돼지 췌장내 Protein Methylase I의 분리정제 및 성질 (Purification and Properties of Protein Methylase I from Hog Pancreas)

  • Protein methylase I has been partially purified from hog pancreas with a 11% yeild. The final preparation is completely free of any other protein-specific methyltransferases and endogenous substrate proteins. The enzyme has an optimum pH of 7.2 and the approximate molecular weight is above 800 thousands dalton. The Km values for S-adenosyl-L-methionine and histone type II-A are 1.32$\times$10$^{-5}$M. The Ki value for S-adenosyl-L-homocysteine is 1.52$\times$10$^{-6}$M. The effect of enyzme concentration on the activity showed a slight sigmoidal curve suggesting the involvement of certain cofactors. Even though the purified enzyme showed two bands on polyacrylamide gel electrophoresis, the enzyme is highly specific for the arginine residues of protein and specifically, highly specific for histone, suggesting histonespecific protein methylase I.

대두 유식물에서 Protein Kinase C의 부분 정제 (Partial Purification of Protein Kinase C in Glycine max)

  • Protein kinase C, a protein related in PI cascade, was partially purified from the cytosol protein of etiolated plants of Glycine max by DEAE-52 cellulose chromatography and phenylsepharose chromatography. When the DEAE column was eluted with 0-0.8 M linear gradient KCl, tow fractions were found that increased the phosphorylation of histon H1 about five and nine-fold in the presence of 5 $\mu\textrm{g}$/mL phosphatidylserine and 0.5 $\mu\textrm{g}$/mL diolein, respectively. These fractions were used as DEAE pool. The reaction eluted with relatively high concentration of KCl was loaded on phyenylsepharose column with 5 mM CaCl2 and eluted with 1 mM EGTA. A fraction contained the protein kinase C, which increased the phosphorylation of the histon H1 was fractionated. To determine the molecular weight of PKC, the fraction eluted from phenylsepharose column was analyzed by 5~15% polyacrylamide gel electrophoresis after concentrated with the Amicon membrane (YM10). That revealed two bands corresponding to 60 and 65 kGy by silver staining of the gel, respectively.

Evaluation of Gelation Properties of Salt-Soluble Proteins Extracted from Protaetia brevitarsis Larvae and Tenebrio molitor Larvae and Application to Pork Myofibrillar Protein Gel System

  • The purpose of this study was to investigate the functional properties of salt-soluble proteins obtained from Protaetia brevitarsis (PB) and Tenebrio molitor (TM) larvae, the interaction between these proteins and pork myofibrillar protein (MP) in a gel system. The gel properties of salt-soluble protein extracts showed that the PB had a higher viscosity than the TM protein. However, the TM protein had higher gel strength compared with the PB protein. The gelation characteristics of the pork MP gel systems added with lyophilized insect salt-soluble protein powder showed to decrease slightly viscosity compared with MP alone. Adding the TM or PB protein powder did not affect the pork MP's hydrophobicity and sulfhydryl group levels. Furthermore, the protein bands of the MP did not change with the type or amount of insect salt-soluble protein. The cooking yields of the pork MP gels containing PB or TM protein powder were higher than those without insect protein. Regardless of the type of insect salt-soluble protein added, the pork MP's gel strength decreased. Furthermore, as the level of insect powder increased, the surface protein structure became rough and porous. The results demonstrated that proteins extracted from PB and TM larvae interfered with the gelation of pork MP in a gel system.

식염첨가 유무에 따른 계육의 부위별 가열온도에 따른 이화학적 성상과 근장 단백질 밴드의 변화 (Physico-chemical Properties and Changes of Sarcoplasmic Protein Bands of Chicken Meat Cuts with or without Salt during Cooking Temperatures)

