A novel calcium binding protein (CaBP) was purified to electrophoretic homogeneity from Dunaliella salina. In the course of purification experiment, this CaBP was identified as a monomer and its molecular weight was about 21 kDand isoelectric point (pI) value was about 4.1 using isoelectrofocusing. This CaBP was able to bind Ca2+ even in the pressence of an excess MgCl2 and KCI both in solution. In the SDS-PAGE, the Ca2+-bound form was slower than the Ca2+-free form in the nondenaturing PAGE. This means that the CaBP undergoes conformational change in the Ca2+-bound condition. Furthermore, UV absorption spectrum and fluorescence intensity of this CaBP was investigated. UV absorption peak was appeared at about 258 nm and decreased somewhat in Ca2+-bound condition. In the measurement of fluorescence, maximum intensity was appeared at 303 nm and decreased in Ca2+-bound state, similarly as UV absorption spectrum. These show distinct changes upon Ca2+-binding, which indicate of structural and/or dynamic changes largely reminiscent of other members of the EF-hand Ca2+-binding protein family.
Toll-like receptors (TLR) are important components of innate immunity in the defense against pathogens. TLRs recognize pathogen-associated common molecular patterns. TLRs are similar to the receptors involved in defense responses in plants. TLR protein is a type 1 membrane protein, consisting of an extracellular domain containing leucine-rich repeats and a cytoplasmic domain. The cytoplasmic domain delivers ligand recognition signals that result in production of anti-microbial agents. The cytoplasmic domain (amino acid 858-1032) of toll-like receptor 9 has been expressed using methylotrophic yeast Pichia pastoris. The protein expression was confirmed by Western-blot, N-terminal sequencing and MALDl-TOF mass spectrometry. The proteins have been purified by nickel affinity, cation exchange and gel-filtration chromatography.
The results of the water purification studies using water parsley (Oenanthe javanica) were obtained as follows. The removal rate of nutrient salts increased with longer plant growth periods. The results of this study use the assumption, log(T+1) = $K(C_0-C)^A$, based on Prakish's Theory. The initial concentration was calculated from experimental data. A and K are closely related to the initial concentration. It is possible to model the concentration of residual salts, as time goes by, if concentration is constant. It was observed that water parsley neutralizes acid and alkali substances promptly. The maximum suitable neutralization period is 48 hours. But water parsley withered up in strong acid and alkali solutions within a week. The removal efficiency of Cd progresses in 2 steps, which are unrelated to the initial concentration of Cd. The first part of the curve shows the concentration rapid rate of Cd removal, followed by a levelling off. The removal rate of $NO_x-N$ in the sample water tank containing 0.5 ppm Cd was between 50~80% but the removal rate was less than 20% for the higher concentration. On the other hand, increased amounts of $PO_4-P$ in the sample water tank from the third day on suspected that $PO_4-P$ was desorbed from the water parsley. The accumulation efficiency of Cd in plant was increased in proportion to the initial concentration of Cd. The accumulation phenomenon was observed in the tanks more than 50~100 times.

Recently, mixers are being widely used in the water purification plant in order to increase the filtration efficiency. The mixer normally consists of impeller, shaft, hub, reduction gear, and the driving motor. It is one of the key design issues to confirm that the vibration caused by the rotation of the shaft should not coincide with the natural vibration frequency of the shaft itself. The vibration characteristics of the hydrofoil type mixer, which is the most widely used in real plants are evaluated through the finite element modeling and verified by experiment using the mock-up facility. The fundamental natural frequency of the mixer's shaft is found to be around 1.8 Hz which showed in good agreement with the experiment. The higher natural frequencies to the 2nd, 3rd, and 4th modes are also verified by the experiment. Thus the developed model is to be utilized for the structural design of the real mixer system.

In order to secure the stability of tap water production and supply, we have built a system that can be pre-verified before applying AI-based composite sensors to the water purification plant, which is a demonstration site. We have collected and analyzed data related to the drug input of the GO-RYEONG water purification plant for about two years from December 2019 to December 2021. The outliers of each tag were removed through data preprocessing such as outliers and derived variable, and the cycle was set as average data for 60 minutes of each one-minute period, and the model was learned using the PLS model.

