Quality Characteristics of Granule Prepared by Protein-Bound Polysaccharide Isolated from Agaricus blazei and Selected Forming Agents (부형제 종류에 따른 아가리쿠스버섯 과립의 품질 특성)

  • The purpose of this study was to prepare the granule using protein-bound polysaccharide isolated from Agaricus blazei Murill. Moisture content was the highest in granule formed with dextrin(DE=9). Sugar content of granule in relation to the forming agent was the highest in granule formed with ${\beta}$-cyclodextrin. pH and protein content were not affected by the forming agent. L and b values were high in granules formed with dextrin(DE=9) and ${\beta}$-cyclodextrin, respectively. Solubility of granule formed with dextrin(DE=23) and ${\beta}$-cyclodextrin was higher than that of formed with dextrin(DE=9), while there was no significant difference between dextrin(DE=23) and ${\beta}$-cyclodextrin. Rate of water absorption was the highest in granule formed with ${\beta}$-cyclodextrin, while the lowest in granule formed with dextrin(DE=9). Overall acceptance of three granules were acceptable in granule formed with ${\beta}$-cyclodextrin.

  • To find antitumor components from higher fungi, the cultured mycelia of Paxillus atrotomentosus were extracted with hot water. The water soluble fraction was purified and separated by DEAE-cellulose ion exchange chromatography and Sepharose CL-4B gel filtration method. The separated fractions(Fr.) were designated CR A, B, C and D. Fr. A showed the highest inhibition ratio of 68.51% among the five tractions at a dose of 20 mg/kg/day. When Fr. A was examined for immunopotentiation activity, it increased the amount of the superoxide anion from activated macrophages to 1.1 fold and the number of plaques in hemolytic plaque assay to 2.3 fold, respectively. Otherwise, it did not show direct cytotoxity in sarcoma 180. Delayed type hypersensitiyity reaction showed that the decreased footpad swelling of tumor-hearing was restored to the normal. These results indicate that antitumor activity was exerted through immunopotentiation. Its chemical analysis showed 86.36% polysaccharide, 1.52% protein and 1.64% hexosamine. The polysaccharide consisted of fucose, galactose, glucose, mannose and xylose. This component was named paxillan.

  • To find antitumor components in the hot water extract of the cultured mycelia of Ganoderma lucidum, protein-bound polysaccharides were purified and fractionated (Fr. I-V) by DEAE-cellulose ion exchange column chromatography and Sepbarose CL-4B gel filtration. When a dose of 20 mg/kg/day of each was, i.p., injected into ICR mice, the inhibition ratios against the solid form of sarcoma 180 were $64.2{\sim}75.8%$. The antitumor component was examined for immunological activity. It increased the amount of superoxide anion released by induced macrophages in peritoneal cavity to 1.8 times and the count of hemolytic plaque-forming cells (PFC) was increased to 4.4 times as compared with those of the control group. It contained 68.6% polysaccharide which consisted of mannose, glucose, galactose, fucose and xylose and 5.1% protein consisting of 17 amino acids. The contents of hexosamine were 0.78%. The molecular weight of Fr. V that showed the highest antitumor activity was $5.8{\times}10^4$ dalton by Sepharose CL-4B gel filtration. It was named lucidan.

  • To find antitumor components with low toxicity from natural resources, antitumor test of the water extract of the carpophores of Trametes sanguinea (L. ex Fr.) Lloyd. was undertaken. The carpophores of this fungus were collected in Gyeong Gi Province and extracted with hot water. The extract was purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained. The fraction was tested for antitumor activity against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction against the tumor was 72.4% at the dose of 10mg/kg/day for the period of ten days. The tumor in one of the eight mice was completely regressed. The antitumor components were found to be a polysaccharide and a protein. The hydrolysis of the polysaccharide moiety yielded three monosaccharides, and from the hydrolysate of the protein moiety 13 amino acids were identified.

  • To investigate antitumor components of Korean higher fungi, the carpophores of Ramaria formosa $(Fr.)Qu\acute{e}l$. were collected in Gang Won Province and extracted with hot water. The extract was concentrated and precipitated by four volumes of ethanol. The precipitate was centrifugated and purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained. The fraction was tested for antitumor activity against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction was 66% in the dose of 50mg/kg/day for the period of ten days. The tumor in two of the eight mice was completely regressed. The components of the fraction were found to be a polysaccharide and a protein. The chemical analysis of the fraction showed that the polysaccharide moiety consisted of four monosaccharides and that the protein moiety contained fourteen amino acids.

  • Antitumor components were obtained from the cultured mycelia of Pleurotus pulmonarius by ethanol precipitation. The protein-bound polysaccharide was subjected to DEAE-­Sephadex column chromatography and Sephadex G-200 gel filtration. The antitumor fraction $C_1$ was isolated. The inhibition ratio of fraction $C_1$ was 81.8 % in the doses of 10 mg/kg/day for 10 days. The antitumor fraction $C_1$ consisted of a polysaccharide and a protein. The protein-moiety was composed of 14 amino acids. From the peritoneal cell populations in the mice given antitumor fraction $C_1$, the injection of the fraction caused the influx of peritoneal macrophages at two days when compared with those of soluble starch. This was named pulmonaran after its species name.

