Pyricularia anguiata Hashioka에 의한 바나나 도열병 (가칭) (Banana Blast Caused by Pyricularia angulata Hashioka)

  • 1985년에서 1986년 지역적으로 식물병해 조사를 실시하던 중 제주도의 바나나 재배온실에서 바나나도열병이 심하게 발생하고 있음을 발견하였다. 어떤 온실에서는 이 병의 발생이 매우 심하여 발병율이 $100\%$에 달하였다. 이 병을 일으킨 균을 균학적인 특성과 병원성에 의해 Pyricularia angulata Hashioka로 동정하였다. 바나나의 잎에 형성된 바나나도열병의 병징은 원형 내지 타원형으로 직경 1-10mm 크기의 암갈색 반점이며 반점주위는 황색을 띄었다. 열매에 형성된 병징은 원형 내지 타원형으로 적갈색 혹은 암갈색의 움푹 들어간 반점이며, 그 크기는 직경이 20mm에 달했다. 병징은 바나나의 잎과 열매뿐만 아니라 엽병, 엽초, 열매의 다발줄기, 열매의 수관에서도 발생하였다. 병원균의 분생포자는 무색, 격막은 2개, 하나의 작은 hium을 가진 달걀모양 내지 서양배모양이고, 분생자경의 작은 돌기 끝에서 단생하며, 크기는 $16.0-34.0\times7.0-12.0{\mu}m$(평균 $22.5\times9.0{\mu}m$)였다. 분생자경은 대부분 격막이 2개, 간혹 둘 혹은 셋으로 분지하고, 정단부위에는 작은 돌기가 있으며, 크기는 $7.5-100.0\times3.0-5.0{\mu}m$(평균 $90.0\times4.0{\mu}m$)였다. PDA에서 이 균의 균사 생육적온 범위는 $26-28^{\circ}C$였다. 병원성검정 결과, P. angulata는 바나나에만 병원성이 있었다. 반면에 Digitaria sanguinalis(L.) Scopo.에서 분리한 Pyricularia grisea(Cke.) Sacc.는 바나나에 병원성이 없었다. 접종시험 결과, 바나나의 여러 부위 중에서 잎이 P. angulata에 가장 감수성이었다. 또한 생육 30일된 열매는 생육 60일된 열매보다 더 감수성이었으며, 열매의 다발줄기는 열매보다 더 수감성이었다.

Evaluation of induced systemic resistance agent, Bacillus subtilis strain BAC02-4 against Magnaporthe grisea in rice in field

  • Bacillus subtilis strain BAC02-4 was tested for its ability induced systemic resistance(ISR) in rice against Magnaporthe grisea We extend these studies to investigate the biological induction of systemic resistance in rice following treatment with the inducer isolate BAC02-4 and naturally infested with Pyricularia oryzae. We also determine levels of ISR activity during the period between disease development and the onset of systemic resistance. Comparition of lesion number according to applied concentration of BAC02-4 to 'Nagdongbyeo' when naturally infested with the conidia of P. grisea. Results from the blast nusery trial using the 'Nagdongbyeo' showed very low rice blast severity with the inducer concentration of 10$\^$8/ cfu level. Considering the low level of treatment and untreated control were observed to have developed typical susceptible lesion type. Highest protection against the rice blast pathogen when applied three times with 5 days interval as root drench at 5 to 6 leaf stage before pathogen challenge. But higher dose of bacterial inducer produced a little stunted plants with less number lesions and delayed disease development. Diseased leaf area of treated with suspension of the isolate which gave about 80% of control efficacy at 20 days later comparable to that in noninfested, inducer-free soil.

벼 주요 품종 및 계통의 지역별, 연도별 도열병 발병 차이(II) (Temporal and Spatial Blast Incidence in Leading Cultivars and Elite Lines of rice in Korea(II))

  • Incidence of blast caused by Pyricularia grisea Sacc. on 21 leading cultivars and elite lines of rice was investigated in different locations such as Icheon, Chuncheon, Jecheon, and Naju during three years from 1991 to 1993. Disease index of leaf blast of Dongjinbyeo was high in the blast nursery with range 4 to 9 at Naju, but low at other locations with range 1 to 4. Percentages of diseased leaf area of Joryungbyeo and Daelipbyeo 1 were 4.3 and 12%, respectively in the fields at Jecheon in 1993, but were much lower in the other locations during three years. The incidences of leaf blast of Daelipbyeo 1 were 0.3 and 12.0% at Chuncheon and Jecheon, respectively in 1993, but no disease occurred at Naju during three years. Percentage of diseased panicles of Joryungbyeo was 100% at Icheon and Jecheon in 1993, 92.4% in Suweon 390 at Icheon on 1991 and 100% in Jinbu 15 at Jecheon on 1993. Race distribution of rice blast fungus at Icheon and Jecheon was more varied than that of the other locations.

