조피볼락(Sebastes schligeli)에 있어 사료내 단백질 사료원으로서의 어분대체품 (Fish Meal Analog as a Dietary Protein Source in Koran Rockfish, Sebastes schlegeli)

  • 본 연구는 조피볼락 사료에 있어서 어분대체품을 결정하고, 상품어분대체품과 유인물질의 첨가 효과를 통하여 조피볼락용 어분대체품개발을 위한 기초자료도 사용하고자 수행되었다. 실험사료는 조단백질 52%로, 가용성 에너지 16.8 KJ/g으로 설계되었고, 주단백질원으로 북양어분, 어분대체품 및 상품어분대체품을 사용하였다. 사료의 성분조성은 다음과 같이 요약된다 : 사료1, 100%FM (control) ; 사료2, 60%FM : 40% CFMA ; 사료3, 60%FM : 40% CFMA+ATT ; 사료 4, 80%FM : 20% FMA ; 사료5, 80% FM: 20%FMA+ATT : 사료 6, ; 사료6, 60%FM : 40%FMA+ATT; 사료7, 40%FM : 60% FMA+ATT. 어분대체품은 혈분(BM), 오징어간분(SLP), 육골분(MBM), 수지박(LM), 가금부산물(PBP), 우모분(FM), 필수아미노산(Met, Lys, Ile) 등을 적절히 배합하여 제작 사용하였다. 성장률, 사료효율, 일간성장률 및 단백질효율에 있어서 사료 1은 사료 4, 사료 5 및 사료 6에 비해 유의적인 차이가 없었지만(P>0.05), 나머지 실험구들은 대조구에 비해 유의적으로 낮게 나타났다(P<0.05). 특히, 상품어분대체품(CFMA)인 사료 2와 사료 3은 실험사료로 제작된 동일한 수준의 사료 6과 비교하여 성장률, 사료효율, 단백질효율에서 유의적으로 낮은 경향을 보였다. 0.5% 유인물질을 첨가한 사료 3과 5는 사료 2와 4에 비해 유의적인 차이는 보이지 않았으며(P>0.05), 간중량지수, hemoglobin 및 비만도에 있어서는 유의적인 차이가 나타났다(P<0.05). 따라서, 조피볼락에 있어서 어분대체품(FMA)으로 어분단백질을 40%정도 대체 가능함을 알 수 있었다.

단백질의 종류가 다른 식이에 L-카르니틴을 첨가했을 때 흰쥐의 지질대사에 미치는 영향 (Effects of Dietary L-Carnitine Supplementation on Lipid Metabolism in Rats fed with Different Types Protein)

  • This study is to investigate the effect of dietary L-carnitine supplementation on lipid metabolism in rats fed with isolated soy protein and casein for their source of protein. Four experimental groups were organized and each group had eight Sprague-Dawley male rats with the initial weight of around 180g. The four groups were CO (casein only supplemented group); CC (casein and 3% L-carnitine supplemented group); ISO (isolated soy protein only supplemented group); ISC (isolated soy protein and 3% L-carnitine supplemented group). All groups were supplemented with the experimental diet for four weeks and carnitine comprised 3% of. their diet. The results were as follows; 1. There was no significant difference in food intake among the groups. 2. Final weight gain was significantly lower in the groups supplemented with isolated soy protein than in the groups supplemented with casein (P<0.05). The groups with supplemented casein and carnitine showed the effect of weight reduction (p<0.05). 3. Food efficiency ratio was lower in the groups supplemented with isolated soy protein than in the groups supplemented with casein (p<0.01). The groups supplemented with casein and carnitine showed low food efficiency ratio. 4. The serum total lipid was higher in the groups supplemented with casein than in the groups supplemented with isolated soy protein (p<0.05). 5. Serum total cholesterol was higher in the groups supplemented with casein than in the groups supplemented with isolated soy protein. 6. There was no significant difference in triglyceride, HDL-cholesterol, and LDL-cholesterol in serum among the groups. 7. Out of the groups supplemented with isolated soy protein the total cholesterol level in liver was low in the groups to which carnitine was supplemented (p< 0.05). However, there was no significant difference of liver total lipid and triglyceride among the groups. 8. There was no difference in TBARS levels and GSH-Px activities in liver among the groups.