  • 본 연구는 계육의 가슴과 다리부위로 식염첨가의 유무에 따라 64부터 74℃까지 2℃ 간격으로 가열하는 동안 pH와 일반성분, 가열감량, 단백질 용해성, 색도 그리고 단백질 밴드의 변화를 비교 분석하였다. 총괄적인 결과에서 pH는 2%의 식염을 첨가 한 것과 가열 온도별로는 차이가 없었지만 부위별로 차이를 보여 다리부위가 가슴부위 보다 높았다. 일반성분 중 수분과 지방 함량은 식염 첨가량에 따른 차이는 없었지만 수분과 지방 함량 모두 다리부위가 높게 나타났으며 반면 단백질 함량과 용해성은 가슴부위가 더 높게 나타났다. 소금첨가에 따른 결과에서 근육부위별 다리와 가슴부위의 가열감량은 유의적 차이를 보이지 않았다. 반면 2 % 식염을 첨가한 처리구는 가열감량이 전혀 나타나지 않았다. 적색도는 대조구의 경우 다리가 가슴보다 높았고 황색도와 함께 가열함에 따라 증가하였다. 단백질의 용해성은 식염 함량에 따른 차이는 없었으나 총괄적인 결과와는 달리 식염을 첨가하지 않은 대조구에서는 단백질 용해성에서 부위별 차이를 나타내지 않았다. pH, 명도와 단백질의 용해성은 식염첨가에 따른 근육부위와 가열온도 상관관계의 유의차를 보임으로써 서로 다른 양상을 보여 주었다. 대조구의 경우 pH는 다리부위가 높았고, 명도는 가열함에 따라 증가하였으며 가열시에 부위별 차이를 보였다. 반면 2%의 식염을 첨가한 처리구는 부위에 상관없이 용해성이 감소하였고 명도는 가슴육에서만 증가하여 부위별 차이를 보였다. 식염을 첨가하지 않은 다리부위에서 가열하지 않은 원료육은 약 7개의 단백질 밴드를 보였는데 내부중심온도를 64℃로 가열했을 시 90, 53, 30.5 kDa의 분자량을 갖는 단백질 밴드가 가열 온도의 영향을 받았으며 가슴부위에서는 원료육의 경우 약 9개의 단백질 밴드가 보였지만 88, 64, 30.5, 30 kDa의 분자량을 갖는 단백질 밴드가 가열에 의해 영향을 받았다. 결과적으로 가열온도와 식염처리에 영향을 받은 단백질 밴드는 다리부위에서 66과 54 kDa 이었으며 가슴부위에서는 66과 54 및 33 kDa의 분자량을 가진 단백질 밴드였다. 또한 54 와 33 kDa의 분자량을 갖는 단백질 밴드는 68℃까지 희미하게 보이다 사라졌으며 그 중 33 kDa의 분자량을 가진 단백질 밴드는 2%의 식염 첨가 시 저해작용을 받아 66℃ 까지만 보이다 사라졌다. 이러한 단백질 밴드는 계육의 최종가열 온도 측정을 위한 지시제로서 사용될 수 있을 것으로 사료된다.

Organ-specific antigens of Clonorchis sinensis

  • This study was carried out to find out specific proteins from different organs of Clonorchis sinensis. Crude extract, organ-specific and excretory-secretory (ES) proteins were analyzed by immunoblot with infected human sera. The bands of 7- and 17 -kDa were main component of intestinal fluid and ES protein and commonly found in all organ-specific proteins. The 17-kDa protein was observed from ES antigen, intestinal fluid, eggs and sperms, 26- and 28-kDa proteins were from the uterus, vitellaria, and ovary, and 34-, 37-, 43- and 50-kDa proteins were mainly from the testis and sperms. Serum of mice immunized with sperms reacted to the 50-kDa protein by immunoblotting and immunohistochemical staining showed a positive reaction at the seminal receptacle and seminiferous tubule. The present results show that the 7-kDa protein is a common antigen of every part or organ of C. sinensis, but different organs express their specific antigenic protein bands.

BALB/c 마우스에서 큰리슈만편모충의 감염부위에 따른 궤양형성과 혈청 면역반응 (Skin ulcer and immunoblot patterns by inoculation sites in BALB/c mice infected with Leishmania major)

  • BALB/c 마우스에서 감염부위와 감염기간에 따른 숙주 체액 면역반응의 변화를 알아보고자 하였다. 배양한 큰리슈만편모충의 전편모형(promastigote)을 BALB/c 마우스의 코. 등 발바닥 부위로 나누어 각각 $3{\;}{\times}{\;}10^6$마리씩 피하 감염 후 10-100일 동안에 궤양의 형성과정을 관찰하고 채혈하여 SDS-PAGE와 면역이적법을 시행하여 각 부위별로 나타나는 항체 반응을 관찰하였다. 외관상으로는 감염 15일부터 코에 감염시킨 마우스에서 먼저 궤양이 형성되기 시작하였고. 코에 궤양이 나타난 후 2-3일 뒤에 발에서 궤양이 형성되었으며 등에서는 감염시킨 후 90일이 되어서야 궤양이 관찰되었다. 감염후 20일에 실시한 면역이적법에 의하면 코 감염군에서는 202, 139, 98, 83, 81, 67, 65, 62, 59, 54, 52, 42, 26, 23 kDa의 항원성 분획이 관찰되었고 발 감염군에서의 항원 분획양상도 코 감염군과 같았으나 등감염군에서는 202, 83, 81, 65 kDa의 희미한 항원성 분획이 관찰되었다. 그러나 감염 후 90일이 경과한 등 감염군에서는 202, 83, 81, 74, 67, 65, 62, 59, 54, 52, 20, 17 kDa의 항원 분획이 관찰되었다 이상의 결과로부터 감염부위와 감염기간에 따라 큰리슈만편모충에 대한 혈청반응이 항원 분획에 따라 다르게 나타남을 관찰하였다. 이 차이는 세 감염부위의 온도차에 의한 결과일 가능성도 있으나 다른 부위에 감염될 경우 한 숙주 내에서도 다른 면역반응이 유발되어 나타날 수도 있다고 추측하였다. 특히 궤양 형성 시기와 혈청 내 67-52 kDa 분획에 대한 항체 출현 시기가 일치하는 것으로 보아 궤양 형성에 이 항체가 관여할 가능성이 있음을 시사한다.

