Effect of Lipid on the Protein Denaturation in Cooking Fish Meat (어육 조리시 지질이 단백질 변성에 미치는 영향)

  • The present work aims to estimate the effect of heat treatment on the in vitro protein digestibility and formation of trypsin inhibitor or trypsin indigestible substrate(TIS) of raw and defatted flounder. It was also carried out to assess the formation of lipid-protein complexes under the conditions of different ratio of lipid addition. The in vitro protein digestibility increased when steamed for 5 min showing $88.09\%$ in raw and $90.56\%$ in defatted samples, respectively. After 40 min steaming, the digestibility decreased by $2{\sim}4\%$. As for microwaving, heating for 1 min resulted in slight increase of digestibility, however, heating for 7 min did decrease of digestibility by $3{\sim}4\%$ for both raw and defatted materials. There was no difference in fatty acid composition found with heat treatment. The major fatty acids of flounder meat were $C_{16:0},\;C_{16:1},\;C_{18:1},\;C_{20:5},\;C_{22:6}$ and the ratio of the unsaturated to saturated was 67.3:32.6. Fat oxidation and nonenzymatic browning were enhanced by heat treatment and protein solubility decreased necessarily as the brown pigment formation increased. On the other hand, the effects on the digestibility and TIS of the complexes formed from interaction of lipid and myofibrillar or meat protein of flounder were examined. The interaction of protein with lipid was considered to mostly contribute to the drop of digestibility of fish products. The digestibility of myofibrillar protein was $93.72\%$ for flounder, and it generally decreased as the amount of lipid added to protein and reaction time increased. Also mixed and heated samples were more active in digestibility decline than those mixed after heating. The result probably indicated that lipid-protein interaction was involved in the drop of digestibility which coincided with protein denaturation.

  • Effect of C/N ratios on the cell growth, cell lipid, cell protein and fatty acids of Rhodotorula sp. was examined. At low concentration of nitrogen in cultural medium the growth of the yeast was retarded, whereas its growth was favorable at high concentration but lipid yield was decreased due to a high content of cell protein. In order to increase lipid yield, the optimum C/N ratio obtained was 45-50 (5% glucose, 0.0414% nitrogen). During the cultivation cell protein synthesis occurred mainly in initia stage and the lipid was accumulated rapidly at the late phase. The highest content of total fatty acid was observed at the optimum C/N ratio. The higher C/N ratio was the more unsaturation degree of fatty acid decreased. It was attributed principally to a decreased oleic acid with palmitic acid increased. No significant changes occurred in content of other fatty acids.

  • Mackerel water-soluble protein fraction produced by washing the mackerel meat were concentrated by isoelectric point shifting precipitation process, and the concentrates were utilized as the alternative feed of fish meal. In the 1st aquaculture diet experiment for Israel common carp, the feed conversion ratio decreased in proportion to the rise in the percentage of the recovered protein containing a residual lipid, which was added to the fish meal. It was supposed that the low feed efficiency was because of lipid oxidation in the recovered protein fraction. In addition, 2nd aquaculture diet experiment for Israel common carp was conducted after removing the oxidized lipid in the recovered protein fish meal. When a portion of the fish meal was substituted by the recovered protein devoid of the residual lipid, the feed conversion ratio increased in proportion to the amount of the substitute being added to the fish meal. Therefore, the recovered protein fraction of the mackerel washing wastewater from mackerel processing factory could be used as the alternative feed of fish meal.

A Study on the Prediction of Parturient Syndrome in Holstein Cows (젖소에서의 산욕기질병 발생예견에 관한 연구)

  • In order to establish a method predicting susceptible cows to the parturient syndrome, various serum chemical parameters (calcium, phosphorus, Ca/P, magnesium, cholesterol, total protein, albumin, globulin, A/G, total lipid, non-esterified fatty acid(NEFA) and aspartate aminotransferase(AST)) were measured during late pregnancy and their relationships with periparturient diseases were investigated during puerpural period. The results obtained were as follows : 1. The factors affecting the prediction of susceptible cows to parturient syndrome were calcium, magnesium, total protein, globulin, A/G ratio and total lipid at 30 day antepartum and the diagnosability was 70.7%. 2. In the experimental cows producing more than 21kg of milk per day, the factors affecting the prediction of susceptible cows to parturient syndrome were calcium, NEFA and A/G ratio at 30 day antepartum and the diagnosability was 66.7%. 3. In the experimental cows calved more than 3 times, the factors affecting the perdiction of susceptible cows to parturient syndrome were calcium, total protein, albumin and NEFA at 30 day antepartum and the diagnosability was 83.3%.

