약독 담배모자이크바이러스 II. RNA 및 외피단백질의 특성 (Studies on Mild Mutants of Tobacco Mosaic Virus II. Biochemical Properties of Ribonucleic Acid and Coat Protein)

  • 액독 TMV, Tw 333 RNA 및 외피단백질에 대한 생화학적 특성을 조사하였다. Tw333-RNA는 $2.03\times10^6$dalton의 분자량을 나타냈고, 고기조성은 guanine 25.4, adenine 29.2, cytosine 17.5, uracil 27.9mol이었다. 열처리에서 얻어진 이 RNA의 농색효과는 $25.1\%$를 나타냈고, 이때 Tm치는 $47^{\circ}C$였다. 한편 Tw 333의 외피단백질은 17,500 dalton의 분자량을 보였으며, 16종의 아미노산으로 구성된 158개의 아미노산잔기를 나타냈다. Trypsin으로 분해한 단백질은 9종의 ninhydrin 양성반응 peptide를 형성하였다. 이들 약독 TMV, Tw333-RNA 및 외피단백질의 생화학적 특성은 원주 OM계통과 전반적으로 매우 유사하였다. 그러나 고기조성, 농색효과, 아미노산조성 및 peptide map에서 약간의 차가 인정되었다.

동결속도에 따른 닭고기의 냉동 저장 중 이화학적 변화 (The Effect of Freezing Rates on the Physico-Chemical Changes of Chicken Meat during Frozen Storage at -2$0^{\circ}C$)

  • 식육을 장기저장 할 때 발생되는 단백질의 변성과 해동시 육량감소를 줄이기 위한 방법의 하나로써 최적동결방법을 모색하고자 동결속도에 따른 저장중 닭고기의 물리화학적 변화를 조사한 결과 동결속도가 빠를수록 pH증가가 지연되어 장기보존의 가능성을 보였다. TBA가 및 유리지방산 생성율은 저장기간이 길어짐에 따라 증가하는 추세를 보이고 있으나 부위육의 종류에 따라 동결속도의 영향이 다르게 나타나고 있다. 그러나 단백질추출성을 비교할 때 염용성단백질과 수용성단백질의 추출성은 부위육의 종류에 관계없이 3cm/hr이상의 동결속도에서 저장기간 중 추출성이 가장 높게 나타났으며, 이러한 경향은 다리살 보다 가슴살의 추출성이 더욱 높게 유지되고 있다. 이상의 결과에서 송풍동결기를 이용하여 닭고기를 동결시킬 경우 3cm/hr이상의 동결속도로 동결시켜 냉동저장 할 때 좋은 품질의 냉동육을 얻을 수 있는 것으로 판단되었다.

  • cAMP수용성 단백질(CRP)은 E. coli의 100가지 이상의 유전자 전자조절에 관계된다. CRP는 dimer로 존재하며 cAMP의 결합으로 활성인 형태로 전환된다. 이중체인 CRP 단백질의 열 안정성과 구조 전이의 연구에 효과적인 온도 기울기 전기영동장치를 이용하여 확인하였다. 본 연구에서 야생형과 S83C CRP 단백질의 melting temperature (Tm)는 산성인 완충용액[89.8 mM Glycine, 24mM Boric acid (pH 5.8)]에서 57$\pm$1(야생형 CRP)과 55$\pm$1$^{\circ}C$ (S83G CRP)였다. 그리고 온도에 따른 CRP 단백질의 구조변화도 protease digestion과 CD spectropolarimeter을 이용하여 확인하였다.

Purification and Characterization of an Antioxidant Protein from Fertilized Eggs

  • Free radicals may attack cells or tissue, leading to chronic diseases, and antioxidant consumption is potentially useful for removing free radicals. Egg proteins may be used as potential sources of antioxidant considering their ability of scavenging free radicals to apply for food or cosmetics industry. In this study, we obtained a natural antioxidant protein from fertilized eggs, which was a dietary supplement in some Asian countries. Meanwhile, antioxidant activities of these proteins were evaluated using different oxidation systems. With increasing incubation time, the antioxidant activity of these proteins increased during 15 d of incubation. The samples on day 15 were performed for isolation of antioxidant protein. The protein, named P4-1 (MW, 45 kDa), was isolated and purified by consecutive chromatographic methods. P4-1 contained 17 amino acids, which was determined by liquid chromatography-mass spectrometry and Amino Acid Analyzer. Moreover, the amino acid sequence was highly similar to that of ovalbumin. Differential scanning calorimetry showed that the denaturation temperature of P4-1 was $57.16^{\circ}C$. Furthermore, P4-1 suggested high oxygen radical-absorbance activity in ${\cdot}OH$ assays, and its antioxidant activity was stable at $30-50^{\circ}C$ in acidic and neutral pH. Thus, these results revealed that P4-1 may be a potential resource as a natural antioxidant.

대두 단백섬유의 제조에 관한 연구 (Soyprotein Fiber Formation)

  • 저자들은 전보(Korean J. Food Sci. Techno., 9,123(1997)에서 분리 대두 단백의 식품학적 성질을 조사하였다. 본 연구에서는 이 성질을 응용하여 대두 단백섬유를 제조하고저 분실 험실에서 고안 설계하여 의뢰제작한 protein spinning apparatus를 사용하여 분리 대두단백으로 대두단백섬유 제조실험을 행한결과 texture가 우수한 제품을 얻었다. 제조조건은 15-18% 단백질용액을 알카리 (0.6%)로 처리하여 50-100 PSI spinning press로 사출한 후 12% Nacl-1 N acetic acid bath에서 응고시켰다. 12% 단백질 용액은 생성된 단백섬유의 texture가 불량하여 일정한 모형을 유지하지 못하였다. 제품의 성질을 Instron기기와 texturometer를 사용하여 측정하였다. 아울러 대두 단백섬유의 형성기작을 제시하였다.

