Diagnostic assessment on vegetation damage due to hydrofluoric gas leak accident and restoration planning to mitigate the damage in a forest ecosystem around Hube Globe in Gumi (구미 휴브글로브 주변 삼림생태계에서 불화수소가스 유출 사고에 기인한 식생피해 진단 및 그 피해를 완화시키기 위한 복원 계획)

  • We obtained the following results from investigation on vegetation damage from 5 to 6 August, 2013, about one year after an accident that hydrofluoric acid leaked from a chemical maker, Hube Globe in Gumi. Pinus densiflora and Pinus strobus showed very severe damage. Ginko biloba, Quercus acutissima, Pinus rigida, Salix glandulosa, Hibiscus syriacus, and Lagerstroemia indica showed severe damage. Quercus variabilis, Lespedeza cyrtobotrya, and Miscanthus sinensis showed moderate damage. Quercus aliena, Smilax china, Arundidinella hirta, Ailanthus altissima, Robinia pseudoacacia, and Paulowinia coreana showed slight damage. We did not find any plants without leaf damage around there. This result means that fluoride damage still persists in this area as was known that fluoride remains for a long time in air, soil and water and exerts negative effects at all levels of an ecosystem. In addition, fluoride content contained in plant leaf depended on the distance from a fertilizer producing factory and vegetation damage tended to proportionate to the concentration in the Yeocheon industrial complex. In these respects, a measure for removal or detoxification of the remaining fluoride is urgently required around the hydrofluoric acid leak spot. Fertilizing of dolomite containing Ca and Mg, which can trap fluoride, was prepared as one of the restoration plans. In addition, phosphate fertilizing was added in order to enhance soil ameliorating effects. Furthermore, we recommend the introduction of tolerant plants as the second measure to mitigate fluoride damage. As the tolerant plants to make a new forest by replacing trees died due to hydrofluoric acid gas damage, we recommended Q. aliena and S. china, A. hirta, etc. were recommended as plant species to add mantle vegetation to the forest margin to ensure stable interior environment of the forest.

Seasonal Sedimentary Characteristics and Depositional Environments after the Construction of seawall on the Iwon Macrotidal Flat (방조제 건설 후 이원 대조차 조간대의 계절별 퇴적학적 특성 및 퇴적환경)

  • In order to elucidate seasonal sedimentary characteristics and depositional environment after construction of seawall on macrotidal flat, a seasonal observations of surface sediments (total 450) and sedimentation rates on 4 transects have been investigated for 2 years. The eastern area of Iwon tidal flat, has been changed from semi-closed coast to open coast by construction of seawall, shows general seasonal changes similar to characteristics of open coast type, which represented both fining and bad sorted distribution due to deposition of fine sediments under low energy condition in the summer, and relatively coarser and better sorted distribution because of erosion of fine sediments in the winter. In considering angles of transects, distribution patterns of surface sediments, the northern and southern parts of eastern tidal flat are dominantly influenced by wave and tidal effects, respectively. As time goes by, the eastern tidal flat shows coarsening-trend of surface sediments caused by direct effect of tidal current, were and typhoon. Meanwhile the western area of seawall, which has been re-formed by construction seawall, is sheltered from northwesterly seasonal wind. The seasonal change pattern of western area of seawall is slightly different from that of eastern tidal flat. Mean grain size and sorting of surface sediments during spring is finer and worse than those during summer. This seasonal change pattern maybe influenced by topographic effects caused from the construction of seawall. In consideration of all result, the transport of fine sediments in the study area, which is supplied to limited sediments, shows clockwise circulation pattern that fine sediments are transported from the eastern tidal flat to the western area of seawall because of blocking of seawall in the winter and are transported reversed direction the summer. As a result, many changes have been observed in the study area after construction of seawall; however, this change is still in progress and is expected to need continuous monitoring.

