Prostaglandin $E_2$ ($PGE_2$) promotes tumor-persistent inflammation, frequently resulting in cancer. Curcumin is a diphenolic turmeric that inhibits carcinogenesis and induces apoptosis. $PGE_2$ inhibits curcumin-induced apoptosis; however, the underlying inhibitory mechanisms in colon cancer cells remain unknown. The aim of the present study is to investigate the survival role of $PGE_2$ and whether addition of exogenous $PGE_2$ affects curcumininduced cell death. HCT-15 cells were treated with curcumin and $PGE_2$, and protein expression levels were investigated via Western blot. Reactive oxygen species (ROS) generation, lipid peroxidation, and intracellular glutathione (GSH) levels were confirmed using specific dyes. The nuclear factor-kappa B ($NF-{\kappa}B$) DNA-binding was measured by electrophoretic mobility shift assay (EMSA). $PGE_2$ inhibited curcumin-induced apoptosis by suppressing oxidative stress and degradation of PARP and lamin B. However, exposure of cells to the EP2 receptor antagonist, AH6809, and the PKA inhibitor, H89, before treatment with $PGE_2$ or curcumin abolished the protective effect of $PGE_2$ and enhanced curcumin-induced cell death. $PGE_2$ activates PKA, which is required for cAMP-mediated transcriptional activation of CREB. $PGE_2$ also activated the Ras/Raf/Erk pathway, and pretreatment with PD98059 abolished the protective effect of $PGE_2$. Furthermore, curcumin treatment greatly reduced phosphorylation of CREB, followed by a concomitant reduction of $NF-{\kappa}B$ (p50 and p65) subunit activation. $PGE_2$ markedly activated nuclear translocation of $NF-{\kappa}B$. EMSA confirmed the DNA-binding activities of $NF-{\kappa}B$ subunits. These results suggest that inhibition of curcumin-induced apoptosis by $PGE_2$ through activation of PKA, Ras, and $NF-{\kappa}B$ signaling pathways may provide a molecular basis for the reversal of curcumin-induced colon carcinoma cell death.
포유류의 자궁조직은 발정주기를 통하여 역동적으로 변화하고 있으며 이러한 자궁조직의 분화는 시상하부-뇌하수체-생식소를 잇는 축에 의해 조절되는 스테로이드 호르몬에 의해 이루어진다. 그러나 에스트로겐 (E)이 어떤 유전자를 발현하여 자궁 내의 변화를 일으키는지는 아직 자세히 알려지지 않고 있다. 본 연구는 난소를 절제한 성숙한 흰쥐에 E을 처리한 후 자궁조직 내에서 c-fos, c-jun 및 hsp25 mRNA의 발현 변화를 Northern blot analysis방법을 사용하여 연구한 것이다. c-fos및 c-jun 암원유전자의 mRNA발현은 E처리 후 1시간 이전부터 증가하기 시작하며, 3시간 이내에 최고치에 도달한 후 급격히 감소하여 기저수준으로 되돌아갔다. 반면에 hsp25 mRNA수준은 E처리 후 3시간 대에서 최고치를 나타내나 증가된 발현량이 서서히 감소하며 12시간이 지난 후 까지도 정상대조군에 비해 높은 수준으로 유지되었다. 이러한 E의 영향이 선별적인지를 검증하기 위하여 E의 길항제인 tamoxifen을 사전처리하고 E을 추가로 처리하여 c-fos, c-jun및 hsp25 mRNA의 발현이 최고치에 이르렀던 3시간대에 자궁조직을 얻어 각각의 유전자 발현량을 조사한 결과 E에 의해 증가되었던 c-fos, c-jun 및 hsp25 mRNA의 수준이 억제됨을 확인하였다. 이러한 결과는 E이 자궁조직에 영향을 미치는데 초기의 일시적인 변화를 보이는 암원유전자인 c-fos 및 c-jun이 중요한 역할을 하리라는 것을 시사하며 hsp25의 경우는 좀더 늦은 반응에 관여하거나 c-fos및 c-jun에 의하여 간접적으로 조절을 받을 수도 있음을 보여 주는 것으로 사료된다.
