인공사료의 단백질 및 탄수화물 수준이 가잠의 실용형질과 Amylase활성에 미치는 영향 (Effects of Level of Dietary Protein or Carbohydrate on the Economic Characters and Amylase Activity of the Silkworm, Bombyx mori)

  • 5령기에 있어서 인공사료 중 탄수화물 함량과 단백질 함양의 다소가 누에의 실용형질 및 소화액 Amylase 활성에 미치는 영향을 알기 위하여 탄수화물 급원으로서 서당과 단백질 급원으로서 대두박 의 수준을 달리한 9종의 입공사료로 사육하여 조사할 결과는 다음과 같다. (1) 사료 중 탄수화물 함량과 단백질 함량의 다소는 유충의 증체량 몇 견질에 미치는 영향이 클뿐만 아니라 사료효율에도 영향을 주므로 적량의 대두박 및 서당의 첨가가 필요하다. (2) 유충의 식하양은 증체량이 많은 유충에서 많았다. (3) 사료 중 단백질 함량이 많을수록 유충의 소화량이 많고 소화율이 높았다. 그러나 탄수화물 함량의 다소와 유충의 소화량 및 소화율과는 일정한 경향이 없었다. (4) 유충의 소화액 Amylase 활성과 사료 중 단백질 및 탄수화물 함량과는 일정한 상관관계가 없었으나 잠체내의 탄수화물 농도를 높일 수 있는 사요에서는 활성이 강하였다. (5) 사료 중 단백질 함량의 다소와 잠분 중 조단백질 함량과는 부의 상관관계가 있었고 가용무질소물 함량과는 부의 상관관계가 있었다. 한편 사료 중 탄수화물 함량이 많으면 않을수록 잠분 중 가용질소물 함량은 많아지나 친단백질 함량은 오히려 적어졌다. (6) 인공사료육잠은 상엽육잠보다 Amylase 활성이 강하여도 유충증체량이 적고 견질이 불량하여 경제적 가치가 적었다. (7) 상엽분말이 44% 함유된 5영기 인공사료에 있어서 서당의 최적 함양은 0g이고 대두박의 최적 함량은 40g이었다. (8) 인공사료에 었어서 Amylase 활성을 강하게 할 수 있으나 실용형질이 불량하여 경제적 가치가 없는 것으로 보아 Amylase 활성을 강하게 하는 인공사료의 개발만으로는 산업적인 육잠목표를 달성할 수 없다고 생각한다.량하나, 빠를수록 급격하게 불량하다. 2) 우리나라의 잠기별 잠작은 춘(1~4기), 추(7~10기), 만추(11기), 하(5기) 초추(6기)잠 순으로 불량하였다. 3) 잠작과 영역접배를 고려한 다회 보통육은 1, 3, 8, 10기 등 4회육이 적합하고 조상 4회육은 10기 대신에 11기 대치로 무난하다. 2. 양잠농가의 추잠작 불안정 요인시험 1) 양잠농가의 수견량을 지배한 치명적인 요인은 잠실 잠구의 소독불충분에 의한 병원축적이며 2) 이차적 요인은 사육온습도 관리불량, 배게치기, 적숙엽 선정기술 불량 및 결상총과소 등이며 잠작과 경영구 요소는 관련이 없는것 같다. 3) 견층비율에는 뽕의 엽질이 가장 크게 영향되고, 다음으로 통원누적으로 인한 누에의 건강도에 영향한 것 같다. 3. 육잠기술 지도효과 1) 지도농가의 상자당 수견량 춘잠기 32.3kg, 추잠기 25.8kg으로서 대조 농가보다 각각 20%, 35%가 증수되어 연평균 29.0kg으로서 26%의 증수를 실증하였다. 2) 지도농가의 3등견 이상의 고치는 춘잠기 77.1%, 추잠기 83.7%로서 대조농가보다 각각 14.3%, 11.3%가 많았다. 3) 지도농가의 지도후년도의 상자당 수견중은 춘잠기 31.2kg, 추잠기 26.9kg, 연평균 28.9kg으로서 대조농가보다 각각 3%. 16%, 8%가 증수되였으므로 이는 기술지도의 잔류효과로 인정하였다. 4) 기술수준에 따른 잔류효과는 기술수준 상위농가는 0% 하위농가는 19%이었다. 4. 상엽질의 계절적 변화추이 1) 장잠기 상엽을 주재로 한 인공사료육의 결과는 춘> 만추≒하>추≒> 초추의 순으로 우수했다. 2) 치잠기 상엽을 주재로한 인공사료육의 결과는 춘, 만추, 추, 하, 초추 순위의 경향을 보였으나 유의차는 인정되지 않았다. 5.

