Puerarin and daidzin have been isolated from Puerariae thunbergiana Benth. Structures were determined by spectroscopic methods. Compounds showed weak antimicrobial activity against S. mutans, S. epidermidis, S. aureus, and C. albicans (MIC, $800{\mu}g/ml$). However, these compounds were not antioxidative. Puerariae thunbergiana Benth. extracts against microorganisms were evaluated in terms of the minimum inhibitory concentrations (MIC). In general, C. albicans was stronger antimicrobial activity than the other microorganisms. The antioxidative activity of was observed in the etyl acetate extract ($IC_{50},\;119.87{\pm}0.16\;{\mu}g/ml$). The DPPH radical scavenging effect ($IC_{50},\;1,673.3{\pm}0.54\;{\mu}M$) of the puerarin was comparable with that of synthetic antioxidant, BHA $(IC_{50},\;88.39{\pm}1.1){\times}10^{3}{\mu}M$. These results suggest that puerarin and daidzin have a potential antimicrobial activity.

Two isoflavone C-glucosides, puerarin (1) and PG-3 (2), a but-2-enolide, $({\pm})-puerol$ B (3), two isoflavone O-glucosides, daidzin (4) and genistin (5), and three pterocarpans, (-)-medicarpin (6), (-)-glycinol (7) and (-)-tuberosin (8), were isolated from a MeOH extract of the roots of Pueraria Iobata, using an in vitro bioassay based on the inhibition of the formation of advanced glycation end products (AGEs) to monitor chromatographic fractionation. The structures of 1-8 were determined by spectroscopic data interpretation, particularly by 1D- and 2D-NMR studies, and by comparison of these data with values in the literature. All of the isolates (1-8) were evaluated for their inhibitory activity on AGEs formation in vitro. Of these, puerarin (1), PG-3 (2), and $({\pm})-puerol$ B (3) exhibited more potent inhibitory activity than the positive control aminoguanidine.

To assess the estrogenicities of Korean arrowroot (Pueraria thunbergiana) the contents of nine phytochemicals which are known to present were analyzed by HPLC. Also the estrogenecities of these phytochemicals were assayed using estrogen receptor dependent transcriptional expression assay. Daidzein and puerarin are major compounds in arrowroot, which were contained in the order of: root>stem>leaf>flower>seed. Among the assayed phytochemicals daidzein, genistein, biochanin A, formononetin, puerarin and genistin were highly estrogenic. The total estrogenicities in different parts of arrowroot and those by regions in Korea were also assessed. Roots had the highest estrogenic effects. The estrogenicities also were shown in stem and leaf. The differences in the estrogenicities of arrowroots by regions in Korea were shown. These results demonstrated that the estrogenic phytochemicals content was the highest in Korean arrowroot and also present in stem and leaf.
This study was designed to evaluated the nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice. The quantitative analytical method for major antioxidants, isoflavones, puerarin, catechine and caffeine in galgun (Puerariae Radix) and green tera extract were established by HPLC. Contents of isoflavones, such as daidzin, genistin, daidzein and genistein were 4.23g/100g, 0.13g/100g, 0.07g/100g, and 0.03g/100g, and puerarin contents was 8.99g/100g, respectively. The total catechins and caffeine contents of green tea extract were 49.24g/100g and 6.53g/100g. The nitrite scavenging ability of galgun extract (pH 1.2, 100mg/ml) was 98.07% and it was higher than those of other extracts. It was the highest at the pH 1.2 and more than 64% in 25~100mg green tea extract, and was dependents on pH and concentration of the samples. The hepatoprotective effects of an aqueous extract from the root of gal gun and standard puerarin were evaluated against lead acetate and cadmium-induced liver damage in mice. Galgun extract and standard at a dose of 100mg/kg and 10mg/kg, 50mg/kg were administered orally once daily for successive 5 days and then a lead acetate and cadmium were orally at 3 hrs after the every day administration of galgun. The substantially elevated serum enzymatic activities of alanine and aspartate aminotransferase were due to lead acetate and cadmium treatment was dose dependently restored to the near normal level. In addition, galgun extract also significantly prevented the elevation of hepatic malon-dialdehyde formation in the liver of lead acetate and cadmium intoxicated mice in a dose-dependent manner. The results of this study clearly indicated that green tea and galgun extracts had nitrite scavenging ability and galgun extract had potent hepato-protective effects against lead acetate and cadmium-induced hepatic damage in mice and standard puerarin was also showed similar to the results of the galgun extracts.
Puerariae Radix is one of the medicinal plants used in oriental medicine for hangover, It has been claimed for several pharmacological effects including anti-alcohol abuse, antidipsotropic activity and anti-alcohol intoxication. In connection with Puerariae Radix effects, an activity-guided purification of active substance on alcohol dehydrogenase (hnH) was carried-out. The most active compound was isolated as puerarin (C$_{21}$H$_{20}$ O$_{9}$ ), molecular weight 416. Puerarin inhibited ADH noncompetitively against ethanol or NAD$^{+}$./.
