transplanting the plant, whereas the concentration of exchangeable $K^{+}$ increased in the plot treated with N 100% and compost. Fresh weight of the plant aerial parts were highest, 492.5kg/10, in the N 100% treatment plot. Correlation equation between N levels treated (X) and yield of the plant aerial parts (Y) presented as $Y=-2.1609X^2+30.082X+344.12$($R^2= 0.7113$) and the optimal rate of N fertilizer application for the plant was 6.6kg/10a. Carbon concentrations in the plant were not different among the different N levels applied. N and K concentrations in the plant were highest in the plot of N 100% with compost applications, the highest P concentration was in N 100% plot, and the highest Ca and S concentrations were in N 200% plot. Quercetin and quercitrin were highest in the N 150% plot and tannin was highest in N 100% or N 100% with compost application plot.