A Sustainable Structured wetland Biotop (SSB) system was constructed on the floodplain of Kyungan stream in December, 2006. It purifies polluted water of Keumeo stream which flows into the stream. Water were sampled once a month at inlet and outlet from December, 2007 to December, 2008. $BOD_5$, SS, T-N and T-P were analyzed. Plant and fish species of the system were monitored twice during the period. Average influent and effluent BOD5 concentration was 6.2 and 2.2 mg/L, respectively and BOD5 removal was 50.8%. SS concentration of influent and effluent was averaged 10.1mg/L and 1.5mg/L, respectively and SS abatement amounted to 77.0%. Average influent and effluent T-N concentration was 4.9mg/L and 2.9 mg/L, respectively and T-N retention was 50.8%. T-P concentration of influent and effluent was averaged 0.386mg/L and 0.107mg/L, respectively and T-P removal amounted to 77.0%. Twenty two plant species were naturally introduced into the system, however, they didn't make up a significant portion of the plant populations compared with the planted species. Dominant plant species were in the following order; Phragmites communis > Typha latifolia > Iris pseudoacorus > Persicaria thunbergii. Five families and 15 species of fish were observed in the system including Chinese minnow (Moroco oxycephalus) which inhabits in clean water. Six more fish species were monitored in the system compared with ones living in Kyungan stream. Amphibia and reptiles accounted for 11 species of 4 orders and 7 families including Korean Salamander (Hynobius leechi) which also lives in cleanwater.
In this study, using soil brick with combined effective microorganisms and emergent plants was identified which it can increase the effect of conservation and improvement of water. Lab-test was consist of four kind of reactors and each of reactors were A(rawwater), B(soil brick), C(emergent plant) and D(soil brick+emergent plant). Iris pseudoacorus, Phargmites australis, Typha angustifolia and Zizania latifolia were used for emergent plant. Evaluation of application on various environment were performed on agricultural waterway and pond. The pH measurement test of soil brick was performed due to evaluate whether a strong alkaline water flows out of the soil brick. Result of lab-test, removal efficiency of D was better than removal efficiency of A presenting 20.9%, 27.9% 21.5%, 33.8% and 58.4% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Removal efficiency of soil brick on agricultural waterway was revealed to be 49.5%, 45.0%, 43.7%, 37.3% and 28.6% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. And removal efficiency of soil brick on the pond was revealed to be 12.7%, 10.5%, 9.32%, 10.4% and 36.3% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Result of pH measurement test of soil brick was neutral which was about 6 to 8.

The objective of this study is to develop a systematic purification plant using the metabolism of aerobic microorganisms. This system is subsequently aerated and continuously removes suspended solids and settling sludges caused by aerating pressure at the bottom of a lower pipe (i.e., Continuous Removal of Suspended solids and Settling sludges, CRSS). The CRSS plants are brought out by introducing fine air bubbles into the liquid phase of a lower pipe in the bio-reactor. These plant uses aeration pipe, with multiple inlets to sweep the floor of bio-reactor tank, instead of the conventional scraper mechanisms. The principal advantage of this system is that it can continuously remove very small or light particles that settles completely within a short time. Once the particles have been floated to the surface, they can be moved into the pipe and collected in the settling tank by sequently aerated pressure. The experimental results shows that about 99.0% of the biochemical oxygen demand(BOD), 99.3% of the suspended solid(SS), 92.3% of the total nitrogen(T-N), 99.0% of the turbidity(TU), 100% of the total coliform(TC)and ammonia was respectively removed during aerobic digestion for 9 days. These result indicates that the CRS S plants are very effective for reduction and deodorization of swine wastewater contaminants, and the efflux from CRS S can either be discharged in the river or used as nutrient solution of formulation for plant growth factories. The developed CRSS plant proved to be flexible and it can simply be adapted to any type of biological waste treatment problem.roblem.
The objective of this study was to quantitatively measure the changes in radon concentration due to ventilation and air purification plants in the lower and upper floors of a building. This study measured and compared radon concentration in the lower and upper floors of the building by using a radon meter when the room was closed, it was ventilated, and air purification plants were installed at a specific time. One-way ANOVA was conducted to evaluate the effect of treatment (i.e., closure, ventilation, and air purification plants) on radon concentration. The results of this study showed that ventilation and air purification plants significantly decreased radon concentration in the lower and upper floors of the building, but the effect of ventilation and that of air purification plants were not significantly different. Therefore, it will be possible to reduce radon concentration effectively when ventilation and air purification plants are used appropriately.

Coenzyme Q10(CoQ10) is a biological quinine compound that is widely found in living organisms including yeast, plants, and animals. CoQ10 has two major physiological activities:(a)mitochondrial electron-transport activity and (b )antioxidant activity. Various clinical applications are also available: Parkinson's disease, Heart disease, diabetes. Because of its various application filed, the market size of CoQ10 is continuously expanding all over the world. A Japanese company, Nisshin Pharma Inc. is the first industrial producer of CoQ10(1974). CoQ10 can be produced by fermentation and chemical synthesis. In several companies, these two methods are used for the production of CoQ10:chemical synthesis - Yungjin, Daewoong, Nishin Parma; fermentation - Kaneka, Kyowa, Yungjin, etc. Researchs in microbial production of CoQ10 have several steps: screening of producing microorganisms, strain development, fermentation process, purification process, scale-up process, plant production. Several strategies are available for the strain development : Random mutation and screening, directed metabolic engineering. For the optimization of fermentation process, various conditions (nutrient, aeration, temperature, culture type, etc.) are considered. Purification is one of the most important step because the quality of final products entirely depends on its purity. The production cost will be reduced and the quality of the CoQ10 will be impoved by continuous researches in strain development, fermentation process, purification process.