We previously reported that the novel isoflavone-free peptide mixture (black soybean peptide, BSP) had several beneficial effects like antiobesity and hypotriglyceridemic effect. However, there are no reports for BSP on anti-hypertensive activity. BSP induced heme oxygenase-1 (HO-1) in HUVECs, thus investigated the HO-1-induced activity in HUVECs and the anti-hypertensive effects in SHR animal model. BSP significantly induced HO-1 expression both at transcriptional and protein levels in a time- and dose-dependent manner as measured by RT-PCR and Western blot analysis, respectively. These inductions were abolished by pretreatment of N-acetyl-cystein (NAC, 1~10 mM), but not by employing Tempol, a superoxide dismutase (SOD) mimetic (1~5 mM). As expected, enzymatic activity (~2 fold) determined by bilirubin formation assay and cGMP concentration (~6 fold) were significantly increased in BSP-treated cells. Based on the numerous evidences on the beneficial effects of HO-1 and our results, we investigated in vivo effects of BSP on the antihypertensive activity. The administration of BSP (1% in drinking water) significantly decreased mean blood pressure (BP) (from $218.6{\pm}6.99$ to $190.0{\pm}3.42$ mm Hg, p<0.01). This result indicates that BSP is strong inducer of HO-1 expression, which may be triggered by oxidative stress, and has anti-hypertensive activity.
포유류 배아의 초기발생과정에서 수란관이 하는 역할을 알아보기 위하여 소의 수란관내액이 생쥐 2-세포기 배아의 체외발생에 미치는 영향을 조사하였다. 대조군의 경우 5%의 배아만이 체외배양과정 중 퇴화한데 비해 5% 혹은 그 이상의 소의 수란관내액 (bOF)이 함유된 배양액 내에서 배양된 배아는 48시간이 지났을 때 모두 퇴화하였다. bOF를 65 \circ C에서 30분간 가열한 후 배양액에 첨가한 결과 bOF의 독성은 변화가 없었으나 90 \circ C에서 30분간 가열하였을때에는 독성이 거의 사라져 95% 의 배아가 정상적으로 발생하였다. bOF를 chymotrypsin으로 1시간 혹은 24시간동안 처리한 후 배양액에 첨가한 경우에도 독성이 사라져 각각 95.5%의 배아가 상실배 혹은 포배로 발생을 진행하였다. 산화방지제인 10mM 농도의 glutathione (GSH)을 bOF와 함께 배양액에 첨가한 결과 마찬가지로 bOF의 독성이 사라져 91.0%의 배아가 상실배 이상으로 발생한 반면 산화 방지능력이 없는 산화형 GSH(GSSG)를 bOF와 함께 배양액에 처리한 경우에는 bOF의 독성은 전혀 제거되지 않았다. 또한 mercaptoethanol, dithiothreitol, cysteamine 등의 다른 산화방지제도 GSSG와 마찬가지로 bOF의 독성을 제거하지 못하였다. 이와 같은 실험 결과로 미루어 소의 수란관내액에는 체외에 노출될 경우 독성을 나타내는 단백질 성분으로 된 물질이 있으며 이 물질의 독성은 대기 중의 산소의 산화작용에 의해 활성화되는 것으로 여겨진다.
오존가스의 젖소 유방염에 대한 치료효과를 구명할 목적으로 본 실험을 수행하였다. 만성유방염에 이환된 비유우 18분방을 대상으로 하였다 실험분방은 대조군(항생제 투여군: norfloxacin 10g/tube, 2회/일, 3일간: 5분방), 실험군 I (오존가스 0.1 ppm, 250m1, 2회/일, 7일간: 5분방) 및 실험군 II(오존가스 1 ppm, 50m1, 2회/일, 3일간: 8분방)로 각각 나누어 공시하였다. 우유 중 체세포수, 혈액충 백혈구수, 호중구수, 림프구수, 호중구/림프구(N/L)비 및 혈청 단백량의 변화를 처치 전 및 처치 후 7일에 각각 비교 검토하였다. 그 결과, 실험군 1에서는 오존가스 주입 후 7일에 주입 전에 비하여 우유 중 체세포수가 감소하였으나 대조군 보다 높은 수치를 나타내었다. 실험군 II에서는 오존가스 주입 후 7일에 주입 전에 비하여 현저한 감소소견을 나타내었으며(p<0.05), 대조군 보다 낮은 수치를 나타내었다. 그러나 대조군 및 실험군 간의 유의성은 인정되지 않았다 또한 말초혈액 백혈구수, 호중구수, 림프구수, N/L비 및 혈청 단백량의 변화에서는 대조군 및 실험군에 있어서 각각 유의한 변화를 나타내지 않았다. 이상의 결과를 종합해 볼 때, 오존가스는 젖소 유방염의 치료에 유효할 것으로 판단된다.
