  • This experiment was conduct to determine the influences of different levels of dietary energy and crude protein on growth performances during growing period (2 to 10 wk of age). A total of 360 egg breeder chicks (Hy-Line Brown) was divided into 5 groups with 4 replicates of 18 birds per replicate and fed the experimental diets containing three levels of energy and three levels of protein. With the increase in dietary crude protein levels, body weight significantly increased (P〈0.05). But dietary energy levels did not greatly affect growth performances. There were no significant differences to flock uniformity and tibia strength among treatments.

The Requirement of Ruminal Degradable Protein for Non-Structural Carbohydrate-Fermenting Microbes and Its Reaction with Dilution Rate in Continuous Culture

  • A continuous culture study was conducted to determine the impact of ruminal degradable soy protein (S-RDP) level and dilution rate (D) on growth of ruminal non-structural carbohydrate-fermenting microbes. Corn starch, urea and isolated soy protein (ISP) were used to formulate three diets with S-RDP levels of 0, 35 and 70% of total dietary CP. Two Ds were 0.03 and $0.06h^{-1}$ of the fermenter volume in a single-effluent continuous culture system. As S-RDP levels increased, digestibilities of dietary dry matter (DM), organic matter (OM) and crude protein (CP) linearly (p=0.001) decreased, whereas digestion of dietary starch linearly (p=0.001) increased. Increasing D from 0.03 to $0.06h^{-1}$ resulted in decreased digestibilities of dietary DM and OM, but had no effect on digestibilities of dietary starch (p=0.77) and CP (p=0.103). Fermenter pH, the concentration of volatile fatty acids (VFA) and daily VFA production were unaffected (p=0.159-0.517) by S-RDP levels. Molar percentages of acetate, propionate and butyrate were greatly affected by S-RDP levels (p=0.016-0.091), but unaffected by D (p=0.331-0.442). With increasing S-RDP levels and D, daily bacterial counts, daily microbial N production (DMNP) and microbial efficiency (MOEFF; grams of microbial N produced per kilogram of OM truly digested) were enhanced (p=0.001). The increased microbial efficiency with increasing S-RDP levels is probably the result of peptides or amino acids that served as a stimulus for optimal protein synthesis. The quantity of ruminal degradable protein from soy proteins required for optimum protein synthesis of non-structural carbohydrate-fermenting microbes appears to be equivalent to 9.5% of dietary fermented OM.

  • This study was conducted to estimate the optimum dietary protein to energy (P/E) ratio in juvenile river puffer. Nine experimental diets were formulated with three energy levels and three protein levels at each energy levels. Three energy levels of 3000, 3750 and 4500 kcal diets were included at 45, 50 and 55% crude protein (CP) levels, respectively (45P3000, 45P3750, 45P4500, 50P3000, 50P3750, 50P4500, 55P3000, 55P3750 and 55P4500). Fish averaging $3.43{\pm}0.02$ g randomly were fed the experimental diets in triplicate groups for 8 weeks. Weight gain of fish fed the 50P4500 diet were significantly higher than that of fish fed the 45P3000, 45P3750, 45P4500, 50P3000 and 50P3750 diets (P<0.05), but there was not significantly different from that of fish fed the 50P4500, 55P3750 and 55P4500 diets. Feed efficiency of fish fed the 50P4500 diet were significantly higher than that of fish fed the 45P3000, 45P3750, 45P4500, 50P3000 and 50P3750 diets (P<0.05), but there was not significantly different from that of fish fed the 50P4500, 55P3000, 55P3750 and 55P4500 diets. Protein efficiency ratio of fish fed the 45P3000 and 45P3750 diets was higher than that of fish fed 50P4500 and 55P4500, but there was not significantly different from that of fish fed the 45P3000, 45P3750, 45P4500, 50P3000, 50P3750, 55P3000 and 55P3750 diets. Based on weight gain, feed efficiency and specific growth rate, diets containing energy levels 4500 kcal/kg diet had an optimum P/E ratio of approximately 111 mg protein/kcal (50% crude protein) in juvenile river puffer.

