단백질 급원과 수준을 달리한 식이가 흰쥐의 납축적에 미치는 영향 (Influence of Dietary Protein Source and Level on Lead Accumulation in Rat)

  • In this experiment forty-eight Sprague Dawley male rats were chosen and used in order to measure the growth rates and to see the effects of lead acumulation in their organs resulting from variously controlled lead protein diet. Protein sources were casein and isolated soyprotein (ISP), and each source was divided into three groups : 7% low protein [LP], 20% standard protein (SP) and 40% high protein (HP) groups. The six experimental groups were given lead acetate(25 mg/kg B.W.) and six control groups were given sodium chloride by oral administration 6 times a week for weeks. The results from this experiment were summeraized as following ; 1) Food consumption, weight gain, organ weight and food efficiency ; Lead acetate administration with protein source had no effects on food consumption, weight gain and organ weight . By their different levels of protein, food consumption of LP group was less the that of SP and HP groups after 3 weeks, weight gain of LP group was less than that of SP and HO groups after 1 weeks. The organ weight in LP group was significantly lower than SP and HP groups except teeth and adrenal s. Effect of lead acetate administration on food efficiency have significantly lower in LP-ISP diet and HP -casein diet than other groups only first week. By their different levels LP group showed significantly lower than SP group until 3 weeks. 2) Hematopoietic effect ; The hematopoieteic effect was not influencec by lead acdtate administration and protein source. But the LP group showed a significantly lowe hematopoietic effect than the SP, HP, groups. 3) Accumulation of lead in the liver, kidney, teeth by protein source showed no significantly differences. Accumulation of lead in blood, heart of LP group, spleen of LP and HP groups. femur of SP and HP groups fed with casein diet groups were significantly higher than fed with ISP diet groups. By their different levels of group showed generally higher than SP and HP groups. But accumulation of lead in teeth of HP group was high also.

ENERGY AND PROTEIN REQUIREMENTS OF KHAKI CAMPBELL × THAI NATIVE GROWING DUCKS

  • The experiment was carried out to estimate the energy and protein requirements of Khaki Campbell $\times$ Thai Native growing ducks. The growing stage was divided into 2: the first was a starting stage from 0 to 4 weeks of age and the second was a growing stage from 4 to 18 weeks of age. In the starting stage, the birds were given diets containing 2.70, 2.75, 2.80, 2.85 and 2.90 Mcal ME/kg and ME/protein ratio of 150/l for the energy requirement trial, and diets containing 17.0%, 18.5%, 20.0% and 21.5% protein and 2.8 Mcal ME/kg for the protein requirement trial. It was found that there were no significant differences in body weight gain of feed efficiency among the energy or protein levels. In the growing stage, no significant differences in body weight gain or feed efficiency were found among the energy levels from 2.70 to 2.90 Mcal ME, or protein levels from 15.5% to 20.0%. The age at first lay was 105-117 days and this is earlier than that observed in purebred Khaki Campbell ducks. The feed cost for raising ducks was lower in the low energy and low protein diets. In conclusion, Khaki Campbell $\times$ Thai native ducks can be raised with a diet low in ME as 2.7 Mcal/kg during the whole growing period. As for the protein. 7.0% in the starting stage and 15.5% in the growing stage is practical.

식이 단백질과 Ca 수준이 흰쥐의 Cd 해독에 미치는 영향 (The Effect of Dietary Protein and Calcium Levels on the Cadmium Detoxication on Rats)

  • This study was performed to investigate the effect of dietary protein and calcium levels on cadmium detoxication in rats. Seventy male Sprague-Dawley rats weighing 208 $\pm$ 19 g were blocked into 10 groups of 7 animals according to body weight. Five groups were fed 15% protein-0.6% calcium diet with 100ppm cadmium in drinking water for first 15days and the other 5groups fed same diet without cadmium in drinking water for same period and served as controls. After this 15-day intoxication period, each one of cadmium intoxication and control groups were fed each of 4 kinds of detoxifying diets different with protein(40%, 15%) and calcium(1.3%, 0.6%) levels without cadmimum in drinking water for following 15 days of detoxifying period. Results were summarized as follows: 1) Food intake, body weight gain, F.E.R. and weights of liver, kidney and femur were increased by detoxifying diets and high protein diet was most effective in weight gains of liver and kidney. 2) When cadmium and metallothionein contents of initial intoxication group and those of all detoxication groups were compared, cadmium and metallothionein contents in the liver were not changed, but those in kidney increased, and those in intestine decreased markedly. 3) Only dietary protein level affected cadmium and metallothionein distribution among organs, and cadmium contents of whole blood, liver, kidney and femur were lower in high protein diet, but metallothionein contents in liver and kidney were higher in high protein diet. 4) Gel filtration chromatogram showed that most of cadmium in the cytosol was bound to metallothionein fractions in high protein-high calcium group. Results obtained indicated that high protein diet was effective in cadmium detoxication by increasing the induction of metallothionein synthesis. But high calcium diet did not play a role in cadmium detoxication.