Interspecific Competition of Paddy Rice Isogenic Lines in Plant Type with Some Perennial Weeds (수도초형(水稻草型)의 Isogenic Line과 다년생(多年生) 잡초(雜草)의 경합특성(競合特性) 연구(硏究))

  • By use of three paddy rice lines as the near-isogenic in plant type (Broom, Open, and Spread type in tillering angle), the interspecific competition patterns of rice plants with three important weed species (Cyperus serotinus, Eleocharis kuroguwai, and Potamogeton distinctus) under the three densities of weed standing (0.25 and 50 percent of the rice plants), were observed. Under the experimented conditions, paddy yields were varied more significantly with weed competition descriptions than with plant types of paddy rice. And spread typed rice was more competitive to the detected weed species, however, the broom and spread typed rice were to Potamogeton SP, among others. The result of the clustering analysis of crop-weed competition patterns, estimated by 1-Q mode correlation coefficients, indicated that the first-order component affecting the competition patterns of crop-weed was rather the plant types of rice than either weed species or weed standing densities.

  • Purpose : In Korea, the school urine screening program is a useful tool for screening urine abnormalities. It is particularly useful in early detection of membranoproliferative glomerulonephritis(MPGN) I, which frequently progresses to chronic renal failure. In this study, we studied the medical history, laboratory findings, and histologic findings of MPGN to gain helpful information on early detection and treatment. Methods : The subjects were 19 children, who were diagnosed with MPGN from kidney biopsies that were performed in ten nationwide university hospitals because of abnormal urine findings from school urine screening programs conducted from July 1999 to April 2004. We divided the patients into 2 groups, a nephrotic range proteinuria group(n=8) and a non-nephrotic proteinuria group(n=11), and retrospectively analyzed the clinical features, laboratory findings, histologic findings, treatment, and clinical course. Results : The mean age at the first abnormal urinalysis was $10.6{\pm}2.2$ years in the nephrotic proteinuria group and $9.6{\pm}3.2$ years in the non-nephrotic proteinuria group. The mean age at the time of kidney biopsy was $11.3{\pm}2.3$ years in the nephrotic range proteinuria group and $10.4{\pm}3.2$ years in the non-nephrotic proteinuria group respectively. There was no significant difference in the mean age and sex between the two groups. In the nephrotic proteinuria group, 6 children had a low plasma C3 level and in the non-nephrotic proteinuria group, 8 children had a low plasma C3 level, but there was no significant difference between the 2 groups. There was no significant difference in the laboratory test results(including WBC count, RBC count, platelet count and other serologic tests) between the 2 groups except for 24 hour urine protein secretion. There was no difference between the 2 groups with regard to the acute and chronic changes in the glomerulus on light microscopic findings, IgG, IgA, Ig M, C1q, C3, C4, fibrogen deposition on immunofluoroscence findings, and mesangial deposits, subendothelial deposits, and subepithelial deposits on electron microscopic findings. The children were treated with corticosteroids, ACE(angiotensin-converting enzyme) inhibitors, dipyridamole and other immunosuppressive agents. During the course of treatment, there were no children whose clinical condition worsened. Among 19 children, 3 children went into remission(2 in the nephrotic proteinuria group, 1 in the non-nephrotic proteinuria group) and 9 children went into a partial remission(4 in the nephrotic proteinuria group, 5 in the non-nephrotic proteinuria group) on urinalysis. There was no significant difference in the treatment results between the two groups. Conclusion : The 73.7% of children who were incidentally diagnosed with MPGN by the school urine screening program had reduced C3. 42.1% of the children had nephrotic range proteinuria. There were no significant differences in clinical features, laboratory test results, light microscopic, immunofluorescence microscopic, and electron microscopic findings between the nephrotic proteinuria group and the non-nephrotic proteinuria group except for the 24 hour urine protein secretion. Therefore, for early detection of MPGN during the school urine screening program, we strongly recommend a kidney biopsy if children have abnormal urine findings such as persistent proteinuria and persistent hematuria, or if the serum C3 is reduced.

