  • 가잠과 천잠 및 작잠의 실크 단백질 분획물로부터 피부섬유아세포에 대한 증식 효과를 알아본 결과 다음과 같은 결과를 얻을 수 있었다. 1. 가잠 및 천${\cdot}$작잠 단백질로부터 다양한 분자량대의 실크 단백질을 순수 분리 분획화 하여 사람 피부세포(human skin fibroblast; CCD-986sk)에 기계적 상처를 준 후 처리한 결과, 평균분자량(Mw)이 300-500 및 950-1500 분획군이 세포 증식에 뛰어난 효과가 있음을 확인${\cdot}$선발하였다. 2. 가잠의 경우 평균 분자량 590(BM-1), 1400(BM-2)에서, 천잠은 340(AY-1), 940(AY-2), 작잠에서는 300(AP-1), 1440(AP-2)에서 $10\;{\mu}g/ml$의 농도로 처리했을 경우 피부섬유 아세포의 증식이 무처리구에 비하여 40% 이상의 증식 효과를 갖았다. 또한 유효 농도 범위에서 대식세포인 RAW 264.7 세포에 대해서는 세포독성을 지니지 않았다. 3. 아미노산 조성에 있어 저분자량 AY-1 분획의 전아미노산 및 유리아미노산은 Tyr 함량이 매우 높게(25-48 g/100g)나타났으며, 작잠의 저분자 분획인 AP-1에서는 Lys 함량이 높게(5.2 g/100g) 나타남을 확인하였다. 4. 동일한 실크 단백질일 경우 저분자량 분획물에서 피부 세포 증식에 좀 더 효과적이였다.

식품단백질 효소분해물의 제조 및 이화학적 특성 (Preparation and Chemical Characteristics of Food Protein Hydrolysates)

  • In this study, food protein hydrolysates were prepared from six types of food protein: purified meat protein, whole egg protein, casein, isolated soy protein, concentrated rice protein, and gluten. Food proteins were hydrolyzed with pepsin and ethanol (80%)-soluble fractions of pepsin hydrolysates were employed for analysis. The products were colorless and odorless powders with low fat content and good solubility. The MW (molecular weight) of the protein hydrolysates was confirmed to be $200{\sim}1,800$ via gel filtration. Free amino acid contents accounted for less than 5% of the samples. The results of our amino acid analysis revealed that all food protein hydrolysates preserved their original amino acid compositions and nutritional values of their source proteins with highly pure oligopeptide mixtures. These results show that the food protein hydrolysates prepared in these investigations should prove excellent dietary nitrogen sources for a variety of applications.

녹두로부터 렉틴 성분의 분리 정제 (Isolation and Purification of Lectin from Phaseolus radiatus)

  • New lectins, lymphoagglutinating lectins from mung beans (MBLA) have been isolated and purified. Mung beans crude extracts were made with 0.15M NaCl and these were purified through anionic exchange chromatography. Four fractions were obtained from DEAE Sephadex A-50 by salt gradients elution. Lectin activity, enzyme activity, protein assay, identification of purity by polyacrylamide gel electrphoresis and immunochemical studies were carried out with these four fractions. Through these results, it can be suggested that 0.2M fraction is newly found potent MBLA. There were some relationships with MBLA and L-PHA but no similarities were observed between MBLA and E-PHA.

Screening and isolation of antibacterial proteinaceous compounds from flower tissues: Alternatives for treatment of healthcare-associated infections

  • Healthcare-associated infection represents a frequent cause of mortality that increases hospital costs. Due to increasing microbial resistance to antibiotics, it is necessary to search for alternative therapies. Consequently, novel alternatives for the control of resistant microorganisms have been studied. Among them, plant antimicrobial protein presents enormous potential, with flowers being a new source of antimicrobial molecules. In this work, the antimicrobial activity of protein-rich fractions from flower tissues from 18 different species was evaluated against several human pathogenic bacteria. The results showed that protein-rich fractions of 12 species were able to control bacterial development. Due its broad inhibition spectrum and high antibacterial activity, the protein-rich fraction of Hibiscus rosa-sinensis was subjected to DEAE-Sepharose chromatography, yielding a retained fraction and a non-retained fraction. The retained fraction inhibits 29.5% of Klebsiella pneumoniae growth, and the non-retained fraction showed 31.5% of growth inhibition against the same bacteria. The protein profile of the chromatography fractions was analyzed by using SDS-PAGE, revealing the presence of two major protein bands in the retained fraction, of 20 and 15 kDa. The results indicate that medicinal plants have the biotechnological potential to increase knowledge about antimicrobial protein structure and action mechanisms, assisting in the rational design of antimicrobial compounds for the development of new antibiotic drugs.

