Immunomodulatory Effect of Pueraria lobata on the Functional Activation of Macrophages by Lipopolysaccharide Treatment (그람음성균 유래 lipopolysaccharide로 활성화된 대식세포의 기능 수행시 갈화 [Pueraria lobata (Willd.) Ohwi]의 조절작용)

  • Pueraria lobata (Willd.) Ohwi was investigated to check its modulatory effects on the activation of macrophages upon inflammatory conditions treatment. For this purpose, we examined several inflammatory responses such as nitric oxide (NO) production, reactive oxygen species (ROS) generation, cytoprotection and phagocytosis under the treatment of methanol extract from P. lobata (Pl-ME). Pl-ME dose-dependently blocked NO production in lipopolysaccharide (LPS)- stimulated RAW264.7 cells but not sodium prusside (SNP)-generated NO release. The NO inhibition seemed to be due to blocking inducible NO synthase (iNOS), since Pl-ME suppressed its expression in a NF-${\kappa}B$-independent manner. Similarly, this extract also effectively protected RAW264.7 cells from LPS-induced cytotoxicity. However, Pl-ME did not block ROS generation and rather it enhanced. Finally, this extract negatively modulated FITC-dextran uptake. Therefore, our data suggested that Pl-ME may be involved in negatively regulating some macrophage-mediated inflammatory responses such as NO production and phagocytic uptake.

Immunostimulatory effects of the Pueraria lobata flower extract via MAPK signaling in RAW264.7 cells

  • Objectives: In this study, we investigated the effects of Pueraria lobata Ohwi flower extracts (PLFE) on macrophages and their underlying mechanism(s) of action. PLFE increased the production of NO and cytokines (IL-6 and TNF-𝛼) in a dose-dependent manner, indicating its immunostimulatory property. Furthermore, PLFE upregulated iNOS, COX-2, and mitogen-activated protein kinase (MAPK) signaling in RAW264.7 cells. Additionally, PLFE enhanced the phosphorylation of I𝜅B𝛼 and subsequent I𝜅B𝛼 degradation, thereby enabling the nuclear translocation of NF-𝜅B. Taken together, these findings demonstrate that the immunostimulatory effects of PLFE are mediated by the nuclear translocation of the p65 subunit of NF-𝜅B and subsequent secretion of cytokines (IL-6 and TNF-𝛼), upregulation of iNOS and COX-2, and stimulation of MAPK signaling (JNK, ERK, and p38). Thus, PLFE may be a potential immunostimulatory therapeutic.

Protective effects against alcoholic liver damage: potential of herbal juice (HJ), blend of Zingiber officinale Roscoe and Pueraria lobata Ohwi extracts

  • Objectives : Alcohol-induced liver disease advances as to reactive oxygen species (ROS) and cellular lipid peroxidation increase. We examined the hepatoprotective effects of Zingiber officinale Roscoe rhizome extract (ZR), Pueraria lobata Ohwi flower extracts (PF), and a newly developed herbal juice (HJ), which was a combination of ZR and PF extracts, against ethanol-induced hepatotoxicity. Methods: The study utilized the human hepatoma cell line HepG2 cells to validate the hepatoprotective effect of HJ (50~200 ㎍/mL) against ethanol (EtOH, 700 mM)-induced liver damage. Results: HJ effectively reduced the protein expression of sterol regulatory element-binding transcription factor 1, adiponectin, and AMP-activated protein kinase in EtOH-induced HepG2 cells. The levels of ROS, total cholesterol, and triglycerides, which are the result of various synthesis and lipogenesis processes induced by EtOH in the liver, were reduced by HJ. Furthermore, the activities of alcohol dehydrogenase and aldehyde dehydrogenase, enzymes linked to alcohol degradation, were more effectively downregulated by HJ treatment compared to treatment with ZR and PF alone, all without causing cytotoxic effects. Conclusions: HJ protects the liver by inhibiting EtOH-induced lipogenesis, lowering ROS generation, and improving alcohol degradation, which is more effective than ZR and PF alone. Further, in vivo experiments can offer additional evidence regarding the effectiveness, safety, and underlying mechanism of action of HJ.

