Local protein synthesis in neuronal axons: why and how we study

  • Adaptive brain function and synaptic plasticity rely on dynamic regulation of local proteome. One way for the neuron to introduce new proteins to the axon terminal is to transport those from the cell body, which had long been thought as the only source of axonal proteins. Another way, which is the topic of this review, is synthesizing proteins on site by local mRNA translation. Recent evidence indicates that the axon stores a reservoir of translationally silent mRNAs and regulates their expression solely by translational control. Different stimuli to axons, such as guidance cues, growth factors, and nerve injury, promote translation of selective mRNAs, a process required for the axon's ability to respond to these cues. One of the critical questions in the field of axonal protein synthesis is how mRNA-specific local translation is regulated by extracellular cues. Here, we review current experimental techniques that can be used to answer this question. Furthermore, we discuss how new technologies can help us understand what biological processes are regulated by axonal protein synthesis in vivo.

노랑초파리의 난자형성과정에서의 단백질 합성의 변화 (Changes of Protein Synthesis during Oogenesis of Drosophila melanogaster)

  • 노랑초파리(Drosophila melanogaster)의 난자형성과정 중 egg chamber 이외의 조직에서 합성, 이입될 수 있는 가능성을 배제한 상태에서 egg chamber에서 일어나는 단백질 합성양상의 변화를 연구하였다. 단백질 합성률은 전반적으로 큰 변화를 보이지 않으나 난황형성, 난황막형성, 그리고 난각형성이 겹쳐 이루어지는 11-12에서는 높고, 성숙이 완료되는 단계 14에서는 낮았다. 누적 및 합성 단백질의 수와 종류가 단계에 따라 크게 변하고 있으며, 노랑초파리의 난자형성이 비록 짧은 기간동안에 이루어질지도 단계에 따라 특이한 단백질 합성 변화를 보이고 있다. 이러한 단백질의 합성은 주로 여포세포에서 이루어지는 것으로 생각되며, 노랑초파리의 난포세포의 성장 및 성숙을 위해서는 egg chamber 이외의 조직 뿐만 아니라 egg chamber 내의 세포 자체에 의한 단백질 합성이 직접적으로 관여하는 것으로 생각된다.

Effect of Intensity of Unconditional Stimulus on Reconsolidation of Contextual Fear Memory

  • Memory reconsolidation is ubiquitous across species and various memory tasks. It is a dynamic process in which memory is modified and/or updated. In experimental conditions, memory reconsolidation is usually characterized by the fact that the consolidated memory is disrupted by a combination of memory reactivation and inhibition of protein synthesis. However, under some experimental conditions, the reactivated memory is not disrupted by inhibition of protein synthesis. This so called "boundary condition" of reconsolidation may be related to memory strength. In Pavlovian fear conditioning, the intensity of unconditional stimulus (US) determines the strength of the fear memory. In this study, we examined the effect of the intensity of US on the reconsolidation of contextual fear memory. Strong contextual fear memory, which is conditioned with strong US, is not disrupted by inhibition of protein synthesis after its reactivation; however, a weak fear memory is often disrupted. This suggests that a US of strong intensity can inhibit reconsolidation of contextual fear memory.

생쥐 배아의 2세포기 분열과정에 있어서의 단백질 합성 분석 : Colcemid와 a-Amanitin의 영향 (Patterns of Protein Synthesis During the Second Cleavage of Mouse Two-Cell Embryos: Effects of Colcemid and a-Amanitin)

