Effects of High Hydrostatic Pressure on Technical Functional Properties of Edible Insect Protein

  • The objective of this study was to determine the effects of high pressure to investigate the technical functional properties of the protein solution extracted from an edible insect, Protaetia brevitarsis seulensis. High pressure processing was performed at 0 (control), 100, 200, 300, 400, and 500 MPa at 35℃. The essential amino acid index of the control was lower (p<0.05) than that of the P. brevitarsis seulensis extract treated with 100 MPa. The SDS-PAGE patterns tended to become faint at approximately 75 kDa and thicker at approximately 37 KDa after high pressure treatment. The protein solubility and pH of the protein tended to increase as the hydrostatic pressure levels increased. The instrument color values (redness and yellowness) of the P. brevitarsis seulensis protein treated with high pressure were lower (p<0.05) than those of the control. The forming capacity of the protein solution with P. brevitarsis seulensis treated with high pressure was higher (p<0.05) than that of the control. In conclusion, we confirmed that the technical functional properties of edible insect proteins extracted under high pressure of 200 MPa are improved. Our results indicate that high pressure can improve the technical functional properties of proteins from edible insects.

Anti-obesity Effect of Hypsizigus marmoreus in High Fat-fed Mice (고지방식이를 섭취한 마우스에서 느티만가닥버섯의 항비만 효과)

  • This study investigated the possible anti-obesity effects of Hypsizigus marmoreus on high fat-fed mice. Thirty male C57BL/6 mice were randomly divided into 3 groups: a normal diet group (N), a high-fat diet group (HF), and a high-fat diet with 5% Hypsizigus marmoreus group (HF-H). After 8 weeks, the body weights in the HF group significantly increased, while those of the HF-H group decreased. Also, liver and adipose tissue weights in the HF-H group significantly decreased. Total serum cholesterol, leptin, and insulin levels were significantly higher in the HF group than those of the N group, but lower than those of the HF-H group. Accumulation of hepatic lipids was apparent in the HF group, as indicated by HE staining and hepatic lipid analysis, while these effects were improved by supplements with Hypsizigus marmoreus in the HF-H group. Also, a reduction in adipocyte size of the epididymal adipose tissue was observed in the HF-H group. $PPAR{\gamma}$, SREBP-1c, and SCD-1 protein expressions were down-regulated in the epididymal adipose tissue of the HF-F group compared to the HF group. Taken together, these results suggest that Hypsizigus marmoreus may an effective anti-obesity treatment.

Analysis of Nutritional Characteristics and Physiological Functionality of Hypsizygus marmoreus (Brown cultivar) (갈색 느티만가닥버섯의 영양특성 및 생리기능성 분석)

  • In order to apply into functional food or medicinal industry, nutritional characteristics and physiological functionality of Hypsizygus marmoreus (brown cultivar) were investigated. Fruiting body of H. marmoreus contained 27.3% of crude protein, 55.8% of total sugar and 11,109.3 mg/100 g dry weight of malic acid. Furthermore, 66.7% of antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity and 37.3% of antigout xanthin oxidase inhibitory activity showed in the water extract from H. marmoreus. The economically ACE inhibitory activity (81.4%) was obtained when the fruiting body of Hysizygus marmoreus was extracted with distilled water of $50^{\circ}C$ for 12 h, even though maximal ACE inhibitory activity (84.4%) was showed the extracts from $70^{\circ}C$ for 12 h.