Chemical Components from the Stems of Pueraria lobata and Their Tyrosinase Inhibitory Activity

  • Phytochemical investigation of the stems of Pueraria lobata (Wild) Ohwi (Leguminosae), led to the isolation of eighteen known compounds: ${\beta}$-amyrone (1), (+)-pinoresinol (2), (+)-syringaresinol (3) $(+)-syringaresinol-O-{\beta}-{\small{D}}-glucoside$ (4), (+)-lariciresinol (5), (-)-tuberosin (6), naringenin (7), liquiritigenin (8), isoliquiritigenin (9) genistein (10), daidzein (11) daidzin (12) daidzein 4',7-diglucoside (13) 2,4,4'-trihydroxy deoxybenzoin (14), S-(+)-1-hydroxy-3-(4-hydroxyphenyl)-1-(4-hydroxy-2-methoxy-phenyl)propan-2-one (15), methyl $2-O-{\beta}-{\small{D}}-glucopyranosylbenzoate$ (16), pyromeconic acid $3-O-{\beta}-{\small{D}}-glucopyranoside$ 6'- (O-4''-hydroxy-3-methoxybenzoate) (17), and allantion (18). The chemical structures of these compounds were elucidated from spectroscopic data and by comparison of those data with previously published results. The effects of isolated compounds on mushroom tyrosinase enzymatic activity were screened. The results indicated that, chloroform extract of P. lobata stems turned out to be having tyrosinase inhibitory effect, and only compounds 5, 8, 9, and 11 showed enzyme inhibitory activity, with $IC_{50}$ values of $21.49{\pm}4.44$, $25.24{\pm}6.79$, $4.85{\pm}2.29$, and $17.50{\pm}1.29{\mu}M$, respectively, in comparison with these of positive control, kojic acid ($IC_{50}\;12.28{\pm}2.72{\mu}M$). The results suggest that P. lobata stems extract as well as its chemical components may represent as potential candidates for tyrosinase inhibitors.

Genetic Variations and Phylogenetic Relationship of and Pueraia lobata Ohwi (Fabaceae) and Related Taxa by RAPD Makers (RAPD분자마커를 이용한 칡(콩과) 및 근연분류군의 유전적 변이 및 유연관계)

  • RAPD analyses were performed to investigate genetic relationships and useful molecular maker for 3 species and their 17 regional populations of the Pueraria lobata and related taxa. The length of amplified DNA fragments ranged from 200 to, 2,800 bp. Two hundred and eight scorable polymorphic makers and three scorable monomorphic makers were found from the PCR reactions with 15 random oligo primers, and those were analyzed by Nei's genetic distance coefficient. Based on the UPGMA phenogram from RAPD analyses, two major groups (9 populations from Korea; 3 populations from foreign countries) were recognized. And it showed distinct genetic differences from related taxa. The RAPD results was very useful to define the samples by geographical distribution and to discuss the relationships among the populations and their related taxa of the Pueraria lobata.

Pueraria lobata Ohwi as an Osteoporosis Therapeutics (칡의 부위별 골다공증 치료효과)