Phenotype-genotype correlations and the efficacy of growth hormone treatment in Korean children with Prader-Willi syndrome (프래더 윌리 증후군의 유전학적 발병 기전에 따른 표현형 및 성장 호르몬 치료 효과에 관한 연구)

  • Purpose : Prader-Willi syndrome (PWS) is a complex genetic disorder, caused by the deletion of the paternally derived 15q11-13 region or the maternal uniparental disomy of chromosome 15 (mUPD(15)). In this study, we compared phenotypic differences between those patients whose disease was caused by microdeletion and those caused by mUPD(15). In addition, a comparison of the efficacy of growth hormone (GH) therapy between these two PWS genotypes was analyzed. Methods : Fifty-three patients were diagnosed as having PWS based on molecular and cytogenetic analyses and clinical features. Data that included maternal age, birth weight, a feeding problem in the neonatal period, cryptorchidism, developmental delay or mental retardation, short stature, hypopigmentation, changes in height, weight, and body mass indexes (BMI) before and after GH treatment were obtained by a retrospective review of medical records. The data from the patients with microdeletion were compared with those from the patients with mUPD(15). Results : Of the 53 patients with genetically confirmed PWS, 39 cases had microdeletion and 14 mUPD(15). Maternal ages were significantly higher in the mUPD(15) group, and hypopigmentation and a feeding problem in the neonatal period were more frequent in the microdeletion group. Growth hormone was administered to 20 patients [14 with microdeletion, 6 with mUPD(15)]. There were no differences between the two groups in height velocity, weight and height SDS, and BMI after GH therapy. Conclusion : Phenotype and genotype correlations were observed in Korean PWS patients, such as more advanced maternal ages in the mUPD(15) group and more feeding problems and hypopigmentations in the microdeletion group. Further long-term prospective studies are needed to correlate other aspects of the phenotypes.

The Comparison of Existing Synthetic Unit Hydrograph Method in Korea (국내 기존 합성단위도 방법의 비교)

  • Generally, design flood for a hydraulic structure is estimated using statistical analysis of runoff data. However, due to the lack of runoff data, it is difficult that the statistical method is applied for estimation of design flood. In this case, the synthetic unit hydrograph method is used generally and the models such as NYMO method, Snyder method, SCS method, and HYMO method have been widely used in Korea. In this study, these methods and KICT method, which is developed in year 2000, are compared and analyzed in 10 study areas. Firstly, peak flow and peak time of representative unit hydrograph and synthetic unit hydrograph in study area are compared, and secondly, the shape of unit hydrograph is compared using a root mean square error(RMSE). In Nakayasu method developed in Japan, synthetic unit hydrograph is very different from peak flow, peak time, and the shape of representative unit hydrograph, and KICT method(2000) is superior to others. Also, KICT method(2000) is superior to others in the aspects of using hydrologic and topographical data. Therefore, Nakayasu method is not a proper in hydrological practice. Moreover, it is considered that KICT model is a better method for the estimation of design flood. However, if other model, i.e. SCS method, Nakayasu method, and HYMO method, is used, parameters or regression equations must be adjusted by analysis of real data in Korea.

An Experimental Study on the Required Performances of Roof Concrete Placed in the In-ground LNG Storage Tank (지하식 LNG 저장탱크의 지붕 콘크리트의 요구성능에 관한 실험적 연구)

  • This study is to derive from the required performances and the optimum mix proportion of the roof concrete placed in the in-ground LNG storage tank with a capacity of 200000 $m^3$, and propose the actual data for site concrete work. The concrete placing work without sliding and segregation in the fresh concrete condition is very important because the slope of domed roof is varied in the large range by its curvature. Also the control of hydration heat and the strength development at test ages are classified with massive section about 1.4 m thick and considered to the pre-stressing work and removal of air support after concrete placing work. Considering above condition, slump range is selected $100{\pm}25$ mm under the slope $20^{\circ}$ and $150{\pm}25$ mm over the slope $20^{\circ}$ s until 60 minutes of elapsed time. Also, the roof concrete is satisfied with compressive strength range including design strength at 91 days (30 MPa), pre-stressing work at 7 days (10 MPa), air support removal work at 21 days (14 MPa). Replacement ratio of limestone powder is determined by confined water ratio test and main design factors include water-cement ratio (W/C), sand-aggregate ratio and dosage of admixture. As test results, the optimum mix proportion of the roof concrete used low heat cement is as followings. 1) Replacement ratio of limestone powder 25% by confined water ratio test 2) Water-cement ratio 57.8% 3) Sand-aggregate ratio 42.0%. Also, test results for the adiabatic temperature rising test is satisfied with its criteria and shown the lower value compared to preceding storage tank (TK-13, 14). These required performances and the optimum mix proportion is to apply the actual construction work.