Presence of Carbonic Anhydrase III in Liver of Flounder, Limanda yokohamae

  • Carbonic anhydrase III was found in liver of flounder, Limanda yokohamae. Protein was isolated from cytosolic extracts and identified using SDS-PAGE and isolectric focusing. Specific protein bands with molecular weight of 30 kDa and pIs of 7.0 and 6.5 were detected by Western blotting. This is the first report of identification of carbonic anhydrase III from L. yokohamae.

Bensulfuron에 대(對)한 내성(耐性) 및 감수성(感受性) 수도품종(水稻品種)의 전기영동(電氣泳動) 표현형(表現型) 차이(差異) (Difference in Electrophoretic Phenotypes of rice Cultivars Selected to Bensulfuron)

  • Bensulfuron에 대표적인 내성군(耐性群)(UCP-28, Chinsurah Boro II, Fukunohama, Fadehpur-2, IR 14252-13-2-2-5)과 감수성품종군(感受性品種群)(HP93(3) FA, HP94(9) FA, Padilabou Alumbis. KH-17854, IR 1846-2841-1)을 공시(供試)하여 종실(種實) 단백질(蛋白質)(SDS-PAGE)과 유묘(幼苗) 동위효소(同位酵素)(malate dehydrogenase, acid phosphatase, peroxidase, esterase: 7% PAGE)의 전기영동(電氣泳動) profile을 분석(分析)하고, bensulfuron 0, $10^{-6}$, $10^{-5}$, $3{\times}10^{-5}M$ 용액에서 생장(生長)한 두 대표 품종(品種)(cv. Chinsurah Boro II와 cv. IR 1846)의 5 일령(日齡) 유묘(幼苗)의 동위효소(同位酵素) profile 변화양상(變化樣相)을 비교(比較)하므로써, bensulfuron에의 내성차이(耐性差異)와 이들 전기영동(電氣泳動) profile 차이(差異)가 갖는 연관성(聯關性)을 알고저 하였다. 결과(結果)를 요약(要約)하면 다음과 같다. 1. 종실단백(種實蛋白)의 전기영동(電氣泳動)으로 16개(個) band를 확인(確認)할 수 있었고, 이를 근거(根據)로 clustering한 결과(結果), 내성(耐性)에서의 M, O, P band가 높게 나타났고 또한 전체 peak 면적(面積)이 크다는 특이성(特異性)에 기인(起因)하여 두 품종군간(品種群間)에 비유사성(非類似性)이 인정(認定)되는 정도(程度)로 분류(分類)되었다. 2. 무처리(無處理)된 유묘(幼苗)의 동위효소(同位酵素) 가운데, peroxidase로는 4개(個) band가 얻어졌고, 내성군(耐性群)의 특이성(特異性)은 D band에서의 활성(活性)이 높은 데 있었다. Malate dehydrogenase는 3개(個) band로 분리(分離)되었고, 내성군(耐性群)만이 A band를 나타내며 B 및 C band도 내성군(耐性群)에서 선명하였다. Esterase는 3~4개(個) band로 구분(區分)되었고 내성군(耐性群)이 A 및 B band에서 높은 활성(活性)을 보였다. Acid phosphatase는 하나의 major band와 2~3개(個)의 minor band로 나타났으며 내성군(耐性群)만은 B band를 갖는 경향(傾向)이었다. 3. Bensulfuron의 처리농도(處理濃度)에 따라, 동위효소(同位酵素) 가운데 peroxidase는 감수성(感受性)에서만 C band의 생성(生成)과 D band의 소실현상(消失現象)이 야기(惹起)되었고, 내성품종(耐性品種)에서는 C band가 나타나지 않았으며, 다른 band에서의 변화(變化)도 없었다. Malate dehyrogenase의 경우, major band인 D, E, F는 감수성(感受性)에서 활성(活性)이 컸고 변화(變化)는 없었으며, minor band인 A, B, C는 감수성(感受性)에서만 민감(敏感)하게 소실(消失)되었다. Esterase는 5개(個) band로 표현(表現)되었으며 내성(耐性)에서는 A, B, C, D 감수성(感受性)에서는 A, B, C, E만 나타났으며, 약처리농도증가(藥處理濃度增加)로 감수성품종(感受性品種)은 A, C, E band의 활성(活性)이 민감(敏感)하게 소실(消失)되었다.