참당귀(Angelica gigas)의 DPPH Radical 소거 활성과 항산화 효과 (DPPH Radical Scavenger Activity and Antioxidant Effects of Cham-Dang-Gui (Angelica gigas))

  • 본 연구의 결과를 요약하면 참당귀추출물 1 mg/mL의 농도에서 열수추출물의 free radical소거 효과는 메탄올추출 물보다 높게 나타났으며 농도 의존적으로 DPPH free radical 소거 효과가 상승하는 것을 관찰하였다. 또한 참당귀 투여에 의하여 화학요법으로 발생된 지질과산화물의 생성을 감소시켰으며, 단백질 산화를 유의적으로 억제함으로써 참당귀추출물은 CYP를 투여로 생성된 free radical을 감소시킴으로써 독성을 감소시켜, 혈액학적 빈혈지표가 더 감소되는 것을 막았다고 사료되며, 항산화제로서의 가능성이 있는 한약재로서 활용성이 높을 것으로 사료된다 항산화효소인 SOD 활성은CYP투여에 의하여 활성이 증가하였으며 참당귀추출물의 투여에 의한 효과가 나타나지 않았으나 감소하는 경향을 보였고 catalase 활성은 CYP 투여에 의하여 활성이 증가하였으나 참당귀의 효과는 차이가 없었다. 결론적으로 참당귀추출물은 free radical 소거능을 갖고, 화학요법으로 발생된 지질과산화물의 생성을 낮춰 화학요법에 의한 부작용으로 발생되는 혈액학적 수치의 감소를 막음으로써 항산화제로의 활용성을 기대해본다.

체감의이인탕(體減薏苡仁湯)이 비만유도 흰쥐의 체중, 지방조직, 혈액변화, leptin과 Uncoupled protein에 미치는 영향 (Effects of Chegameyiin-tang extract on the change of the weight, tissue in epididymal fat, blood, leptin and uncoupled protein in visceral fat of obesity rats induced by high fat diet)

  • In Oriental medicine, there has been a theory that the deficiency of the Qi(氣) and the Phlegm(濕)-Damp(痰) bring Obesity. And so a clinically representative herb-medicine of the obesity treatments is Chegameyiin-tang . We observed the effects of Chegameyiin-tang on the fat tissues and what the function of Chegameyiin-tang is. These experimental studies were designed to investigate the effects of Chegameyiin-tang on the weight and lipid metabolism of obesity rats induced by high fat diet. And what is changed in the blood and how the leptin and uncoupled protein is affected. The measurement has been performed on (1) the weight of obese rats fed high fat diet, (2) the average size and number of epididymal fat cells, (3) the total cholesterol, triglyceride, glucose. and free fatty acids in the blood. and (4) the leptin and uncoupled protein in the blood are observed. The results are as follows; 1. In the sample group, the weight decrease occured significantly throughout the whole research period than that of control group. 2. In the sample group, epididymal fat weight showed significantly decrease in the 8th and 14th weeks than that of control group.3. In the sample group, epididymal fat cell size was decreased significantly in the 8th and 14th weeks than that of control group. 4. In the sample group, total cholesterol. triglyceride and glucose increased rather than control group in 8 weeks, those decreased significantly in 14 weeks. 5. In the sample group, free fatty acids and insulin increased rather than control group in 8 weeks. those showed some decrease in 14 weeks . 6. In the sample group, leptin decreased significantly than control group in 8, 14 weeks. Uncoupled protein showed some decrease in 8 weeks. that decreased significantly in 14 weeks.