  • It is reported that Pueraria Radix contains phtoestrogens whereas flower, and bud of Pueraria lobata Ohwi were not known. In the present study, we determined the amount of phytoestrogen in each portion of P. lobata Ohwi and carried out therapeutic effects of osteoporosis. The amounts of genistein, daidzein, and formononetin in Pueraria Radix (PR), Pueraria Flos (PF), and young Pueratia Folium (PL) were quantitated using a HPLC system. Proliferation of osteoblast and growth inhibitory effect on osteoclast were measured in order to screen their effects on osteoporosis. Proliferation of osteoblast-like cells (Saos-2) was analyzed by both MTT methods and alkaline phosphatase (ALP) assays. Growth inhibitory effect on osteoclast was also detected as Tartrate resistant acid phosphatase (TRAP) assay. Ovariectomized rat as an in vivo animal model was selected and administrations of PR were 1 g/kg/day (PR-1) and 5 g/kg/day (PR-5) for 9 weeks, respectively. Trabecular bone areas (TBAs) of tibia and lumbar were analyzed usibg histomorphological methods. Results show that PR contains the highest level of daidzein ($10435{\pm}2143\;mg/kg$ of dried herb) and stimulated ALP activity, approximately 160% of the control. Growth inhibitory effect on osteoclast by both PR and daidzein were almost identical with control although $IC_{50}$ of genistein was $5.81{\times}10^{-7}$ M. Increases in body weight of OVX rats were suppressed by administration of PR but wet weights of uterus in PR-5 group were increased (p<0.05). Plasma ALP and HDL-cholesterol levels were decreased following ages (p<0.01), and LDL-cholesterol level was also decreased in PR-5 group at 20 week of age (p<0.01). TBAs of tibia and lumbar in PR-1 and PR-5 groups were higher than those of the control although the values were less than those of the sham group (each p<0.01) In conclusion, administrations of PR prevented loss of TBAs of tibia and lumber in OVX rats, while PL and PF did not (p<0.01).

  • The purpose of this study is to investigate the effect of Sungkangwon (Astragalus membranaceus Bunge + Pueraria lobata Ohwi + Salvia miltiorrhiza Bunge) on fatty liver induced by alcohol diet. Rats were either fed control diet or alcohol diet. On 6 weeks, alcohol group were randomly assigned to groups as follows; supplemented without SKW and with SKW (.033g/kg, 1g/kg, and 3g/kg). all of five groups were fed the chow diet for 4 weeks. After 6 weeks. the blood and liver concentration of TG and total-C is comparably high in alcohol group. (omitted)

Short-term Response of Vegetation to Cattle Grazing in an Abandoned Orchard in Southwestern Japan

  • An abandoned mandarin orange orchard in southwestern Japan was set-stocked by Japanese Black cows at two stocking rates (1.0 and 2.0 animals/ha), and vegetation dynamics and diet selection by cattle were monitored for two years, in an effort to obtain information on effective use of abandoned agricultural fields for low-cost animal production and environmental conservation. Two dominant species at the commencement of grazing, kudzu (Pueraria lobata Ohwi) and tall goldenrod (Solidago altissima L.), showed different responses to grazing during the two years; the composition of kudzu decreased, contrasting with that of tall goldenrod which increased at both stocking rates. This was caused by high preference for kudzu and avoidance or low preference for tall goldenrod by cattle. Retrogression of vegetation due to cattle disturbances occurred at both stocking rates, with the high stocking rate leading to a lower degree of succession than the low stocking rate. It was shown that cattle grazing, particularly at a high stocking rate, was effective in the management of vegetation of an abandoned orchard.

  • Now many sustitution and false articles is used in korea instead of gegen. To use gegen correctly, we will make a quilitative certificational plan of gegen to investigate all of lieraturea, records and documents. And we could reach conclusions as folloews. 1) Source As a source of gegen, we think all of Pueraria lobata Ohwi and Pueraria thomsonii Benth can be used in korea and china. 2) Harvesting time. According to a data, content of effective components is high when harvested in January and February. Though there is no problem because domestic harvesting and working time is between January and February, we think it need to sure well-founded datas. 3) Standars of goods According to cutting methods, gegen divide into 'jiaogegen' and 'piangegen' from users So there is no problem at cutting methods because it is not a affective factor. 4) Processing A processing method is a best important case in express quility of herb. According to a data, effective components of peeled gegen is much less than that of non-peeled geeen. A tested samples is non-peeled gegen at a researh, they received suitable judgments in all of items. Especially we got a result content of puerarin is much more than 2.0%. But they were not washed in water, it is a problem that there is bleaching effect by almost drying at a briquat. So a processing method is to wash in water with unavoidable turnning to brown, to prevent this grgrn must be dried in bulk at low temporature. And we think that content of puerarin must be looked upward.
