Antioxidative Activities and Whitening Effects of Solvent Fraction from Prunus davidiana (Carriere) Franch. Fruit (산복사나무(Prunus davidiana (Carriere) Franch.) 열매 분획 추출물의 항산화 및 미백 효능)

  • In this study, methanol extract and its organic solvent fractions were prepared from Prunus davidiana (Carriere) Franch. fruit. Antioxidative activities, polyphenolic and flavonoid contents, and tyrosinase inhibitory activities were evaluated. Among the fractions, ethyl acetate fraction showed the highest antioxidative activity with $IC_{50}$ values of 5.8 ${\mu}g/mL$ and 88.1 ${\mu}g/mL$ for DPPH radical and nitrite scavenging, respectively, which were comparable to those of ascorbic acid. Further, total flavonoid and polyphenolic contents were highest in the ethyl acetate fraction with $IC_{50}$ values of 244.5 mg/g and 210.2 mg/g, respectively. $IC_{50}$ values for tyrosinase inhibitory activity were 0.310 mg/mL and 0.329 mg/mL for the ethyl acetate and n-butanol fractions, respectively. Based on these results, it is suggested that the ethyl acetate fraction of Prunus davidiana (Carriere) Franch. fruit could be used as a functional material in the food and pharmaceutical industries.

Optimization of Hyaluronidase Inhibition Activity from Prunus davidiana (Carriere) Franch Fruit Extract Fermented by its Isolated Bacillus subtilis Strain SPF4211

  • Strain SPF4211, having hyaluronidase (HAase) inhibition activity, was isolated from P. davidiana (Carriere) Franch fruit (PrDF) sugar extract. The phenotypic and biochemical properties based on 16S rDNA sequencing and an API 50 CHB kit suggested that the organism was B. subtilis. To optimize the HAase inhibition activity of PrDF extract by fermentation of strain SPF4211, a central composite design (CCD) was introduced based on three variables: concentration of PrDF extract (X1: 1-5%), amount of starter culture (X2: 1-5%), and fermentation time (X3: 0-7 days). The experimental data were fitted with quadratic regression equations, and the accuracy of the equations was analyzed by ANOVA. The statistical model predicted the highest HAase inhibition activity of 37.936% under the optimal conditions of X1 = 1%, X2 = 2.53%, and X3 = 7 days. The optimized conditions were validated by observation of an actual HAase inhibition activity of 38.367% from extract of PrDF fermented by SPF4211. These results agree well with the predicted model value.

Comparative Analysis of Three Different Types of Gami-Saengmaeksan in Enhancing the Immune System (산림자원을 가미한 생맥산 3종의 면역 증진 효능 비교분석)

  • With infectious diseases and an aging population on the rise worldwide, there is a growing interest in boosting immunity to combat them. We aim to identify forest resources that can enhance immune function and scientifically prove their efficacy. We examined three different types of gami-saengmaeksan can increase the immune function in cyclophosphamide-induced immunosuppression mice. First, we injected cyclophosphamide (50 mg/kg) twice to induce immunosuppression. Then, three different types of gami-saengmaeksan (Prunus davidiana Carrière gami-saengmaeksan; PDS, Chaenomeles sinensis Koehne gami-saengmaeksan; CSS, Pyrus pyrifolia Nakai gami-saengmaeksan; PPS) 200 mg/kg/day were oral administered for 14 days. In order to confirm the immune-enhancing effect three different types of gami-saengmaeksan, we analyzed body weight chagne, the number of immune cells in blood, cytokines, and immunoglobulins levels in serum. Cyclophosphamide was decreased the mice body weight and the number of immune cells in blood. However, PDS and PPS have significantly increased the body weight, white blood cell, neutrophil, lymphocyte and monocyte count. Also, they were increased immune-related cytokines (IFN-γ, IL-2, IL-4, IL-6, IL-10, and TNF-α) and immunoglobulins (IgA, IgG, and IgM) levels in serum. But, CSS were slightly increased body weight, immune cells, cytokines, and immunoglobulins levels, non-significance was observed. The results suggest that the three types of gami-saengmaeksan can be used as ingredients in dietary supplements to immune enhancing.

  • 본 연구는 경남 함양군 삼봉산의 관속식물을 조사한 후 식물상 현황과 특정식물종 및 귀화식물 등을 정리하였으며, 차후 식물상의 보호 관리에 있어 기초 자료로 활용하고자 실시하였다. 삼봉산은 전북 남원시 산내면 중황리, 인월면 상우리와 경남 함양군 마천면 구양리, 함양읍, 죽림리가 접하는 지역으로 남쪽으로는 국립공원 지리산의 연봉이 길게 늘어서 있고 북쪽으로는 광주대구 고속도로 건너 함양군의 백운산과 괘관산 등 1,000m가 넘는 산이 많은 지역내에 위치하고 있다. 삼봉산지역에 자생하는 관속식물은 총 80개과 182개속 254개 종이 생육하는 것으로 나타났다. 삼봉산 지역의 특정식물종은 식물구계학적 특정식물 V등급인 승마 Cimicifuga heracleifolia Kom.)와 IV등급인 도깨비부채(Rodgersia podophylla A.Gray), 점현호색 (Corydalis maculata B.U.Oh & Y.S.Kim), 솔붓꽃 (Iris ruthenica KerGawl.), 산복사나무 [Prunus davidiana (Carriere) Franch.] 등이 분포하는 것으로 확인되었다. 조사결과 가흥지역에서 환경부 멸종위기종은에 해당하는 분류군은 없었으며 특정식물종 V등급에 해당되는 승마와 IV등급에 해당되는 4개 분류군이 확인되었다. 국립공원지리산 근처의 1,000m이상의 산지임에도 전체적으로 50년 전부터이루어진 조림과 능선 바로 아래까지 경작지가 존재하는 등 식생의 파괴가 심하여 전체적인 관속식물의 다양성은 낮은 것으로 확인되었다.