  • In this study, we attempted to determine the precise patterns of protein synthesis during the second cleavage in mouse. F1 hybrid 2-cell embryos showing a highly synchronized cell cycle and outbreed ICR strain 2-cell embryos were used. The patterns of protein synthesis during the second cleavage showed the sequential changes in the F1 hybrid and ICR strain 2-cell embryos. Moreover, we examined the effects of mitotic and transcriptional inhibitors such as colcemid and a-amanitin on the protein synthesis in the late 2-cell embryos of ICR strain. Treatment of colcemid (0.1mg/ml) blocked the second cleavage, but did not affect on the change of protein synthesis. However, treatment of a-amanitin induced the synthesis of two set of polypeptides without affecting on synthesis of other proteins and cleavage. It thus seems that the appearance of a-amanitin-sensitive proteins may be not involved in the second cleavage. Therefore, these results indicate that the second cell cycle in mouse embryos appears to be regulated at post transcriptional level, presumably independent on the expression of embryonic genome. 본 연구는 생쥐 배아의 2세포기 분열과정중 단백질 합성양상과 단백질합성에 미치는 colcemid와 $\alpha$-amanitin의 영향을 조사하였다 이를 위하여 체내 수정된 ICR strain의 2세포기 배아와 매우 일치된 초기배아 분열양상을 보여주는 체외 수정된 F1 (C57BL x CBA) hybrid 2세포기 배아를 사용하였다. 두 종류의 2세포기 배아에서 단백질 합성은 분열단계에 따라서 매우 일치된 변화를 보여 주었다. 또한 유사분열 억제제인 colcemid (0.1mg/ml)의 처리는 2세포기 배아분열을 억제하였으나, 단백질 합성에는 아무런 변화를 주지 못하였다. 그리고 후기 2세포기 배아에 전사 억제제인 a-amanitin (100mg/ml)을 처리하였을 때 세포분열이나 다른 단백질의 합성에는 아무런 영향이 없이 단지 두개의 단백질의 합성만을 유도하였다. 이는 아마도 a-amanitin의 stress효과에 기인하는 것으로 추측된다. 따라서 생쥐 2세포기 배아의 분열과정은 배아게놈의 유전자 발현과는 무관하게 이미 합성되어 존재하는 mRNA에 의하여 조절되는 것으로 사료된다.

Cricket (Gryllus bimaculatus) meal pellets as a protein supplement to improve feed efficiency, ruminal fermentation and microbial protein synthesis in Thai native beef cattle

  • Objective: Replacing soybean meal (SBM) with cricket (Gryllus bimaculatus) meal pellets (CMP) in concentrate diets was investigated for feed efficiency, ruminal fermentation and microbial protein synthesis in Thai native beef cattle. Methods: Four male beef cattle were randomly assigned to treatments using a 4×4 Latin square design with four levels of SBM replaced by CMP at 0%, 33%, 67%, and 100% in concentrate diets. Results: Results revealed that replacement of SBM with CMP did not affect dry matter (DM) consumption, while digestibilities of crude protein, acid detergent fiber and neutral detergent fiber were significantly enhanced (p<0.05) but did not alter digestibility of DM and organic matter. Increasing levels of CMP up to 100% in concentrate diets increased ruminal ammoniacal nitrogen (NH3-N) concentrations, blood urea nitrogen, total volatile fatty acids and propionate concentration (p<0.05), whereas production of methane and protozoal populations decreased (p<0.05). Efficiency of microbial nitrogen protein synthesis increased when SBM was replaced with CMP. Conclusion: Substitution of SBM with CMP in the feed concentrate mixture at up to 100% resulted in enhanced nutrient digestibility and rumen fermentation efficiency, with increased volatile fatty acids production, especially propionate and microbial protein synthesis, while decreasing protozoal populations and mitigating rumen methane production in Thai native beef cattle fed a rice straw-based diet.

Effect of inclusion of different levels of Leucaena silage on rumen microbial population and microbial protein synthesis in dairy steers fed on rice straw

  • Objective: Leucaena leucocephala (Leucaena) is a perennial tropical legume that can be directly grazed or harvested and offered to ruminants as hay, silage, or fresh. However, Leucaena contain phenolic compounds, which are considered anti-nutritional factors as these may reduce intake, digestibility and thus animal performance. Therefore, the objective of this experiment was to determine effects of Leucaena silage (LS) feeding levels on rumen microbial populations, N-balance and microbial protein synthesis in dairy steers. Methods: Four, rumen fistulated dairy steers with initial weight of $167{\pm}12kg$ were randomly assigned to receive dietary treatments according to a $4{\times}4$ Latin square design. Treatments were as followings: T1 = untreated rice straw (RS; Control), T2 = 70% RS+30% LS, T3 = 40% RS+60% LS, and T4 = 100% LS. Dairy steers were fed rice straw and LS ad libitum and supplemented with concentrate at 0.2% of body weight/d. Results: Results revealed that the rumen microbial population, especially cellulolytic, proteolytic bacteria and fungal zoospores were enhanced in steers that received 60% of LS (p<0.05), whereas the amylolytic bacteria population was not affected by treatments (p>0.05). Protozoal population was linearly decreased with increasing level of LS (p<0.05). Moreover, N-balance and microbial protein synthesis were enhanced by LS feeding (p<0.05) and were the highest in 60% LS group. Conclusion: Based on this study, it could be concluded that replacement of RS with 60% LS significantly improved microbial population and microbial protein synthesis in diary steers.