지방 함량에 따르는 흰쥐의 체내 대사 연구 (A study of metabolic effect in high and low fat diet on Albino Rat)

  • male female 80 마리의 흰쥐를 저지방식, 고지방식, 무지방식, 표준식이를 주어 16주간 동안 사육한 결과는 다음과 같다. 동물의 체중증가율 질소의 이용율, 간과 serum 내의 성분의 변화를 분석 시험한 결과를 요약하연 다음과 같다 지방의 식이내 함량에 따라서 체내 질소 대사에 미치는 영향으로 총 질소 보유율과 serum 내의 총단백질 albumin, globulin 의 함량, 각 장기 내의 질소 함유량들을 보았으나 대체적으로 현저한 변화를 가져오지 않는 것으로 본다. 그러나 각 장기에 있어서 총 질소 함유량을 보면 liver 와 spleen 이 식이 성분에 대해서 예민한 반응을 보였으며, brain 이 가장 적은 변화를 나타내었다. 한편 serum 내 glucose함량에는 식이 함유된 지방 함량에 따라서 큰 변화를 나타내지는 않았다. 각 군간의 유의적인 차이는 산발적으로 나타났지만 저지방군과 표준군을 male female에서 비교해 보면 다 같이 유의적인 차이를 나타내지는 않았다 . 이로 미루어 보면 2% 저지방군이 serum 내의 glucsoe 양에는 영향을 크게 미치지는 않는다고 볼 수 있다. serum 내 lipid 와 cholesterol 의 함량은 식이내 함유된 지방량에 따라서 현저한 차이를 나타냈다. 고지방군인 30% 지방 첨가로 serum 내의 lipid 와 cholesterol 의 함량이 증가되었으며 저지방군인 2% 의 식이내 지방 첨가는 표준군과 큰 차이를 나타내지 않는 것으로 보아 2%의 저지방군이 지방질 체내 축척에 있어서는 유리하다고 볼 수 있다 . 한국 영양 섭취 실정으로 미루어 보면 지방의 섭취량을 우선 일정량으로 올리는 것이 중요한 일이라고 보는데 본 실험의 결과에 따르면 식사내 2% 지방 첨가가 표준군과 모든 면에서 유사하였다. 반면 대부분의 구미 각국에서는 총열량에 대한 지방량의 비율이 $30{\sim}40%^{7)}$로서 상당히 많은 량의 지방을 섭취하고 있으나, 이들 각국은 지방 섭취량을 저하시키는 경향이므로 지방 섭취량의 대한 권장을 고려해야 할 필요가 있는 것으로 본다.

알코올 및 고콜레스테롤 식이로 유도된 흰쥐의 지방간에서 지방산 결합단백질의 역할 및 특성 (The Role of Fatty Acid Binding Protein in the Fatty Liver Induced by Alcohol or High Cholesterol Diet in Rats)

  • There is a marked increase in geriatric disease, especially liver disease, due to the continuous increase in alcohol and fat consumption. Since the fatty liver, induced by alcohol or fat, is basically from abnormalities in the lipid metabolism, it is possible that fatty acid binding protein(FABP) which is related to the fatty acid metabolism may also be abnormal in these livers. FABP is a small molecular weight protein family present in cytosol in high concentration. It has been proposed as a fatty acid transfer protein and as a binding protein responsible for controlling intracellular free fatty acid concentration. In this research, we have examined the relationship between liver FABP and fatty liver induced by alcohol or high cholesterol diet. Rats were fed one of either semipurified liquid diets; control diet containing 65% carbohydrate, 20% protein, and 15% fat or high cholesterol diet containing 1%(w/w) cholesterol or alcohol diet containing 37% of alcohol instead of carbohydrate. After 5 weeks of feeding period, all rats received commercial chow diet for 5 weeks to examine recovery effect. Liver and blood samples were collected at 0, 1, 3, 5 and 10 weeks to analyze lipid compositions. FABP was purified from liver cytosol and injected to rabbit to obtain antiserum. Liver FABP amount was determined by SDS-PAGE and western blotting methods. Fatty acid binding capacity was determined by binding of 14Cpalmitate with the delipidated liver cytosol. Consumption of alcohol increased serum cholesterol, triglyceride concentration and decreased HDL-cholesterol concentration after 5 weeks. Serum apolipoprotein B concentration increased after 3 weeks and LDL-cholesterol and apolipoprotein A concentration changed after 1 week. Liver cholesterol and triglyceride concentration increased after 3 weeks. Consumption of high cholesterol diet changed liver and serum lipid composition after 3 weeks. Swiching to normal diet for 5 weeks did not normalize most of lipid composition in serum and liver except serum and liver except serum cholesterol, triglyceride and liver cholesterol. Liver cytosol FABP content and the fatty acid binding capacity decreased dramatically after 1 week with alcohol consumption. This results indicate that FABP content changes before the changes before the changes of blood or liver lipid composition, suggesting changes of FABP may cause development of the fatty liver induced by alcohol and can be used as an index of detecting a early development of fatty liver.

