Purification and Characterization of Paclitaxel from Plant Cell Cultures of Taxus chinensis in Large-Scale Process (식물세포 Taxus chinensis 배양으로부터의 Paclitaxel 대량 정제 및 특성)

  • In developing a HPLC purification process, it was hoped that a single chromatographic system would be sufficient to abtain pure paclitaxel in high yield. However, no such system was found, due in part to the complex taxoid profile of crude paclitaxel and to the rigorous nature of the product specification. A two step HPLC purification was adopted using reverse-phase separation on C(sub)18 as a first step, and normal-phase separation on silica as the final polishing step. Impurity profiles were established and maintained for paclitaxel, which identified and quantified each impurity observed in purified paclitaxel from these two steps, all impurities at or above 0.1% were identified. Results provide information for improving the quality control of paclitaxel production.

Air Purification of Smoking Booth Using Photocatalytic Process and Air Filter (광촉매공정과 필터를 이용한 흡연부스 공기정화연구)

  • The current study evaluated the air quality of the smoking booth equipped with the air purification system consisting of photocatalysts and air filters by measuring the concentrations of hazardous substances of tobacco smoke such as CO, HCHO, $CH_3CHO$, PM10 and PM2.5. To enhance the removal efficiency of hazardous substances, an infrared ray was exposed to improve the reactivity of OH radical generated from the photocatalyst toward environmental tobacco smoke (ETS) gas phase hazardous materials. It was found that the smoking booth with the air purification system improved the removal efficiency of hazardous substances containing formaldehyde by 85.2% compared to that of the smoking booth without any purification systems. In addition, the removal efficiency of the fine dust after treatment was enhanced up to 89.4%.

Microbial Activity of Gravel Intertidal Zone for Purification of Polluted Near Shore Water

  • Microbial activity of biofilm formed on the surface of gravels from intertidal zone was estimated using an aerobic respirometer system, and compared with that of suspended marine microorganisms contained in a near shore water. The maximum oxygen uptake rate of the suspended marine microorganisms was 0.15mg O$_2$/L/hr, indicating the potential of purification of polluted near shore water. For the gravels from the intertidal zone, the maximum uptake rate of oxygen was affected by the vertical positions, but their gross value was 0.77mg O$_2$/L/hr, which was around 5.1 times higher than the purification potential of polluted near shore water by the microorganisms contained in the near shore water. The nitrogen removed by the gravels from the intertidal zone and the marine microorganisms was about 1/20-1/39 times of the total consumption of oxygen, which was similar to that of the phosphate. The gravel intertidal zone contained lots of particulate organics, over than that in the near shore water, and this was confirmed from the large difference between total oxygen consumption and the removed soluble COD in the microbial activity test. This indicates that the gravel intertidal zone plays an important role in controlling the non-point source pollutants from land, as well as self-purification of polluted near shore water by trapping and degrading the particulate organics.

An Experimental Study on the Pond Sediment and Water Quality Purification using Oxygen Solubilization Device(OSD) System (산소용해수를 이용한 호소 저질 및 수질개선에 관한 실험적 연구)

  • The pollution in closing water can be caused by not only artificial factor like sewage but also natural factor like elution from sediment. In this study we analyzed Sediment Oxygen Demand (SOD) for verification of sediment purification effect and sediment elution experiment as well as general items like COD, TN, TP, SS to complement and assess the effect of sediment and water quality. The experiment result showed that the release rate of OSD system were 4 times and 3 times as large as control for P and Fe respectively. SOD for operated OSD system and control were $12.18gO_2{\cdot}m^{-2}{\cdot}d^{-1}$ and $47.95gO_2{\cdot}m^{-2}{\cdot}d^{-1}$. From water qualities analyzed by COD, TN, TP, SS, chlorophyll-a, the removal efficiency increase of TN, TP, chlorophyll-a and COD were about 10~20%, 40~50% and 10% respectively. In conclusion, OSD can contribute to improvement of both the waterbody and the sediment environment effectively.

Improved recovery of active GST-fusion proteins from insoluble aggregates: solubilization and purification conditions using PKM2 and HtrA2 as model proteins

  • The glutathione S-transferase (GST) system is useful for increasing protein solubility and purifying soluble GST fusion proteins. However, purifying half of the GST fusion proteins is still difficult, because they are virtually insoluble under non-denaturing conditions. To optimize a simple and rapid purification condition for GST-pyruvate kinase muscle 2 (GST-PKM2) protein, we used 1% sarkosyl for lysis and a 1 : 200 ratio of sarkosyl to Triton X-100 (S-T) for purification. We purified the GST-PKM2 protein with a high yield, approximately 5 mg/L culture, which was 33 times higher than that prepared using a conventional method. Notably, the GST-high-temperature requirement A2 (GST-HtrA2) protein, used as a model protein for functional activity, fully maintained its proteolytic activity, even when purified under our S-T condition. This method may be useful to apply to other biologically important proteins that become highly insoluble in the prokaryotic expression system.