Effects of Preparation Method and Evaluations on Structural Integrity in Model Antigen-Containing Biodegradable Microspheres for Vaccine Delivery

  • To demonstrate the effect of formulation conditions and evaluations of structural integrity from ovalbumin containing poly lactide glycolide copolymer (PLGA) microspheres for Vaccine delivery, OVA microspheres were prepared by a W/O/W multiple emulsion solvent extraction technique. Dichloromethan (DCM) and Ethyl acetate (EA) were applied as an organic phase and poly vinyl alcohol (PVA) as a secondary emulsion stabilizer. Microspheres were characterized for particle size, morphology (optical microscopy and Scanning Electron Microscope (SEM)). Protein denaturation was evaluated by size exclusion chromatography (SEC), SDS-PAGE and isoelectric focusing (IEF). Residual organic solvent was estimated by gas chromatography (GC) and differential scanning calorimetry (DSC). Optical photomicrograph and SEM revealed that micro spheres were typically spherical but various morphologies were observed. Mean particle size $(d_{vs})$ of microspheres were in the range of $3{\sim}50{\mu}m$. Also, The protein stability was not affected by the fonnulation process and residual organic solvent was beyond the detection below 0.1ppm. These results demonstrated that micro spheres might be a good candidate for the parenteral vaccine delivery system.

경구용 항원 수송체 모델로서 폴리락티드-글리콜리드 마이크로스피어의 입자도 조절 (Particle Size Control of Poly(Lactide-co-Glycolide) Microspheres for Oral Antigen Delivery Systems)

  • Poly (lactide-co-glycolide) (PLGA) microspheres containing ovalbumin (OVA) as a model protein drug were prepared by double emulsification method, and various conditions such as mixing rate, volume of outer phase and isopropyl alcohol concentration in outer phase during secondary emulsification were observed to control the size of microspheres. In addition, entrapment efficiency of OVA and protein denaturation were also evaluated. As the rate of stirring was increased, the size of particles was decreased. But excessive stirring increased the particle size of microspheres. In a preparation condition of small volume of outer phase, the particle size was decreased but the entrapment efficiency was increased. Adding isopropyl alcohol to outer phase decreased the size of particles, but increased the entrapment efficiency. Microparticles should have smaller size than $10\;{\mu}m$ to be uptaked by Peyer's patch in small intestine. High speed of mixing and relatively small volume of outer phase are needed to reduce the size. In addition, appropriate amount of isopropyl alcohol in outer phase also plays an important role in size reduction of PLGA microspheres.

Rebinding Dynamics of CO Following Photodissociation of 4.0 M Guanidine HCl-Denatured Carbonmonoxyhemoglobin

  • Femtosecond vibrational spectroscopy was used to probe the dynamics of CO rebinding to hemoglobin (Hb), denatured by 4.0 M GdnHCl in $D_2O# at 283 K, after photolysis of HbCO. The stretching mode of $^{13}CO$ bound to the denatured $Hb^{13}CO$ showed a single band centered at 1922 $cm^{-1}$, indistinguishable from that of denatured $Mb^{13}CO$. Geminate rebinding of CO to the denatured Hb was accelerated more than 1000 times, suggesting that the native structure of the Hb is required to suppress efficient geminate rebinding of CO, as is the case in Mb. The geminate yield and rate for CO rebinding are almost the same in both the denatured Hb and Mb. Similarity in the equilibrium spectrum and rebinding dynamics of CO indicates that the state of the denatured Hb is very similar to that of the denatured Mb. In the denatured Hb, quaternary contact of the protein is likely severed, with the denatured protein existing as an independent subunit much like Mb.

  • 0.05 M 중탄산 나트륨 및 0.04 M 인산염 완충용액 (pH 8.3)으로 처리한 노계육 가슴살 및 다리살 시료의 경우 근섬유 단백의 불안정화를 시사하는 열변성 온도의 저하가 관찰되었다. 이러한 현상을 근섬유 단백중 95 Kdalton 및 55 Kdalton protein의 추출과 관련 가능성이 있는 것으로 판단되었으며 처리 시료의 근섬유구조 변화(Z-line의 파괴)와도 관련 가능성이 있는 것으로 판단되었다.

분리균 Erwinia carotovora subsp. carotovora LY34의 병원성 및 CMCase Isozymes 생성

  • Soft-rot bacterial pathogen, Erwinia sp., was isolated from chinese cabbage tissue showing soft-rot symptom. This bacterial strain caused soft-rot to chinese cabbage and potato, and it was identified as Erwinia carotovora subsp. carotovora LY34 (E. c. subsp. carotovora LY34). Erwinia carotovora subsp. carotovora LY34 did not have hemicellulase but extracellular cellulase, pectinase, polygalacturonase, protease activity. The results of the microscopy showed that chinese cabbage tissue and potato tissue were macerated by infection of E. c. subsp. carotovora LY34. In analysis of the cellulases activity of the isolated cellulose-degradation enzymes from E. c. subsp. carotovora LY34 total protein, three cellulase activity bands were detected by non-denaturation gel electrophoresis method and five cellulase activity bands were detected by CMC-SDS-PAGE direct stain method.
