Changes in taste Characteristics of Traditional Korean Soy Sauce with Ripening Period (I) - Analysis of General Compounds and Sugars - (숙성 기간에 따른 전통 간장의 맛 특성 변화(I) -일반 성분과 당류 분석-)

  • This study was carried out in order to analyze the changes in general compounds and sugars of traditional Korean soy sauce with varying Meju concentrations (Meju-water ratios of 1 : 4 and 1.3 : 4) and ripening periods (up to 2 years). Along with ripening period, pH decreased and titratable acidity, total acidity and crude protein contents increased, which were higher in 1.3 : 4 than in 1.4. Buffering power increased significantly with increasing ripening period. Crude fat content was higher in 1.3 : 4 than 1 : 4. Moisture content decreased and pure extract content increased significantly with increasing ripening period, which was high in 1.3 : 4. Salt contents increased with increasing ripening period, but decreasd in soy sauce ripened for 2 years. As the result of sugar analysis, reducing sugar showed slow increase with increasing ripening period. Among detected free sugars, galactose was present in the highest concentration and the rest were in the sequence of glucose, fructose. Total free sugar content was highest in soy sauce ripened for 150days in 1 : 4 and 210 days in 1.3 : 4.

A Study on the Electrochemical Properties of Porous Carbon Electrode according to the Organic Solvent Contents (유기용매의 함량비에 따른 다공성 탄소전극의 전기화학적 특성 연구)

  • In order to increase the surface area of electrodes for electrosorption, porous carbon electrodes were fabricated by a wet phase inversion method. A carbon slurry consisting of a mixture of activated carbon powder (ACP), polyvinylidene fluoride (PVdF), and N-methyl-2-pyrrolidone (NMP) as a solvent was cast directly on a graphite sheet. The cast film was then immersed in pure water for phase inversion. The physical and electrochemical properties of the electrodes were investigated using scanning electron microscopy (SEM), porosimetry, and cyclic voltammetry. The SEM images verified that the pores of various sizes were formed uniformly on the electrode surface. The average pore sizes determined for the electrodes fabricated with various NMP contents ranged from 64.2 to 82.4 nm and the size increased as the NMP content increased. All of the voltammograms showed a typical behavior of charging and discharging characteristic at the electric double layer. The electrical capacitance ranged from 3.88 to $5.87F/cm^2$ depending on the NMP contents, and the electrical capacitance increased as the solvent content decreased. The experimental results showed that the solvent content is an important variable controlling pore size and ultimately the capacitance of the electrode.

  • It has been well known that the alkali-aggregate reaction between the aggregates and cement paste is one of the reasons of a concrete siructre expansion. Because of a serious demage on the concrete stnicture from the expansion, in many countries, the safety of the materials is checked in laboratory by mortar-bar test and the upper limit of expansion in length is 0.1%. The prescriptions are presented in the ASTM C227 and 490 of U.S. which has been international currency and in the KS Handbook F2503, F2546 and L5107 of Korea published by Korean Standards Association. Both of the prescriptions are almost same in their contents. Actually, in the process of preparing and measuring the mortar-bar according to the prescription mentioned above, it seems that there are no problems for its own purpose but a few points are found to be improved upon the methods to increase the accuracy for laboratory work as follows. 1. The prescription of blending ratio(aggregate, cement and water) should be noted by volume not by weight. 2. It is unreasonable to measure the initial length of mortar bars after 24$\pm$2 hours at once regardless the kind of aggregates. 3. It may bring about errors in calculating the expansion ratio under the condition of the denominator value fixed as 254mm. 4. The measuring methods of specific gravity are selected according to the purposes but the pure specific gravity displays the highest accuracy among them.

  • Selective catalytic oxidation of $H_2S$ to elemental sulfur using $TiO_2/SiO_2$ catalysts was investigated in this study. The reaction test with pure $TiS_2$ and $Ti(SO_4)_2$ and cyclic temperature operation revealed that $TiO_2$ had a good resistance to sulfation and sulfidation, which are known as the main cause of catalytic deactivation in sulfur recovery process. With the increase of $TiO_2$ loading amount in $TiO_2/SiO_2$ catalysts, the conversion of $H_2S$ increased and the selectivity of elemental sulfur was very slightly decreased. As the ratio of $O_2/H_2S$ increased, the selectivity to elemental sulfur was drastically decreased. In the presence of 10 vol.% water vapor to a stoichiometric mixture of $H_2S$ and $O_2$($H_2S$= 5 vol.% O=2.5 vol.% ), both activity and selectivity of 10 wt.% $TiO_2/SiO_2$ catalyst are decreased, but it still showed more than 80% of sulfur yield.