에탄올을 투여한 흰쥐에서 단백질 섭취수준이 단백질 대사에 미치는 영향 (Effects of Dietary Protein Levels on Protein Metabolism in Ethanol-Administered Rats)

  • 에탄올섭취 수준을 5%로 고정하고 식이 단백질의 급여수준을 8%, 16%, 24%로 달리한 흰주의 혈액 및 간, 분, 뇨중의 단백질대사에 미치는 영향을 알아보기 위하여 생후 15주된 Sprague-Dawely계 숫쥐를 모두 8군으로 나누어 4주 및 10주로 실험기간을 달리하여 실험한 결과 체중증가량, 혈당농도 및 간중 단백질 농도는 에탄올과 단백질섭취 수준에 따른 영향은 없었으며, 장기의 무게는 10주 급식군에서 폐의 경우 대조군과 에탄올 섭취군(16PE)사이에는 유의성이 없었으나 에탄올 투여시 단백질 수준이 가장 높은 24PE군에 유의하게(p<0.05) 증가되었다. 혈청중 총단백질은 10주 급식군에서 대조군에 비해 에탄올을 첨가한 16PE, 24PE군에서 유의적으로(p<0.05) 증가하였으며, 알부민농도는 10주 급식군에서 대조군에 비해 에탄올을 첨가한 군에서 유의적으로(p<0.05) 높았으며 단백질 수준에 따른 차이는 없었다. 분과 뇨중 질소대사에서는 에탄올에 의한 영향은 나타나지 않았다.

Effect of different levels of fiber and protein on growth performance and fecal characteristics in weaning pigs

  • This experiment was conducted to evaluate the growth performance, fecal score, and fecal microbial shedding in weaning pigs fed diets with different levels of fiber and protein. A total of 96 weaning piglets ($7.41{\pm}0.71kg$) were used in a 5-week trial. Pigs were allotted to dietary treatments based on initial body weight in a $2{\times}2$ factorial design with the following factors: dietary fiber (100 and 200 g/kg, respectively, during days 0 to 14; 175 and 300 g/kg, respectively, during days 14 to 35) and dietary protein (170 or 200 g/kg). There were 6 replicates with 4 pigs per pen. On day 14, pigs fed high protein or high fiber diets had heavier body weight (p < 0.05). During days 0 to 14, pigs fed high protein or high fiber diets grew faster (p < 0.05). Additionally, during days 14 to 35, an interactive effect of fiber and protein was found (p < 0.05) on average daily gain. The different levels of protein and fiber in diet did not affect the pigs' fecal scores (p > 0.05). However, feces from the high fiber group showed lower concentration of Escherichia coli (p < 0.05). In conclusion, the results of the present study indicate that a high protein diet improves the growth of weaning pigs especially during the first two weeks. Moreover, the increments in fiber level, even in the high protein diet, favorably decreased the number of E. coli.

Effect of Pulsed Electromagnetic Field on MMP-9 and TIMP-1 Levels in Chondrosarcoma Cells Stimulated with IL-1β

  • Chondrosarcoma, the second most common type of bone malignancy, is characterized by distant metastasis and local invasion. Previous studies have shown that treatment by pulsed electromagnetic field (PEMF) has beneficial effects on various cancer cells. In this study, we investigated the effects of PEMF applied for 3 and 7 days on the matrix metalloproteinase (MMP) levels in chondrosarcoma SW1353 cells stimulated with two different doses of $IL-1{\beta}$. SW1353 cells were treated with (0.5 and 5 ng/ml) $IL-1{\beta}$ and PEMF exposure was applied either 3 or 7 days. MMP-9 and TIMP-1 levels were measured in conditioned media by enzyme-linked immunosorbent assay. The results were relative to protein levels. Statistical analyses were performed using one-way analysis of variance (ANOVA). P<0.05 was considered significant. PEMF treatment significantly decreased MMP-9 protein levels in human chondrosarcoma cells stimulated with 0.5 ng/ml $IL-1{\beta}$ at day 7, whereas it did not show any effect on cells stimulated with 5 ng/ml $IL-1{\beta}$. There was no significant change in TIMP-1 protein levels either by $IL-1{\beta}$ stimulation or by PEMF treatment. The results of this study showed that PEMF treatment suppressed $IL-1{\beta}$-mediated upregulation of MMP-9 protein levels in a dual effect manner. This finding may offer new perspectives in the therapy of bone cancer.