  • Incidences of blast caused by Pyricularia grisea Sacc. on 24 leading cultivars and elite lines of rice were investigated in the fields at Icheon, chuncheon, Jecheon, Naju from 1990 to 1992. In the blast nursery, disease index of leaf blast on Jinmibyeo and Ilpumbyeo were very low as 1 to 3 at Naju, but as high as 6 to 9 at Icheon and other Chuncheon in 1990, but the disease did not occur in other locations and years. The most variable incidence of temporal and spatial leaf blast was observed on Nagdongbyeo, which was 30.6% at Icheon and 2.1% at Chuncheon on 1990, but the disease did not occur at Naju during the investigation. Percentages of diseased panicles on Chucheongbyeo were 11.6% in 1990 and 4.3% in 1992 at Icheon. Odaebyeo and Sobaekbyeo revealed more severe blast occurrences at Chuncheon and Sangju where the elevation was higher than the other places. Regional race distributions of rice blast fungus were more variable at Icheon and Chuncheon than the others.

황련 추출물의 고추역병에 대한 In-vitro 항진균 활성 및 고추 생육촉진 효과 (Bioactivity of the Extract of Coptis chinensis: In-vitro Antifungal Activity against Phytophthora capsici and Growth-promotion Effect in Red-pepper)

  • 황련의 식물병원성 곰팡이에 대한 항진균 활성을 평가하기 위해 황련 메탄올 추출물 및 유기용매 분획물들을 조제하여 Colletotrichum gloeosporioides(고추탄저 병), Phytohpthora capsici(고추역병), Pyricularia grisea(벼도열병), Rhizoctonia solani(잎집무늬마름병), Botryosphaeri dothidea(겹무늬썩음 병), Glomerella cingulata(사과과실 탄저병) 및 Botrytis cinerea(잿빛곰팡이병)의 7종에 대해 $0{\sim}1,000{\mu}g/mL$ 농도로 처리한 후, 포자발아억제 활성 및 균사체 생육억제 활성을 평가하였다. 그 결과 황련 메탄올 추출물은 $500{\mu}g/mL$ 농도에서 B. dothidea를 제외한 6종의 곰팡이의 포자발아를 억제하였으며, P. grisea 및 B. cinerea에 대해 $300{\mu}g/mL$의 MIC를, P. capsici에 대해서는 $500{\mu}g/mL$의 MIC를 나타내었고, 다른 4종에 곰팡이에 대해서는 균사체 성장억제 활성이 나타나지 않았다. 고추 재배지의 실제적 적용을 위해 대량 추출조건을 선정한 후, 상업적 시설에서 열수 추출물($1,000{\mu}g/mL$)을 조제하고, 이를 6월부터 한달 간격으로 3회에 걸쳐 엽면살포한 결과 병해와 약해가 전혀 나타나지 않았다. 황련 살포에 의해 고추의 전반적인 생육, 잎, 뿌리 발육이 촉진되었으며, 고추는 평균중량이 119%, 과육의 두께가 평균 117% 증가되어 생육촉진 및 수확량 증가효과를 확인하였다. 본 연구결과는 황련 추출물이 고추 관련 질병 방제에 유용하며, 고추생육 촉진효과를 가짐을 시사하고 있다.

Fungicidal Activity of Oriental Medicinal Plant Extracts against Plant Pathogenic Fungi

  • Methanol extracts from 53 species of oriental medicinal plants in 34 families were tested for their fungicidal activities against Pyricularia grisea, Rhizoctonia solani, Phytophthora capsici, Phytophthora infestans, Collectotrichum dematium, Botryospaeria dothidea, Fusarium oxysporum f. sp. cucumerinum, Botrytis cinerea, Puccinia recondita, and Erysiphe graminis. In in vitro study using impregnated paper disc method, the efficacy varied with both plant pathogen and plant species tested. Methanol extracts of Asarum sieboldii roots, Sinomenium acutum roots, Pinus densiflora leaves, Rheum undulatum root barks, Coptis japonica roots, and Phellodendron amurense barks showed potent fungicidal activities against the various pathogens when treated with 10 mg/disc. In a whole plant test, methanol extracts of P. densiflora leaves and roots and C. japonica roots were highly effective against a variety of plant pathogens. As a naturally occurring fungicide, P. densiflora- and C. japonica-derived materials could be useful as new fungicidal products against various plant diseases induced by plant pathogenic fungi.