  • Several authors reported that vegetable proteins cause to decrease serum cholesterol and reported the effect of limiting amino acid balance in dietary protein on the blood component. This study was designed to observe growth rate and blood component in 1% cholesterol-fed male rats intake pure isolated soy proteins and methionine for 4 weeks. The diets were supplemented with either 20% casein or isolated soy protein as dietary protein sources and methionine as limiting amino acid of isolated soy protein and casein protein. The results were as follows; 1) The body weight gain was the greatest in casein groups. 2) Serum total cholesterol concentration was significantly increased by casein groups and slightly decreased by isolated soy protein groups. 3) Serum HDL-cholesterol concentration was significantly increased by isolated soy protein groups than control group. 4) Serum total lipid and triglyceride concentrations were significantly decreased by isolated soy protein groups than control group. 5) Serum glucose concentration was significantly increased by isolated soy protein plus 0.8% methionine than control group. 6) Serum total protein and albumin concentrations were significantly increased by all experimental diets groups than control group. Especially, casein groups were the most increased in all experimental diets groups.

포유모돈에 있어 복합효소제 함유 식물성 단백질 공급원이 생산성과 돈유성상에 미치는 영향 (Effects of Plant Protein Source Containing Multienzyme on Performance and Milk Characteristics in Sow)

  • 본 시험은 복합효소제 함유 식물성단백질공급원이 포유모돈의 생산성, 영양소 소화율 및 돈유성상에 미치는 효과를 알아보고자 실시하였다. 모돈은 건강상태 및 산차를 고려하여 3처리, 처리당 10두씩 30두를 공시하였다. 사료는 1차로 분만 일주일 전부터 급여하고 2차로 분만 후부터 21일령 이유시까지 급여하였다. 시험설계는 1)CON(basal diet), 2)CGLT(included corn gluten) 및 3) FSPM(included fermented soy protein containing multienzyme)로 하였으며, 분만후 자돈은 모돈에 복당 10마리씩 균일하게 배치하였다. 포유모돈의 영양소 소화율에서는 질소 소화율에서 FSPM 처리구가 CON 처리구와 비교하여 유의적으로 높았다(P<0.05). 포유모돈의 혈중 요소태질소 함량에서는 종료시에 FSPM 처리구가 CGLT 처리구보다 유의적으로 높았으며(P<0.05) 변화량에 있어서는 FSPM 처리구가 CON 및 CGLT 처리구보다 유의적으로 높았다(P<0.05). 포유모돈의 모유내 지방 함량에서는 종료시에 FSPM 처리구가 CON 처리구보다 유의적으로 높았으며(P<0.05), 모유내 단백질 함량에서는 개시시에 FSPM 처리구가 CGLT 처리구와 비교하여 유의적으로 높았다(P<0.05). 포유모돈의 직장온도 변화에 있어서는 세 처리 모두 유사한 변화를 보였다. 포유자돈의 성장률에서는 종료시체중, 증체량 및 일당증체량에서 FSPM 처리구가 CON 처리구와 비교하여 유의적으로 높았으며(P<0.05) CGLT 처리구는 CON 처리구보다 높았으며 FSPM 처리구보다 낮았다.포유자돈에서의 설사발생에 있어서는 0~5일째에 CGLT 처리구에서 한 마리가 발생하였다. 본 시험결과 사료내 0.25% 복합효소제 함유 식물성단백질공급원의 급여는 포유모돈의 질소 소화율과 혈중 요소태질소 함량, 모유내 단백질과 지방 함량을 향상시켰으며 포유자돈의 증체량 또한 향상시켰다.