Prealbumin and Retinol Binding Proteins Are Not Usable for Nutrition Follow-Up in Pediatric Intensive Care Units

  • Purpose: Feeding children is a problem in pediatric intensive care units (PICU) and it is difficult to know the correct amount. The purpose of this study is to evaluate if prealbumin or retinol binding proteins (RBP) are effective relative to daily enteral nutrition, without being affected by severity of diseases or infections and can be used to follow up nutritional amount. Methods: This is a prospective observational study that includes 81 patients admitted to PICU in Akdeniz University with estimated duration >72 hours, age between 1 month and 8 years. Daily calorie and protein intake were calculated and prealbumin, RBP and C-reactive protein (CRP) levels were measured on the first, third, fifth and seventh mornings. Results: We find moderate correlation between daily calorie intake and prealbumin levels (r=0.432, p<0.001), RBP levels and daily protein intake (r=0.330, p<0.001). When we investigated the relationship between changes of prealbumin, RBP, CRP, calorie and protein intake during intensive care stay, we found that increase of Prealbumin and RBP levels are explained by decrease of CRP levels (r=-0.546 and -0.645, p<0.001) and not with increase of nourishment. Conclusion: Even adjusted for PRISM3, age and CRP, prealbumin and RBP are correlated with last 24 hours' diet. However, it is not convenient to use as a follow up biomarker because increase of their levels is related with decrease of CRP levels.

한우 송아지의 초유섭취에 의한 수동면역이 포유기간 중의 질병발생에 미치는 영향 (Effects of colostrum-conferred passive immunity on disease incidence in Korean native calves during the suckling period)

  • The effects of the serum total protein and immunoglobulin levels of the colostrumconferred Korean native calves at 2 days of age on the disease incidence during the suckling period were studied. The results obtained were summerized as follows: 1. Serum total protein, total immunoglobulin, IgG, and IgM levels of 6 Korean native claves died from septicemia were lower than those of calves survived (p<0.05). 2. Korean native calves having low serum total protein and immunoglobulins were affected with diarrhea earlier in life, when compared with calves having high serum levels (p<0.05). 3. Korea native calves having low serum total protein were affected with bronchopneumonia earlier in life, when compared with calves having high serum levels (p<0.05). 4. Korean native calves having low serum neutralizing antibody titers to bovine viral diarrhea virus had high incidence rate of BVD, when compared with calves having high serum neutralizing antibody titers (p<0.05).

Tumor necrosis factor-α converting enzyme (TACE) increases RANKL expression in osteoblasts and serves as a potential biomarker of periodontitis

  • Periodontitis is a very prevalent disease. Therefore, biomarkers for the early and standard diagnoses of periodontitis are urgently needed. TACE is a membrane-bound metalloprotease. Although a recent study suggested that TACE levels in GCF are elevated during periodontal disease, the levels of TACE in GCF at different stages of chronic periodontitis have not been determined. Here, we analyzed the protein levels of TACE in GCF from periodontal disease subjects and confirmed that the protein levels of TACE were higher in the moderate periodontitis groups. TACE is known to be a NF-${\kappa}B$ ligand that stimulates RANKL secretion in osteoblasts. To understand the effects of TACE on RANKL and OPG in osteoblasts, we treated MG63 cells with TACE. We observed an increase in RANKL protein expression but a decrease in OPG protein expression. Our data suggest that TACE can induce RANKL expression and promote osteoclastogenesis, thus worsening the outcome of periodontitis.

Expression of Tumor Necrosis Factor-alpha-induced Protein 8 in Pancreas Tissues and its Correlation with Epithelial Growth Factor Receptor Levels

  • Tumor necrosis factor (TNF)-alpha-induced protein 8 (TNFAIP8 or TIPE) is a recently identified protein considered to be associated with carcinogenesis. To investigate its expression pattern in pancreatic cancer patients and to analyse its correlation with clinicopathological significance and the expression levels of epithelial growth factor receptor (EGFR), immunohistochemistry was performed to detect the TNFAIP8 and EGFR proteins in pancreatic cancers, pancreatitis tissues, and healthy controls. The results showed stronger staining of TNFAIP8 protein in pancreatic cancer tissues compared with normal pancreas tissue. Furthermore, in 56 patients with pancreatic cancer, the expression levels of TNFAIP8 in patients with low tumor stage was higher than that with high tumor stage, and correlated with tumor staging and lymph node metastasis (P<0.05). Furthermore, TNFAIP8 expression positively correlated with EGFR levels (r=0.671135, P<0.05). These results indicate that TNFAIP8 may play important roles in the progression of pancreatic cancer.