  • To determine contents of inorganic elements of Ganoderma lucidum, the horn-shaped carpophores and the pileus of Ganoderma lucidum were incinerated and analyzed by inductively coupled plasma atomic emission spectrophotometry. The ash contents of the pileus and the horn-shaped carpophore were 1.48% and 1.40%, respectively. The pileus contained calcium, magnesium, sodium, manganese, iron, zinc and germanium in that order. The horn-shaped carpophore contained magnesium, calcium, zinc, manganese, iron, copper and germanium in that order. To examine the protein-bound polysaccharide from Ganoderma lucidum for immunopotentiating activity, its fruit bodies were extracted with hot water. Purification of the extract was carried out by acetone precipitation and dialysis. The fraction obtained during the purification procedure consisted of a polysaccharide moiety (51%) and a protein moiety (5%). When the compound was administered intraperitoneally to the mice at a dose of 50mg/kg, it enhanced the accumulation of the peritoneal exudate cells, macrophage and polymorphonuclear leucocytes, thereby indicating immunopotentiation.

  • To find antitumor components from higher fungi, the mycelia of Collybia confluens (Pers. ex Fr.) Kummer were cultured in artificial media. For efficient production of the mycelia, the influences of various modifications of culture conditions were examined. A water-soluble protein-bound polysaccharide fraction, Fr. A, was obtained from the mycelia by hot water extraction. When Fr. A was purified and fractionated by DEAE-cellulose and Sepbadex G-200 gel filtration chromatographies into four fractions which were designated B, C, C-I and C-II. The tumor inhibition ratios of these fractions ranged from 46% to 75% against the solid forms of sarcoma 180 in ICR mice at doses of 20 and 50 mg/kg/day when given intraperitoneally. Especially, Fr. C which was named Collyban(CB) exhibited a marked life-prolonging effect of the mice against ascitic forms of sarcoma 180 at a dose of 50 mg via i.p. administration. To extend spectra of the antitumor activities and eliminate the effects of allograft rejection, the characterization of antitumor effects of CB was performed in syngeneic host-tumor systems. It did not show any antitumor activity against L1210 murine leukemia in $CD_2Fl$ mice but prolonged their life span against ascitic forms of $MM_{46}$ carcinoma in $C_3H/He$ mice. Also it exhibited antitumor activity against human cervical cancer HeLa cells that were xenografted into nude mice having BALB/c genetic backgrounds by the i.p. injection at a dose of 100 mg/kg/day. In order to characterize the antitumor components, CB was examined by chemical analysis. It was acidic protein-bound polysaccharides composed of 31% polysaccharide, 27% protein and 3% hexosamine. CB was fractionated into two fractions, Fr. C-I(M.W.: 500 Kd) and Fr. C-II(M.W.:30 and 8 Kd) by Sephadex G-200 gel filtration chromatography.

  • On the five interspecific protoplast fusants of Ganoderma lucidum and G. applanatum was the antitumor test performed. The fusant F-2 was selected, to examine the cultured mycelia (protein bound polysaccharide) as antitumor components. When a dose of 20 mg/kg/day of each components purifed from F-2 fusant was, i.p., injected into ICR mice, the inhibition ratio of Fr. II against the solid form of sarcoma 180 increased to 1.5 times as compared with that of their parents. When Fr. II was examined for immunopotentiation activity, it increased the amount of the superoxide anion in activated macrophages to 1.2 times and the count of hemolytic plaque forming cells in the spleen to 4.3 times as compared with that of each control group. Its chemical analysis showed 85.2% polysaccharide which consisted of glucose, galactose, mannose, fucose and xylose, and 0.39% protein of 15 amino acids. The content of hexosamine was 0.39% and the molecular weight of Fr. V was $5.6{\times}10^4$ dalton.

Protective Effect of Polysaccharide Fractions from Radix A. Sinensis against tert-Butylhydroperoxide Induced Oxidative Injury in Murine Peritoneal Macrophages

  • Three Angelica sinensis polysaccharide fractions (APFs), named APF1, APF2 and APF3, were isolated and purified from Radix A. sinensis and their antioxidant activities were evaluated in isolated mouse peritoneal macrophages by pretreatment with APFs before exposure to 0.2 mM tertbutylhydroperoxide (t-BHP). The results showed that pretreatment of the macrophages with APFs as low as $10{\mu}g$/ml could significantly enhance t-BHP-decreased cell survival, intracellular glutathione (GSH) content and superoxide dismutase (SOD) activity, and also inhibited t-BHP-increased lactate dehydrogenase (LDH) leakage and malondialdehyde (MDA) formation (p < 0.05), and APF3 was the most active fraction, followed by APF2 and APF1 in decreasing order. Furthermore, we found for the first time that the bound-protein in APF3 was associated closely with the protective effects and the polysaccharide inhibited the excess NO release from t-BHP-activated macrophages to protect host cells.