  • 본(本) 연구(硏究)는 에너지와 단백질(蛋白質) 공급원(供給源)에 따른 반추위내(反芻胃內) 영양소(營養素) 이용성(利用性) 및 반추위(反芻胃) 미생물(微生物)과의 관계(關係)를 조사하고자 에너지원중 반추위에서 느리게 분해되는 옥수수와 빠르게 분해되는 보리와 beet pulp를 사용하고 단백질원중 빠르게 분해되는 채종박과 느리게 분해되는 옥수수 글루텐과 면실박을 이용 하고, $2{\times}2$요인 시험으로 설계하여 처리당 면양4두를 공시하여 소화율, 반추위 성상 및 미생물 수를 조사한 바 다음과 같이 요약되는 결과를 얻었다. 1. 반추위내(反芻胃內) pH는 사료급여후(飼料給與後) 경과시간에 관계없이 F-F구가 가장 높았고, F-S구가 가장 낮았다 2. 반추위내(反芻胃內) 암모니아 농도는 S-F구가 다른 처리구보다 높았으며, 모든 처리구에서 사료급여 직후 급격히 증가하여 1시간째 최고에 달하였다가 이후 급격히 떨어지는 경향을 보이다가 사료(飼料) 급여 직전에 다시 상승하였다 3. 반추위내(反芻胃內) 총 휘발성지방산, 초산, 프로피온산 및 뷰틸산 농도는 F-S구가 가장 높았으며, 시간의 경과에 따른 경향은 1~3시간대에서 1차 peak를 이룬 후 9시간대에 2차 peak를 형성하였다. 4 시험사료(試驗飼料)의 일반성분 및 ADF소화율을 처리간에 영향이 없었으나 NDF소화율은 F-F구가 가장 높았다. 5. 질소축적율(窒素蓄積率)은 S-F구가 처리구중 가장 높았으며 F-F 및 F-S구가 가장 낮았다. 6. 시험사료(試驗飼料)의 대사율(代謝率), DCP, TDN, DE 및 ME 함량(含量)은 처리구간에 영향이 없었다. 7. 총균수(總菌數)는 사료급여후 3시간때 까지 전처리구 공히 증가하였다가 점차 감소하였고, 전체적으로 보면 F-F급여구가 $7.16{\times}10^{11}$ 으로 가장 많았으며 F-S구가 $2.85{\times}10^{11}$ 으로 가장 적었다. 8. Cellulolytic박테리아 수(數)는 S-S급여구가 다른 급여구보다 많았으며, 그 외 기능별 미생물수(微生物數)는 처리간에 차이를 볼 수 없었다.

Effects of The Soy Protein Level on Plasma Glucose, Lipids, and Hormones in Streptozotocin-Diabetic Rats

  • The number of diabetics in Korea is about 3 to 5 percent of the population, and the incidence is increasing yearly due to changes of life style and food intake. Diet is a key element in the management of diabetes, yet the appropriate diet for diabetes remains controversial. We have recently shown that a diet rich in protein of animal origin(casein) seems beneficial to controling plasma glucose and lipids in streptozotocin-induced diabetic rats. It therefore seemed desirable to find out whether the beneficial effect of high casein diet in experimental diabetes could also be reproduced with a vegetable source of protein(soy). The purpose of this study is to compare these results with the results of our previous study. In the present study, non-diabetic and streptozotocin-induced diabetic rats were studied in order to examine the effects of altering the level(20% vs 60%) of dietary soy protein on blood glucose, lipids, and hormones. Results of the present study showed that a high soy protein diet decreased triglyceride concentration in diabetic rats. However, diabetic rats fed a high soy protein diet were not hypocholesterolemic compared to rats fed a control diet. Moreover, diabetic rats fed a high soy protein diet had significantly increased plasma glucose concentration compared to rats fed a control diet. This study was not able to discern a specific effect of dietary protein level on insulin, glucagon, or insulin/glucagon ratio. Except for the hypotriglyceridemic effect, the results were not similar to the findings of our previous study which showed a beneficial effect on streptozotocin-induced diabetic rats fed a high casein diet.

어린병아리에서 저단백질사료내 제한아미노산의 규명과 함유황아미노산의 요구량 결정 (Identification of Limiting Amino Acids and Determination of Requirement of Total Sulfur-containing Amino Acids in a Low Protein Diet in Young Chicks.)

  • 저단백질(13% C. P.) 사료는 어린 병아리에서 단백질급원의 Net Protein Utilization가 조사하거나 또는 아미노산의 불균형을 연구할 때 적절한 방법으로 인정되고 있다. 이때의 기준 단백질로서 정제된 대두단백을 많이 이용하고 있으나 이런 사료의 제한 아미노산에 대한 연구가 되어 있지 않다. 본 연구에서는 사료내 유일한 단백질급원으로서 isolated soy protein을 15% 수준으로 함유한 purified-type의 기초사료에서 제한 아미노산을 규명하고 이의 요구량을 조사하였다. 어린병아리의 기초사료에 methionine, lysine, threonine 및 tryptophan 등을 단독으로 또는 두개 이상을 같이 첨가해서 급여한 결과. methionine만이 결핍된 것으로 나타났다. 총함유황아미노산(TSAA)의 요구량을 dose-response curve 방법에 의해 반응선과 plateau선과의 상호교차점에서 구하였는데, 최대의 성장과 사료섭취량을 위한 요구량은 각각 사료단백질의 4.73%와 3.73% 수준이였다. 이들 수치를 TSAA의 섭취량을 기준으로 표시해 보면 최대성장율, 사료섭취량 및 사료효을을 위해 각각 167.7, 136.8 및 159.7mg/bird/day의 양이 필요하였다.

