For comparing with flavones/flavonols, isoflavonoids isolated from Pueraria radix and chemically synthesized from biochanin A were evaluated for the antiinflammatory activity using mouse ear edema test, Isoflavonoids such as daildzein and puerarin hsowed the significant antiinflammatory activity at a dose of 2 mg/mouse, although their activity was generally less than that of flavones/flavonols. 7-O-Substitution of biochanin A was not facorable for the antiinflammatory activity.
This study was designed to investigate the synergetic hepatoprotective effects from a mixture of Korean Red Ginseng and Pueraria Radix on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity in mice. Liver toxicity was induced by intraperitoneal administration of $CCl_4$ (0.6 mg/kg) in 12 groups of ICR mice. The negative control group was given $CCl_4$ without test samples and the normal group was given no treatment. Among treatment groups, the RGAP treatment (Korean Red ginseng acetic acid extract : Pueraria radix water extract, w/w, 38.4:57.6) decreased $CCl_4$-elevated ALT (101.60 IU/L), AST (833.89 IU/L), and LDH (365.02 IU/L) levels in the serum, and increased the SOD (11.03 unit/mg protein) and CAT (0.37 unit/mg protein) levels and the LPO levels ($59.09{\mu}M/g$ tissue) more than that in the mice group with $CCl_4$-induced control group hepatotoxicity. These results suggest that administration of a mixture of Korean Red ginseng and Pueraria radix decreases $CCl_4$-induced liver damage and enhances antioxidant activity in mice and imply that administration of the mixture in a certain ratio is more effective than single administration of either Korean Red ginseng or Pueraria radix alone.

The present study was investigated effect of each water extract from Pueraria flos (PF) and Pueraria radix (PR) on serum several biomarkers in ethanol-treated rats. Male Sprague-Dawley rats were randomly divided into six groups: Normal (None-treated group); Ethanol (only ethanol-treated group); EPF I (ethanol-treated, supplemented group); EPR (ethanol-treated, PF II-supplemented group); EPR I (ethanol-treated, PR I-supplemented group) ; EPR (ethanoltreated, PR II-supplemented grou). Five groups of male Sprague-Dawley rats were orally administered 25% ethanol (5 g/kg body weight/day) and sacrified 5 weeks post treatment. Aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and ${\gamma}-glutamyl$ transpeptidase activities were significiantly lowered by feeding of PF or PR than those of only ethanol-treated group. Whereas serum glucoseand liver glycogen contents were significantly decreased (p<0.05) by ethanol administration and increased decreased (p<0.05) by PF or PR supplement. This results indicate that Pueraria flos and radix water extract supplement improves alcoholic disorder.
This study was designed to investigate the effects of korean pueraria radix extract in Pb administered rats. Forty-two male Sprague-Dawley rats weighing $100{\pm}10g$ were used for this experiment and divided into following 6 groups; control group, 3% pueraria radix group(3% pue.), 100 ppm and 200 ppm alone pb group, 100 ppm and 200 ppm alone pb group with 3% pue. Tissue weight of liver, lung, stomach, heart, kidney and spleen of pb exposed rats were reduced by 3% extracts of pueraria radix group. The Pb content in the rats tissue of pb alone administered group was lower than in the rats tissue of pb group with 3% pue. GPT and GOT were increased in pb-administered group and lower in the 3% extracts of pueraria radix group. LDH was lower in the 3% extracts of pueraria radix-pb group than in the pb group. ChEase was higher in the 3% extracts of pueraria radix group than in the pb alone administered group.

The phytochemical compounds of Pueraria, a medicinally important leguminous plant, include various isoflavones that have weak estrogenic activity and a potential role in preventing chronic disease, cancer, osteoporosis, and postmenopausal syndrome. However, the major isoflavones are derivatives of puerarin and occur mainly as unabsorbable and biologically inactive glycosides. The bioavailability of the glucosides can be increased by hydrolysis of the sugar moiety using ${\beta}$-glucosidase. In this study, we investigated the antioxidant effects of a Pueraria extract after fermentation by Lactobacillus rhamnosus BHN-LAB 76. The L. rhamnosus BHN-LAB 76 strain was inoculated into Pueraria powder and fermented at $37^{\circ}C$ for 72 hr. The total polyphenol content of the Pueraria extract increased by about 134% and the total flavonoid content increased around 110% after fermentation with L. rhamnosus BHN-LAB 76 when compared to a non-fermented Pueraria extract. Superoxide dismutase-like activities, DPPH radical scavenging, and ABTS radical scavenging increased by approximately 213%, 190%, and 107%, respectively, in the fermented Pueraria extract compared to the non-fermented Pueraria extract. Fermentation of Pueraria extracts with L. rhamnosus BHN-LAB 76 is therefore possible and can effectively increase the antioxidant effects. These results can be applied to the development of improved foods and cosmetic materials.