A Study on the Growth Rate, Nutritional Effects and Serum Lipid Contents of Rats by Feeding with Leaf and Stem Extract of the Ginseng Radix (인삼잎과 줄기 혼합 추출액의 첨가급식이 흰쥐의 성장, 영양효과 및 혈청 지질에 미치는 영향)

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from the mixture of leaf and stem of ginseng radix(Panax ginseng CA. Meyer). This study measured the intake levels and efficiency ratio, growth rate, absorption ratio of carbohydrate, lipid and protein of rat by feeding with LSE during 6 weeks. We analyse the hematological and serum metabolic variables, serum lipid concentrations. Total diet and protein intake levels were low, but efficiency ratios were significantly high in LSE administered groups than the control group. Weight gain, liver and kidney weight of LSE groups were significantly higher than the control group. Blood RBC, Hct, Hb, total protein and albumin concentrations were reasonable levels in LSE administered groups compared to the basal diet group. Also serum total cholesterol, LDL-cholesterol, triglyceride contents of LSE groups were low, but HDL-cholesterol level was higher than the basal diet group. These results imply that leaf and stem of ginseng radix could be used as possible food resources, functional food material and feed stuff.

Optimum Dietary Protein and Lipid Levels on Growth in Parrot Fish (Oplegnathus fasciatus) (돌둠사료의 적정 단백질 및 지질 함량)

  • A feeding trial was conducted to investigate optimum dietary protein and lipid levels in diets for parrot fish (Oplegnathus fasciatus). Three groups of 25 fish averaging 7.0g were fed each of the experimental diets containing 30, 40, 50 or 60% crude protein (CP) with 8 or 16% crude lipid level ($4{\times}2$factorial desing) for 8 weeks. Weight gain and feed efficiency increased significantly (P<0.05) with dietary protein level up to 50% CP when diets contained 16% lipid level, and increased with dietary protein level up 60% CP when diets contained 8% lipid level (P<0.05). The broken-like model showed that an optimum dietary protein level was 46% when the diet contained 16% dietary lipid. Fish fed diets containing 16% lipid level showed significantly (P<0.05) higher weight gain, feed efficiency and protein efficiency ratio than did fish fed diets containing 8% lipid level when diets contained 40 or 50% dietary protein (P<0.05).

Effects of Dietary Lipid Level on Growth and Body Composition of Juvenile Red Sea Bream (Pagurus major) during Winter (저수온기 참돔 치어의 성장 및 체조성에 미치는 사료내 지질의 영향)

  • This study was conducted to investigate effects of dietary lipid level on growth and body composition of juvenile red sea bream in low temperatures. Duplicate groups of fish (initial body weight of 79 g) were fed one of three isonitronic diets (47% crude protein) containing different lipid levels (10%, 17% and 22%) for 25 weeks during the winter season. Weight gain and survival of fish fed 17% lipid diet were significantly (P<0.05) higher than those fish fed the 10% or 22% lipid diet. Protein efficiency ratio, daily feed intake, condition factor, hepatosomatic index and viscerasomatic index were not affected by dietary lipid level, but feed efficiency of fish fed 10% lipid diet was significantly (P<0.05) lower than those fish fed the 17% or 22% lipid diet. Proximate composition of the whole body, liver, viscera and dorsal muscle were not significantly different among all groups except for crude protein content of dorsal muscle. The contents of 16:0, 18:0, 20:4n-3 and 20:5n-3 of the whole body were significantly (P<0.05) affected by dietary lipid level. The results of this study suggest that an increased dietary lipid level from 10% to 17% can improve growth of juvenile red sea bream in low temperature periods.

Utilization of dietary protein, lipid and carbohydrate by flounder (Paralicthys olivaceus)