In vitro cytotoxic activities of cisplatin combined with Chungpesagan-tang or puerarin, which were treated with or without human intestinal bacteria, were measured. When cisplatin was combined with Chungpesagan-tang and its ingredient treated without intestinal bacteria, they did not affect the in vitro cytotoxicity of cisplatin against tumor cell lines. However, when cisplatin was combined with intestinal bacteria-treated Chungpesagan-tang and its ingredients, the cytotoxicities against SNU C4, L1210, A549 and P388 tumor cell lines were synergistically increased. Puerarin, which was isolated from Puerariae Radix, did not show in vitro cytotoxicity. However, its metabolite, daidzein, showed potent cytotoxicity against tumor cell lines and was synergistic by the combined usage of cisplatin. These results suggest that natural glycosides are not only prodrugs which can be transformed to active compounds by intestinal microflora, but the combined usage of cisplatin with natural components, such as daidzein, and herbal medicinal polyprescriptions, such as Chungpesagan-tang, may be a new method for prevention and minimization of the toxicity of cisplatin.
Isoflavonoid production in cell cultures of Pueraria tuberosa as influenced by an angiospermic parasite, Cuscuta reflexa, was studied. During the time course, maximum isoflavonoid content was recorded when Cuscuta elicitor was added on day 15 of culture. Among various concentrations of elicitor tried, $1g\;l^{-1}$ of Cuscuta elicitor was found to be the most effective. The optimized elicitation conditions were used in vessels of varying capacity where maximum yield of ${\sim}91mg\;l^{-1}$ of isoflavonoid was recorded in a 2-l bioreactor which was about 19% higher than the control cultures. In this case, puerarin content increased up to $11mg\;l^{-1}$ which was 580% higher that the value recorded in the control cultures. In the bioreactor, 8 days of elicitation was optimal for the high accumulation of isoflavonoid, giving productivity of ${\sim}4mg\;l^{-1}\;day^{-1}$. The study showed persistent high isoflavonoid yield even during scale-up. Use of a preparation of Cuscuta reflexa as an elicitor is reported for the first time. The increase in isoflavonoid content was elicitor dose-dependent and can be explored to trigger high yields of isoflavonoid/secondary metabolites in production.
Pueraria lobata is a perennial legume plant that produces a variety of isoflavones, such as puerarin, daidzin and daidzein. These are metabolized to equol via dihydrodaidzein and tetrahydrodaidzein by the bacterial fermentation of natural isoflavone sources in the human intestines. In this study, we described the growth and accumulation of isoflavone in the hairy root of the Korean wild arrowroot according to the culture period, as well as dihydrodaidzein biosynthesis in hairy root extracts fermented with Pediococcus pentosaceus. Daily proliferation was best in DY1 cultured for 1 week. DY1 showed significant differences in daily production of puerarin and daidzin+daidzein, as compared to DJ7; furthermore, both were best in DY1 cultured for 1 week. The hairy root extract was fermented successfully with P. pentosaceus with confirmed production of dihydrodaidzein, an equol precursor formed by biotransformation. The results indicated that the growth of hairy roots and isoflavone accumulation in the hairy roots is best 1 week after culture. These results are expected to contribute to the mass production of hairy root and isoflavones as equol precursors from the Korean wild arrowroot and provide a basis for equol production by biotransformation in vitro.
For comparing with flavones/flavonols, isoflavonoids isolated from Pueraria radix and chemically synthesized from biochanin A were evaluated for the antiinflammatory activity using mouse ear edema test, Isoflavonoids such as daildzein and puerarin hsowed the significant antiinflammatory activity at a dose of 2 mg/mouse, although their activity was generally less than that of flavones/flavonols. 7-O-Substitution of biochanin A was not facorable for the antiinflammatory activity.
This study was performed to investigate the effect of Puerariae radix-ethanol extracts rich in isoflavone on the antio-xidative system of rats. For this purpose, first, Puerariae radix was extracted with ethanol, and its total isoflavone and puerarin contents were analysed. Second, female Sprague Dawley rats were fed for 6 weeks with four diets which were based on AIN96G diet and supplemented with Puerariae radix-ethanol extracts to contain isoflavone. The isoflavone contents of four experimental diets were 0 mg, 500 mg, 1,000 mg, 2,000 mg per kg diet, respectively (control, P0.05%,P0.1%, P0.2%). Liver and erythrocyte activities of antioxidative enzyme such as superoxide dismutase (SOD), catalase,glutathione peroxidase (GSHpx) were measured. Also, plasma and liver malondialdehyde (MDA) concentrations, liver glutathione (GSH) and oxidized glutathione (GSSG) concentrations were measured. The total isoflavone content of Puerariae radix-ethanol extract was 3067.6 mg per 100 g extract and the content of puerarin was 2557.4 mg per 100 g extract. The erythrocyte activities of GSH-Px and catalase were higher in group P0.1% and P0.2%. But SOD activity of erythocyte did not show any difference by the Puerariae radix-ethanol extract supplementation in diet. The activity of SOD in liver increased significantly by the supplementation of extract, showing highest level in P0.1% group. The liver GSH concentration increased significantly in group of P0.05%, P0.1%, and P0.2% compared with control group (p <0.05). The GSSG concentration in liver showed no difference by the supplementation of Puerariae radix extract from the control group, except P0.2% group. The plasma MDA concentration did not show any significant differences by the extract supplementation. But the liver MDA concentration decreased by the extract supplementation, showing the lowest level in P0.1 % diet group. These results suggest that the supplementation of Puerariae radix-ethanol extract can inhibit lipid peroxidation in liver and enhance the antioxidative defense competence of rats.