  • Biofloc technology (BFT) systems provides an additional protein source for fish by converting waste in microbial flocs. Therefore, we aimed to determine the difference in the protein levels of the diet of Far Eastern catfish Silurus asotus reared in BFT and FW systems. We applied a two-way (six protein levels × two aquaculture systems) ANOVA design. We formulated six experimental diets to contain different protein levels (CPs)- 20%, 25%, 30%, 35%, 40% and 45%-referred to as the CP20, CP25, CP30, CP35, CP40 and CP45 diets, respectively. We randomly distributed 900 fish into 36 (18 each for BFT and FW), 300 L tanks (25 fish/tank) with three replications of each diet for 4 weeks. The weight gain (WG) and specific growth rate (SGR) considerably improved as the dietary protein level increased in the formulated diets. The BFT fish showed higher growth performance with any experimental diet than the FW fish. The growth performance of the fish fed the BFT-CP35 diet was higher than that of the fish fed the FW-CP45 diet. In conclusion, the cultivation of Far Eastern catfish (S. asotus) in BFTs can be carried out under reduced dietary protein levels.

  • In this third report influence of vibration on growth and on some metabolism of young growing rats fed on varying levels of protein was investigated. Forty eight (48) young growing male rats weighing about 60 grams were used, grouping to four (4) groups, twelve (12) rats each group. They were fed on 8%, 13%, 19%, and 26% casein diet respectively (See the table 1) for the period of 10 weeks experiment. During the experimental period the half number of the rats of each group were subjected to a given degree of vibration for two (2) hours daily. Observations for growth rate, food and protein efficiency ratios, organs development, cholesterol levels in aorta, total nitrogen, urea nitrogen and creatinine levels in urine may be summarized as follows 1. Growth was impaired by the vibration in all groups including 26% easein diet. 2. There is tendeney that in higher protein diets, the organs (See table 3) developed more well. And also the impairment of the organs development by the vibration revealed less in higher protein diets. 3. Food and protein efficiency ratios were generally decreased under vibration and the food efficiency was improved by increasing the protein level in diet. 4. Total and free cholesteral levels in aorta were increased by the vibration. Ester from/tatal ratio was 17.7% and 17.3% respectively at 8% and 13% protein diets and 54.8% and 54.2% at 19% and 26% protein diets. These show that, in higher protein diets, the vibration doesn't influence the cholesterol ratio. 5. Total nitrogen, urea nirogen and creatinine levels in urine were increased by increasing the protion level in diet and also increased by the vibration. 6. It seems that, according to the above observations, the vibration used in this experiment influenced, in certain extent, on physical development, physiological availability of nutrients, and on body metabolism. And it is also thought that higher protein diets act some good role in protecting body from suffering from vibration.

미꾸리(Misgurnus anguillicaudatus) 치어 성장을 위한 사료의 적정 단백질 및 지질 함량 (Optimal Dietary Protein and Lipid Levels for Growth of Juvenile Muddy Loach Misgurnus anguillicaudatus)

  • A feeding trial was conducted to determine the optimum dietary protein and lipid levels for the growth of juvenile muddy loach Misgurnus anguillicaudatus. Eight experimental diets (designated as P20L7, P20L14, P30L7, P30L14, P40L7, P40L14, P50L7 and P50L14) were formulated to contain 20%, 30%, 40% and 50% protein with 7% and 14% lipid. Three replicate groups of muddy loach (1.5 g/fish) were fed one of the experimental diets ad libitum for 8 weeks. Survival of fish fed the P20L14 diet was lower than in other groups (P<0.05). Specific growth rate (SGR) and feed efficiency (FE) of fish were affected by dietary protein (P<0.05) and lipid (P<0.005) levels. SGR and FE of fish tended to decrease with increasing dietary lipid level at the same protein level. There were no significant differences in SGR and FE among fish fed diets containing different protein levels ranging from 30% to 50% with the same lipid level. Whole body moisture and lipid contents were affected by dietary protein and lipid levels. Crude lipid content of fish fed the 14% lipid diets was higher than that of fish fed 7% lipid diets at each protein level. The results of this study indicate that a diet containing 30% protein with 7% lipid would be suitable for the optimum growth and effective feed utilization of juvenile muddy loach.

한국여성의 단백질 섭취수준이 질소대사에 미치는 영향 (The Effect of Dietary Protein Levels on Nitrogen Metabolism in Young Korean Women)

  • This study was performed to investigate the effect of dietary levels on protein metabolism in eight healthy Korean adult females. The 20-day metabolic study consisted of 2 day adaptation period and three 6-day experimental. Three experimental diets were low protein (LP : protein 44g), high protein(HP : protein 85g) and high animal protein (HAP : protein-84g). The apparent absorption and balance on nitrogen were significantly higher in high protein than in low protein diet. Nitrogen, absorption rate was about 75% for low protein and about 85% for high protein intake. The mean values of nitrogen balance were -1.28% for low protein and 0.78% for high protein diet. All the subjects were in negative nitrogen balance at the low protein intake while they were in positive nitrogen balance at the high protein intake. The mean daily urinary nitrogen excretion increased with increased level of protein intake. Urea nitrogen was the largest part of the urinary nitrogen. The ratio of urea nitrogen to total urinary nitrogen increased significantly for 79 to 85% as protein intake was doubled.

