The Effect of Coptidis Rhizoma, Feaxini Cortex, Jinpisan(秦皮散) on Experimental Pseudomonas aeruginosa Keratitis. (黃連, 秦皮, 秦皮散이 綠膿菌性 角膜炎에 미치는 效果에 관한 實驗的 硏究)

  • Coptidis Rhizoma, Fraxini Cortex, Jinpisan(秦皮散) have been as eye washes of inflammatory eye disease in the oriental medicine. Especially Jinpisan(秦皮散) has been used for the disease which is similar to Peudomonas aeruginisa keratitis. This research was attempted to investigate the effect of Coptidis Rhizoma, Fraxini Cortex, Jinpisan(秦皮散), on Peudoronas aeruginisa keratitis. Pseudomonas aeruginosa keratitis causes a deep rapid intense ulceration which often leads to perforation of the cornea within 48 hours. In this research, we induced keratits in the rabbits by inoculating Pesudomonas aeruginosa(9027) and observed the effect on the keratitis and the irritation against the external eye. Also we mesured the minimum inhibitory consentration(MIC) of Coptidis Rhizoma, Fraxini Cortex, Jinpisan(秦皮散) by agar diliution method and the anti-bacterial activites by disk method. The tested bacteria were as follows : a) Pseudomonas aeruginosa (9027), b) Streptococcus pneumoniae(6303), c) Staphylococcus epidermidis(12228), d) Staphylococcus aureus(6538P). The results were as follows ; 1. The groups which were applied eye washes of Fraxini Cortex, Jinpisan reavealed a significant effect, but the group applied eye wash of Coptidis Rhizoma reveaded no effect on Pseudomonas aeruginosa keratitis. 2. Applying eye washes of Coptidis Rhizoma, Fraxini Cortex, Jinisan revealed an irritation against external eyes. 3. Coptidis Rhizoma showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylucoccus aureus by agar diliution method 4. Coptidis Rhizoma showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by disk method. 5. Fraxini Cortex showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by agar diliution method 6. Fraxini Cortex showed an anti-bacterial activity on Pseudomonas aeruginosa, Sireptococcus pneumoniae, Staphylococcus epidermidis, Staphy1ococcus aureus by disk method. 7. Jinpisan showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by agar diliution method. 8. Jinpisan showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by disk method. According to the above results, Fraxini Cortex, Jinpisan(秦皮散) are recognized to have an effective treatment on the Pesudomonas aeruginosa keratitis, so this experiment is thought to be a basic ingredient in proving the effect of Fraxini Cortex, Jinpisan which is applied many in documents and clinical medicine. In the comparison of anti-bacterial activity and results of treatment on the Pesudomonas aeruginosa keratitis, Jinpisan(秦皮散) was more effective than Coptidis Rhizoma, Fraxini Cortex.

The Experimental Study on Anti-bacterial Potency of Sean-tang on Three species of causative bacteria of Keratitis (세안탕(洗眼湯)이 다종(多種)의 각막염(角膜炎) 유발균(誘發菌)에 미치는 효과(效果)에 관한 실험적 연구)

  • Objective : This experimental study was performed to investigate the effect of herbal eye drops, Sean-tang and Coptidis rhizoma on Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa keratitis. Methods : After administering herbal eye drops (Sean-tang, Coptidis rhizoma) on Staphylococcus ausreus, Staphylococcus. epidermidis and Pseudomonas aeruginosa, I measured MIC and the size of inhibition zone. MIC was measured by dropping to $50{\mu}{\ell}$ according to density(l00%, 10%, 1%, 0.1%). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Result : On Staphylococcus aureus, Staphylococcus epidermidis, Coptidis rhizoma showed the highest anti-bacterial potency and on Pseudomonas aeruginosa, Sean-tang, Coptidis rhizoma was nor anti-bacterial potency. Conclusions : The present author think that the herbal eye drops, Sean-tang and Coptidis rhizoma can be used to cure Staphylococcus aureus, Staphyloccus epidermidis keratitis and if further study is performed, the use of the herbal eye drops will be valuable and beneficial in the clinical medicines.

  • Objectives : This experimental study was performed to investigate the effectiveness of Fel Ursi Pharmacopunture solution(FUPS) manufactured by using alcohol/water extraction method. To identify the use of it as eyedrops, antibacterial test on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans was performed. Methods : After treatment FUPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, we investigated anti-bacterial effects of FUPS on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans in terms of measuring MIC and size of inhibition zone respectively. Results : After FUPS was treated, significant changes of MIC on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans were not observed at all. Conclusions : The present study suggests that FUPS doesn't have anti-bacterial effects on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans which cause Keratitis. Perhap These results recommend that FUPS doesn't have anti-bacterial effects but have other mechanism which suppress inflammation.