Components of Pine Needles Extract and Functionality of the Dyed Fabrics (솔잎 추출물의 성분 분석 및 염색물의 건강안전 기능성 평가)

  • The pine needles can be used for four seasons in normal living and it can be taken friendly everywhere as it is distributed over 50% in Korea. The pine needles consist of vitamins, protein, minerals, essential oil and enzyme related to antimicrobial activity. It has effect like high blood pressure, neuralgia and hanged over by terpene, glucokinin, rutin, apigenic acid and tannin. Also the extract of them can be used for dyeing of fabrics. However, the extract components and effects of them are not well known yet. Therefore, the purpose of this study was to investigate the volatile components of the pine needles extract and functionality. The pine needles extract was dyed into various fabrics(nylon, silk, wool and soybean) and mordanted with Al, Cu, Cr, Fe and Sn. The extracted aroma compounds were compared by gas chromatography-mass spectrometry. The major volatile compounds of pine needles verified by using SPME were alpha-pinene, beta-pinene, beta-phellandrene, caryophyllene, ethanon, benzen. A total of 15 compounds were identified by using the SPME fibers. In the UV-visible spectra, the maximum absorption of wavelength of the pine needles ethanol extract appeared at 460, 630nm for chlorophyll component and at 237, 281nm for tannin component with the pine needles distilled water extract. Most of sample showed high antibacterial effect in none mordant but wool fabric showed high antibacterial effect in mordants. The result of UV block test showed a superior ability of blocking ultraviolet ray infiltration in all sample.

The Current Status and Prospect of Sericultural Byproduct Industry in China

  • Sericulture is a traditional agro-industry, which involves mulberry cultivation and silkworm rearing, has made great contributes to the human civilization. With the development of national economy and modem technology, mulberry and silkworm are being used to develop products with functionality besides the traditional cocoon production in China. In this paper, we brief the current developing situation of sericultural byproducts with functionality in the following aspects. (1) Functional products from silkworm larvae: silkworm powder, white muscardine silkworm, isolation and purification of anti-bacterial proteins from the larvae and production of medically valuable substances by Bombyx mori nuclear polyhedrosis virus (BmNPV) vector. (2) Utilization of silkworm feces: for pillow and for isolation of chlorophyll etc. (3) Production of valuable Chinese traditional medicine like Cordyceps sinensis with pupae, functional utilization of pupa protein and chitin. (4) Silk as additives to cosmetics, silk food and medical materials. (5) Functional utilization of mulberry: cultivation of edible fungus on mulberry shoots as medium, mulberry fruit drinks, mulberry tea, etc. The prospect of sericultural byproduct industry in China is also discussed.

Protein Separation in Functional Rice Grains Using Two-Dimensional Gel Electrophoresis

  • The proteins from functional rice cultivars (Nogwonchalbyeo, Giant embryonic, Arhyangchalbyeo, and Goamibyeo) and general white rice were extracted and separated using two-dimensional (2D) gel electrophoresis. A wide variation in the molecular weight (MW) and pH range of the expressed proteins in rice samples were observed. The green-kerneled rice (Nogwonchalbyeo) exhibited proteins with MW of 9-57 kDa and appeared at a pH range of 4-7. The Giant embryonic contained proteins with MW of 31-63 kDa and a pH range of 5-6. The aromatic glutinous rice (Arhyangchalbyeo) showed proteins with MW of 24-28 and pH of 5.8-6.8. The high-amylose rice (Goamibyeo) exhibited proteins with MW of 3-63 and pH of 5.2-5.6. The identified proteins uniquely found and highly expressed in each cultivar may have a significant role on rice functionality. The results illustrate that the 2D gel electrophoresis is a valuable method in the determination of the protein expression profiles in functional rice grains and may be useful in the identification of specific marker proteins associated with the functional property of rice.

Novel Vectors for the Convenient Cloning and Expression of In Vivo Biotinylated Proteins in Escherichia coli