Identification of Molecular Markers Linked to Ti Locus in Soybean

  • Soybean is a major source of protein meal in the world. Kunitz trypsin inhibitor (KTI) protein is responsible for the inferior nutritional quality of unheated or incompletely heated soybean meal. The objective of this research was to identify RAPD markers linked to KTI protein allele using bulked segregant analysis. Cultivar Jinpumkong2 (TiTi) was crossed with C242 (titi, absence of KTI protein) and F. seeds were planted. The $F_1$. plants were grown in the greenhouse to produce $F_2$ seeds. Each $F_2$ seed from $F_1$. plants was analysed electrophoretically to determine the presence of the KTI protein band. The present and absent bulks contained twenty individuals each, which were selected on the basis of the KTI protein electrophoresis, respectively. Total 94 $F_2$ individuals were constructed and 1,000 Operon random primers were used to identify RAPD primers linked to the Ti locus. The presence of KTI protein is dominant to the lack of a KTI protein and Kunitz trypsin inhibit protein band is controlled by a single locus. Four RAPD primers (OPAC12, OPAR15, OPO12, and OPC08) were linked to the Ti locus. RAPD primer OPO12 was linked to Ti locus, controlling kunitz trypsin inhibitor protein at a distance of 16.0 cM. This results may assist in study of developing fine map including Ti locus in soybean.

고들빼기 김치가 단백질 소화율에 미치는 영향 (Effect of Godulbaegi(Korean Lettuce, Ixeris sonchifolia H.) Kimchi on the in vitro Digestibility of Proteins)

  • Extent of reduction in protein digestibility by Godulbaegi(lxeris sonchifolia H.) kimchi powder appeared to be related to the kimchi weight-to-protein weight ratio, the kind of protein sources, part of plants and fementation period. In vitro digestibilities of protein were significantly(p<0.05) reduced as the freeze dried kimchi powder weight-to-protein ration increased from 0.5 : 1.0 to 1.0 : 1.0 for all Godulbaegi samples and protein sources. Overall digestibility of protein sources in the presence of kimchi powder, the reduction ranked in the following order : soybean>casein>beef>squid. Lower(p<0.005) reduction occurred for each protein source when raw plant was exchanged for kimchi products. Some greater reduction of digestibility was noted in young plants and leaf samples than ripe or root samples. Trypsin inhibitor, which expressed as soybena trypsin inhibitor, was inversely related(r=0.8437) to in vitro protein digestibility of casein in the presence of Godulbaegi kimchi powder. More than three times of total polyphenols contained in leaves than in roots. Young leaves had 30% more total polyphenols(37.64mg/g sample) than that in ripe ones. Soaking in 5% NaCl solution for 24 hrs was markedly reduced in total polyphenols as 73% for leaves and 33% for roots. Remarkable reduction in total polyphenols was not checked during fermentation followed after soaking. Trypsin inhibitor content correlated well(r=0.8873) with total polyphenols in all of Godulbaegi samples.

WPC 분말이 첨가된 설기떡의 품질 특성 (Quality Characteristics of Seolgiddeok added with Whey Protein Concentrate (WPC) Powder)

  • The effects of substituting whey protein concentrate (WPC) powder for rice flour in the preparation of seolgiddeok were determined by objective and subjective tests. Milk whey is drained from milk curd as a by-product of the cheese manufactureing process. Whey protein is known as a good nutritional source and is a functional material for many processed foods. WPC contains more than 80% whey protein. The moisture content decreased gradually during storage and the decrease in moisture was less in the control than in the WPC powder substituted groups. The color lightness (L) decreased significantly as the amount of WPC powder increased, wherease redness (a) and yellowness (b) both increased. Texture analyses revealed that the hardness, chewiness, gumminess and adhesiveness of seolgiddeok tended to increase in proportion to the amount of WPC powder in the formula. Seolgiddeok gelatinization was investigated by amylographing. Initial pasting temperature, peak viscosity, hot pasting viscosity and breakdown were low in seolgiddeok prepared with WPC powder substituted for rice flour. Setback had the lowest value in the control. Sensory evaluations revealed that, seolgiddeok prepared with 3% WPC powder had the highest overall acceptability score. These results indicated that WPC seolgiddeok with 3% WPC powder has the best quality.