The Protein and Isozyme Patterns During in vitro Plant Regeneration of Yooja (Citrus junos Sieb.) and Trifoliate Orange (Poncirus trifoliata Rafin.)

  • In this study, plant regeneration through in vitro culture from plantlet stems of Yooja (C. junos Sieb.) and trifoliate orange (P. trifoliata Rafin.) was attempted to make mass-production system of virus-free plants having the same genotype with mother plant. In order to investigate physiological change depending on the developmental stage of plant regeneration, the changes of total protein, peroxidase and esterase activity and their isozyme patterns as well were examined in 1/2 MS medium. The results are as follows : 1. The MS medium for the optimal callus induction and shoot formation was utilized. The medium was supplemented either with 2,4-D and Kinetin or with BA and NAA. The optimal concentrations were the combination of 1.0mg/ 2,4-D +0.3mg/ Kinetin and 1.0mg BA +0.3mg NAA in callus induction and shoot formation, respectively. 2. For the plant regeneration from somatic embryos, 1/2 MS medium was used with supplements of growth regulators (free, 1.0mg/ IBA +1.0mg/ BA ,0.5mg/ IBA +0.5mg/ BA). Shooting and rooting were the best in the treatment of 0.5mg/ IBA and 0.5mg/ BA combination. 3. The total protein content has a tendency of increase with the developmental stage of embryo, but it was decreased at the plantlet. Also it was the highest at 8 and 6 weeks stage in C. junos Sieb. and P. trioliata Rafin, respectively. In the SDS-PAGE pattern of protein, C. junos Sieb. showed bands of 29.0 and 40kDa at 10 weeks. The 45,66 and 97.4 kDa bands at 10 weeks of culture were shown in P. trifoliata Rafin. 4. The highest esterase activity was shown at the 6 and 8 weeks of culture in C.junos Sieb. and P. trifoliata Rafin.., respectively. 5. Esterase isozyme patterns were shown difference according to the developmental stage. In C. junos Sieb. a new band was observed at pl 7.7 following 4 weeks culture. On the other hand, new bands in P. trifoliata Rafin. were observed at pl 7.5~6.5 following 4 and 6 weeks culture, respectively.

생장조절제와 프라이밍 처리에 의한 우슬종자의 발아특성과 단백질 발현 양상에 관한연구 (Studies on Seed Germination Characteristics and Patterns of Protein Expression of Achyranthes japonica by Treating Plant Growth Regulators and Seed Primings)

  • This study was conducted to improve the postharvest storage techniques of managing and storing seeds, to test qualities and viabilities of the seeds and to examine the germination rate and the protein expression of Achyranthes japonica Nakai. The seeds collected from different areas of Je-Cheon and Gwang-Ju were stored with different temperatures and durations. Two plant growth regulators and two seed priming were treated to investigate their effect on the germination rates and the days required for germination. The weight of one hundred seed collected in Gwang-Ju was heavier than those in Je-Cheon. Seed length collected in Gwang-Ju was also longer about 5.12 mm than those in Je-Cheon about 4.90 mm and seed width was longer in Gwang-Ju than those in Je-Cheon. The rates of seed germination in two different collection areas were higher about 2.9 to 13.0% in Gwang-Ju compared to those in Je-Cheon. Comparing its rates with the storing temperatures and durations, they were not clearly different in between $4^{\circ}C$ and $25^{\circ}C$ and they also were gradually decreased with getting longer storing durations. The germination rates treated by plant growth regulators were higher with $GA_3$ than those with Kinetin. The highest seed germination rate was appeared at 50 ppm of $GA_3$. Comparing its rates with different seed priming, they were relatively higher with $KNO_3$ than those with PEG6000. In protein expression patterns between before the germinating and after the germinating of seeds, more and clear bands were appeared in the seed after the germination compared to those before the germination of seeds, especially 10 ~ 20 kDa. These results showing more and clear bands were more clearly appeared in Gwang-Ju compared to Je-Cheon. Comparing the protein expression with plant growth regulators and seed primings, $GA_3$ was better expression than those with Kinetin and $KNO_3$ was better than those with PEG6000. More and clear bands were closely related to the germination rates of seeds and more detailed studies would be required.