  • A radioprotective ginseng protein fraction was obtained from Korean white ginseng powder by the following isolation and purification procedures: Tris-HCI buffer extraction, 70% ammonium sulfate fractionation, CM-rellulosr column chromatography, heat inactivation and Sephadex G-75 column chromatography. This fraction was further purified by Sepharose 4B and Sephadex G-150 column chromatographies. Three fractions obtained were subjected to Native-PAGE and SDS-PAGE using gradient gels and the silver staining method. Molecular weights of the native proteins and their subunits were estimated.

Adsorption of $\textrm{Pb}_{2+}$ in the components of bacterial cell membrane

  • S. epidermidis cell was fractionated into cell wall, cell membrane and cytoplasm. The cell membrane adsorbed the most abundant $\textrm{Pb}_{2+}$ per unit dry weight of the three fractions tested. Adsorption behavior of $\textrm{Pb}_{2+}$ in lipid and protein, which are the main components of the cell membrane, indicated that phosphatidylethanolamine and phosphatidylinositol having phosphoryl group and gangliosides containing carboxyl groups adsorbed much more $\textrm{Pb}_{2+}$ than triglycerides lacking any chargeable functional groups. Protein purified from cell membrane adsorbed larger amount of $\textrm{Pb}_{2+}$ than total native cell membrane or cell membrane lipid.

  • Toxoplasma-killing activities of mouse peritoneal macrophages activated by the extracts of Tetrahymena pyriformis (Korean and Chinese strains) were evaluated, and the active protein fractions from both strains were partially characterized by a method including chromatographies and SDS-PAGE. The first peak in Korean strain and the second peak in Chinese strain of T. pyriformis obtained by DEAE-Sephadex A-50 chromatography were most effective in the activation of macrophages to kill Toxoplasma gondii tachyzoites in vitro. Subsequent fractionations of obtained peak fractions were performed on a Sephadex G-200 gel. The first peaks fractionated from both strains of T. pyrtyomis had the highest toxoplasmacidal activities, and when subjected to the SDS-PAGE, one prominent band was visualized for each of the strains showing the same molecular weight of ca. 52.6 kDa. This active protein is suggested to be related to non-specific activation of mouse peritoneal macrophages.

인삼 단백질분획에 대한 폴리아크릴아미드 전기영동 (Polyacrylamide Gel Electrophoresis on Ginseng Proteins)

  • Korean ginseng was purified to obtain radioprotective protein fractions by buffer extraction, ammonium sulfate fractionation, CM-cellulose column chromatography, heat inactivation and Sephadex G-75 column chromatography. The final three fractions, GI, GII and GIII were subjected to Disc-polyacrylamide gel electrophoresis (PAGE) and SDS-PAGE. The molecular weights(M.W.) of native and denatured proteins were estimated by using regression line equations obtained from the mobilities of standard proteins. As the results, in Disc-PAGE, the GI fraction showed two protein bands with M.W. of above 213, 000 and 55, 000, GII showed one band with M.W. of 44, 000 and GIII, also one band with M.W. of 19, 000. In SDS-PAGE, GI fraction gave four subunit bands with M.W. of above 114, 000, 27, 000, 24, 000 and 19, 000, GII gave two bands with M.W. of 46, 000 and 22, 000, and GIII, one band of 19, 000.

탄광부 진폐증자의 흉부 방사선 사진상 소음영의 밀도에 따른 혈청 단백 농도 및 전기영동 분획상 (Serum Protein Concentrations and Electrophoretic Patterns by Categories of Small Opacity Profusions in Coal Workers' Pneumoconioses)

  • The concentrations of serum proteins fractions and their electrophoretical patterns were investigated in 135 patients with coal workers' pneumoconiosis who participated in confirmative examination for pneumoconiosis in December 1989. Their radiographical profusions were classified as 1/0 or more. Agarose film and phosphoric acid-sodium hydroxide buffer(pH 9.6) were used for electrophoresis. Concentration of each protein fractions and electrophoretical patterns seemd to be equivalent to reference values. Serum ${\alpha}_1$- and $\beta$-globulin concentrations, however, were significantly different (p<0.50) among categories of small opacity profusions and showed the lowest level in the group of category 1. Albumin concentratins decreased and ${\alpha}_2$- globulin concentrations increased significantly (p<0.05) in the group of complicated with pulmonary tuberculosis. $\gamma$-globulin concentrations were not varied by category of profusions nor by pulmonary tuberculosis complication.