A Fundamental Study on the Auditory Characteristics of Amberjack Seriola dumerili in the Coast of Jeju Island (제주 연안산 잿방어의 청각특성에 관한 기초적 연구)

  • In this paper, We examined auditory threshold and critical ratio of amberjack seriola dumerili, in the Jeju Island coastal waters, to find out hearing ability of the fish. The auditory threshold level, critical ratio and hearing index of amberjack were determinded by conditioning method using a sound coupled with electric shock in the condition of ambient noise or white noise in an experimental water tank. The audio-signals of pure tone and electric shock were from 80 HZ to 800 Hz and DC 7 V, respectively. Values for the critical ratios were calculated in terms of the masked thresholds using the noise projected to stable spectrum levels at all measurement frequencies of background noise. Masking noises were in the spectrum level range of 65 dB∼75 dB $(re 1{\mu}Pa\sqrt{Hz})$. The auditory thresholds of amberjack within the test the frequencies were most sensitive at 300HZ as 94.5 dB. The critical ratios of fishes ranged from 36.4 to 52.8 dB. The noise spectrum level that started masking was about 58∼72 dB within frequencies.

  • In order to elucidate the taste compounds of Korean catfish, Parasilurus asotus, free amino acids, nucleotides and their related compounds, organic bases, sugars, organic acids and minerals in the extracts of Korean catfish were analyzed, and then evaluated by sensory test of synthetic extract prepared on the basis of the analytical data. Taste panel assessments of synthetic extracts prepared with each extractive component omitted were carried out by a triangle difference test, and changes in taste profile were assessed. In free amino acid composition, glycine was dominant occupying $25\%$ of total free amino acids. The other abundant free amino acids were lysine, taurine and alanine. Judging from the result of analysis of nucleotides, IMP was dominant showing about $70\%$ of total nucleotides while ATP, ADP, AMP, inosine and hypoxanthine were low in content. Among organic bases, total creatinine was abundant and its nitrogen content occupied more than $50\%$ of the total extractive nitrogen. The amount of betaine was 18mg/100g and trace amount of trimethylamine and trimethylamine oxide were detected. The main organic acids were succinic, butyric, propionic and valeric acid. On the other hand, oxalic, fumaric, maleic, tartaric and citric acids were analyzed in trace. As for the sugars, glucose was found to be the most abundant monosaccharide. Extremely small amounts of fructoae, inositol were also detected and ribose and arabinose were trace in content. $K^+,\;Na^+,\;PO_{4}^{3-}$ and $Cl^-$ were found to be the major ions and small amount of $Ca^{2+}$ were defected. The synthetic extract, prepared with about 40 pure chemicals based on the analytical data satisfactorily revealed the natural taste of the original extract except slight difference in meaty taste and mildness. From the results of omission test the major components which contribute to produce the taste were serine, IMP, succinic acid and $PO_{4}^{3-}$.

EFFECTS OF TITANIUM SURFACE COATING ON CERAMIC ADHESION (타이타늄 표면 코팅이 도재 결합에 미치는 영향)

  • Statement of problem: The adhesion between titanium and ceramic is less optimal than conventional metal-ceramic bonding, due to reaction layer form on cast titanium surface during porcelain firing. Purpose: This study characterized the effect of titanium-ceramic adhesion after gold and TiN coating on cast and wrought titanium substrates. Material and method: Six groups of ASTM grade II commercially pure titanium and cast titanium specimens$(13mm{\times}13mm{\times}1mm)$ were prepared(n=8). The conventional Au-Pd-In alloy served as the control. All specimens were sandblasted with $110{\mu}m\;Al_2O_3$ particles and ultrasonically cleaned for 5min in deionized water and dried in air before porcelain firing. An ultra-low-fusing dental porcelain (Vita Titankeramik) was fused on titanium surfaces. Porcelain was debonded by a biaxial flexure test at a cross head speed of 0.25mm/min. The excellent titanium-ceramic adherence was exhibited by the presence of a dentin porcelain layer on the specimen surface after the biaxial flexure test. Area fraction of adherent porcelain (AFAP) was determined by SEM/EDS. Numerical results were statistically analyzed by one-way ANOVA and Student-Newman-Keuls test at ${\alpha}=0.05$. Results: The AFAP value of cast titanium was greatest in the group 2 with TiN coating, followed by group 1 with Au coating and the group 3 with $Al_2O_3$ sandblasting. Significant statistical difference was found between the group 1, 2 and the group 3 (p<.05). The AFAP value of wrought titanium was greatest in the group 5 with TiN coating, followed by the group 4 with Au coating and the group 6 with $Al_2O_3$ sandblasting. Conclusion: No significant difference was observed among the three groups (p>.05). The AFAP values of the cast titanium and the wrought titanium were similar. However the group treated with $Al_2O_3$ sandblasting showed significantly lower value (p<.05).

