Anti-inflammatory Activity of Complex Saponin Separated from Pueraria flos and Adzuki Beans Mixture Extract

  • Pueraria flos and Adzuki Beans contains several bioactive compounds, such as saponin, oleanolic acid, and flavone. Pueraria flos and Adzuki Beans has traditionally been used to treat disorders of antioxidant activity, diabetes and liver detoxication, and it has antinociceptive and anti-inflammatory properties. However, complex saponin were validity of the anti-inflammatory activity has not been scientifically investigated. In this study, to determine anti-inflammatory activity of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract on nitric oxide and prostaglandinE2 assay. The anti-inflammatory activities of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract were evaluated for inhibitory activities against lipopolysacchride induced nitric oxide and prostaglandinE2 production protein expressions in RAW264.7 cell lines. The complex saponin separated from Pueraria flos and Adzuki Beans mixture extract inhibitory activity for both tests with protein high depressions(%) values showed in the ranges of 50~100 ㎍/ml. Overall, prostaglandinE2 tests had a higher inhibitory effect on inflammation than nitricoxide tests. Theseis result suggest a potential role of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract as source of anti-inflammation agent.

갈화와 갈근 열수추출물들이 에탄올 투여 흰쥐의 혈청성분에 미치는 영향 (Effects of Pueraria flos and radix Water-extracts on levels of Several Serum Biomarkers in Ethanol-treated Rats)

  • 갈화와 갈근의 에탄올성 간손상 흰쥐의 혈액학적 성분에 미치는 영향을 구명하기 위하여 에탄올을 투여한 흰쥐에게 갈화와 갈근을 수준별로 5주간 급여한 후 효과를 관찰하였다. 간손상 지표 효소인 혈청 AST, ALT, ALP와 ${\gamma}-GTP$ 활성은 에탄올 투여로 증가되었으나 칡추출물 급여로 감소 되였으며, ${\gamma}-GTP$의 경우 갈화추출물이 갈근추출물에 비하여 유의적이지는 않지만 활성 감소효과가 큰 것으로 관찰되었다. 혈청 혈당치와 간조직의 글리코겐 함량은 에탄올 투여시 정상군에 비하여 유의적인 감소가 관찰되었으며 힘추출 물급여시 증가되었다. 힘의 부위에 따른 차이는 갈근추출물에 비해 갈화추출물이 함량 증가효과가 큰 것으로 나타났다. 알부민 함량은 대조군이 정상군에 비하여 유의적으로 감소되였으며 칡추출물 급여에 의한 유의적인 차이는 나타나지 않았다. 총빌리루빈, 직접 빌리루빈, 간접 빌리루빈, 크레아 티닌과 요산 함량은 정상군에 비하여 에탄올군에서 유의적으로 증가되였으며 갈화와 갈근급여군에서 감소되었다. 이상의 결과에서 갈화와 갈근추출물이 에탄올 투여로 인한 간조직 손상 지표효소의 활성을 감소시켜 에탄올성 간손상의 예방 및 치료에 효과적일 것으로 사료된다.

  • Two isoflavonoids were isolated from the flowers of Pueraria lobata (Flos puerariae) guided initially fractionation based on brine shrimp lethality assay. The structures were identified as 4',5,7-trihydroxy-6-methoxyisoflavone (tectorigenin) and 4',7-dihydroxy-6-methoxyisoflavone (glycitein), respectively, on the basis of their spectroscopic and physicochemical evidences.

갈화(葛花)의 함유성분 분석과 항산화 활성 (Component Analysis and Antioxidant Activity of Pueraria flos)

  • 본 연구는 갈화를 대상으로 약이성 음식으로의 활용을 위한 가능성을 타진하고자 계획 수행되었다. 따라서 갈화의 영양성분 분석을 통한 식품영양학적 접근, 생리활성 기능을 기대할 수 있는 관련 물질 함량의 분석하였다. 식품영양학적 접근에서의 갈화의 일반성분은 건량기준으로 당질 69.2%, 조단백질 19.9%, 조지방 2.2% 및 조회분 8.9%이었고 갈화 100 g의 함유 열량은 340.4 kcal로 분석되었으며, 총 식이섬유소 함량은 건량기준으로 총 당질 중 85.1%이었고 수용성 및 불용성 식이섬유소 함량은 각각 12.0%, 46.7%로 나타났다. 또한, 총 18종의 아미노산으로 구성되었으며 필수아미노산과 비필수아미노산 함량은 각각 5.0 g, 6.7 g이었고, 무기질 중 칼슘의 함유량이 가장 높았고 그 다음이 칼륨, 마그네슘, 인 순으로 나타나 알칼리성 재료임을 알 수 있었으며, 지방산 함량의 경우 총 포화지방산 25.37%, 단일불포화지방산 33.61% 및 다가불포화지방산 35.68%로 구성되어 있어 다른 식물류에 비해 불포화지방산의 함량이 높은 것으로 나타났다. 갈화 70% 에탄올, 헥산, 클로로포름, 에틸아세테이트, 부탄올과 물 추출물에서 DPPH 라디칼 소거능의 항산화 효과 결과는 $RC_{50}$값이 217.3, 134.5, 116.7, 109.9, 129.8, 473.5 ${\mu}g$으로 나타나, 에틸아세테이트 분획물에서 가장 높은 항산화 효과를 나타내었다.