Methodologies for the Determination of Endogenous Phosphorus Losses in Growing Pigs

  • A study was conducted to compare the endogenous basal losses of phosphorus (EBLP) in pigs fed diets containing gelatin (GEL) or spray-dried porcine plasma (SDPP) as protein sources and to determine the standardized total tract digestibility (STTD) of phosphorus (P) in SDPP. The trial was carried out at the Federal University of Santa Maria, Brazil. Twelve castrated pigs with an initial body weight of 55 kg were individually allotted in metabolic crates during two 12-day periods, each with 7 days of adaptation and 5 days of total fecal collection. The beginning and the end of the collecting periods were determined according to the marker-to-marker approach, using ferric oxide as an indigestible marker. Pigs were submitted to four semi-purified diets, one being a P-free diet with 30% of GEL as the protein source and three were diets with 10%, 20%, and 30% inclusion of SDPP respectively. Data were subjected to analysis of variance and the model included the effects of period, animal and treatments; the results of the three diets with increased levels of SDPP were subjected to linear regression analysis. The intercept of the relation of between ingested P and absorbed P represented the EBLP, while the slope indicated the STTD of P in SDPP. The EBLP means obtained by P-free diet and regression method were compared with the Student t test. The EBLP were 128.95 mg/kg dry matter intake (DMI) and 153.63 mg/kg DMI (standard error = 77.0; p<0.06) using the P-free diet with GEL as the protein source and the regression method, obtained with diets containing increased levels of SDPP, respectively. The apparent digestibility of P was 87.9%, 94.2%, and 92.9% for the treatments containing 10%, 20%, and 30% inclusion of SDPP, respectively. The estimated STTD of P obtained with the linear regression was 97.4%. When the EBLP estimated by the P-free diet was used to corrected the apparent digestibility of P in diets containing SDPP, the STTD of P in SDPP was 96.9%, 98.8%, and 95.9% for 10%, 20%, and 30% SDPP, respectively. Therefore, it can be concluded that SDPP can replace GEL to estimate the endogenous losses of P. In addition, the STTD of P in SDPP estimated with the P-free diet was 97.2% and it was 97.4% by the regression method, utilizing SDPP.

Effect of Lowering Dietary Protein with Constant Energy to Protein Ratio on Growth, Body Composition and Nutrient Utilization of Broiler Chicks

  • A trial was conducted to determine the effect of low crude protein (CP) diets with constant metabolizable energy to crude protein (ME:CP) ratio on growth, body composition and nutrient utilization of broiler chicks from 1 to 26 days of age. Four dietary treatments having four levels of CP and ME as 23, 22, 21 and 20% and 3,036, 2,904, 2,772 and 2,640 kcal/kg, respectively, were formulated and a ME:CP ratio of 132 was maintained in all the diets. Digestible lysine was maintained at 1.10 of the diet. A total of 1,760 day-old Hubbard broiler chicks were randomly divided into 16 experimental units and each diet was offered to four experimental units at random. Feed intake was increased (p<0.05) while weight gain and feed conversion ratio were adversely affected (p<0.05) when the diets with low CP and ME were fed to broilers. Total protein intake and total ME intake were linearly decreased (p<0.05) and protein efficiency ratio and energy efficiency ratio were lower (p<0.05) than in the chicks fed dietary regimen with 22% CP and 2,904 kcal/kg ME. The whole body analysis of the birds revealed that chicks fed the lowest dietary regimens retained less (p<0.05) nitrogen and more ether extract than chicks fed the control diet, however, body dry matter, total body ash and fat free body protein were not affected. Similarly, protein and energy utilization were also unaffected by the dietary treatments. In summary, chicks fed low CP diets with constant ME:CP ratio grew slower, used feed less efficiently and retained less protein and more body fat than chicks fed the control diet.

