LIPUS Promotes Endothelial Differentiation and Angiogenesis of Periodontal Ligament Stem Cells by Activating Piezo1

  • Background and Objectives: Low-intensity pulsed ultrasound (LIPUS) promotes differentiation and regulates biological functions of various stem cells, but its effect on the endothelial differentiation of periodontal ligament stem cells (PDLSCs) is unclear. This study investigated the effect of LIPUS on endothelial differentiation and angiogenesis in PDLSCs and the role of the mechanically sensitive ion channel Piezo1 in this process. Methods and Results: PDLSCs obtained from healthy people were used for endothelial induction, and 10 ㎍/ml lipopolysaccharide (LPS) was used to simulate the inflammatory state. The induced cells were treated with LIPUS (50 mW/cm2, 1.5 MHz) to study its effect on the endothelial differentiation of PDLSCs and the tube formation of differentiated cells. PCR, flow cytometry, immunofluorescence, and Matrigel tube formation assays were used to detect the differentiation and tube formation of PDLSCs. GsMTx4 was used to inhibit the expression of Piezo1, and the role of the Piezo1 pathway in the endothelial differentiation and microvascular formation of PDLSCs after LIPUS treatment was studied. The data showed that LIPUS increased endothelial differentiation and angiogenesis in PDLSCs under inflammatory or noninflammatory conditions. The use of an inhibitor weakened the effect of LIPUS. Conclusions: This study demonstrated that LIPUS can activate the expression of Piezo1 and promote the endothelial differentiation and microvascular formation of PDLSCs.

Low-Intensity Pulsed Ultrasound Promotes Healing with Increases Collagen Deposition and Collagen mRNA Expression in Skin Wound of Rat (저강도 맥동초음파에의한 피부 상처 치유 촉진과 아교질 축적 및 아교질 mRNA 발현 증가)

  • 목적: 본 연구는 저강도 맥동 초음파적용이 흰쥐의 전층 상처 치유와 아교질 축적 및 아교질 mRNA발현에 미치는 영향을 규명할 목적으로 시행하였다. 방법: 12마리의 Sprague-Dawley계 흰쥐를 저강도 맥동 초음파군(n=6)과 대조군(n=6)에 무작위 배정하고 등에 $19.63mm^2$ 크기의 전층 적출 상처를 만든 다음 저강도 맥동 초음파군은 3 MHz, 순환주기 20%, SATA 강도 $0.4W/cm^2$로 1일 1회, 1회 5분씩 초음파를 적용하고, 대조군은 가짜 초음파를 적용하였다. 7일간 처치 후 초음파군과 대조군의 아교질 축적, 아교질 mRNA 발현, 상처치유율, 절반치유시간을 비교하였다. 결과: 초음파군의 아교질 축적(p<05)과 아교질 mRNA 발현(p<.01)이 대조군보다 유의하게 증가하였고, 상처치유율(p<.05)과 절반치유시간(p<.0)도 초음파군의 대조군보다 유의하게 빨랐다. 결론: 본 연구에서 전층 상처에 저강도 맥동 초음파를 적용한 결과 상처 치유가 촉진되었고 육아조직에 아교질 축적이 증가하였다. 이러한 결과는 맥동 초음파의 기계적 자극이 제 1형 아교질 mRNA 전사활동을 촉진시키는 것으로 사료된다.

Effects on Functional Recovery of Eccentric Exercise-Induced Muscle Damage by Phonophoresis of Piroxicam Gel (피록시캄 겔의 음파영동이 원심성 운동-유발 근 손상의 기능회복에 미치는 효과)

  • This study was investigated the effects on functional recovery of eccentric exercise-induced muscle damage by phonophoresis transdermal permeation of piroxicam gel and observed the change of amplitude at muscle action potential. Through eccentric exercise-induced muscle damage, performed healthy men and women take eccentric resistance exercise and measured action, potentials. The subjects were divided into three groups of four men each 24 hour, 48 hour, 72 hour. The results of this were as follows: 1. Change of maximal action potential at maximal voluntary contraction : The phonophoresis group was increase more than control group and gel group. 2. Change of average action potential at maximal voluntary contraction : The gel group was increase more than control group and phonophoresis group. 3. Change of maximal action potential at pain subthreshold voluntary contraction : The phonophoresis group was increase more significantly than control group and gel group. 4. Change of average action potential at pain subthreshold voluntary contraction : The phonophoresis group was increase more significantly than control group and gel group. In conclusion, the change of muscle action potential amplitude by eccentric exercise-induced muscle damage showed that the phonophoresis by pulsed ultrasound of piroxicam gel was improved the recovery of muscle function.