  • Biotinylation of recombinant proteins is a powerful tool for the detection and analysis of proteins of interest in a large variety of assay systems. The recent development of in vivo biotinylation techniques in E. coli has opened new possibilities for the production of site-specifically biotinylated proteins without the need for further manipulation after the isolation of the recombinantly expressed proteins. In the present study, a novel vector set was generated which allows the convenient cloning and expression of proteins of interest fused with an N-terminal in vivo biotinylated thioredoxin (TRX) protein. These vectors were derived from the previously reported pBIOTRX vector into which was incorporated part of the pBluescript II+phagemid multiple cloning site (MCS), amplified by PCR using a pair of sophisticated oligonucleotide primers. The functionality of these novel vectors was examined in this system by recombinant expression of rat transforming growth factor-$\beta$. Western-blot analysis using TRX-specific antibodies or peroxidase-conjugated streptavidin confirmed the successful induction of the fusion protein and the in vivo conjugation of biotin molecules, respectively. The convenience of molecular subcloning provided by the MCS and the effective in vivo biotinylation of proteins of interest makes this novel vector set an interesting alternative for the production of biotinylated proteins.

Defective Interfering HIV-1 Pseudotypes Carrying Chimeric CD4 Protein

  • Chimeric CD4 proteins were assembled. They contained the entire CD4 ectodomain that is linked to different membrane anchors. Membrane anchors consisted of either glucosyl phosphatidyl inositol (gpi), the transmembrane and cytoplasmic regions of HIV-1 Env protein, or the vesicular stomatitis virus G glycoprotein, respectively. The HIV-1 co-receptor CXCR4 and CD4 were independently inserted into viral envelopes. We compared the insertion of six different CD4/CXCR4 constructs into HIV-1 envelopes, as well as their functionality in targeting and specific infection of cells that constitutively express the HIV-1 Env protein. All of the six different HIV-1 (CD4/CXCR4) pseudotypes were able to transduce Env (+) cells at similar efficiency. In addition, stable transduction of the Env (+) recipient cells demonstrated that all chimeric proteins were functional as receptors for Env when inserted into HIV-1 envelopes. In fact, these results demonstrate for the first time a stable transduction by a targeted HIV-1 pseudotype virus.

Effect of succinylation on functional properties of soy protein isolate and qualities of soy bean curd (Succinyl화가 대두 단백질의 기능성과 두부의 품질에 미치는 영향)

  • Soy protein was isolated from Korean soy bean 'Chang ryub' and chemically modified with succinic anhydride. Functionality of the soy protein isolate(SPI), succinylated SPI(SPPI), and PP590(commercial) at various pH were investigated. The mechanical and sensory properties of soy bean curds made from several mixing ratio of succinylated soy bean milk were observed. The solubility of SPI significantly increased with succinylation. The solubility of PP590 was lower than that of SSPI. The solubility of SPPI increased significantly in 0.03M CaCl2 solution. The emulsifying activity of SSPI increased. On the range of pH above pI the emulsifying activity of PP590 was higher than that of SPI. There was no difference in emulsion stability among the groups. The foam expansion capacity of SPPI increased at higher pH than pI but the foam stability decreased significantly above pH 9. Mechanical texture profile analysis revealed the modified soy bean curds had the lower hardniss, chewiness and cohesiveness with increased modification. The mechanical characteristics of modified soy bean curds revealed generalized Maxwell Model of 7-elements or 5-elements. In sensory evaluation, the hardness, the springiness and acceptability of modified soy bean curds were lower significantly than those of control soy bean curd.

Functionality Changes of Rapeseed Protein upon Proteolysis (유채단백질의 Proteolysis에 의한 기능성 변화)

  • purified rapeseed(Brassica napus var. Youngsan) protein was hydrolyzed by pronase. The hyrolysate protein was investigated for the some physicochemical and functional properties. UV and intrinsic fluorescence spectra of the hydrolysate showed the maximum absorption at 274nm and 360nm respectively. Intensity of yellow color decreased in the process of hydrolysis and the surface hydrophobicity decreased up to fourfold. The main bands of hydrolysate by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) were observed at 14,000 to 12,000 dalton molecular weight. Solubilities of hydrolyzed protein increased by 10~15% compared to those of unhydrolyzed protein at acidic pH. In the hydrolysate, while absorption of both water and oil, foam expansion and emulsion stability were increased, absolute viscosity, heat coagulation, calcium coagulation, foam stability and emulsion activity were decreased.