  • To investigate the effects of dietary protein and calcium levels on calcium and bone metabolism Sprague-Dawley male growing rats weighting approximately 91.4g were divided into four groups and fed one of the following four experimental diets-15% protein 0.2% calcium ; 15% protein 0.5% calcium ; 30% protein 0.2% calcium ; 30% protein 0.5% calcium-for five weeks. Calcium intake and excretion, apparent calcium absorption were measured and bone densities and mineral contents of femur and scapula were analyzed. Calcium excretion through feces and urine was significantly greater in animals receiving diets of higher calcium. Fecal calcium but not urinary calcium excretion was greater when the protein level was increased from 15% to 30%. Apparent calcium absorption rate was significantly higher with lower calcium intakes. Serum alkaline phosphatase activity was significantly higher in 0.2% calcium group than in 0.5% calcium group, while urinary hydroxyproline excretion was essentially same among all experimental groups. Weights and mineral contents or protein. Bone weights were greater, but calcium and ash contents of femur and scapula were lower in animals on the diet containing low calcium and high protein, which suggests that bone metabolism may be affected by the interaction between calcium and protein intake. These results indicate that during growth high protein intake might be beneficial to bone health if the diet is sufficient in calcium, however, if the diet fails to provide an optimum amount of calcium, such practice might be detrimental.

Comparisons of Recombinant Protein Expression in Diverse Natural Isolates of Escherichia coli

  • We assessed heterologous protein expression in 64 strains obtained from the Escherichia coli Reference (ECOR) collection, a collection representing diverse natural E. coli populations. A plasmid generating a glutathione S-transferase and plant carbonic anhydrase fusion protein (GST-CA) under the control of the tac promoter was introduced into the ECOR strains, and the quantity of the fusion protein was determined by SDS-PAGE. The foreign protein was generated at various levels, from very high (40 strains, high producers) to very low (six strains, low producers). Immunoblotting showed that the high producers expressed approximately 250-500 times more GST-CA protein than the low producers. The results of semi-quantitative RT-PCR showed that the low producers generated mRNA levels comparable to those of the high producers, thereby suggesting that, at least in this case, inefficient translation is a major cause of the low production. We introduced a different plasmid, which expressed a maltose binding protein and plant guanylate kinase fusion protein (MBP-GK) into the six low producers. Interestingly, five of these expressed MBP-GK at very high levels. Thus, we conclude that the production of a particular protein from an expression vector can vary considerably, depending on the host strain. Strains in the ECOR collection could function as useful alternative hosts when a desired level of protein expression is not obtained from commonly used strains, such as E. coli K12 or B derivatives.

Gamma-Irradiation Enhances RECK Protein Levels in Panc-1 Pancreatic Cancer Cells

  • Radiotherapy is an important treatment for many malignant tumors, but there are recent reports that radiation may increase the malignancy of cancer cells by stimulating expression of type IV collagenases. In this study, we examined changes in matrix metalloproteinase (MMP) inhibitors, such as the tissue inhibitors of metalloproteinase (TIMP)-1, TIMP-2 and RECK, in response to irradiation in Panc-1 pancreatic cancer cells. Irradiation increased RECK protein levels but not mRNA levels, whereas no significant changes were found in TIMP-1 and TIMP-2. The enhanced RECK protein levels were associated with an increase in MMP inhibitory activity. However, irradiation slightly but reproducibly increased the invasiveness of the Panc-1 cells. Like irradiation, treatment of Panc-1 cells with transforming growth factor $(TGF)-{\beta}1$ led to a 2-fold increase in RECK protein levels. Transient transfection with Smad3 also increased RECK protein levels, but transfection with Smad7 markedly reduced them. Stable expression of Smad7 and treatment with SB431542, an inhibitor of $TGF-{\beta}$ receptor I kinase, abolished $TGF-{\beta}1$- and radiation-mediated effects on RECK. Furthermore, irradiation increased levels of phosphorylated Smad3. We conclude that radiation post-transciptionally enhances RECK protein levels in Panc-1 cells, at least in part, via $TGF-{\beta}$ signaling, and that irradiation increases Panc-1 invasiveness via a mechanism that may not be linked to MMP-2 activity.