Phase Behavior and Structural Analyses of the THF + H2 Binary Clathrate Hydrate (THF + H2 이성분계 크러스레이트 하이드레이트의 상거동 및 구조 분석)

  • In this study, we investigated that hydrate formation and phase behavior of the $THF+H_2$ binary clathrate hydrates. In order to confirm the binary clathrate hydrate formation we employed the Raman and NMR spectroscopies that are known to be quite powerful tools, particularly for examining the cage occupancy pattern of guest molecules. In addition, we traced the P-T profiles from clathrate hydrate formation and dissociation process, which made it possible for the resulting phase equilibrium boundary to be clearly established. In the binary clathrate hydrate matrix we further identified that the relatively large THF molecules can only occupy the large $5^{12}6^4$ cavities, while the small $H_2$ molecules are entrapped in the empty $5^{12}$ cavities in structure-II, making the hydrate to be stable above 273 K even at relatively low pressure condition. Considering that pure $H_2$ hydrate can be produced at the extreme pressures higher that 1,000 bar, we can conclude that the water-soluble second guest inclusion induces $H_2$ storage and transportation to be readily achievable under much milder conditions.

An in vitro study of immune activity by β-1,3/1,6-glucan isolated from Aureobasidium pullulans (Aureobasidium pullulans으로 부터 분리한 β-1,3/1,6-glucan의 면역활성의 연구)

  • ${\beta}$-Glucan is a natural compound contained in cell walls of yeast or fungi, and cereal's fiber. It is also known to boost the immune system in human. Aureobasidium is a producer of water-soluble ${\beta}$-1,3/1,6-glucan. In this study, natural killer (NK) cell and macrophage activity were tested to investigate the effects of ${\beta}$-1,3/1,6-glucan isolated from A. pullulans on immune activity. Activation of NK cell was increased about 63-39% by the treatment of $10-200{\mu}g/mL$ ${\beta}$-1,3/1,6-glucan than control. Besides, only $10{\mu}g/mL$ of ${\beta}$-1,3/1,6-glucan was enough to boost activation of NK cell. Phagocytosis of macrophage was increased to 15~21% by the treatment of $10{\sim}200{\mu}g/mL$ of ${\beta}$-1,3/1,6-glucan than zymosan-treatment. In LP-BM5 proliferating inhibition test, relative mRNA level of LP-BM5 virus was decreased in ${\beta}$-1,3/1,6-glucan-treated cell about 36~74% than control. The decline of LP-BM5 mRNA level appeared to depend on the concentration of ${\beta}$-1,3/1,6-glucan. These results suggest that pure ${\beta}$-1,3/1,6-glucan from A. pullulans might be contributing to enhancement of immune activity through the activation of NK cell and phagocytosis of macrophage. Moreover, treatment of the ${\beta}$-1,3/1,6-glucan could increase the resistance to virus infection such as LP-BM5 through the restraining of the multiplication.

Effect of Solubilizing and Microemulsifying Excipients in Polyethylene Glycol 6000 Solid Dispersion on Enhanced Dissolution and Bioavailability of Ketoconazole

  • Polyethylene glycol (PEG) 6000-based solid dispersions (SDs), by incorporating various pharmaceutical excipients or microemulsion systems, were prepared using a fusion method, t o compare the dissolution rates and bioavailabilities in rats. The amorphous structure of the drug in SDs was also characterized by powder X-ray diffractometry (XRD) and differential scanning calorimetry (DSC). The ketoconazole (KT), as an antifungal agent, was selected as a model drug. The dissolution rate of KT increased when solubilizing excipients were incorporated into the PEG-based SDs. When hydrophilic and lipophilic excipients were combined and incorporated into PEG-based SDs, a remarkable enhancement of the dissolution rate was observed. The PEG-based SDs, incorporating a self microemulsifying drug delivery system (SMEDDS) or microemulsion (ME), were also useful at improving the dissolution rate by forming a microemulsion or dispersible particles within the aqueous medium. However, due to the limited solubilization capacity, these PEG-based SDs showed dissolution rates, below 50% in this study, under sink conditions. The PEG-based SD, with no pharmaceutical excipients incorporated, increased the maximum plasma concentration (C$_{max}$) and area under the plasma concentration curve (AUC$_{0-6h}$) two-fold compared to the drug only. The bioavailability was more pronounced in the cases of solubilizing and microemulsifying PEG-based SDs. The thermograms of the PEG-based SDs showed the characteristic peak of the carrier matrix around 60$^{\circ}C$, without a drug peak, indicating that the drug had changed into an amorphous structure. The diffraction pattern of the pure drug showed the drug to be highly crystalline in nature, as indicated by numerous distinctive peaks. The lack of the numerous distinctive peaks of the drug in the PEG-based SDs demonstrated that a high concentration of the drug molecules was dissolved in the solid-state carrier matrix of the amorphous structure. The utilization of oils, fatty acid and surfactant, or their mixtures, in PEG-based SD could be a useful tool to enhance the dissolution and bioavailability of poorly water-soluble drugs by forming solubilizing and microemulsifying systems when exposed to gastrointestinal fluid.