Effect of Elastic and Non-elastic Taping on Pain, Balance, and Left / Right Step Time Difference in Patients with Acute Ankle Sprain (급성기 발목 염좌 환자에게 탄력, 비탄력 테이핑이 통증, 균형, 좌우 걸음 시간 차이에 미치는 영향)

  • PURPOSE: This study was conducted to investigate the effects of pain, balance, and left / right step time difference between elastic taping and non-elastic taping in patients with acute ankle sprain. METHODS: The subjects were patients with acute ankle sprains who had been injured within 1 day. A total of 30 subjects were divided into three groups (CG: control group, EG1: experimental group 1, EG2: experimental group 2) of 10 people. The intervention period was 3 days. In the CG, only physical therapy (cryotherapy+pulsed ultrasound) was performed, while physical therapy and elastic taping were applied in EG 1 and physical therapy and non-elastic taping were applied in EG 2. RESULTS: Changes in pain, balance and left / right step time difference following intervention decreased significantly in all three groups. After intervention, the balance between the groups differed significantly between the CG and the EGs. In addition, the left / right step time difference differed significantly between the CG and EGs, as well as between the EG 1 and the EG 2. CONCLUSION: Physiotherapy is helpful for treatment of pain associated with acute ankle sprain. Taping is considered to be a way to provide more balance and gait ability.

  • Synthetic zinc wave employs Pulsed plane wave as transmit beam with linear time delay curve. The received echoes in different transmit directions at different transmit times are superposed at imaging Points with Proper time delay compensation using synthetic focusing scheme. This scheme. which uses full aperture in transmit, obtains a high SNR image, and also features high lateral resolution by using two way dynamic focusing at all imaging depths. In this Paper, we consider the Problems in realization of synthetic zinc wave. Also. we have applied the scheme to obtain phantom and in-vivo images using a linear array of 5 MHz. In phantom test. experimental images show high resolution over a more extended imaging depth than conventional fixed Point transmit and receive dynamic focusing schemes In-vivo images show that the resolution could not overcome conventional focusing systems because of motion blurring and(or) aberration of tissue. but the frame rate tan be increased by a factor of more than 5 compared to conventional focusing schemes. with competitive resolution at all imaging depths .

Signal-to-noise Ratio in Time- and Frequency-domain Photoacoustic Measurements by Different Frequency Filtering (주파수 필터링 함수에 따른 시간 및 주파수 영역 광음향 측정에 대한 신호 대 잡음비 분석)

  • We investigate the signal-to-noise ratios (SNRs) of time-domain (i.e. pulsed illumination) and frequency-domain (i.e. chirped illumination) photoacoustic signals measured by a spherically focused ultrasound transducer for spherical absorbers. The simulation results show that the time-domain photoacoustic SNR is higher than that of frequency-domain photoacoustic signals, as reported in the previous literature. We understand the reason for this SNR gap between the two measurement modes by analyzing photoacoustic-signal spectra, considering the incident beam energy controlled by the maximum permissible exposure. As the result of this approach, we find that filtering off the DC term in the chirped signal's spectrum improves frequency-domain photoacoustic SNRs by up to approximately 5 dB. In particular, it is observed that photoacoustic SNRs are highly sensitive to an upper-frequency value of frequency filtering functions, and the optimal upper-frequency values maximizing the SNR are different in time- and frequency-domain photoacoustic measurements.

A Study on the Fetal Umbilical Artery Doppler Blood Flow Velocity Waveforms in Normal Pregnancy (정상 임신에서 Doppler 초음파를 이용한 제대동맥 혈류속도 파형에 관한 연구)

  • Noninvasive techniques for antenatal detection of the fetal development and well-being such as biophysical profile, non-stress and stress test remain major challenges in modem obstetric practice. To obtain and analyze umbilical artery velocity waveform by pulsed-wave doppler ultrasound, a total of 160 determinations were carried out on 157 normal pregnant women between 16th to 41st week gestation. The ratio of peak systolic to end-diastolic flow velocity(S/D ratio), pulsatility index and resistance index were measured as indices of the resistance in feto-placental circulation. The results were as follows : As gestation advances, the, mean values for peak systolic and end-diastolic velocities raised progressively. As gestation advances, the mean values for the S/D ratio declined progressively, exhibiting high diastolic flow velocity caused by low resistance. Pulsatility index, and resistance index were also declined progressively, as gestation advances. The analysis of umbilical artery blood flow velocity waveforms provides a new noninvasive technique to evaluate fetal development and well-being, and may be expected a reliable method for assessment of fetal life.