Fibroblast growth factor(FGF)는 세포의 성장과 이동, 분화와 생존과 관련된 여러가지 과정을 조절하는 것으로 알려져 있다. 이들의 prototype인 FGF-1과 FGF-2는 FGF receptors (FGFRs)를 통해서 세포내로 신호를 전달하는데, 두개봉합부의 조기융합을 보이는 craniosynostosis는 FGFRs중, 특히 FGFR-2의 point mutation에 의해서 야기된다. 최근 여러 보고에서 FGF/FGFR 신호전달은 골아세포의 분화에 있어 필수적인 역할을 한다고 하였으며, bFGF soaked bead를 쥐 두개골의 시상봉합부의 mid-mesenchyme과 osteogenic front부위에 적용하였을 때 osteopontin(OPN) 유전자의 발현을 유도한다고 하였다. 이에 본 연구에서는 ST-2 cell line를 이용한 in vitro실험에서 bFGF가 OPN 유전자 발현에 미치는 영향과 그 기전을 Northern blot analysis를 통해서 연구하고자 하였다. 1 ng/ml bFGF의 투여는 OPN, fibronectin 유전자 발현을 증가시켰으며, type I collagen 유전자 발현은 감소시켰으나 alkaline phosphatase 유전자 발현에는 영향을 미치지 않았다. OPN은 그 발현양상이 bFGF의 농도 증가에 따라 증가하는 양상으로 나타났으며, 시간결과에 따른 발현양상도 bFGF 투여 3시간째부터 bFGF를 투여한 군에서 대조군에 비해 높게 나타났으며 이는 24시간까지 시간의 경과에 따라 증가하는 양상을 보였다. 단백질 합성 억제제인 cycloheximide를 처리한 군에서는 OPN의 증가 양상을 보이지 않았는데 이는 bFGF에 의한 OPN유전자 발현이 새로운 전사조절 단백질 합성 등의 여러 단계를 거쳐서 일어남을 의미한다. 결론적으로 bFGF는 새로운 전사조절 단백질의 합성을 통해서 OPN 유전자 발현을 농도 및 시간 의존적으로 증가시킨다.
Objective: The purpose of this study was to examine the role of programmed cell death 4 (PDCD4) expression in predicting tumor response to neoadjuvant chemoradiotherapy and outcomes for patients with locally advanced rectal cancer. Methods: Clinicopathological factors and expression of PDCD4 were evaluated in 92 patients with LARC treated with nCRT. After the completion of therapy, 4 cases achieved clinical complete response (cCR), and thus the remaining 88 patients underwent a standardized total mesorectal excision procedure. There were 38 patients (41.3%) with a good response (TRG 3-4) and 54 (58.7%) with a poor one (TRG 0-2). Results: Immunohistochemical staining analyses showed that patients with high expression of PDCD4 were more sensitive to nCRT than those with low PDCD4 expression (P=0.02). High PDCD4 expression before nCRT and good response (TRG3-4) were significantly associated with improved 5-year disease-free survival and 5-year overall survival (P<0.05). Multivariate analysis demonstrated that the pretreatment PDCD4 expression was an independent prognostic factor. Conclusion: Our study demonstrated that high expression of PDCD4 protein is a useful predictive factor for good tumor response to nCRT and good outcomes in patients with LARC.
Objectives : Oxidative stress due to excessive accumulation of reactive oxygen species (ROS) is one of the risk factors for the development of several chronic diseases, including neurodegenerative diseases. Methods : In the present study, we investigated the protective effects of cheongnoemyeongsin-hwan (CNMSH) against oxidative stress‑induced cellular damage and elucidated the underlying mechanisms in neuronal-derived SH-SY5Y cells. Results : Our results revealed that treatment with CNMSH prior to hydrogen peroxide (H2O2) exposure significantly increased the SH-SY5Y cell viability, indicating that the exposure of the SH-SY5Y cells to CNMSH conferred a protective effect against oxidative stress. CNMSH also effectively attenuated H2O2‑induced comet tail formation, and decreased the phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V‑positive cells. In addition, CNMSH exhibited scavenging activity against intracellular ROS generation and restored the mitochondria membrane potential (MMP) loss that were induced by H2O2, suggesting that CNMSH prevents H2O2‑induced DNA damage and cell apoptosis. Moreover, H2O2 enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase, a typical substrate protein of activated caspase-3, as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with CNMSH. Furthermore, CNMSH increased the levels of heme oxygenase-1 (HO-1), which is a potent antioxidant enzyme, associated with the induction of nuclear factor-erythroid 2-related factor 2 (Nrf2). According to our data, CNMSH is able to protect SH-SY5Y cells from H2O2-induced apoptosis throughout blocking cellular damage related to oxidative stress through a mechanism that would affect ROS elimination and activating Nrf2/HO-1 signaling pathway. Conclusions : Therefore, we believed that CNMSH may potentially serve as an agent for the treatment and prevention of neurodegenerative diseases caused by oxidative stress.