Effects of Active Immunization against Somatostatin or its Analogues on Milk Protein Synthesis of Rat Mammary Gland Cells

  • Effects of active immunization against native 14-mer somatostatin (SRIF, somatotropin releasing inhibiting factor) and its two 14-mer-somatostatin analogues on the milk production in rat mammary cells were studied. Native SRIF, Tyr11-somatostatin (Tyr11-SRIF), and D-Trp8, D-Cys14-somatostatin (Trp8Cys14-SRIF) were conjugated to bovine serum albumin (BSA) for immunogen preparation. Twenty-four female Sprague-Dawley rats were divided into four groups and immunized against saline (Control), SRIF, Tyr11-SRIF, and Trp8Cys14-SRIF at five weeks of age. Booster immunizations were performed at 7, 9, and 11 weeks of age. SRIFimmunized rats were mated at 10 weeks of age. The blood and mammary glands were collected at day 15 post-pregnancy and -lactation. To measure the amount of milk protein synthesis in the mammary gland, mammary cells isolated from the pregnant and the lactating rats, were cultured in the presence of $^3H$-lysine. No significant differences in growth performance, concentration of growth hormone in the circulation, and the amount of milk protein synthesis were observed among the groups. Inductive levels of serum anti-SRIF antibody in the SRIF and Tyr11-SRIF groups but not in the Trp8Cys14-SRIF group, were significantly higher than that of the control group during the pregnancy and lactation periods. The result suggests that active immunization against native 14-mer SRIF and Tyr11-SRIF was able to induce anti-SRIF antibodies, but did not affect the milk protein synthesis.

  • Biosynthesis of nanoparticles has acquired particular attention due to its economic feasibility, low toxicity and simplicity of the process. Extracellular synthesis of nanoparticles by bacteria and fungi has been stated to be brought about by enzymes and other reducing agents that may be secreted in the culture medium. The present study was carried out to determine the underlying mechanisms of extracellular silver nanoparticle synthesis by Pseudomonas hibiscicola isolated from the effluent of an electroplating industry in Mumbai. Synthesized nanoparticles were characterized by spectroscopy and electron microscopic techniques. Protein profiling studies were done using Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (1D-SDS PAGE) and subjected to identification by Mass Spectrometry. Characterization studies revealed synthesis of 50 nm nanoparticles of well-defined morphology. Total protein content and SDS PAGE analysis revealed a reduction of total protein content in test (nanoparticles solution) samples when compared to controls (broth supernatant). 45.45% of the proteins involved in the process of nanoparticle synthesis were identified to be oxidoreductases and are thought to be involved in either reduction of metal ions or capping of synthesized nanoparticles.

  • 20-hydroxvecdysone (20-HE) seems to be related in the regulation of vitellogenesis in Drosophifa sp. (robwta species sroup). Although yolk proteins (YPs) synthesis does not occur at a high rate in fat body cells of one daw-old female after eclosion, application of 20-HE to isolated abdomens deprived of anterior endocrine glands stimulated the synthesis and secretion of YPs into the hemolvmph. An injection of 0.3 $\mul$ of a 10-s M 20-HE was sufficient topromote synthesis and secretion of YPs in isolated abdomens. The response of isolated automens to hormones was first detected between 2 hr and 3 hr after treaDent of 10-s M 20-HE. Transcript analysis showed that the effect of 20-HE on yolk protein synthesis was mediated at the level of transcription.

Effects of Whole Crop Corn Ensiled With Cage Layer Manure on Nutritional Quality and Microbial Protein Synthesis in Sheep

  • An experiment was conducted to study the nutritional quality of whole crop corn silage ensiled with cage layer manure in sheep. Treatments were designed as a $3{\times}3$ Latin square with 16-day periods. Sheep were allotted in one of three diet-treatments, which were whole crop corn silage (CS), whole crop corn+30% cage layer manure (CLM) silage (based on DM; MS) and rice straw+concentrate (SC) mixed at 8:2 ratio (on DM basis). Silage ensiled with CLM significantly increased (p<0.05) digestibilities of crude protein, NDF and ADF, TDN over the other treatments. Ruminal pH in sheep fed SC was significantly (p<0.05) higher than that of the other diets at 0.5, 1, 2, 4 and 8 h after feeding. Ruminal ammonia nitrogen concentration of the MS treatment was significantly (p<0.05) higher than that of the other treatments at 0, 1, 2 h after feeding. The MS treatment highly increased (p<0.05) feed intake, digestibility of organic matter and crude protein, nitrogen intake and retained nitrogen. The MS treatment highly increased (p<0.05) purine derivative (PD) excretion leading to higher microbial protein synthesis.