황련의 증량에 따른 전탕액이 Pseudomonas aeruginosa에 미치는 항균효과에 대한 실험적 연구

  • Objective : This experimental study was performed to investigate the effect of Coptidis rhizoma extract compared with quantity on Pseudomonas aeruginosa that induce keratitis. Methods : MIC was measured by dropping to 50 ${\mu}l$ according to density Coptidis rhizoma extract(100%, 10%, 1%, 0.1%) compared with quantity(40g, 80g, 120g, 160g). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results : MIC on Pseudomonas aeruginosa in Coptidis rhizoma extract compared with quantity(40g, 80g, 120g, 160g) was not showed all samples(100%, 10%, 1%, 0.1%). Conclusions : Coptidis rhizoma extract compared with quantity was not showed anti-bacterial potency on Pseudomonas aeruginosa. The results differ that Coptidis rhizoma extract was showed anti-bacterial potency in 1999 year. Anti-bacterial Potency of Coptidis rhizoma extract compared with quantity on Pseudomonas aeruginosa must be studied.

The Experimental Studies on Anti-bacterial Effect of Tangpo-san on Three species of causative bacteria of Keratitis (탕포산(湯泡散)이 삼종(三種)의 각막염(角膜炎) 유발균들에 미치는 영향)

  • This experimental study was performed to investigate the effect of herbal eye drops, Tangpo-san and Coptidis rhizoma on Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa keratitis. The following results were obtained by using Minimum inhibition Concentration(MIC) and inhibition Zone. 1. MIC on Staphylococcus aureus in Tangpo-san was 100%, in Coptidis rhizoma was 100% and in Cravit was 0.1% 2. MIC on Staphylococcus epidermidis in Tangpo-san was 100%, in Coptidis rhizoma was 10% and in Cravit was 0.1%. 3. MIC on Pseudomonas aeruginosa in Tangpo-san, Coptidis rhizoma was not showing and in Cravit was 0.1%. 4. The size of inhibition zone on Staphylococcus aureus for Tangpo-san was 13.3mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 26mm in $50{\mu}{\ell}$ and for Cravit was 31mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 5. The size of inhibition zone on Staphylococcus epidermidis for Tangpo-san was 16mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 25mm in $40{\mu}{\ell}$ and for Cravit was 34mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 6. The size of inhibition zone on Pseudomonas aeruginosa for Tangpo-san, Coptidis rhizoma was not and for Cravit was 24.7mm in $50{\mu}l$, showing the antibacterial effect. In addition, the results shows that the herbal eye drops, Tangpo-san and Coptidis rhizoma can be used to cure Staphylococcus aureus, Staphylococcus epidermidis keratitis and if further study is performed, the use of herbal eye drops will be valuable and beneficial in the clinical medicines.

The Experimental Study of Safety and Efficacy in Using Bovis Calculus Pharmacopuncture Solution as Eye Drop (점안용 우황약침액의 안전성 및 유효성 평가)

After administering Bovis Calculus pharmacopuncture solution on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : 1. After Bovis Calculus pharmacopuncture solution was administered in the left eye of the rabbits, it was found that none of nine rabbits have abnormal signs and weight changes. 2. After Bovis Calculus pharmacopuncture solution was medicated in the left eye of the rabbits, no eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 3. There was no response to MIC on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Bovis Calculus pharmacopuncture solution was medicated. Conclusions : The present study suggests that Bovis Calculus pharmacopuncture solution is a nontoxic and non-irritant medicine, which does not cause eye irritation in rabbits, but dosen't have antibacterial effects on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.

Effects of natural and chemical disinfectants on soft contact lens infected with Pseudomonas aeruginosa : Scanning Electron Microscopic study (Soft contact lens에 있어서 천연보존제와 합성보존제의 살균효과 : 주사 전자현미경적 관찰)

  • The effects of grapefruit seed extract and $H_2O_2$ disinfectants on Pseudomonas aeruginosa were investigated by antimicrobial assay and scanning electron microscopy. Pseudomonas aeruginosa is a Gram negative bacteria and a common causative microorganism of bacterial keratitis. After incubation of soft contact lens in Pseudomonas aeruginosa suspension for 120 minutes containing the disinfectants, the grapefruit seed extract showed antimicrobial activity as good as the chemical disinfectant. The living rates of Pseudomonas aeruginosa in grapefruit seed extract and $H_2O_2$ disinfectants were $1{\times10^3CFU/m{\ell}$ and $0.5{\times}10^3CFU/m{\ell}$, respectively showing no major difference in the killing activity. However, the mechanisms of the antimicrobial activity were quite different as indicated by scanning electron microscopy of the bacterial surface morphology. Considering chemically or other toxins induced keratitis, the natural disinfectant isolated from grapefruit seed extract shows promising results as a soft contact lens disinfectant.

Methods: FUBCPS was manufactured by using alcohol/water extraction method. Measure the size of inhibition zone and MIC(Minimum Inhibition Concentration) after administering FUBCPS on bacterial species. Staphylococcus aureus. Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans, which cause keratitis. Administering cravit(Levonoxacin medicine) on bacterial species also performed to compare the anti-bacterial potency of this material, measured by using the size of inhibition zone Results : After administering FUBCPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans). there was no response to MIC and there was no anti -bacterial potency also. Conclusions : This study suggests that FUBCPS dose not have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that we need to research more on herbal medicines of eye drop which have anti-bacterial effects on keratitis