Improved Acroparesthesia During Enzyme Replacement Therapy in a Patient Lately Diagnosed with Fabry Disease (진단이 지연된 Fabry 병 환자에서 효소대체요법을 통한 사지 말단 동통의 호전을 보인 1례)

  • Fabry disease (FD) is an X-linked lysosomal storage disorder caused by an ${\alpha}$-galactosidase A (GLA, MIM 300644) enzyme deficiency due to pathogenic variants in the ${\alpha}$-galactosidase A gene (GLA). The disease leads to accumulation of globotriaosylceramide (Gb3) and related glycophospholipids affecting nearly all major organ systems, with the primary sites damaged by Gb3 including renal glomeruli, myocardium, neurons of the dorsal ganglion and autonomic nervous system, and vascular endothelial and smooth muscle. Progressive deposition in these organ systems present with various clinical manifestations including acroparesthesia, renal failure and heart failure. Here, we report a Chinese male diagnosed with Fabry disease in his late $4^{th}$ decades showing improvement of acroparesthesia during enzyme replacement therapy (ERT). A 48-year-old Chinese man who presented with chronic recurrent severe burning pain in his fingers and toes since the age of 10, with worse involvement of the former visited to our clinic for further evaluation. His medical history included a transient ischemic attack aged 40 and diagnosed with stage 4-5 chronic kidney disease aged 47. In the family history, the patient's brother was found to be have Fabry disease 1 month before his visit. Except for his brother, all other members of the family are healthy. Based on his medical history and family history, he was strongly suspicious for Fabry disease. He was found to have a galactose-alpha-1,3-galactose level 4.96 (Reference range, 42.5-67.9) suggestive of Fabry disease. The followed sequencing of GLA coding region in our patient revealed hemizyosity for the mutation c.988C>T (Q330X) in Exon 7. Since ERT start, he showed significant improvement in his symptoms of burning sensation of fingers and toes. On the contrary, due to deteriorating kidney function even with ERT, he is considered for kidney transplantation. Despite of diagnostic delay until late 4th decades, ERT showed a potential improvement of acroparesthesia in our patient. However, late start of ERT can lead to poor outcome in multiorgan function. Therefore, early diagnosis with high index of suspicion followed by continuous ERT with regular monitoring have an impact on quality of life in Fabry disease.

Effects of Experimental Warming on Growth of Quercus variabilis Seedlings (실외 실험적 온난화 처리가 굴참나무 묘목의 생장에 미치는 영향)

  • Climate change affects all biological processes in terrestrial ecosystems including photosynthesis, plant growth and productivity. This study was conducted to investigate the effects of experimental warming on the growth of Quercus variabilis seedlings. One-year-old Q. variabilis seedlings were planted in control and warmed plots in April 2010. The air temperature of warmed plots was increased by $3^{\circ}C$ compared to control plots using the infrared lamp from November 2010. Shoot height and root collar diameter were measured in March 2011 and June 2012, respectively, and aboveground and belowground biomass were also measured in March 2011 and 2012, respectively. Shoot height and root collar diameter were significantly higher in warmed plots than in control plots, except for root collar diameter in March 2011. Increment (mm) of shoot height and root collar diameter were also higher in warmed plots ($529{\pm}30$, $5.6{\pm}0.5$) than in control plots ($464{\pm}28$, $4.5{\pm}0.4$). However, there were no significant differences between warmed and control plots except for root collar diameter. Increment (g/year) of total, aboveground and belowground biomass were higher in warmed plots ($36.88{\pm}6.52$, $11.91{\pm}3.44$, $24.97{\pm}3.73$) than in control plots ($30.59{\pm}5.51$, $8.73{\pm}1.66$, $21.86{\pm}3.88$), however, the differences were not statistically significant. Higher seedling growth and biomass of warmed plots might be related to the enhanced net photosynthetic rates in spring and the extended growing season.

