Toxicity Decrease of Cadmium by the Pigment Produced by Azomonas agilis PY101 in the Culture of Bacterial Cells and Vero Cells

  • The morphological patterns and the cytopathogenicity time of the Vero cells induced by free $Cd^{2+}$ and pigment-bound $Cd^{2+}$ were observed by inverted microscope in order to investigate the difference of cadmium toxicity. The Vero cells induced by Hee $Cd^{2+}$ of 0.1 mM were shown to have a fatal toxic effect and the cytopathogenicity could be seen early after 6$\pm$2 hours of incubation. Partially affected cells induced by pigment-bound $Cd^{2+}$ of 0.1 mM were shown and the cytopathogenicity could be seen after 20 hours of incubation. The Vero cells grown with free 0.001 mM $Cd^{2+}$ were also affected and the cytopathogenicity could be seen after 17 hours of incubation, whereas the Vero cells grown with 0.001 mM pigment- bound $Cd^{2+}$ were unaffected. The sensitivity of Escherichia coli DH5$\alpha$ bacterial cells was also examined after a short treatment with free $Cd^{2+}$ or pigment-bound $Cd^{2+}$. About 5% of cells survived after 0.01 mM of free $Cd^{2+}$ treatment, while about 68% of cells survived after 0.01 mM of pigment- bound $Cd^{2+}$.

  • The nucleophilic substitution reactions of p-substituted benzyl bromide with l-substituted N,N-dimethylanilines in methanol and acetonitrile binary solvent mixture which is known to an isodielectric solvent system kinetically and the results are as follows. The positive charge is developed on the reaction center of the substrate and it means that the bond cleavage is preceded more than bond formation in the transition state on the analogy of Hammett px values. The bond form3tlon is not progressed in the case of electron donating substituent of substrate. However, the bond formation is much developed in the case of electron withdrawing substituent of substrate on the analogy of Hammett py values. The nucleophilic attacking ability is shown a highest at 80% (V/V) methanol content and the bond formation is well progressed at the same methanol composition on the result of a cross interaction coefficient, pxy. The result of transition state structure that is applicated to the potential energy surface model is in accord with the result that Is applicated to the reaction susceptibilities. The reaction Is subject to the polarity-polarizability term more than the hydrogen bond donor acidity term by application to the solvatochromic parameter eouation.

Characteristics of Concentration and Size Distribution of PAHs of Total Suspended Particulates in urban air (도시대기부유분진중 다환방향족 탄화수소의 농도 및 입경분포 특성)

  • In order to comprehension of the behaviour of PAHs in air which is known as carcinogens and/or mutagens suspended Particulates in ambient air were collected by Anderson air sampler from 1992. 6 to 1993. 5 in Seoul. Ten species of PAHs( Phen, An, Fl, R, Py, BaAn, BaP, Chry, BeP, DiB(a, h)An, I123p) were analyzed to understand monthly variations of PAHs distribution of PAHs concentration according to particle size, and correlation between PAHs and independent charactierstics of PAHs The highest concentration of TSP was 155.58$\mu\textrm{g}$/㎥ in May and the lowest was 60$\mu\textrm{g}$/㎥ in August. Concentration of TSP was more affected by coarse particles in spring, otherwise which was more affected by fine particles in winter. According to results of anaylsis of samples that were collected by Anderson air sampler, concentration of PAHs was more high in winter than that in summer. In almost samples collected by Anderson air sampler, concentration of PAHs was more high in coarse particles than in fine particles, but BaP well known as carcinogenic matter had more high concentration in fine particles(56-97.5%) than that in coarse particles(2.5-46%). Correlation between concentrations of TSP and PAHs was more high in fine Particles than in coarse Particles. Both fine particles and coarse particles have negative correaltion with radiation.

Characterization of a Photosynthetic Mutant Selected by Increased Formation of Poly-3-Hydroxybutyrate in Rhodobacter sphaeroides

  • Various mutants either lacking or having decreased levels of light-harvesting complexes and reaction center complex were obtained with a high frequency by an increased formation of poly-3-hydroxybutyrate (PHB) in Rhodobacter sphaeroides. One of the photosynthesis-defective mutants, PY-17, which was devoid of any of the light-harvesting complexes (B800-850, B875) as well as the reaction center complex, was analyzed further. The mutant showed substantial transcription of the puhA, pufKBALMX, and pucBAC operons coding for the structural proteins of the photosynthetic complexes although each of the activities was lower than that of the wild type. Translation of the pufKBALMX and pucBAC operons were also active in the mutant although with activities different from the corresponding one of the wild type. From these results the mutation appears to exert its effect at the post-translational level of the photosynthetic complex assembly. Complementation of the photosynthesis-defective phenotype of the mutant was achieved with an about 12-kb DNA region containing the puhA gene. The relationship between the formation of PHB and photosynthetic complexes is discussed.