Objectives : The purpose of this study was to evaluate the neuroprotective effect of Pueraria lobata extract on focal cerebral ischemia in mice. Methods : Focal cerebral ischemia was induced by occlusion of the right middle cerebral artery using the intraluminal filament model. ICR male mice underwent 90 minutes of middle cerebral artery occlusion (MCAo) followed by 24 hours of reperfusion. Mice were administered Pueraria lobata extract orally at the dose of 300mg/kg just prior to reperfusion. Rotarod test and balance beam test were practiced to assess sensory-motor function 23 hours after MCAo. In rotarod test, the latency to fall on the accelerating rotarod was recorded for 5 min. In balance beam test, the score was graded according to number of slips and latency to cross. The infarct volume was measured 24 hours after MCAo using 2% 2,3,5-triphenyltetrazolium chloride (TTC) staining. Results : Pueraria lobata extract treated group showed significant reduction in infarct volume by 27.3% compared to control group (p<0.05). In rotatod test, it also showed significant extension of latency time compared to control group ($67.82{\pm}15.08$ vs. $5.62{\pm}1.06$, p<0.001). In contrast to performance in rotarod test, that in balance beam test did not improve with Pueraria lobata extract treatment. Conclusions : We conclude that Pueraria lobata extract has a significant neuroprotective effect and reduces damage of sensory-motor function in MCAo model. These findings suggest that Pueraria lobata could be a potent neuroprotective agent.
Pueraria lobata (Willd.) Ohwi was investigated to check its modulatory effects on the activation of macrophages upon inflammatory conditions treatment. For this purpose, we examined several inflammatory responses such as nitric oxide (NO) production, reactive oxygen species (ROS) generation, cytoprotection and phagocytosis under the treatment of methanol extract from P. lobata (Pl-ME). Pl-ME dose-dependently blocked NO production in lipopolysaccharide (LPS)- stimulated RAW264.7 cells but not sodium prusside (SNP)-generated NO release. The NO inhibition seemed to be due to blocking inducible NO synthase (iNOS), since Pl-ME suppressed its expression in a NF-${\kappa}B$-independent manner. Similarly, this extract also effectively protected RAW264.7 cells from LPS-induced cytotoxicity. However, Pl-ME did not block ROS generation and rather it enhanced. Finally, this extract negatively modulated FITC-dextran uptake. Therefore, our data suggested that Pl-ME may be involved in negatively regulating some macrophage-mediated inflammatory responses such as NO production and phagocytic uptake.
The objective of this study was to determine antioxidant effect of ether and ethylacetate fractions of 70% ethanol extract of some food (acid treated or not) on perilla oil. Each fraction of food extract was added to perilla oil and stored for 0,3,6.9,11 days at 60$^{\circ}C$. Then, the peroxide value (POV) of perilla oil samples were analyzed. Perilla oil contained ${\gamma}$-tocopherol 0.6800 $\mu\textrm{g}$/mg, ${\alpha}$-tocopherol 0.3189 $\mu\textrm{g}$/mg. $\delta$-tocopherol 0.0463 $\mu\textrm{g}$/mg, but it was easily oxidized due to high linolenic acid content. To increase yield of ether and ethylacetate fractions from each food extract, the 70% ethanol extract was treated with 0.2% H$_2$SO$_4$ and fractionized by ether and ethylacetate. Among ether and ethylacetate fractions of 70% ethanol extracts of some food, the yield of ethylacetate fraction of acid treated Pueraria thunbergiana extract was 5 times more than that of ethylacetate fraction untreated with acid. Perilla oil which added 100 ppm ethylacetate fraction of acid treated Pueraria thunbergianan extract showed low POV (44.8 meq/kg) compared to POV (80.0 meq/kg) of control.
The diurnal changes of the stomatal conductance, transpiration and leaf water potential were measured in order to assess the water relations characteristics of Pueraria thunbergiana in August of 1995 and 1996. The results showed two different responses depending on the duration of rainless days. The microclimatic conditions were highly stressful on 2 August. Daily maximum temperature reached to $39.0{\circ}C$ and vapor pressure deficit was 3.55 KPa. During this time the leaf water potential decreased to -1.02 MPa and a marked reduction of stomatal conductance was shown. However, on 15 August the stomatal conductance increased with increment of photon flux density, and transpiration was highly maintained during the day time. Minimum leaf water potential was only -0.47 MPa in spite of high transpiration rate. Furthermore, on 15 August reduced leaf water potential during the day time was recovered rapidly with decrease of photon flux density, whereas recovery of leaf water potential on 2 August was delayed. However, reduced leaf water potential on 2 August was recovered untile the next dawn. Osmotic potential at turgor loss point of Pueraria thunbergiana on 2, 3 and 15 August was -1.79, -1.70 and -1.60 MPa, respectively. The vapor pressure deficit is more contributive to the regulation of stomatal conductance than leaf water potential.