The Effects of Supplementing Methionine plus Cystine to a Low-protein Diet on the Growth Performance and Fat Accumulation of Growing Broiler Chicks

  • This experiment was conducted to determine the effects of a low-protein diet supplemented with DL-methionine plus L-cystine (Met + Cys) on the growth performance and fat accumulation of female broiler chicks during the growing period (3-6 wks old). A low-protein diet (17% CP; 3,200 ME kcal/kg) was supplemented with Met + Cys (1.1 : 1.0) at levels 0.75, 0.94, 1.25, 1.31 or 1.50% of diet, respectively. Another diet with 21% CP and 3,200 ME kcal/kg served as the control group. All essential amino acids were adjusted to meet the National Research Council (1984) requirement for chicks. Feed and water were given ad libitum. Body weight of the chicks fed the low-CP diets supplemented with Met + Cys were heavier than those of the control birds. Feed conversion ratio and feed intakes were not significantly different between and among the treatment groups. Similary, abdominal fat content was not significantly different among the various treatments except that of the chicks fed the low CP diet with 1.25% Met + Cys which was higher than that of the control group. Fatty acid synthetase (FAS), acetyl-CoA carboxylase (ACC) activities and carcass protein content were not influenced by dietary treatments. Carcass fat content was lowest in chicks fed low CP diet with 0.75% Met + Cys and highest in the group that received 1.50% Met + Cys supplementation. Liver triglyceride increased as Met + Cys supplementation level increased. Various lipid fraction concentrations (cholesterol ester, free cholesterol, and phospholipid) in the serum went up as Met + Cys increased up to 1.25% after which it levelled off. Results of this experiment suggest that it is possible to reduce dietary protein level from 21% to 17% for growing broiler chicks by the supplementation of Met + Cys when other EAA were sufficient.

  • This experiment was designed to observe some influence on the serum components of growing rats maintained with basal diet supplemented with sausage. Forty male Albino rats were used as the experimental animals and the subjects were divided into four diet groups (control group; feeding of basal diet containing 10.8% and 10.4% of protein and lipid respectively, A group; feeding of sausage containing 12.5% and 27.6% of protein and lipid respectively supplemented with basal diet every day, B group; feeding of sausage supplemented with basal diet once a three-day and C group; feeding of sausage supplemented with basal diet once a five-day) and each group was fed on the corresponding diet for forty-days. The contents of protein, lipid and cholesterol in their serum were determined. The result obtained are summarized as follows: 1) The final body weight and liver weight of the A group were increased more than other groups. 2) The protein contents in the serum of the B group was increased more than other groups. 3) The total lipid contents in the serum of the A group and B group were decreased compared with control group. 4) The total cholesterol, ester form cholesterol and free form cholesterol in the serum of the A group were decreased, but the total cholesterol and ester form cholesterol in that of the C group were increased compared with control group.

Comparison of the Efficiency of Absorbed Nitrogen Use from Different Protein Sources in Diets Having Similar Amino Acid Balance

  • Nine crossbred female pigs fitted with the bladder catheters were used to investigate the effects of dietary protein form on the efficiency of absorbed nitrogen for nitrogen retention in growing pigs. Combinations of the main protein sources were corn-soybean meal (CSM; slow + slow absorption rate form), corn-hydrolyzed casein (CAS; slow + rapid absorption rate form) and corn-porcine plasma (CPL; slow + intermediate absorption rate form). All experimental diets were formulated to be isonitrogenous (CP 11%) and isocaloric (3.5 Mcal/kg) and synthetic amino acids were added to the diet as required to maintain an equivalent amino acid profile among diets. Fecal digestibility of nitrogen was not different among treatments (p > 0.10). Ingested nitrogen was absorbed with an apparent efficiency of 82% to 84%. Mean nitrogen retention in pigs fed the CSM diet was as high as for pigs fed the CPL diet (0.74 g N/kg $BW^{0.75}$ per d), which was higher than the N retention rate in pigs fed CAS diet (0.68 g/kg $BW^{0.75}$ per d; P < 0.05). Apparent biological values (ABV = 100 ${\times}$ N retention/absorbed nitrogen) were 63.3%, 58.0% and 61.6% for CSM, CAS, and CPL groups, respectively (p < 0.05). There was no difference in mean energy digestibility among treatments. The efficiency of absorbed lysine utilization was significantly different among treatments (p < 0.05). Pigs fed the CAS diet were inferior to counterparts on the other diets in utilizing absorbed lysine. The ratios of free (and small peptide-bound) to protein-bound amino acids in CSM diet differed considerably from the CAS diet. This may affect the efficiency of amino acids utilization for nitrogen retention if hydrolyzed and intact amino acid pools reach the blood at different times.