Purification and Identification of an Antifungal Agent from Streptomyces sp. KH-614 Antagonistic to Rice Blast Fungus, Pyricularia oryzae

  • The actinomycete strain KH-6l4 possessed strong antifungal activity, especially antagonistic to the rice blast fungus, Pyricularia oryzae. Diaminopimelic acid (DAP) type and morphological and physiological characteristics, examined by scanning electron microscopy (SEM), indicated that KH-614 belonged to the genus Streptomyces. Antifungal agent produced by this strain was found to be most active, when the strain was cultured in the presence of glucose, polypeptone, and yeast extract (PY) medium for 6 days at $27^{\circ}C$. Based on the spectral report data, MS and NMR, the antifungal agent was identified as cyclo(L-leucyl-L-prolyl). According to the antimicrobial activity test measured by minimal inhibitory concentration (MIC), the cyclo(1eu-pro) exhibited the activity against Candida albicans IAM 4905, Mucor ramannianus IAM6218, Rhizoctonia solani IFO 6218, Aspergilus fumigatus ATCC 42202, Glomerella cingulata IFO 9767, Trichophton mentagrophytes ATCC 18749, and Trichophyton rubrum ATCC 44766, the order of MIC values were 50, 12.5, 5, 50, 25, 5, $5\;\mu\textrm{g}/ml$, respectively. Specifically, cyclo(1eu-pro) was one of the most effective elements against Pyricularia oryzae IFO 5994 with the MIC value of $2.5\;\mu\textrm{g}/ml$, thus indicating that cyclo(leu-pro) is a potential antifungal agent.

  • Dendrimers represent a new class of synthetic macromolecules characterized by a regularly branched treelike structure. Dendrimer can be made with high regularity and controlled molecular weight. Peculiar features of the dendritic geometry are the large number of end groups as well as the shape persistence in higher generations, approaching spherical geometry. One of the most peculiar characteristics of dendritic macromolecules is their controlled molecular structure and orientation, which means that they have a practical application in achieving a highly organized molecular arrangement. We attempted to fabricate a G4-48PyA dendrimer LB films containing 48 pyridinealdoxime functional end group that could form a complex structure with metal ions. Also, we investigated the surface activity of dendrimer films at air-water interface. And we have studied the electrical properties of the ultra-thin dendrimer LB films. The electrical properties of the ultra-thin dendrimer LB films were investigated by studying the current-voltage(I-V) characteristics of metal/dendrimer LB films/metal (MIM) structure. And rectifying behavior of the devices was occurred in applied field.

  • The Korea Atomic Energy Research Institute (KAERI) has been developing pyroprocessing technology for recycling useful resources from spent fuel since 1997. The process includes pretreatment, electroreduction, electrorefining, electrowinning, and a waste salt treatment system. This paper briefly addresses unit processes and related innovative technologies. As for the electroreduction step, a stainless steel mesh basket was applied for adaption of granules of uranium oxide. This basket was designed for ready handling and transfer of feed material. A graphite cathode was used for the continuous collection of uranium dendrite in the electrorefining system. This enhances the throughput of the electrorefiner. A particular mesh type stirrer was designed to inhibit uranium spill-over at the liquid Cd crucible. A residual actinide recovery system was also tested to recover TRU tracer. In order to reduce the waste volume, a crystallization method is employed for Cs and Sr removal. Experiments on the unit processes were tested successfully, and based on the results, engineering-scale equipment has been designed for the PRIDE (PyRoprocess Integrated inactive DEmonstration facility).

Dependence of Light-Emitting Characteristics of Blue Phosphorescent Organic Light-Emitting Diodes on Electron Injection and Transport Materials

  • We investigate the light-emitting performances of blue phosphorescent organic light-emitting diodes (PHOLEDs) with three different electron injection and transport materials, that is, bathocuproine(2,9-dimethyl-4,7-diphenyl-1,10-phenanthroline) (Bphen), 1,3,5-tri(m-pyrid-3-yl-phenyl)benzene (Tm3PyPB), and 2,6-bis(3-(carbazol-9-yl)phenyl)pyridine (26DCzPPy), which are partially doped with cesium metal. We find that the device characteristics are very dependent on the nature of the introduced electron injection layer (EIL) and electron transporting layer (ETL). When the appropriate EIL and ETL are combined, the peak external quantum efficiency and peak power efficiency improve up to 20.7% and 45.6 lm/W, respectively. Moreover, this blue PHOLED even maintains high external quantum efficiency of 19.6% and 16.9% at a luminance of $1,000cd/m^2$ and $10,000cd/m^2$, respectively.

  • For the development of antifungal agents, modification of naftifine which exhibits significant antimycotic activity was performed by replacing the naphthalene ring of it to hete ro cyclic rings such as morpholine. benzothiazole, piperidine and pyridine derivatives. The synthesized compounds were tested in vitro antifungal activity against five different fungi with naftifine as a comparative antimycotic molecule. From the biological evaluation two compounds, (E)-N-(3-phenyl-2-propenyl)-N-(4-piperidinylmethyl)amine(3d) and (E)-N-(3-phenyl-2-propenyl)-N-(3-pyridylmethyl)amine(3f) showed relatively noticeable activity(MIC=50${\mu}g$/ml). On the other hand, the other compounds had no activity.

The Effects of Multiple Recycling on Deinkability and Properties of Recycled Paper

  • This study examined the effects of multiple recycling on deinkability and properties of recycled paper from laser computer printout (LCPO). First, alkaline paper with a 20% printed area was disintegrated by TAPPI standard disintegrator at room temperature. After dewatering, the pulp was flotated and dried in oven at80$^{\circ}C$ for 24 hrs. A sequence of wetting, disintegrating, flotating and drying was one recycling cycle and this cyclic treatment was repeated from zero to five times. The recycled handsheet dropped to 90% of the original brightness after five cycles, and lost the most brightness after five cycles, and lost the most brightness in the first two cycles. However, it had a gain of 10% in opacity after five cycles as the same as the case of nonprinting. And, in this study, the method for determining residual ink(toner) content in recycled handsheets ere established by means of SEM-EDX and Py-GC. The change of residual ink percentage on recycled paper showed the effect of recycling numbers on deinkability of waste paper. A slight decrease in deinkability was noted for the recycled handsheets, which may be due to the change of fiber surface free energy connected with fiber swelling.