Identification and Characterization of Lactobacillus salivarius subsp. salivarius from Korean Feces

  • This study was conducted to isolate lactobacilli having probiotic characteristics to be used as health adjuncts with fermented milk products. Acid tolerant strains were selected in Lactobacilli MRS broth adjusted to pH 4.0 from 80 healthy persons (infants, children and adults). And bile tolerant strains were examined in Lactobacilli MRS broth in which 1.0% bile salt was added. By estimation above characteristics, the strains No. 27, which was isolated from adult feces, was selected and identified as Lactobacillus salivarius subsp. salivarius based on carbohydrate fermentation and 16S rDNA sequencing. It was used as a probiotic strain in fermented milk products. The pH of fermented milk decreased from pH 6.7 to 5.0 and titratable acidity increased from 0.3% to 1.0% by L. salivarius subsp. salivarius (isolation strain 20, 35, and 37), when incubated for 36 h at $37^{\circ}C$. The number of viable cell counts of fermented milk was maximized at this incubation condition. The SDS-PAGE evidenced no significant change of casein but distinct changes of whey protein were observed by isolated L. salivarius subsp. salivarius for titratable acidity being incubated by $0.9{\sim}1.0%$ at $37^{\circ}C$. All of the strains produced 83.43 to 131.96 mM of lactic acid and 5.39 to 26.85 mM of isobutyric acid in fermented products. The in vitro culture experiment was performed to evaluate ability to reduce cholesterol levels and antimicrobial activity in the growth medium. The selected L. salivarius subsp. salivarius reduced $23{\sim}38%$ of cholesterol content in lactobacilli MRS broth during bacterial growth for 24 hours at $37^{\circ}C$. All of the isolated L. salivarius subsp. salivarius had an excellent antibacterial activity with $15{\sim}25$ mm of inhibition zone to E. coli KCTC1039, S. enteritidis KCCM3313, S. typhimurium M-15, and S. typhimurium KCCM40253 when its pH had not been adjusted. Also, all of the isolated L. salivarius subsp. salivarius had partial inhibition zone to E. coli KCTC1039, E. coli KCTC0115 and S. enteritidis KCCM3313 when it had been adjusted to pH 5.7. The selected strains were determined to have resistances of twelve antibiotic. Strains 27 and 35 among the L. salivarius subsp. salivarius showed the highest resistance to the antibiotics. Purified ${\alpha}$-galactosidase was obtained by DEAE-Sephadex A-50 ion exchange chromatography, Mono-Q ion exchange chromatography and HPLC column chromatography from L. salivarius subsp. salivarius 27. The specific activity of the purified enzyme was 8,994 units/mg protein, representing an 17.09 folds purification of the original cell crude extract. The molecular weight of enzyme was identified about 53,000 dalton by 12% SDS-PAGE. Optimal temperature and pH for activity of this enzyme were $40^{\circ}C$ and 7.0 respectively. The enzyme was found to be stable between 25 and $50^{\circ}C$. ${\alpha}$-galactosidase activity was lost rapidly below pH 5.0 and above pH 9.0. This enzyme was liberated galactose from melibiose, raffinose, and stachyose, and also the hydrolysis rate of substrate was compound by HPLC. These results indicated that some of the L. salivarius subsp. salivarius (strain 27 and 35) are considered as effective probiotic strains with a potential for industrial applications, but the further study is needed to establish their use as probiotics in vivo.