효모세포벽 용해효소의 일종인 Zymolyase$(endo-{\beta}-1\;,3-glucanase)$와 더불어 Arthrobacter luteus로 부터 생산되었고, 또한 Zymolyase의 조효소중에서 발견된 바 있는 염기성 AL-protease의 효모세포벽 용해작용을 S. sake의 생세포와 그 세포벽 표품(標品)을 사용하여 조사하였다. AL-protease의 효모 생세포에 대한 용해활성은 그 단독작용만으로는 지극히 미약하였으나, Zymolyase와의 복합작용에 의해서 용해활성은 고도로 상승하였다. 효모생세포를 AL-protease와 Zymolyase로써 단계적인 처리를 할 경우, 효모세포는 AL-protease로 전처리된 뒤에 Zymolyase로 처리되는 처리 순서에 한하여 효과적으로 용해되었다. 이러한 AL-protease의 촉진적 작용은 AL-protease처럼 염기성이고 serine protease로 알려진 몇가지 시판의 효소들 중에서는 발견되지 않았으며, 또한 AL-protease의 이러한 작용은 실험에 사용된 효모들의 배양조건 및 균종에 따라서 커다란 영향을 받는 것으로 밝혀졌다. AL-protease는 효모세포벽 표품(標品)으로부터 일정량의 peptide와 상당량의 당을 유리시키고 있으나, 그 세포벽을 66% 이상은 용해시키지 못하였다. 반면에, Zymolyase는 그 단독작용으로도 효모세포벽을 거의 완전히 용해시킬 수 있었다. 이상의 실험결과를 기초로 하여, S. sake 세포벽의 용해에 있어서 AL-protease의 효소적 작용은, 먼저 AL-protease가 mannan과 단백질로 구성되는 세포벽 표층부에 결합하고, 이어서 그들의 구조를 변화시킴으로써, Zymolyase를 세포벽의 외부로 부터 알카리 불용성 glucan으로 구축되고 있는 세포벽 내부의 골격구조로까지의 침투를 촉진시키는 것으로 추론되었다.
The importance of the kidney in the development of hypertension was first demonstrated by Goldblatt and his colleagues more than fifty years ago. Many hormones and other regulatory factors have been proposed to play a major role in the development of hypertension. Among these factors angiotensia II (ANG II) is closely involved in renal hypertension development since it directly regulates $Na^+$ reabsorption in the renal proximal tubule. Thus the aim of the present study was to examine signaling pathways of low dose of ANC II on the $Na^+$ uptake of primary cultured rabbit renal proximal tubule cells (PTCs) in hormonally defined seum-free medium. The results were as follows: 1) $10^{-11}$ M ANG II has a significant stimulatory effect on growth as compared with control. Alkaline phosphatase exhibited significantly increased activity. However, leucine aminopeptidase and ${\gamma}-glutamyl$ transpeptidase activity were not significant as compared with control. In contrast to $10^{-11}$ M ANG II stimulated $Na^+$ uptake $(108.03{\pm}2.16% of that of control)$, $10^{-9}$ M ANG II inhibited ($92.42{\mu}2.23%$ of that of control). The stimulatory effect of ANG II on $Na^+$ uptake was amiloride-sensitive and inhibited by losartan (ANG II receptor subtype 1 antagonist) and not by PD123319 (ANG II receptor subtype 2 antagonist). 2) Pertussis toxin (PTX) alone inhibited $Na^+$ uptake by $85.52{\pm}3.52%$ of that of control. In addition, PTX pretreatment prevented the AMG II-induced stimulation of $Na^+$ uptake. 8-Bromoadenosine 3',5'-cyclic monophosphate (8-Br-cAMP), forskolin, and isobutylmethylxanthine (IBMX) alone inhibited $Na^+$ uptake by $88.79{\pm}2.56,\;80.63{\pm}4.38,\;and\;84.47{\pm}4.74%$ of that of control, respectively, and prevented the ANG II-induced stimulation of $Na^+$ uptake. However, $10^{-11}$ M ANG II did not stimulate cAMP production. 3) The addition of 12-O-te-tradecanoylphorbol-13-acetate (TPA, 0.01 ng/ml) to the PTCs produced significant increase in $Na^+$ uptake ($114.43{\pm}4.05%$ of that of control). When ANG II and TPA were added together to the PTCs, there was no additive effect on $Na^+$ uptake. Staurosporine alone had no effect on $Na^+$ uptake, but led to a complete inhibition of ANG II- or TPA-induced stimulation of Na'uptake. ANG II treatment resulted in a $111.83{\mu}4.51%$ increase in total protein kinase C (PKC) activity. In conclusion, the PTX-sensitive PKC pathway is the main signaling cascade involved in the stimulatory effects of ANG II on $Na^+$ uptake in the PTCs.

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