An Experimental Study on the Air Sterilization Performance of A Reflective Electro Magnetic Energy System (Reflective Electro Magnetic Energy(REME)를 이용한 공기살균시스템의 성능평가에 관한 실험적 연구)

  • From the point of view to prevent airborne infection-related diseases such as H1N1, SARS, and MERS, an actual application of air cleaning and purification systems including technologies like UVGI has become increasingly important. Recently, an air purification system using REME (Reflective Electro Magnetic Energy) developed in the U.S. is applied for indoor air purification and sterilization technology to counteract the outbreak of new airborne infections. In this study, an air sterilization performance experiment using REME was carried out. The results verified that air sterilization performance in the case of installing a REME system in a medical center was 31%, namely the number of floating bacteria decreased by 31% after only a five-day operation. In addition, the number of culture collections in the REME operating air conditioning systems using nonpathogenic Geobacillus stearothemophilus as a biological indicator decreased maximally to 67%. A field application of REME technology will be useful to prevent airborne infection-related diseases, especially in response to public health crises due to the advent of emerging diseases.

  • The radioactivity in the spent fuel storage pool is calculated to ensure to maintain its concentration below the permissible limit, when the storage capacity of Uljin nuclear power plant unit 1&2 is extended from 9/3 to 32/3 core using consolidated fuels in maximum density rack (MDR). For this evalulation, two models to calculate the spent fuel pool activities on the continuous and intermittent operating its purification system are developed and these results compared, The results of above two cases show that the current water purification system can not guarantee the radioactivity concentration below the design limit, 5$\times$10$^{-4}$ $\mu$Ci/ml, for the extention to 32/3 core. Therefore, it has been concluded that a modification of the current purification system is necessary to extend the spent fuel storage capacity with the above method. The alternative way suggested in this study is to increase the number of cation bed demineralizers.

Heterologous Expression and Optimized One-Step Separation of Levansucrase via Elastin-like Polypeptides Tagging System

  • Elastin-like polypeptides (ELPs) undergo a reversible inverse phase transition upon a change in temperature. This thermally triggered phase transition allows for a simple and rapid means of purifying a fusion protein. Recovery of ELPs-tagged fusion protein was easily achieved by aggregation, triggered either by raising temperature or by adding salt. In this study, levansucrase has been used as a model enzyme in the development of a simple one-step purification method using ELPs. The levansucrase gene cloned from Pseudomonas aurantiaca S-4380 was tagged with various sizes of ELPs to functionally express and optimize the purification of levansucrase. One of two ELPs, ELP[V-20] or ELP[V-40], was fused at the C-terminus of the levansucrase gene. A levansucrase-ELP fusion protein was expressed in Escherichia coli $DH5{\alpha}$ at $37^{\circ}C$ for 18 h. The molecular masses of levansucrase-ELP[V-20] and levansucrase-ELP[V-40] were determined as 56 kDa and 65 kDa, respectively. The phase transition of levansucrase-ELP[V-20] occurred at $20^{\circ}C$ in 50 mM Tris-Cl (pH 8) buffer with 3 M NaCl added, whereas the phase transition temperature ($T_t$) of levansucrase-ELP[V-40] was $17^{\circ}C$ with 2 M NaCl. Levansucrase was successfully purified using the phase transition characteristics of ELPs, with a recovery yield of higher than 80%, as verified by SDS-PAGE. The specific activity was measured spectrophotometrically to be 173 U/mg and 171 U/mg for levansucrase-ELP[V-20] and levansucrase-ELP[V-40], respectively, implying that the ELP-tagging system provides an efficient one-step separation method for protein purification.

Submerged Type Water Purification System using Hollow Fiber Microfiltration Membrane (중공사 정밀여과막을 이용한 상수처리용 일체형 시스템 개발)

  • Membrane separation process is considered as an alternative of conventional water purification system using coagulationㆍsedimentation+sand filtration. In this study, it was examined that the application possibility of Hollowfiber Microfiltration membrane for water purification process. A $20m^3/day$ scale pilot plant was used for studying the possibility of long-term operation and the stability of water quality under the optimum conditions, 0.03m/h permeate flux, filtration for 10 minutes, pause for 2 minutes(including air-scrubbing for 30 seconds), obtained by lab-scale experiment. As a result, it was proved stability of pilot plant over one year and filtrate quality(Turbidity. SS etc). Therefore, it was proved that membrane separation process using Hollowfiber Microfiltration membrane can be applied for water purification system