  • Although flounder is one of the most important marine fish for aquaculture in Korea, feeding the flounder in commercial farms depends mainly on moist pellet in which over 70% frozen fishes (e.g. frozen horse mackerel) are incorporated in its formulation. Therefore, for further expansion of flounder farming, it is essential to employ practical formulated feeds that can support reasonable growth. Development of nutritionally balanced and cost-effective feeds is dependant on the information about nutritional requirement and feed utilization of the species. Nutrient and energy source in feed are needed for the growth and maintenance of fish. Protein is probably the most important nutrient affecting fish growth and feed cost. Therefore, it is essential to determine the optimum dietary protein level for the growth of fish, both its high proportion in the feed and because it is the main factor in determining feed cost. Dietary energy level is also critical because protein source in the feed is utilized as an energy source when the feed deficient in energy is fed to fish, whereas when the feed excess in energy is fed to fish, feed consumption decreased and resulted in growth reduction due to lack of other necessary nutrients for normal growth. Improper dietary protein, energy levels and/or their ratio will lead to an increase of fish production cost and deterioration of water quality resulting from wasted feed; thus, they are important in formulating commercial feed. Dietary lipids play important roles in providing energy and essential fatty acid for normal growth and survival of fish. Although carbohydrates are not essential nutrients for carnivorous fish, these compounds play important roles as a low-cost energy source for protein sparing and also as a feed binder. Nutrition researches for flounder have identified its requirements of protein, lipid and essential fatty acid, vitamin, and minerals for normal growth. Other studies have also been carried out to investigate the utilization of the protein, lipid and carbohydrate sources. Based on these nutritional information obtained, practical feed formulations have been studied for improve aquaculture production of flounder. The results of the researches on utilization of dietary protein, lipid and carbohydrate by flounder are discussed in this review.

Curcumin represses lipid accumulation through inhibiting ERK1/2-PPAR-γ signaling pathway and triggering apoptosis in porcine subcutaneous preadipocytes

  • Objective: Excessive lipid accumulation in adipocytes results in prevalence of obesity and metabolic syndrome. Curcumin (CUR), a naturally phenolic active ingredient, has been shown to have lipid-lowering effects. However, its underlying mechanisms have remained largely unknown. Therefore, the study aims to determine the effect of CUR on cellular lipid accumulation in porcine subcutaneous preadipocytes (PSPA) and to clarify novel mechanisms. Methods: The PSPA were cultured and treated with or without CUR. Both cell counting Kit-8 and lactate dehydrogenase release assays were used to examine cytotoxicity. Intracellular lipid contents were measured by oil-red-o staining extraction and triglyceride quantification. Apoptosis was determined by flow cytometry and the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-nick end labelling assay. Adipogenic and apoptosis genes were analyzed by quantitative polymerase chain reaction and Western blot. Results: The CUR dose-dependently reduced the proliferation and lipid accumulation of PSPA. Noncytotoxic doses of CUR (10 to 20 μM) significantly inhibited extracellular signal-regulated kinase 1/2 (ERK1/2) phosphorylation and expression of adipogenic genes peroxisome proliferation-activity receptor-γ (PPAR-γ), CCAAT/enhancer binding protein-α, sterol regulatory element-binding protein-1c, adipocyte protein-2, glucose transporter-4 as well as key lipogenic enzymes fatty acid synthase and acetyl-CoA carboxylase, while ERK1/2 activation significantly reversed CUR-reduced lipid accumulation by increasing PPAR-γ. Furthermore, compared with differentiation induced media treated cells, higher dose of CUR (30 μM) significantly decreased the expression of AKT and B-cell lymphoma-2 (BCL-2), while increased the expression of BCL-2-associated X (BAX) and the BAX/BCL-2 expression ratio, suggesting triggered apoptosis by inactivating AKT and increasing BAX/BCL-2 ratio and Caspase-3 expression. Moreover, AKT activation significantly rescued CUR inhibiting lipid accumulation via repressing apoptosis. Conclusion: These results demonstrate that CUR is capable of suppressing differentiation by inhibiting ERK1/2-PPAR-γ signaling pathway and triggering apoptosis via decreasing AKT and subsequently increasing BAX/BCL-2 ratio and Caspase-3, suggesting that CUR provides an important method for the reduction of porcine body fat, as well as the prevention and treatment of human obesity.

Genetic Ana1ysis for Rice Grain Properties Using a Doubled Haploid Population

  • Demand for high quality rice has always been a major factor in the international rice marketing. In the present study, doubled haploid (DH) population derived from anther culture of a Tongil/japonica hybrid was used for genetic analysis of rice grain quality. The average values of DH lines for grain weight, grain length and the ratio of grain length to width were near the mid-parent value. More than 40% DH lines showed transgressive segregation for grain weight, length, amylose and lipid content, but less than 10% DH lines observed on ratio of length to width and grain thickness were transgressive segregation. Correlation analysis between appearance qualities and physicochemical characters indicated that grain width and grain thickness both significantly and negatively correlated to protein and lipid content. A highly significant negative correlation between protein content and amylose content was observed.
