Estimation and Analysis Methods for Trastuzumab Deamidation Levels Using Mass Spectrometry

  • We aimed to develop a suitable quantification method for detecting asparagine deamidation and aspartic acid isomerization in peptide mapping using LC-MS. Our assessment of its validity and suitability involved comparing its quantitative findings with those obtained from cation-exchange chromatography and capillary electrophoresis methods. By subjecting trastuzumab to rigorous conditions to induce these modifications, we validated the efficacy of this new analytical method in peptide mapping via LC-MS, evaluating both qualitative and quantitative aspects of asparagine deamidation and aspartic acid isomerization. Our investigation underscored the significance of enzyme selection and the presence of miss-cleaved or non-specific peptides in achieving accurate quantitative results. The experimental results demonstrated a strong correlation with results from cation-exchange chromatography and capillary electrophoresis analyses, confirming the reliability of the LC-MS based peptide mapping approach.

  • A number of different thin-film analytical method using electron spectroscopies are reviewed and evaluated. Results from various kinds of films (e.g. superconductive, electron-emissive, amorphous, rare-earth containing, etc.) are given for illustration. The existing quantification problems from this technology and some approaches for solving them are also discussed.

  • In this study, to specify an evaluation of human sensibility, the types of color, intensity of illuminations and lights consisting work environmental condition are decided, and image data from examining the change of human sensibility followed by changes of the above three conditions are obtained. Using the factor analysis and quantification theory in multi-variate analysis type of Sensibility Ergonomics, determinating the structure of factors, specifying the relations of environmental conditions and factors can be done so that the structure of image on human sensibility space with the change of environmental conditions is analyzed.

HPLC에 의한 Domoic acid 분석법 검정 (Validation of the Analysis of Domoic Acid using High Performance Liquid Chromatography)

  • To prevent amnesic shellfish poisoning (ASP) resulting from the consumption of shellfish contaminated with domoic acid, the quantitative analysis of domoic acid is very important. We validated a high performance liquid chromatography (HPLC) method for accurate and precise quantification of domoic acid. A clear peak and the isolation of domoic acid resulted on injecting a 50% methanol extract of CRM-ASP-Mus-c mussel reference material using HPLC. The limit of detection of domoic acid under the established HPLC conditions was $0.10\;{\mu}g/g$, and the limit of quantification of the toxin under the same conditions was $0.25\;{\mu}g/g$. The intra-accuracy and precision for domoic acid in CRM-ASP-Mus-c were 90.7-95.7% and 0.28-22.25%, respectively. The inter-accuracy and precision for domoic acid were 89.1-97.1% and 1.7-4.1%, respectively. The mean recovery of domoic acid in methanol extracts from ten species of marine invertebrates was 88.6-1105.1%.

Development and Validation of a Robust LC-MS/MS Method for the Simultaneous Quantification of Doxifluridine and its Two Metabolites in Beagle Dog Plasma

  • A reverse-phase HPLC method with detection by mass spectrometry is described for the simultaneous determination of doxifluridine and its two active metabolites, 5-fluorouracil (5-FU) and 5-fluorouridine (5-FUrd), in beagle dog plasma. The optimal chromatographic separation was achieved on a Waters $Xterra^{(R)}$ $C_{18}$ column ($4.6{\times}250\;mm$ i.d., $5\;{\mu}m$ particle size) with a mobile phase of 0.1% formic acid in a mixture of 99% methanol and purified water (99:1, v/v). The developed method was validated in beagle dog plasma with a lowest limit of quantification of $0.05\;{\mu}g/mL$ for both doxifluridine and 5-FU, and $0.2\;{\mu}g/mL$ for 5-FUrd. Doxifluridine and its two metabolites were stable under the analysis conditions, and intra- and inter-day accuracies exceeded 92.87%, with a precision variability ${\leq}11.34%$ for each analyte. Additionally, the method for quantifying doxifluridine and its two metabolites, 5-FU and 5-FUrd, in beagle dog plasma was applied successfully to the analysis of pharmacokinetic samples.

LC-MS/MS를 이용한 곽향정기산(藿香正氣散) 추출물 중 20종 성분의 함량분석 (Quantitative Analysis of the Twenty Marker Components in Gwakhyangjeonggi-san using Ultra-Performance Liquid Chromatography with Mass Spectrometer)

  • Generally, Gwakhyangjeonggi-san has been used for treatment of diarrhea-predominant irritable bowel syndrome. In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer method was established for the simultaneous quantification of marker compounds 1-20 in Gwakhyangjeonggi-san water extract. All analytes were separated by gradient elution using two mobile phases on a UPLC BEH $C_{18}$ ($100{\times}2.1mm$, $1.7{\mu}m$) column and maintained at $45^{\circ}C$. The injection volume was $2.0{\mu}L$ and the flow rate was 0.3 mL/min with detection at mass spectrometer. Regression equations of the compounds 1-20 were acquired with $r^2$ values ${\geq}0.9950$. The values of limit of detection and quantification of all analytes were 0.01-2.79 ng/mL and 0.03-8.37 ng/mL, respectively. The amounts of the compounds 1-20 in Gwakhyangjeonggi-san water extract were not detected $-3,236.67{\mu}g/g$. The established LC-MS/MS methods will be valuable to improve quality control of traditional herbal formula, Gwakhyangjeonggi-san.

Quantitative Analysis of Leuconostoc mesenteroides and Lactobacillus plantarum Populations by a Competitive Polymerase Chain Reaction

  • A multiplex competitive polymerase chain reaction (PCR) method was developed for the rapid identification and quantification of Leuconostoc mesnteroides and Lactobacillus plantarum populations which are the key microorganisms in kimchi fermentation. The strain-specific primers were designed to selectively amplify the target genes encoding 165 rRNA of L. plantarum and dextransucrase of L. mesenteroides. There was a linear relationship between the band intensity of PCR products and the number of colony forming units of each model organism. The PCR quantification method was compared with a traditional plate-counting method f3r the enumeration of the two lactic acid bacteria in a mixed suspension culture and also applied to a real food system, namely, watery kimchi. The population dynamics of the two model organisms in the mixed culture were reliably predictable by the competitive PCR analysis.