Selection of KYC 3270, a Cellulolytic Myxobacteria of Sorangium cellulosum, against Several Phytopathogens and a Potential Biocontrol Agent against Gray Mold in Stored Fruit

  • During 2002-2008 in Korea, 455 extracts from myxobacteria consisting of 318 cellulolytic and 137 bacteriolytic myxobacteria were isolated, which were then screened for antifungal activity against the phytopathogens Botrytis cinerea, Colletotrichum acutatum, Penicillium sp., Pyricularia grisea, and Phytophthora capsici. 204 isolates had antifungal activity, causing both a clear zone due to blocked spore germination and inhibition of mycelial growth; most (199) were from cellulolytic (Sorangium cellulosum) and only five were from bacteriolytic myxobacteria. B. cinerea, the best controlled among the five tested pathogens, had a unique group of antifungal isolates of myxobacterial extracts compared to the other pathogens' groups. Among seventy-nine bioactive myxobacteria, four isolates, KYC 3130, KYC 3247, KYC 3248 and KYC 3270, were selected and all were cellulolytic. Liquid culture filtrates of these four myxobacteria were applied to tomato, cherry tomato, strawberry, and kiwi fruits 5 h before inoculation with gray mold conidia; then the treated fruits were placed in an airtight container and the experiment was repeated six to eight times. Incidence (%) of gray mold on fruit of the infected control treatment was 84-98%, whereas it was only 5-21% after the KYC 3270 treatment. After KYC 3270 treatment of the four fruits, mold control was 79-95%, which was highest among the filtrates and statistically the same as treatment with fludioxonil, a registered chemical against gray mold of stored fruits.

Biocontrol with Myxococcus sp. KYC 1126 Against Anthracnose in Hot Pepper

  • Antifungal activity of three Myxococcus spp., KYC 1126, 1136, and 2001, was tested in vitro against three phytopathogenic fungi (Botrytis cinerea, Colletotrichum acutatum, and Pyricularia grisea). Spore germination and mycelial growth of the three pathogenic fungi were completely inhibited by bioactive substances from a myxobacterium KYC 1126. In addition, the activity of KYC 1126 was fungicidal, but liquid culture filtrate of KYC 1126 did not affect protoplast reversion in C. acutatum. A bioassay of KYC 1126 filtrate against anthracnose in hot pepper was conducted in the greenhouse and field at 2009 and 2010. The incidence of anthracnose in control seedlings was 74%, but was reduced to 29% after KYC 1126 treatment. The control value with KYC 1126 was 60% while that with the fungicide dithianon was 42%. In the greenhouse, disease incidence with KYC 1126 was consistentely 10-35% lower than with fungicide as a positive control. The control value with KYC 1126 was 13.4% and 41.0%, whereas that with the fungicide was 52.3% and 63% in 2009 and 2010, respectively. Although anti-anthracnose activity of KYC 1126 was not maintained for long time in the field, the bacteriolytic myxobacterium KYC 1126 could be a prospective biocontrol agent.

도열병균 새로운 레이스 KI-1117a에 의한 저항성 품종의 이병화 및 레이스 분포변동(1999~2000) (Breakdown of Resistance of Rice Cultivars by New Race KI-1117a and Race Distribution of Rice Blast Fungus During 1999~2000 in Korea)

  • 최근(1999∼2000년도) 남부지방에서 벼도열병이 심하게 발생된 원인을 분석한 결과, 잎도열병과 이삭도열병의 발병정도는 1998년에 비해 각각 1.5, 2.9배 증가되었다. 이시기는 밀양95호를 모본으로 육성된 대산, 동안, 일미벼의 재배면적 비율이 전국평균 11%(1998)에서 38.4%(2000)로 급격히 증가되었으며, 특히 전남지역에서는 2000년도 벼재배면적의 85%이상을 차지하였다. 농가포장의 도열병균 레이스 분포는 1998년의 우점레이스 KJ-301이 1999∼2000년도에는 KI-1117, KI-1113, KJ-105 레이스로 바뀌었으며, 남부지방에서의 세 레이스 분포비율은 1998년 1.7%에서 1999년에는 30%로 증가되었다. 대산, 동안, 일미벼는 기존의 주요 레이스에 대하여 저항성이었으나, KI-1117등 3개 레이스에 대해서는 균주에 따라 이병성과 저항성 반응을 나타내고 있었다. 따라서, 대산벼를 참고품종으로 하여 저항성 반응을 보이는 균주는 기존의 KI-l117, KI-1113, KJ-105로 분류하였으며, 이병성 반응을 보이는 균주는 새로운 레이스인 KI-1117a, KI-1113a, KJ-105a로I 명명하였다. 따라서, 최근 남부지방의 도열병 격발은 KI-1117a, KI-1113a, KJ-l05a 레이스의 급격한 증가에 따른 대산벼 등 밀양95호 모본 품종의 이병화에 의한 것이었다.