  • Puerariae Flos is a traditional herbal medicine that has long been used as a treatment for colds, diabetes, and hangovers. The herbal medicine contains a wide variety of isoflavones such as kakkalide, tectoridin, and tectorigenin. This study demonstrates that the substances undergo a certain degree of change depending on the storage period by the method of HPLC and $^{13}C-NMR$, and that the HPLC analysis can be used to determine the freshness of Puerariae Flos.

칡 열수추출물이 흰쥐의 알콜올 대사효소계에 미치는 영향 (Effects of Pueraria thunbergiana Bentham Water Extracts on Hepatic Alcohol Metabolic Enzyme System In Rats)

  • 갈화와 갈근이 알코올성 간손상 흰쥐의 알코올 대사효소계에 미치는 영항을 구명하기 위하여 알코올을 투여한 흰쥐에게 갈화와 갈근 열수추출물을 2수준으로 나누어 5주간 급여한 후 그 효과를 관찰하였다. 체중증가량, 식이섭취량과 식이효율은 알코올 투여시 감소하였으며 갈화와 갈근추출물 급여로 회복되는 경향을 보였다. 장기 무게에 미치는 칡추출물의 영향은 알코올 투여로 증가된 간, 뇌와 심장 무게를 감소시켰는데 갈화가 갈근에 비하여 뇌와 심장 무게의 감소효과가 큰 것으로 나타났다. 알코올 대사효소 활성 변화에서 알코올 투여시 유의적으로 증가된 ADH와 MEOS 활성은 칡추출물 급여시 감소된 반면 AlDH 활성은 대조군에서 유의적으로 감소되었는데 칡추출물 급여시 그 활성이 증가되었다. MAO 활성과 과산화수소 함량도 알코올 투여시 증가되었으며 갈화와 갈근 열수추출물 급여시 유의적으로 감소되었다. 이상의 결과에서 갈화와 갈근 열수추출물 급여는 알코올 중독된 흰쥐의 알코올 대사계 효소 활성 변화에 영향을 미쳐 알코올성 간손상 완화에 효과적일 것으로 사료된다.

  • We investigated the effect of tectorigenin (1) with hypoglycemic and hypolipidemic effects on Phase I and II enzymes and TF activity to elucidate the action of an immunosuppressive compound (1) in the diabetic rat. Compound 1 was obtained from the hydrolysis of tectoridin easily isolated from the flower of Pueraria thunbergiana(Leguminosae). Puerariae Flos has been used as therapeutics for diabetes mellitus in traditional medicine of Korea. (omitted)

Quantitative Analysis of the Isoflavone Content in the Flower and the Root of Pueraria thunbergiana before and after Acid Hydrolysis

  • The contents of isoflavones and 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging activities were determined in flowers (Puerariae Flos) and roots (Puerariae Radix) of Pueraria thunbergiana to differentiate both pharmacochemical or pharmacognostical usage of both oriental medicinal drugs. The tectorigenin contents in flower before and after acid hydrolysis were shown to be 17.10 ${\mu}mole\;g^{-1}$ and 49.58 ${\mu}mole\;g^{-1}$, respectively, analyzed by HPLC. However, the root displayed much less content of tectorigenin even after acid hydrolysis (6.56 ${\mu}mole\;g^{-1}$) than in the flower. Acid hydrolysis made glycitin and tectoridin of the isoflavone glycosides almost disappear in the flower and root. DPPH assay results demonstrated that acid hydrolysis of both extracts or both glycosides should increase free radical-scavenging activities due to the increase of isoflavone contents. It is also suggested that the flower had much higher concentration of isoflavonoids than in the root.

  • Tectorigenin has an apoptosis-inducing ability and immunosuppressive activity. We investigated the effect of tectorigenin on Phase I and II enzyme activities to elucidate the pharmacological action of the immunosuppressive tectorigenin in the diabetic rat. This compound was obtained from the hydrolysis of tetoridin isolated from the flower of Pueraria thumbergiana (Leguminosae). This crude drug (Puerariae Flos) has been used as a therapeutic for diabetes mellitus in traditional Korean medicine. Tecotrigenin inhibited the formation of malondialdehyde (MDA) and hydroxy radicals in serum and liver but promoted superoxide dismutase (SOD) activity. Low MDA contents and low xanthine oxidase and aldehyde oxidase activities were observed in the tectorigenin-treated rats, suggesting that such Phase I enzyme activities are the major source of lipid peroxidation. However, tectorigenin increased Phase II enzyme activities such as SOD, glutathione peroxidase and catalase, suggesting the activation of free radical-scavenging enzymes. The activities of tectorigenin were comparable to those of glibenclamide, which was employed as a positive control. These results suggest that tectorigenin may share some biological properties with glibenclamide in insulin-dependent-diabetes mellitus (IDDM).

Puerariae flos inhibits inflammatory responses in interferon-γ and lipopolysaccharide-stimulated mouse peritoneal macrophages

  • In macrophages, nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation. In this study, we have examined the inhibition effects of NO production by 85% methanol extract of the flower of Pueraria thunbergiana (PF) in mouse macrophages. Extract of PF (1, 10, 100 ${\mu}g/ml$) inhibited NO production, inducible NO synthase and cyclooxygenase-2 expression in interferon-g and lipopolysaccharide-stimulated mouse peritoneal macrophages and it had no cytotoxicity. These data suggest that 85% methanol extract of PF might be useful in controlling macrophages mediated inflammatory disease.
